화장품 소재는 안전성이 무엇보다 중요시된다. 화장품에 사용되는 기존의 보존제인 PE에 대장균 효소 𝛽-gal을 이용하여 안전성이 증가된 PE-gal을 생합성하였다. 화장품 소재로 사용하기 위해 생합성된 생성물인 PE-gal의 피부 흡수도를 기존의 보존제인 PE와 비교하기 위해 Franz Diffusion cell Assay 시스템을 이용하여 경피투과도를 측정하였다. 같은 질량농도의 시료를 사용하였을 때 PE의 Flux 값, Kp 값은 시간이 지날수록 증가하는 것으로 나타났으나 PE-gal은 투과도를 측정할 수 있을 만큼 투과되지 못하였다. 이는 PE의 피부투과도가 생합성된 PE-gal 배당체보다 높다는 것을 나타낸다. Marzulli 등에 따라 Kp 값을 이용하여 투과 정도를 확인하였을 때 PE의 투과속도는 1mg/mL의 농도에서 느림(slow)으로 측정되었다. 따라서 배당체 형태의 PE-gal은 PE에 비해 경피 투과도가 현저히 낮게 나타났다.
Transplantation is considered to be a very useful approach to improve human welfare and to prolong life-span. Heterologous organ transplantation using pig organs which are similar to human beings and easy to make mass-production has known as one of the alternatives. To ensure potential usage of the pig organ for transplantation application, it is essentially required to generate transgenic pig modifying immuno-related genes. Previously, we reported production of heterozygous ${\alpha}1,3$-galactosyltransferase (GalT) knock-out and human membrane cofactor protein (MCP) expressing pig ($GalT^{-MCP/+}$), which is enforced for suppression of hyperacute and acute immunological rejection. In this study, we reported generation of homozygous pig ($GalT^{-MCP/-MCP}$) by crossbreeding $GalT^{-MCP/+}$ pigs. Two female founders gave birth to six of $GalT^{-MCP/-MCP}$, and seven $GalT^{-MCP/+}$ pigs. We performed quantitative real-time PCR, western blot, and flow cytometry analyses to confirm GalT and MCP expression. We showed that fibroblasts of the $GalT^{-MCP/-MCP}$ pig do not express GalT and its product Gal antigen, while efficiently express MCP. We also showed no expression of GalT, otherwise expression of MCP at heart, kidney, liver and pancreas of transgenic pig. Taken together, we suggest that the $GalT^{-MCP/-MCP}$ pig is a useful candidate to apply xenotransplantation study.
Galanin (Gal) is a 29-amino-acid neuropeptide which is expressed in superior cervical ganglion (SCG) neurons and plays a trophic role in the adult animal and acts as an inhibitory modulator of cholinergic and noradrenergic neurotransmission. Whether activation or inhibition of alpha-adrenoreceptors infl uences Gal mRNA expression in SCG neurons remains unknown. Here, we have evaluated the possible regulation of Gal mRNA expression with acute (4 h) and chronic (4 days) stimulation of alpha 1- and alpha 2-adrenoreceptor agonists or antagonists in primary cultured SCG neurons. The results showed that the amount of Gal mRNA expression in cultured SCG neurons increased signifi cantly after chronic stimulation with alpha 2-adrenoreceptor antagonist yohimbine compared with control SCG neurons at the same time point, whereas the amount of Gal mRNA expression decreased signifi cantly after chronic stimulation with alpha 2-adrenoreceptor agonist clonidine as compared with that in control group. All these effects were not dose-dependent on the administration of alpha 2-adrenoreceptor agonist clonidine or alpha 2-adrenoreceptor antagonist yohimbine. Alpha 1-adrenoreceptor agonist phenylephrine or antagonist prazosin chronic stimulation did not have effects on Gal mRNA expression. Acute exposure of these agents did not have effects on Gal mRNA expression. The present study showed that Gal may be regulated by activation or inhibition of alpha 2-adrenoreceptors, but not alpha 1-adrenoreceptors in sympathetic neurons.
본 연구에서 갈락토즈를 거의 사용하지 않고 glucose repression 정도가 줄어든 변이주를 이용하여 발현최적화를 수행하였다. 두 균주에서의 GAL promoter에 의한 외래단백질 생산시 glucose repression 정도에 대해 조사하였는데 대조해 Y2805는 glucose가 다 소비된 후 2~3시간 지난 후 발현이 시작되나 변이주 Y334는 약 0.5g/L 글루코즈 농도에서 25%정도의 발현이 이루어짐에 따라 변이주 Y334는 GAL promoter에 미치는 glucose repression정도가 매우 약한 장점을 확인하였다. GAL promoter에 의한 외래 단백질 생산시 발현을 위한 최적 갈락토즈 농도를 조사하였는데, Y3805는 3% 까지의 높은 갈락토즈 농도에서, 변이주 Y334는 1%정도의 낮은 갈락토즈 농도에서 각각 최대 발현량을 보였으며 변이주 Y334는 특히 0.01%정도의 낮은 갈락토즈 농도에서도 최대 발현의 60%량을 발현하였고 오히려 높은 갈락토즈 농도에서는 성장장애 현상을 보였다. 두 균주를 이용하여 배지중 pH가 외래 단백질 생산에 미치는 영향을 조사하였는데 두 균주 모두 pH 5근처에서 최대 발현량을 보임을 알 수 있었다. GAL promoter에 의한 외래 단백질 생산시 글루코즈와 갈락토즈, 에탄올의 소비속도를 조사하였는데, 글루코즈와 에탄올의 소비속도는 거의 비슷하였으나 갈락토즈 소비속도는 Y2805는 0.1232 g/L/hr/O.D.이고, 변이주 Y334는 0.0131g/L/hr/O.D. 이다. 또한 두균주의 분비효율을 조사하였는데 Y2805는 발효후반부에 총 생산 albumin중 약 70%는 분비되었고 30%는 cell 내 위치하는 것을 알 수 있었고 Y334의 경우에는 발효후반부에 총 생산 albumin중 50%가 cell 밖으로 분비되고 50%는 cell 내에 존재하는 것으로 확인되었다. 이는 Y334의 해결해야 할 단점으로 생각 되어지며 이러한 문제의 해결을 위해 albumin 생산성이 2~3배 증가된 초분비 S. cerevisiae 돌연변이주 개발이 현재 진행중이다.
대장균 효소 𝛽-gal를 이용하여 합성된 phenylethanol galactoside (PhE-gal)의 분자구조를 NMR (1H-와 13C-)과 고성능 mass spectrometry를 이용하여 분석하였다. 그 결과 PhE-gal은 1H NMR에서 2-phenylethanol (PhE)에 갈락토실기가 도입되었음을 나타내는 피크가 나타났다. 방향족 고리에서 오는 𝛿H 7.30~7.21 ppm의 피크와 𝛿H 2.88 ppm에 나타난 벤질기 위치의 CH2에서 오는 피크는 PhE가 존재함을 나타낸다. 지방족 사슬 영역인 𝛿H 4.31 ppm, 4.07 ppm과 𝛿H 3.86~3.38 ppm에서 나타나는 7개의 proton 피크로부터 단당류가 도입되었음을 확인할 수 있었다. 13C NMR 스펙트럼에서 나타난 12개의 탄소 피크 중 4개의 피크는 방향족 고리인 페닐기로부터, 또한 단당류에서 기인한 6개의 탄소피크가 존재하므로 PhE에 단당이 도입되었음을 알 수 있다. PhE-gal의 분자량을 확인하기 위하여 질량분석기로 분석한 결과 m/z가 307.1181인 PhE-gal의 sodium adduct ion ([M+Na]+)이 나타나 생성물이 PhE-gal임을 알 수 있었다. 따라서 본 연구결과 E. coli 𝛽-galactosidase에 의한 촉매반응으로 PhE에 갈락토즈가 첨가된 생성물인 PhE-gal이 성공적으로 생합성 되었음을 확인하였다.

Background: Bovine pericardium is one of the most widely used materials in bioprosthetic heart valves. Immunologic responses have been implicated as potential causes of limited durability of xenogenic valves. This study aimed to determine the effectiveness of decellularization and ${\alpha}$-galactosidase (${\alpha}$-gal) to remove major xenoreactive antigens from xenogenic tissues. Materials and Methods: Recombinant Bacteroides thetaiotaomicron (B. thetaiotaomicron) ${\alpha}$-gal or decellularization, or both were used to remove ${\alpha}$-gal from bovine pericardium. It was confirmed by ${\alpha}$-gal-bovine serum albumin-based enzyme-linked immunosorbent assay (ELISA), high-performance anion exchange chromatography, flow cytometry, 3,3'-diaminobenzidine-staining, and lectin-based ELISA. The mechanical properties of bovine pericardium after decellularization or ${\alpha}$-gal treatment were investigated by tests of tensile-strength, permeability, and compliance. Collagen fiber rearrangement was also evaluated by a 20,000${\times}$ transmission electron microscope (TEM). Results: Recombinant B. thetaiotaomicron ${\alpha}$-gal could effectively remove ${\alpha}$-gal from bovine pericardium B. thetaiotaomicron (0.1 U/mL, pH 7.2) while recombinant human ${\alpha}$-gal removed it recombinant human ${\alpha}$-gal (10 U/mL, pH 5.0). There was no difference in the mechanical properties of fresh and recombinant ${\alpha}$-gal-treated bovine pericardium. Furthermore, the TEM findings demonstrated that recombinant ${\alpha}$-gal made no difference in the arrangement of collagen fiber bundles with decellularization. Conclusion: Recombinant B. thetaiotaomicron ${\alpha}$-gal effectively removed ${\alpha}$-gal from bovine pericardium with a small amount under physiological conditions compared to human recombinant ${\alpha}$-gal, which may alleviate the harmful xenoreactive immunologic responses of ${\alpha}$-gal. Recombinant ${\alpha}$-gal treatment had no adverse effects on the mechanical properties of bovine pericardium.
We here demonstrate the evidence of increased anti-tumor and immunostimulating activities of crude extracts (GAL) from Galium. aparine L. In experimental lung metastasis of colon26-M3.1 carcinoma or B16-BL6 melanoma cells, prophylactically intravenous (i.v) administration of GAL significantly inhibited lung metastasis in a dose-dependant manner. In an in vitro cytotoxicity analysis, GAL at the concentration up to $500\;{\mu}g/ml$ did not affect the growth of B16-BL6 melanoma cells. In contrast, GAL showed the enhancement of splenocyte proliferating activity in a dose-dependent manner. Peritoneal macrophages stimulated with GAL produced various cytokines such as $1L-1{\beta},\;TNF-{\alpha},\;IFN-{\gamma}$ and IL-12. These data suggest that GAL has an antitumor activity to inhibit tumor metastasis, and its antitumor effects is associated with activation of nonspecific immnune related cells.
We have previously reported that genetically modified tumor cells with 4-1BBL have anti-cancer effects in a CT26 mouse colorectal tumor model. In this study, genetically modified tumor cells with 4-1BBL were evaluated for their potential as candidates for preventive and therapeutic cancer vaccine. To identify the effect of preventive and therapeutic vaccine of genetically modified tumor cells with 4-1BBL, tumor growth pattern of CT26-4-1BBL as a cancer vaccine was examined compared to CT26-beta-gal. In therapeutic vaccination, CT26-WT was inoculated into mice and then vaccinated mice with doxorubicin (Dox)-treated CT26-beta-gal and CT26-4-1BBL (single or three times). Triple vaccination with Dox-treated tumor cell inhibited tumor growth compared to single vaccination. Vaccination with CT26-4-1BBL showed an efficient tumor growth inhibition compared to vaccination with CT26-beta-gal. For preventive vaccination, Dox-treated CT26-beta-gal and CT26-4-1BBL was vaccinated into mice with three times and then administered mice with CT26-WT. Preventive vaccination with CT26-4-1BBL showed no tumor growth. Preventive vaccination with CT26-beta-gal also led to tumor-free mice. These results suggest that genetically modified tumor cells with 4-1BBL can be used as therapeutic or preventive cancer vaccine.
Endocrine disruptors (EDs) are the chemicals that affect endocrine systems through activation or inhibition of steroid hormone response. It is necessary to have a good system to evaluate rapidly and accurately endocrine-disrupting activities of suspected chemicals and their degradation products. The key targets of EDs are nuclear hormone receptors, which bind to steroid hormones and regulate their gene transcription. We constructed a co-expression system of Gal4p DNA binding domain (DBD)- ligand binding domain of human estrogen receptor $\alpha$ or $\beta$, and Gal4p transactivation domain (TAD)-co-activator AIB-1, SRC-1 or TIF-2 in Saccharomyces cerevisiae with a chromosome-integrated lacZ reporter gene under the control of CYC1 promoter and Gal4p binding site (GAL4 upstream activating sequence, GAL4$_{UAS}$). Expression of this reporter gene was dependent on the presence of estrogen or EDs in the culture medium. We found that the two-hybrid system with combination of the hER$\beta$ LBD and co-activator SRC-1 was most effective in the xenoestrogen-dependent induction of reporter activity. The extent of transcriptional activation by those chemicals correlated with their estrogenic activities measured by other assay systems, indicating that this assay system is efficient and reliable for measuring estrogenic activity. The data in this research demonstrated that the yeast detection system using steroid hormone receptor and co-activator is a useful tool for identifying chemicals that interact with steroid receptors.s.
T-flask와 air-lift 생물반응기를 이용하여 Autographa californica nuclear polyhedrosis virus (AcNPV)의 Tn5B1-4 세포에의 감염시 fatty acid 및 lipid, mannose, folic acid, $CaCl_2$등의 배지 첨가물이 재조합 ${\beta}-galactosidase\;({\beta}-gal)$ 생산에 미치는 영향을 조사하였다. Cholesterol, tocopherol, tricaprylin 또는 mannose를 첨가하거나 folic acid를 보강첨가 했을 때 AcNPV의 재조합 ${\beta}-gal$ 생산은 향상되었으나 $CaCl_2$의 보강 첨가는 ${\beta}-gal$ 생산을 높이는데 효과적이지 못했다. Air-lift 생물반응기에서 0.34 mM cholesterol, 2.2 mM mannose 및 0.045 M folic acid를 보강 첨가한 배지를 이용하였을 때 재조합 ${\beta}-gal$ 생산은 basal medium에 비해 약 2배 정도의 증진 효과가 있었다.
