가루녹차를 첨가한 설기떡이 저장중 품질 변화 (Quality Changes of Sulgiduk Added Green Tea Powder during storage)

  • This study was quality changes of sulgiduk added green tea powder during storage. Sulgiduk prepared with various concentrations of green tea powder; 0%(control group), 0.5%(GT 0.5 group), 1%(GT 1.0 group), 1.5%(GT 1.5 group), 2%(GT 2.0 group), and the changes of its quality during 7 days of storage were investigated. Total microbe numbers, the acidity and pH in sulgiduk during storage were decreased with increasing the added amounts of a green tea powder, and especially those of GT 1.0 and GT 1.5 groups had relatively the lower than other groups. The "L" value(lightness) of the control group was the highest among five groups, and its value was decreased throughout storage, and especially GT 0.5 groups had the lowest brightness. The "a" value(reddness) of the control group was higher than other four groups, and increased GT 2.0>GT 1.5>GT 1.0>GT 0.5 groups, in that order. The "b" value(yel lowness) was increased in the order; GT 2.0>GT 1.5>GT 1.0>GT 0.5 groups. These results indicated that GT 1.0 group showed the best quality than other groups of sulgiduk during storage. of sulgiduk during storage.

가루녹차를 첨가한 설기떡의 관능적 품질특성 (Quality Characteristics of Seolgiddeok added with Green Tea Powder)

  • The purpose of this study was to find out the optimal mixing ratio of green tea powder, sugar, and water for the preparation of Seolgiddeok through Response Surface Methodology based on the color, sensory, and texture test. The oganoleptic and textural properties of Seolgiddeok prepared with various concentrations of green tea powder (0% (control group), 0.5% (GT-0.5 group), 1% (GT-1.0 group), 1.5% (GT-1.5 group), 2% (GT-2.0 group)), and their quality changes during storage were also investigated. The optimal mixing ratio of green tea powder, sugar, and water for preparing the best quality Seolgiddeok was 1.0%, 12%, and 22%, respectively. The proximate composition of green tea powder was 21.70% of crude protein, 8.49% of crude lipid, 2.95% of reducing sugar, and 6.40% of ash. The contents of crude lipid, reducing sugar, and catechins in Seolgiddeok added with a green tea powder were increased with increasing the content of green tea powder. The hardness and gumminess of GT-1.0 group were the lowest among four groups, whereas GT-1.0 and GT-2.0 groups had the lowest cohesiveness. While the control group was the lowest in adhesiveness, the springness was not significantly different among all groups. GT-0.5 and GT-1.5 groups were the highest in sweet taste and colorfulness, respectively. However, GT-1.0 group had the best overall quality. The“b”value (yellowness) was increased with the increase of green tea powder content. Above results indicated that GT-1.0 group showed the best quality of Seolgiddeok through organoleptic and rheology tests.

Role of plastidic glucose transporter in source metabolism of Arabidopsis

  • To study the biochemical and physiological role of the plastidic glucose transporter (pGlcT) in carbohydrate metabolism, we characterized transgenic plants with mutations in the pGlcT gene (GT), gt-1 and gt-2, as well double mutants of GT and the maltose transporter (MEX1) and GT and the triose phosphate/phosphate translocator (TPT), GT and the cytosolic fructose-1,6-bisphosphatase gene (cFBP), and MEX1 and TPT, gt-1/mex2, gt-1/tpt-2, gt-1/cfbp-1, mex1-1/tpt-2, respectively. Compared to the wild type, all mutants except the gt-1/cfbp-1 mutant lines displayed higher starch accumulation and higher levels of maltose. Starch accumulation is due to a decrease in starch turnover, leading to an imbalance between the rates of synthesis and degradation. Sucrose levels of gt alleles were higher than those in wild-type plants during the light period, suggesting possible nightly supplementation via the maltose transport pathway to maintain proper carbohydrate partitioning in the plant leaves. The gt plants displayed less growth retardation than mex1-1 mutant and gt-1/mex2 double mutant displayed accumulativesevere growth retardation as compared to individual gt-1 and mex1-1 mutants, implying that the maltose transporter-mediated pathway is a major route for carbohydrate partitioning at night. The gt-1/tpt-2, mex1-1/tpt-2 and gt-1/cfbp-1 double mutants had retarded growth and low chlorophyll content to differing degrees, indicating that photosynthetic capacity had diminished. Interestingly, the gt-1/tpt-2 line displayed a glucose-insensitive phenotype and higher germination rates than wild type, suggesting its involvement not only in carbon partitioning, but also in the sugar signaling network of the pGlcT and TPT.

  • Objectives : Gal-Geun-Tang (GT) has been described from SANGHAN in Korean traditional medicine and known to act against cold, fever, hypertension, and nasal catarrh. However, little has yet been learned about the effect of GT on immune function. In the current study, in vitro and in vivo immunomodulatory activity of GT (water extract) was investigated.Methods : Water extract of GT induced in vitro proliferation of spleen cells and significantly increased their proliferative responses during anti-CD3 activation. Using purified splenic T and B cells, it was revealed that GT has a mitogenic activity to B cells and promotes their proliferation induced by lipopolysaccharide, whereas T cell proliferation was not triggered and GT was rather inhibitory to T cell activation caused by anti-CD3 antibody. In the presence of antigen presenting cells (APC), GT addition resulted in a significant increase of IFNγ and IL-4, but not IL-2, production. However, addition of high concentration (1,000㎍/㎖) of GT led to a marked reduction in T cell cytokine production and under such condition, GT facilitated apoptosis of T cells when examined by flow cytometry with propidium iodide staining.Results : In vivo immunomdulation of GT was also investigated using a mouse model. Following keyhole limpet hemocyanin (KLH) immunization, GT (1 ㎎/day) was orally administered for 9 days. Cell numbers in thymus, spleen and peripheral blood were not altered by GT administration, indicating that such dose is not immunotoxic. Cell numbers in draining lymph nodes (LN) and ex vivo Ag-specific proliferation of LN cells were significantly elevated by GT administration. However, any preferential stimulation of T or B and CD4+ or CD8+ T cell subpopulations was not observed in a flow cytometric analysis of LN cells. This result shows that GT does not promote in vivo B cell proliferation while GT enhances Ag-specific proliferation of LN cells, unlike what was observed in vitro.Conclusions : For a further understanding of in vivo immunomodulatory activity of GT, ex vivo cytokine production of LN cells obtained from KLH-immunized mice was evaluated. Ag-specific IFNγ production was significantly higher in GT-treated mice when compared to PBS-treated control mice. In contrast, IL-4 production in GT-treated group was comparable to control group unlike to in vitro data. In addition, GT administration did not result in any significant differences in serum levels of Ig (IgM, IgG1 and IgG2a) between GT-treated and control groups. Taken together, these data strongly support that GT promotes immune response, more profoundly type 1 helper T cell (Th1) activity and GT may be applicable for treatment of intracellular parasite infection such as viral diseases.

성장 온도에 따른 InP/GaP SPS 구조의 광학적 특성

  • 초격자는 동종의 III-V 삼원 합금층을 성장하는 동안 스피노달 분해(spinodal decomposition)로 인한 원자배열(atomic ordering)과 상분리phase separation)에 의해서 발생하는데 MBE (molecular beam epitaxy)과 MOVPE (metalorganic vapor phase epitaxy)를 이용하여 성장시킬 때 주로 발생한다. 본 논문에서는 성장온도에 따른 InP/GaP SPS (short-period superlattices) 구조의 광학적 특성 변화를 시료의 온도와 여기광의 세기를 이용하여 분석하였다. 시료는 MBE 장비를 이용하여 성장하였으며, SPS층은 659쌍의 GaP(2.9 ${\AA}$)과 InP(3.1 ${\AA}$)로 이루어져 있고, GaP 층을 처음에 증착한 뒤, InP 층을 증착 하였다. 성장시 온도를 $400^{\circ}C$, $425^{\circ}C$, $460^{\circ}C$ 그리고 $490^{\circ}C$로 변화를 주어 성장하였다. 이들 시료를 GT400, GT425, GT460 그리고 GT490이라 하였고 이에 대한 광학적 특성을 PL (photoluminescence)를 이용하여 분석하였다. 10 K에서 PL 피크는 GT400 시료는 634 nm, GT425 시료는 636 nm, GT460 시료는 680nm, 그리고 GT490 시료는 692 nm에서 나타났으며, GT425 시료의 PL 세기가 가장 강하게 나타나고 GT400 시료의 PL 세기가 가장 약하게 나타났다. 그러나 260 K에서 PL 세기는 GT460 시료가 가장 강하게 나타나고 GT425 시료가 가장 약하게 나타났다. 성장온도가 증가함에 따라 밴드갭이 감소하는 것은 특정 성장온도($460^{\circ}C$) 이상에서 LCM (lateral composition modulation)이 형성되는 것으로 설명할 수 있다. GT400 시료와 GT425 시료의 PL 피크가 1.94 eV와 1.95 eV로 비슷하고, GT460 시료와 GT490 시료의 PL 피크가 1.82 eV과 1.79 eV로 비슷하게 나타난 것은 $460^{\circ}C$이상에서 성장한 시료에서 LCM 구조 형성으로 설명할 수 있다.

Comparison of Catalyzing Properties of Bacterial 4-α-Glucanotransferases Focusing on Their Cyclizing Activity

  • A newly cloned 4-α-glucanotransferase (αGT) from Deinococcus geothermalis and two typical bacterial αGTs from Thermus scotoductus and Escherichia coli (MalQ) were investigated. Among 4 types of catalysis, the cyclization activity of αGTs that produces cycloamylose (CA), a valuable carbohydrate making inclusion complexes, was intensively studied. The new αGT, DgαGT, showed close protein sequence to the αGT from T. scotoductus (TsαGT). MalQ was clearly separated from the other two αGTs in the phylogenetic and the conserved regions analyses. The reaction velocities of disproportionation, cyclization, coupling, and hydrolysis of three αGTs were determined. Intriguingly, MalQ exhibited more than 100-fold lower cyclization activity than the others. To lesser extent, the disproportionation activity of MalQ was relatively low. DgαGT and TsαGT showed similar kinetics results, but TsαGT had nearly 10-fold lower hydrolysis activity than DgαGT. Due to the very low cyclizing activity of MalQ, DgαGT and TsαGT were selected for further analyses. When amylose was treated with DgαGT or TsαGT, CA with a broad DP range was generated immediately. The DP distribution of CA had a bimodal shape (DP 7 and 27 as peaks) for the both enzymes, but larger DPs of CA quickly decreased in the DgαGT. Cyclomaltopentaose, a rare cyclic sugar, was produced at early reaction stage and accumulated as the reactions went on in the both enzymes, but the increase was more profound in the TsαGT. Taken together, we clearly demonstrated the catalytic differences between αGT groups from thermophilic and pathogenic bacteria that and showed that αGTs play different roles depending on their lifestyle.

고농도 감마 토코페롤 보충식이가 흡연에 노출된 쥐의 혈액 및 조직 비타민 E와 대사산물 농도에 미치는 영향 (Effects of Gamma-Tocopherol (GT) Supplementation on Vitamin E Concentration in Cigarette Smoke (CS) Exposed Mice)

  • 흡연은 활성 산소/질소종의 생성을 증가시켜 체내 산화적 스트레스를 증가시키고 폐의 염증을 유발한다. 이는 흡연자들의 체내 항산화 영양소들의 감소와 밀접한 관계를 가지는 것으로 알려져 있다. 본 연구에서는 항산화, 항염증 기능을 가진 비타민 E 중 식이에 많이 포함되어 있는 GT를 이용하여 흡연에 의한 항산화, 항염증 작용을 알아보기 위한 선행 연구로 고농도의 GT 식이가 혈액과 간, 폐의 AT, GT농도와 이들의 대샤산물인 CEHC 농도에 미치는 영향에 대하여 알아보고자 실시하였다. 연구 결과 고농도 GT 식이는 체내 혈액과 간, 폐 조직에 GT를 축적시키고 G-CEHC의 배설을 증가시켰다. 흡연은 대조군의 혈액과 폐 AT 농도는 증가시켰지만, 고농도 GT군의 혈액과 폐의 GT농도와 간의 G-CEHC의 농도를 감소 시켰다. 이러한 변화는 흡연에 의한 산화적 스트레스 상태에서 각기 다른 기능을 가진 조직의 요구량에 따라, 폐의 이용률을 높이기 위해 혈액으로 운반되는 양을 증가시키고 폐로의 운반을 임시적으로 증가시켜 조직의 AT, GT 농도를 선택적으로 조절하고 GT의 생체 이용률의 증가 때문이라 사료된다. 하지만 정확한 기전에 대한 연구들이 부족한 실정이므로 고농도의 GT 식이가 흡연에 의해 유도된 체내 산화적 스트레스와 폐의 염증 반응에 긍정적인 효과를 알아보기 위한 후속 연구가 절설히 필요한 것으로 사료된다.

Effect of FC-GT Supplement on Body Fat and Lipid Metabolism in Rats

  • The objective of this study was to investigate the effects of supplementation of an antiobese functional formula (FC-GT) on body weight and lipid metabolism in rats fed a high-fat diet. Three groups of male Sprague-Dawley rats were fed different diets for 6 weeks: normal control (NC), high-fat (HF), and high-fat supplemented with powdered antiobese functional formula (FC-GT) (5% wt/wt) groups. Although body weight was not significantly different among the groups, relative weights of epididymal and perirenal white adipose tissues were significantly lower in the FC-GT group than in the HF group. FC-GT supplementation significantly lowered the plasma total cholesterol and triglyceride concentrations, whereas it elevated the ratio of HDL-C/total-C and improved the atherogenic index. Hepatic cholesterol and triglyceride concentrations were significantly lowered in the FC-GT group compared to the HF group. The accumulation of hepatic lipid droplets and the epididymal white adipocyte size of the FC-GT group were diminished compared to the HF group. Hepatic HMG-CoA reductase activity was significantly lower in the FC-GT group than in the HF group. Plasma GPT activity was significantly lowered in the FC-GT group compared to the HF group. Additionally, fecal weight was significantly increased in the FC-GT group than in the HF group. In addition, contents of fecal triglyceride and cholesterol were significantly higher in the FC-GT group compared to the other groups. The antioxidant activities of hepatic SOD, CAT, and GR were significantly increased in the FC-GT group compared to the HF group. Hepatic mARS and plasma mARS levels were significantly lowered in the FC-GT group compared to the NC group. Accordingly, we conclude that supplementation of FC-GT improves plasma and hepatic lipid levels in high-fat fed rats.
