Glucosyltransferase Inhibition of Streptococcus mutans by Dioscorea batatas and Prunella vulgaris extract (마와 꿀풀 추출물에 의한 Streptococcus mutans의 산 생성 및 Glucosyltransferase 저해효과)

  • Streptococcus mutans is the major causative factor in dental caries and has been known to induce dental caries by the process of initial attachment proliferation and acid production. The pH of Streptococcus mutans(S. mutans) media and glucosyltransferase(GTase) activity were determined to evaluate the anticariogenic activity of Dioscorea batatas and Prunella vulgaris hexane fraction. In the experiment of hexane fraction of Dioscorea batatas, the relative growth ratio(RGR) against Streptococcus mutans (S. mutans) was determined as 93% in concentration of 0.125 mg/ml, 100% in 0.25 mg/ml. The hexane fraction of Prunella vulgaris revealed relative growth ratio(RGR) against Streptococcus mutans (S. mutans) as 86% in concentration of 0.25 mg/ml, 100% in 0.5 mg/ml. The relative growth ratio(RGR) against of hexane fraction were determined as 92% in concentration of 0.25 mg/ml, 100% in 0.5 mg/ml. The acidities were pH 7.2 on Dioscorea batatas and 7.0 on Prunella vulgaris in 2.0 mg/ml in contrast to pH 5.6 on Dioscorea batatas and 5.2 on Prunella vulgaris in control. The inhibitory effect to activity revealed 35% on Dioscorea batatas and 25% on Prunella vulgaris in 2.0 mg/ml. The inhibitory effect of Dioscorea batatas was more potent than Prunella vulgaris.

Anti-cariogenic Activities of Mushroom Extracted with Various Solvent Systems (용매 변화에 따른 버섯추출물의 항충치활성)

  • Various mushroom extracts were examined for their anti-Streptococcus mutans and anti-glucosyltransferase (GTase) activities. Mushrooms were extracted with chloroform, methanol, acetone, ethyl acetate, and distilled water. The chloroform, methanol, and ethyl acetate extracts showed higher antimicrobial activity than that of the others. The inhibitory effects of the chloroform, methanol, and ethyl acetate extracts of Lyopyllum ulmarium, which have high anti-microbial and GTase activities, on the growth of S. mutans were examined and the optimal ratio of solvents was also evaluated to investigate the influence of solvents using a simplex centroid design. The anti-cariogenic effects of the L. ulmarium extract on S. mutans was influenced by the solvent, and the optimum anti-cariogenic activity of the extract was obtained with the ratio of chloroform:methanol:ethyl acetate=1:1:1 (v/v/v). These results suggest that solvent selection is an important factor to extract anti-cariogenic materials effectively from mushrooms.

Anticariogenic Activity and Glucosyltransferase Inhibition of Phenolic Compounds (페놀성 화합물의 항충치활성 및 Glucosyltransferase 억제효과)

  • Fourteen phenolic compounds(benzoic acid, p-hydroxybenzoic acid, protocatechuic acid, vanillic acid, syringic acid, gallic acid, caffeic acid, ferulic acid, (+)-catechin, quercetin, rutin, catechol, chlorogenic acid and L-ascorbic acid) were examined for their effects on the anticarigenic activity. Among tested samples, catechol was significantly inhibited the S. mutans, exhibiting an clear zone 18.5-19.5mm by 10 mg/disc level. The minimal inhibition concentration(MIC) of the phenolic compounds for Streptococcus mutans, M1 and M2 strain were determined as 2,000 ppm, whereas catechol was 1,000 ppm. The activity of glucosyltransferase(GTase) was significantly inhibited by catechol, at 10 ppm(58.7%), 50 ppm(60.7%) and 100 ppm(88.4%) and 500 ppm(89.6%), respectively. Among them, catechol showed most significant anticariogenic activity as well as inhibition of GTase activity.

Isolation of an Anticariogenic Compound from Magnoliae Bark (후박피(Magnoliae bark)로부터 항충치활성을 갖는 물질의 분리)

  • We have screened total 32 herbal drugs to find the highest activity against human cariogenic enzyme, glucosyltransferase (GTase) from the extracts of Magnoliae bark. The extracts were separated into three phases, i. e. water, n-butanol and ethylacetate according to their solvent polarity. Among them, ethylacetate fraction had approximately more than 70% of total activities, and the active principle was further isolated by prep. HPLC following silicagel column chromatography to yield single compound as white powder. The chemical structure of the compound was finally elucidated to be 4,4'-dihydroxy-3,3'-dimethoxylignan from the spectral data of FAB-MS. $^1H-\;and\;^{13}C-NMR$ spectrometries. The compound was also shown to have relatively strong antibacterial activity against ten types of cariogenic oral bacteria and one kind of Actinomyces sp.

Antibacterial Effects of Sophora flavescens on Streptococcus mutans (고삼의 Streptococcus mutans에 대한 항세균 효과)

  • The purpose of this study is to investigate antibacterial activities of Sophora flavescens Ait. extracts for the prevention of dental caries caused by Streptococcus mutans. The fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. Showed strong growth inhibition activity against Streptococcus mutans (MIC, 3.13 ${\mu}$g/ml). The glucosyltransferase activity inhibited the formation of glucan by the fraction 5-4-3 and showed 77% of the antiproliferative effect at 100 ${\mu}$g/ml. This showed a significant activity (p<0.05). These results suggest that the fraction 5-4-3 of the ethyl acetate soluble extract of Sophora flavescens Ait. may be a valuable material as the prevention of dental caries.

  • Objectives : The purpose of the study is to investigate the activities of Nelumbo nucifera leaf extracts on Streptococcus mutans, Streptococcus mitis, Streptococcus sobrinus, Porphyromonas gingivalis, Prevotella intermedia, and Treponema denticola. Methods : The inhibitory effect of lotus leaf extracts on the growth of oral bacteria was assessed in experiments with extracts from freshly harvested and pulverized lotus leaves and bacterial cultures of dental caries. Results : The results showed that N. nucifera extracts possess antimicrobial activity on all bacterial strains. The minimal inhibitory concentration (MIC) values varied from 4 mg/ml to 10 mg/ml against antimicrobial activity. The relative growth ratio (RGR) against of N. nucifera extracts were determined as 50% in concentration of 4.0 mg/ml. The extract of N. nucifera was effective in reducing on the glucosyltransferase (GTase) activity of six strains in vitro. Conclusions : Methanol extracts of lotus leaves showed antimicrobial effects on three bacterial species causing dental caries and three bacterial species causing periodontitis, as well as inhibitory effects on GTase activity.

  • In the course of our studies, novel flavan-3-ol structure isolated from Theobroma cacao L. husk was established by the thiolysis, desulfurization and spectroscopic method. The structure was identified for cinnamtannin A-2 containing the tetrameric epicatechin and molecular weight was [1153] by FAB-MS negative ion. The inhibitory effect on the glucosyltransferase activity was investigated. Cinnamtannin A-2 showed complete inhibition at 0.03 mM and inhibited on the glucosyltransferase noncompetitively. The hydroxyl group of flavan-3-ol was supposed to be the essential element for inhibition on the glucosyltransferase.

Effects of Dietary Xylooligosaccharide on Hepatic UDP-Glucuronyl Transferase Activity and Compositions of Fecal Sterols in Rat Fed High Cholesterol Diets (Xylooligo당이 고콜레스테롤 식이 횐쥐 간조직의 UDP-Glucuronyl Transferase 활성과 분변중 Sterol류 조성에 미치는 영향)

  • This study was conducted to examine the effects of dietary xylooligosaccharide on UDP-glucuronyl transferase (UDP-GTase) activity and excretion of fecal sterols in rat fed high cholesterol diet. Sprague-Dawley male rats weighing 100$\pm$10 g were randomly divided into five groups, one with normal diet and four with high cholesterol diets containing 1% cholesterol (w/w). The high groups with cholesterol diet groups were classified into xylooligosaccharide free diet (C), 5% xylooligosaccharide diet (C5XO), 10% xylooligosaccharide diet (C10XO), and 15% xylooligosaccharide diet (C15XO) group according to the five groups of dietary xylooligosaccharide by weights. The experimental diets were fed ad libidum for 4 weeks. Fecal weights were increased 86% by xylooligosaccharide. Fecal total lipid contents including fecal neutral and acidic sterols in xylooligosaccharide groups were significantly higher than those of the normal and C groups, and especially that of C10XO group was the highest among all experimental groups. Activity of UDP-glucuronyl transferase (UDP-GTase) in liver in C group was 35% higher than that of normal group and the activities in C5XO, C10XO and C15XO groups were 15%, 41%, and 21% higher than in C group, respectively. Fecal bile acid excretions per day were increased 3.1, 3.6 and, 2.8 folds in C5XO, C10XO, and C15XO groups, respectively, compared with that of C group. Contents of neutral sterol, coprostanol, and coprostanone were higher in xylooligosaccharide groups than in C group. These results suggest that dietary xylooligosaccharide may act as potential substitute for a dietary fiber capable of improving a gastrointestinal function and lipid metabolism.

  • The optimal condition for the production of a glucan and a fructan synthesizing enzymes from Leuconostoc mesenteroides NRRL B-1149 were studied based on the different medium compositions. Response surface methodology was applied to find the optimistic condition showing the relationship between the fermentation response (enzyme activities) and the fermentation variable concentrations of yeast extract, peptone concentration, K2HP04 concentration and sucrose. Optimum medium composition for both enzymes production was $0.75\%$ yeast extract, $0.72\%$ peptone, $1\%$ K2HP04 and $2.17\%$ sucrose. Using this medium, the activities produced in culture was 0.90 U/m~ for glucosyltransferase (GTase) and 0.96 U/ml for fructosyltransferase (FTase). After purification of 1149FTase by consecutive chromatographies using Sephadex G-150 and DEAE-Sepharose, a 1149FTase of 210 kDa on $7\%$ polyacrylamide gel was isolated and it synthesized soluble fructan. The 1149GTase showed a band of 180 kDa on $8\%$ polyacrylamide gel after purification using Bio-Gel P-100 gel chromatography and DEAE-Sepharose ion exchange chromatography and it synthesized insoluble glucan. The linkages of polymers were determined by methylation using Hakomori reagent and following NMR analysis. The glucan was composed of a(1~6) and a(1~3) linkages and the fructan was levan.

Inhibitory effect of n-hexane extract from Korean red ginseng marc against Streptococcus mutans causing dental caries (홍삼박 n-hexane 추출물의 충치를 유발하는 Streptococcus mutans 저해 효과)

  • In this study, to investigate the anticariogenic effect of red ginseng, the antibacterial effect, cell adherence and biofilm formation inhibitory effect of n-hexane extract of red ginseng marc (HERGM) against Streptococcus mutans, the causative bacteria of caries, were measured. The growth of S. mutans was inhibited in proportion to the concentration of HERGM, and was hardly observed at a concentration above 125 ㎍/mL (MIC =125 ㎍/mL). It was found that HERGM acts on the cell membrane and the nucleic acid component of the cell was leaked. In addition, HERGM inhibited the adherence and biofilm formation of S. mutans by more than 90% at a concentration of 125 ㎍/mL. GTase activity was completely inhibited at a concentration of 50 ㎍/mL of HERGM. In conclusion, it was found that HERGM commonly inhibited the growth and biofilm formation of S. mutans.
