덕태산(전북 진안)의 관속식물상 (Floristic Study of Mt. Deoktaesan, Jinan, Jeonbuk)

  • 덕태산(1,113 m)은 전라북도 진안군의 백운면과 장수군의 천천면에 걸쳐 위치해있다. 조사지로 덕태산과 선각산을 비롯한 인근을 선정하였으며, 조사는 2015년도 3월부터 10월까지 총 12일에 걸쳐 실시하였다. 결과의 정확성을 위해 꽃, 열매, 포자 등의 생식기관을 지닌 개체를 수집해 증거 표본으로 활용하여 관속식물목록을 작성하였다. 조사 결과 확인된 관속식물은 108과 341속 525종 4아종 65변종 14품종 총 608분류군으로, 한반도 관속식물의 약12.45%에 해당한다. 한반도 특산식물은 은사시나무, 키버들, 병개암나무, 제주큰물통이, 참개별꽃 등 15분류군의 분포가 확인되었다. 이 중 은사시나무, 섬초롱꽃, 개나리 3분류군은 식재된 것으로, 이를 제외한 12분류군이 조사지역에 자생하는 것으로 확인되었다. 산림청에서 제시한 희귀식물목록집에 의거한 멸종위기종(CR)은 백운기름나물 1분류군, 취약종(VU)은 백작약과 식재에 의한 모감주나무, 가시오갈피 3분류군, 약관심(LC)종은 너도바람꽃, 태백제비꽃, 섬초롱꽃 등 6분류군이 분포하는 것이 확인되었다. 환경부 지정 특정식물종에는 V등급1분류군, IV등급3분류군, III등급12분류군이 각각 조사되었다. 이중 IV등급 식물인 당마가목과 고로쇠나무는 각각 선각산 능선부, 덕태산 북사면 계곡부에 자생하는 것이 확인되었다.

리소좀 교통 이상을 초래하는 뮤코지방증 2형과 3형 환자의 섬유아세포를 이용한 신규 유전자 탐색 및 돌연변이에 대한 연구 (A Study on the Screening of the Novel Genes Associated with Lysosomal Trafficking and Mutation Detection in Fibroblasts of the Patients with Mucolipidosis type II and III)

  • 목적: 뮤코지방증 유형 II와 III은 리소좀 효소인N-acetylglucosaminyl-1-phosphotransferase (UDP-N-acetylglucosamine, GlcNAc-phospho-transferase)의 결손에 의해 초래되며, 상염색체 열성으로 유전되는 질환이다. ${\alpha}/{\beta}/{\gamma}$ subunit로 구성되는 이 효소의 결핍으로 인해 리소좀으로 운반되는 수십 종류의 효소들에 mannose-6-phosphate(M6P)를 부착하는 과정에 장애가 생겨 분해되지 않은 물질이 축적되어 질병이 초래된다. 이 질환에 있어서 산전 진단과 유전 상담을 위해서는 상기 효소의 해당 유전자가 밝혀져 있어야 하나, 현재 이 효소의 ${\gamma}$ subunit를 암호화하는 유전자가 GNPTAG에 해당된다고 밝혀져 있을 뿐 ${\alpha}/{\beta}$ subunit을 암호화하는 GNPTA에 해당하는 유전자는 밝혀져 있지 않고 돌연변이 역시 보고된 적이 없다. 본 연구는 리소좀 효소의 인산화에 관여하는 N-acetylglucosamine-1-phospho-transferase의 결함이 있는 환자의 섬유아세포를 이용하여 리소좀 연관 신규 유전자를 찾아내고, 그 유전자를 대상으로 돌연변이를 규명하고자 하였다. 방법: 이를 위해 5명의 환자와5명의 연령, 성별이 일치하는 정상아의 섬유아세포를 계대 배양하여 이 세포를 이용하여 수행한 subtractive hybridization을 통해 신규 유전자를 탐색하고, 신규 유전자를 대상으로 돌연변이 분석을 수행하였다. 결과: 연구 결과 환자에서 발현이 증가된 유전자 73개와 발현이 감소된 유전자 50개를 밝혀냈다. 분석된 유전자 중에서 MGC4170이환자에서는 발현되지 않으나 정상인에서는 발현됨을 발견하였고, 이 유전자가 아직 밝혀지지 않은 GNPTA로 확인되어 환자를 대상으로 돌연변이 분석을 시행하였다.분석 결과 기존에 돌연변이가 보고되었던 GNPTAG에는 돌연변이가 없었으나, MGC4170에는 7개의 돌연변이가 발견되었다. 본 연구는 GlcNAc-phosphotransferase의 ${\alpha}/{\beta}$ subunit에 해당되는 MGC4170의 최초 돌연변이 보고이다. 결론: 본 연구를 통해 뮤코지방증 II형과 III형에서 발현되는 유전자 군을 파악할 수 있었으며, 동시에 MGC4170 돌연변이를 규명함으로써 이 질환의 병리 기전 연구와 산전 진단에 기여하고자 한다.

Enhancement of Antibacterial Activity of Short Tryptophan-rich Antimicrobial Peptide Pac-525 by Replacing Trp with His(chx)

  • Trp residue is considered as one of the important constituents in antimicrobial peptides (AMPs) as it presence secured good activities in many cases. However, it is preferable to be changed because of their sensitivity towards light. We have synthesized the short Trp-rich AMP Pac-525 and its analogues to investigate the possibility of His(chx) as possible replacement analogue for Trp in AMPs. Based on the assay result of the antibacterial activity including anti-MRSA activity, His(chx) is considered as good candidate for the Trp replacement. Through these study, we found that His(chx) had several merits to design therapeutic antimicrobial agents compare to Trp in terms of i) increasing antibacterial activity without hemolytic activity, ii) successful in designing the short peptide (only four residues), iii) having anti-MRSA activity, iv) overcoming the light sensitivity. Furthermore, transmission electron microscopy (TEM) and dye leakage experiments suggested that P11 and P16 containing His(chx) kill bacteria via forming pore/ion channels on bacterial cell membranes.

한방병원에 내원한 암환자 52명에 대한 NK cell Activity 특성 분석 (Analysis of Natural Killer Cell Activity in 52 Cancer Patients at a Korean Medicine Hospital)

  • Objectives: This study was aimed to analyze the correlation of clinical characteristics of cancer patients treated with Korean Medicine and their natural killer cell activity (NKA). Methods: The medical records of 52 cancer patients who conducted NKA test at Cheonan Korean Medicine Hospital of Daejeon University from March 2016 to May 2018 were reviewed. NKA was determined at Seegene Medical institute using NK Vue Gold(R) kit, evaluating activated $IFN-{\gamma}$ by enzyme immunoassay after incubation of whole blood. Careful investigations were conducted by categorizing the patients by their sex, origin, stage, ECOG scale, and conventional treatment type. We used data from the medical records and statistical analysis using SPSS 25.0 V. Results: The average NKA of all patients was $623.1{\pm}722.0pg/mL$. The NKA value was higher for females than for males. No correlation was evident between age and NKA, but patients aged over 70 years had the lowest NKA average result. Stage III and IV patients showed clearly lower NKA values when compared to stage I and II patients. NKA and ECOG scale values showed no evident relationship. Conclusions: This study presented the characteristics of NKA in cancer patients treated with Korean medicine. The NKA result differed by gender and stage. Based on these findings, further study is needed regarding the clinical meaning of NKA.

변수화 모델을 통한 $InAs_xSb_{1-x}$ 화합물의 유전함수 분석

  • 적외선 영역에서의 밴드갭 에너지를 가지고 있는 III-V 족 화합물 반도체 물질인 $InAs_xSb_{1-x}$는 좋은 성장 안정성과 높은 전자, 홀 이동도를 가지며, 제작 비용이 적게 드는 등 적외선 광소자 제작에 많은 이점을 가지고 있기 때문에 그에 관한 연구가 최근 활발히 진행 되고 있다. 하지만 이러한 $InAs_xSb_{1-x}$를 소자 제작에 이용하기 위해서는 임의의 As 함량에 따른 InAsSb의 물질의 광학적 특성 정보가 필요하다. 본 연구에서는 1.5~6.0 eV 에너지 구간에서 $InAs_xSb_{1-x}$ ($0{\leq}x{\leq}1$) 화합물의 임의의 As 함량에 따른 유전함수를 분석하고 그 분석 변수들을 보고하고자 한다. 기성박막층착장치 (molecular beam epitaxy)를 이용하여 GaAs 기판 위에 성장 시킨 $InAs_xSb_{1-x}$ (x = 0.000, 0.127, 0.337, 0.491, 0.726, 1.000) 박막의 순수한 유전함수 $\varepsilon$을 화학적 에칭을 통해 산화막 층을 제거하여 타원편광분석법을 이용하여 얻었다. 측정된 유전율 함수는 Gaussian-broadened polynomial 들의 합으로서 반도체 물질의 유전함수를 정확히 기술하는 변수화 모델을 이용하여 재현하였다. 변수화 모델을 통해 얻어진 각각의 변수들을 As 조성비 x 에 대한 다항식으로 피팅하여 임의의 As 조성비에 대한 변수 값을 얻었다. 그 결과 임의의 조성비에 따른 $InAs_xSb_{1-x}$ ($0{\leq}x{\leq}1$) 의 유전율 함수를 얻어낼 수 있었다. 우리는 이러한 결과가 물질의 실시간 성장 모니터링이나 다층구조 분석, 광소자의 제작 등에 유용한 정보를 제공할 것으로 확신한다.

기능성 소화불량 환자 97명의 변증유형별 특성 (Pattern Identification of 97 Functional Dyspepsia Patients and the Characteristics of Each Pattern Type)

  • Objective: This study was designed to identify and explore the pathological patterns of functional dyspepsia (FD) patients. We also evaluated the usefulness of the Pattern Identification Questionnaire by comparing it with other assessment tools for FD. Methods: We recruited 97 FD patients based on the Rome III criteria for FD diagnosis. The pathological patterns of the subjects were determined by the Pattern Identification Questionnaire. Their dyspepsia-related symptoms were assessed using the Gastrointestinal Symptom Questionnaire (GIS) and the Pyeongwi-san (Pingwei-san) Patternization Questionnaire. Depressive symptoms were evaluated with the Beck Depression Inventory (BDI) and quality of life with the Functional Dyspepsia-Related Quality of Life (FD-QoL) Questionnaire. Tongue coating was measured by the Digital Tongue Diagnosis System (DTDS). Results: The male to female ratio was 1:1.1, and the forties and fifties age groups were largest in number. The spleen deficiency and phlegm-dampness pattern was the most common pattern found among the FD patients. No significant differences in the GIS, BDI, FD-QoL, and DTDS scores were found among the five pattern types. All pattern types showed significant correlation with GIS, Pyeongwi-san Patternization Questionnaire, and FD-QoL scores. Conclusions: Pattern Identification Questionnaire can not only identify the pathological pattern types of FD patients but also evaluate the severity of their symptoms. Compared to conventional assessment tools for FD, it could enable a more dynamic evaluation of FD patients reflecting the severity of dyspeptic symptoms and the quality of life. Further studies on the Pattern Identification of FD patients are anticipated in order to improve the diagnosis and therapy for Korean FD patients.

저강도레이저 조사가 근육압좌손상 후 척수분절의 EGF 발현에 미치는 영향 (Effects of Low Power Laser for the Expression of EGF after Muscle Crush Injury)

  • Low energy laser irradiation(LELI) therapy in physical therapy is widespread but the mechanisms are not fully understood. The purpose of the present study was to examine the epidermal growth factor(EGF)'s expression within lumbar spinal cord which corresponding with crushed extensor digitorum longus(EDL) of rats after low-power laser irradiation applied. After a crushed injury on the right EDL, low-power laser irradiation was applied by using 2000mW, 2000Hz, 830nm GaAlAs(Gallium-aluminum-arsenide) semiconductor diode laser. The laser treatment was performed with 10 minutes daily for 3days. After EDL crush injury, EGF immunoreactive positive neurons in experimental group were progressively decreased from the first to third days. Especially 1 day subgroup is highly expressed in dorsal horn(Lamina I, II, III) and around of central cannal of spinal cord(Lamina VII). Control group was only expressed slightly at 3 days. This study suggests that LELI stimulate that release and migration of EGF in spinal cord, which distict to wound site, therfore promote wound healing of EDL crush injury.

비 무기산 세척제에 의한 중금속 오염 토양 세척효과 평가 (Evaluation on Soil Washing of Metal-contaminated Soil using Non-Inorganic Acids)

  • Inorganic acids such as HCl, HNO3, and H2SO4 have been commonly applied to soil washing of heavy metals-contaminated soil due to their cost-effectiveness. However, implementing the 'Chemical Substance Control Act' requires off-site risk assessment of the chemicals used in the soil washing. Therefore, in this study, organic acids or Fe(III)-based washing agents were evaluated to replace commonly used inorganic acids. Ferric removed heavy metals via H+ generated by hydrolysis, which is similar to the HCl used in the control group. Oxalic acid and citric acid were effective to remove Cu, Zn, and Cd from soil. Organic acids could not remove Pb because they could form Pb-organic acid complexes with low solubility. Furthermore, Pb could be adsorbed onto the iron-organic acid complex on the soil surface. Ferric could remove exchangeable-carbonate, Fe-Mn hydroxide, and organic matter and sulfides bound heavy metals (F1, F2, and F3). Organic acids could remove the exchangeable-carbonate and Fe-Mn hydroxide bound metals (F1&F2). Therefore, this research shows that the fractionation of heavy metals in the soil and the properties of washing agents should be considered in the selection of agents in the process design.

Muscle Radiation Attenuation in the Erector Spinae and Multifidus Muscles as a Determinant of Survival in Patients with Gastric Cancer

  • Purpose: To determine the prognostic role of muscle area and muscle radiation attenuation in the erector spinae (ES) and multifidus (MF) muscles in patients undergoing gastrectomy. Methods: Patients with stage I-III gastric cancer undergoing gastrectomy were retrospectively enrolled in this study. Clinicopathologic characteristics were collected and analyzed. Both paraspinal muscle index of ES/MF muscles (PMIEM) and paraspinal muscle radiation attenuation in the same muscles (PMRAEM) were analyzed at the 3rd lumbar level using axial computed tomographic images. Cox regression analysis was applied to estimate overall survival (OS) and disease-free survival (DFS). Results: There was only a weak correlation between PMIEM and PMRAEM (r= 0.28). Multivariate Cox regression revealed that PMRAEM, but not PMIEM, was an important determinant of survival. PMRAEM along with age, tumor-node-metastasis (TNM) stage, perineural invasion, and serum albumin level were significant determinants of both OS and DFS that constituted Model 1. Harrell's concordance index and integrated area under receiver operating characteristic curve were greater for Model 1 than for Model 2 (consisting of the same covariates as Model 1 except PMRAEM) or Model 3 (consisting of only TNM stage). Conclusion: PMRAEM, but not PMIEM, was an important determinant of survival. Because there was only a weak correlation between PMIEM and PMRAEM in this study, it was presumed that they were mutually exclusive. Model 1 consisting of age, TNM stage, perineural invasion, serum albumin level, and PMRAEM was greater than nested models (i.e., Model 2 or Model 3) in predicting survival outcomes.

Bonding Temperature Effects of Robust Ag Sinter Joints in Air without Pressure within 10 Minutes for Use in Power Module Packaging

  • Ag sintering technologies have received great attention as it was applied to the inverter of Tesla's electric vehicle Model III. Ag sinter bonding technology has advantages in heat dissipation design as well as high-temperature stability due to the intrinsic properties of the material, so it is useful for practical use of SiC and GaN devices. This study was carried out to understand the sinter joining temperature effect on the robust Ag sintered joints in air without pressure within 10 min. Electroplated Ag finished Cu dies (3 mm × 3 mm × 2 mm) and substrates (10 mm × 10 mm × 2 mm) were introduced, respectively, and nano Ag paste was applied as a bonding material. The sinter joining process was performed without pressure in air with the bonding temperature as a variable of 175 ℃, 200 ℃, 225 ℃, and 250 ℃. As results, the bonding temperature of 175 ℃ caused 13.21 MPa of die shear strength, and when the bonding temperature was raised to 200 ℃, the bonding strength increased by 157% to 33.99 MPa. When the bonding temperature was increased to 225 ℃, the bonding strength of 46.54 MPa increased by about 37% compared to that of 200 ℃, and even at a bonding temperature of 250 ℃, the bonding strength exceeded 50 MPa. The bonding strength of Ag sinter joints was directly influenced by changes in the necking thickness and interfacial connection ratio. In addition, developments in the morphologies of the joint interface and porous structure have a significant effect on displacement. This study is systematically discussed on the relationship between processing temperatures and bonding strength of Ag sinter joints.
