Variations of Gonadotropin Subunits mRNA Levels at Different Stages of Ovarian Development in Masu Salmon, Oncorhynchus masou

  • The variations of gene expression and pituitary contents of GTH subunits during the ovarian development of masu salmon, Oncorhynchus masou, were investigated. The pituitary GTHs contents was measured by radioimmunoassays (RIAs) using purified GTH subunits and their antibodies. Pituitary contents of GTH $I\beta$ gradually increased from April through August, and reached the maximum in October. On the other hand, pituitary contents of GTH $II\beta$ remained low until August, but they rapidly increased in October. Total RNAs were prepared from pooled pituitaries and the GTH subunits mRNA in pituitaries was quantified by Northern blot hybridization using masu salmon cDNA probes for each GTH subunit. GTH $I\beta$ mRNA level increased with the progression of ovarian maturity. However, GTH $II\beta$ mRNA was detected only at a more advanced stage, and were extremly high at ovulation. A high levels for GTH a mRNA was detected only at ovulation stage. The synchronous increase in pituitary contents and mRNA levels suggested that ovarian maturity in masu salmon was regulated by both GTH I and GTH II.

Effect of Testosterone on the mRNA Levels of Gonadotropin Subunits in the Immature Rainbow Trout Pituitary

  • In order to clarify the role of gonadal sex steroids in the synthesis of gonadotropin (GTH) subunits in immature rainbow trout, we examined in vitro and in vivo effects of testosterone (T) on the pituitary mRNA levels of GTH I $\beta$, GTH II$\beta$ and a subunits by Northern blot analysis and on the pituitary content levels of GTH I$\beta$ and GTH II$\beta$by radioimmunoassay (RIA). The mRNA levels of the a subunit in T-treated fish were not changed more dramatically than those in control fish both in vivo and in vitro. Interestingly, the mRNA levels of GTH I$\beta$ in T-treated fish were shown to be slightly lower than those in the control fish under these experimental conditions, but no differences were observed in pituitary GTH I$\beta$ contents. In contrast, the mRNA levels and pituitary contents of GTH II$\beta$ subunit were strongly increased by T both in vivo and in vitro. These results demonstrate that the expressions of GTH I$\beta$ and II$\beta$ subunit genes in immatue rainbow trout pituitary are subjected to differential regulation by T.

Isolation of cDNAs for Gonadotropin-II of Flounder (Paralichthys olivaceus) and Its Expressions in Adult Tissues

  • Gonadotropin (GTH) is a pituitary glycoprotein hormone that regulates gonadal development in vertebrates. In teleosts, two types of gonadotropins, GTH-I and GTH-II, are produced in the pituitary, and they comprised of common ${\alpha}$ and distinct ${\beta}$ subunits. In the present study, the cDNAs encoding GTH ${\alpha}\;and\;GTH-II{\beta}$ subunits were cloned and sequenced from flounder (Paralichthys olivaceus) pituitary cDNA library. The nucleotide sequence of the a subunit was 619 bp long, encoding 124 amino acids, and that of the $GTH-II{\beta}$ subunit was 538 bp long, encoding 145 amino acids. GTH subunits had well conserved cysteines, when aligned with other members of the glycoprotein family. The ${\beta}$ subunit of gonadotropin II ($GTH-II{\beta}$) had a different N-linked glycosylation site. RT-PCR analysis showed an increase of GTH II mRNA levels in association with gonadal development, and also showed that the mRNA expression of the ${\alpha}$ subunit was detected only in tissues from pituitary glands.

  • We cloned and sequenced the cDNA encoding the $FSH{\beta}$ and $LH{\beta}$ subunits from the pituitary of the blacktip grouper Epinephelus fasciatus, which regulate vitellogenesis and maturation in vertebrates, to achieve stable and healthy gametes. The full-length cDNAs of $FSH{\beta}$ and $LH{\beta}$ were 571 bp and 617 bp, encoding 120 amino acid (aa) and 147 aa proteins, respectively. The deduced amino acid sequences of $FSH{\beta}$ and $LH{\beta}$ were highly homologous (68-97%) to those of other Perciformes; E. bruneus, Dicentrarchus labrax, Thunnus thynnus, and Pseudolabrus sieboldi. Phylogenetic analysis showed that the deduced $FSH{\beta}$ and $LH{\beta}$ amino acid sequences were categorized as a distinct subunit in the $GTH{\beta}$ family, and are closely related to the teleostei $FSH{\beta}$ and $LH{\beta}$, respectively. $FSH{\beta}$ and $LH{\beta}$ mRNA exhibited high abundance in the pituitary gland and low in other brain areas, but were not present in peripheral tissues, as determined by RT-PCR.

Neurokinin B-related Peptide Suppresses the Expression of GnRH I, Kiss2 and tac3 in the Brain of Mature Female Nile tilapia Oreochromis niloticus

  • Neurokinin B (NKB) and neurokinin B related peptide (NKBRP) belong to tachykinin peptide family. They act as a neurotransmitter and/or neuromodulator. Mutation of NKB and/or its cognate receptor, NK3R resulted in hypogonadotropic hypogonadism in mammals, implying a strong involvement of NKB/NK3R system in controlling mammalian reproduction. Teleosts possess NKBRP as well as NKB, but their roles in fish reproduction need to be clarified. In this study, NKB and NKBRP coding gene (tac3) was cloned from Nile tilapia and sequenced. Based on the sequence, Nile tilapia NKB and NKBRP peptide were synthesized and their biological potencies were tested in vitro pituitary culture. The synthetic NKBRP showed direct inhibitory effect on the expression of GTH subunits at the pituitary level. This inhibitory effect was confirmed in vivo by means of intraperitoneal (ip) injection of synthetic NKB and NKBRP to mature female tilapia (20 pmol/g body weight [BW]). Both NKB and NKBRP had no effect on the plasma level of sex steroids, E2 and 11-KT. However, NKBRP caused declines of expression level of GnRH I, Kiss2 and tac3 mRNAs in the brain while NKB seemed to have no distinct effect. These results indicate some inhibitory roles of NKBRP in reproduction of mature female Nile tilapia, although their exact functions are not clear at the moment.

Characterization of Pubertal Development Phases in Female Longtooth Grouper, Epinephelus bruneus via Classification of Bodyweight

  • Puberty is the developmental period which animals obtain the ability of reproducing sexually for the first time in life. In commercially important aquaculture fish species, the onset of puberty is a matter of major interest due to controlling of sexual maturation to improve broodstock management. To investigate pubertal characteristics of female longtooth grouper (Epinephelus bruneus), specimens were classified into three groups by the bodyweight, including 1, 2, and 3 kg group. Thereafter, we focused on ovarian development and level changes of endocrine regulation factors (GnRH, GTHs, steroid hormone). In the non-breeding season (April), the levels of endocrine regulation factors showed increasing trends in accordance with bodyweight gaining; nevertheless, the oocytes were growth phase belongs to almost peri-nucleous stages in all groups. In the breeding season (June), the levels of endocrine regulation factors were fluctuated that decreases in levels of sbGnRH and $FSH{\beta}$ mRNA expressions along with serum $E_2$ concentrations in 3 kg of group. However, $LH{\beta}$ mRNA expression levels sustained increasing trends by the bodyweight. Moreover, the oocytes developed that 2 kg and 3 kg groups obtained plentiful vitellogenic oocytes while 1 kg group was still composed with greater part of pre-vitellogenic oocytes. Especially, the oocytes of 3 kg group reached over 450 ${\mu}m$ of diameters that indicating possibility to enter the final maturations. These results suggest that the progress of pubertal development in female E. bruneus could be classify into three phases via bodyweight, including pre-puberty (1 kg), early-puberty (2 kg) and puberty (3 kg).

Expression Profiles of Kiss2, GPR54 and GnRH Receptor I mRNAs in the Early Life Stage of Nile Tilapia, $Oreochromis$ $niloticus$

  • Kisspeptin has been implicated in the process of puberty onset in various animal groups. This peptide is encoded by a gene, Kiss1 in avian and mammalian species. Contrary to these higher vertebrates, however, fish appeared to have another gene, Kiss2 that also codes for the precursor peptide of kisspeptin. To figure out biological significance of this gene during the puberty onset in fish, the expression profile of Kiss2 gene was investigated in the brain of Nile tilapia together with genes of GPR54, GnRH receptorI (rGnRHI) and GTH subunits ($LH{\beta}$ and $FSH{\beta}$). Expression of Kiss2 mRNA significantly increased at 2 weeks post hatch (wph) and 13 wph ($P$<0.05). This increase coincided with the increases of GPR54 and rGnRH I gene expression. Detection of $LH{\beta}$ and $FSH{\beta}$ subunit gene expression was possible later than 13 wph, indicating the activation of gonadotrophs in the pituitary. Data obtained from this study strongly suggest that, in addition to Kiss1 gene, Kiss2 gene is deeply associated with the onset of puberty by the activation of hypothalamus pituitary gonadal axis in Nile tilapia.
