Fabrication and Characterization of Porous PLLA Scaffolds with Gentamicin Sulfate Release System (겐타마이신 설페이트를 서방화한 다공성 PLLA 지지체의 제조와 물성평가)

  • PLLA scaffold loaded with gentamicin sulfate (GS) was prepared by emulsion freeze-drying method for the prevention of infection and the improvement of wettability. i.e., the cell- and tissue-compatibility. GS-loaded PLLA scaffolds were characterized by scanning electron microscopy (SEM), mercury porosimetry and blue dye intrusion, and the GS release pattern was analyzed by high performance liquid chromatography (HPLC). GS-loaded PLLA scaffolds with porosity above 50%, medium pore size ranging from 30 to 57 ${\mu}{\textrm}{m}$ (with larger pore diameters greater than 150 ${\mu}{\textrm}{m}$), and specific pore area in the range of 35 to 75($m^2$ /g )were manufactured by varying processing parameter as GS concentration. It was observed that GS-loaded PLLA scaffolds were highly porous with good interconnections between pores for allowing cell adhesion and growth. These scaffolds may be applicable for scaffold as structures that facilitate either tissue regeneration or repair during reconstructive operations.

Authentication of Traded Traditional Medicine Ogapi Based on Nuclear Ribosomal DNA Internal Transcribed Spacers and Chloroplast DNA Sequences (nrDNA ITS 및 엽록체 DNA 염기서열 분석에 의한 유통 한약재 오가피 판별)

  • Background : Plants belonging to 5 species of the genus Eleutherococcus are currently distributed in the Korean peninsula. The traditional medicine 'Ogapi', derived from Eleutherococcus sessiliflorus and other related species, and 'Gasiogapi', derived from Eleutherococcus senticosus, are frequently mixed up and marketed. Therefore, accurated identification of their origins in urgently required. Methods and Results : Candidate genes from nuclear ribosomal DNA (nrDNA) and chloroplast DNA (cpDNA) of Eleutherococcus plants were analyzed. Whereas the nrDNA-internal transcribed spacer (ITS) regions were useful in elucidating the phylogenetic relationships among the plants, the cpDNA regions were not as effective. Therefore, a combined analysis with nrDNA-ITS was performed. Various combinations of nrDNA and matK were effective for discriminating among the plants. However, the matK and rpoC1 combination was ineffective for discriminating among some species. Based on these results, it was found that OG1, OG4, OG5, OG7, GS1, GS2, and GS3 were derived from E. sessiliflorus. In particular, it was confirmed that GS1, GS2, and GS3 were not derived from E. senticosus. However, more samples need to be analyzed because identification of the origins of OG2, OG3, OG6 and GS4 was not possible. Conclusion : The ITS2, ITS5a, and matK combination was the most effective in identifying the phylogenetic relationship among Eleutherococcus plants and traditional medicines based on Eleutherococcus.

Genetic regulation for the biosynthesis of glutamate family in Corynebacterium glutamicum (Corynebacterium glutamicum에서의 glutamate계 아미노산 생합성의 유전적 조절)

  • The regulation of three ammonia assimilatory enzymes, GDH (glutamate dehydrogenase), GS (glutamine synthetase) and GOGAT (glutamate synthase), has been examined in C. glutamicum. Three kinds of arginine auxotrophs blocked in each step of arginine biosynthetic pathway from glutamate were selected as arg 5, arg 6, arg 8. Histidine and tryptophan auxotrophs were also selected because histidine and tryptophan repressed GS biosynthesis in E. coli. These strains were cultured on the media containing nitrogen-excess and limited conditions, to compare the specific activities of ${\alpha}$-ketoglutarate dehydrogenase(${\alpha}-KGDH$), GDH, GS, GOGAT from the cell-free extracts. These results showed that enzyme levels of ${\alpha}-KGDH$ and GDH from 3 kinds of arginine auxotrophs, histidine and tryptophan auxotrophs in nitrogen-excess condition and those of GS and GOGAT in nitrogen limited condition were increased compared with opposite condition. The tryptophan and histidine auxotrophs showed higher level of glutamate and glutamine than parental strains and other mutants. it is assumed that the higher levels of ${\alpha-KGDH}$ and GDH from mutants in nitrogen-excess condition promoted the accumulation of glutamate and glutamine in fermentation broth. The inhibition of GS activities by ADP suggested that GS is regulated by energy charge in C. glutamicum. The results with histidine, tryptophan, glycine, alanine, serine and GMP implied that a system of feedback inhibition were effective. The GDH, GS and GOGAT biosynthesis in culture broth was markedly repressed by the nature and kinds of available nitrogen sources such as tryptophan, proline, glycine, alanine, serine and tyrosine.

A Study on the Dry Matter Yield and Nutritive Values of Wild Korean Lespedeza(Lespedeza Stipulacea Maxim.) (야생 코리언 레스페데자(Lespedeza stipulacea Maxim.)의 건물수량 및 사료가치에 관한 연구)

  • This study was conducted to evaluate the dry matter yield, quality and utilization of nutrients of wild Korean lespedeza as a forage legume sources. Dry matter(DM) yield of stages, chemical composition and DM digestibility were observed, also DM intake, digestibility and utilization of nitrogen and energy by Korean native goats were determined. Experimental diets include three treatrnents ; mixture grass hay(GS), GS 80% + lespedeza(LA) 20% and GS 60% + LA 40%, respectively. The DM yield of Korean lespedeza(LA) with advancing the growing stage tended to significantly increase(P<0.05). The average DM yield was observed 7,336kg/ha. CP content of LA tended to decrease, but fiber content tended to increase at growing stage advanced(P<0.05). The tannin content of LA was highest in bud stage (54.5mg/g), but tended to decrease at growing stage advanced, lowest in seed ripe stage(26.8mg/g) (P<0.05). With increasing the level of LA supplementation, DM intake by Korean native goats slightly increased, but no differences were observed in all diets. The digestibility of DM, cellular constituents and NDF were slightly lower for GS 100% diets than LA 20% and 40% supplemented diets(P<0.05). Apparently digested N% was not detected significant differences in all diets, but retained N and biological value were higher for LA supplemented diets than for GS 100% diets (P<0.05). Utilization of DE and ME was not detected significant differences in all diets. Above the results, it could be suggested that LA have a potential to provide as forage legume sources considering the DM yield, DM digestibility and nitrogen utilization.

Correlation between Low Gleason Score and Prostate Specific Antigen Levels with Incidence of Bone Metastases in Prostate Cancer Patients: When to Omit Bone Scans?

  • Background: To identify correlation and incidence of bone metastases in prostate cancer patient with low Gleason scores (GS) and prostate specific antigen (PSA) levels. Materials and Methods: This descriptive restrospective study covered patients with prostate cancer in Cipto Mangunkusumo Hospital in 2006-2011. Of a total of 478, those who had PSA values, histological examination, and bone scan were included, resulting in 358 eligible cases. PSA values were measured using the sandwich electrochemiluminescent immunoassay. Histological examination was graded according to Gleason's grading system and divided into 3 categories: well differentiated ($GS{\leq}6$), moderately differentiated (GS 7) and poorly differentiated (GS 8-10). Bone scans were performed using a radiopharmaceutical agent ($T_c$ 99m methylenen diphosphonate) with images captured by gamma camera. Results: The mean age was $67.5{\pm}7.8$, mean GS was $7.7{\pm}1.3$ and median PSA was 56.9 (range: 0.48-17000 ng/mL). There were 11 patients (3.0%) with positive bone scan with PSA<20 ng/mL and GS<8. Furthermore, there were 2 patients (0.6%) with $GS{\leq}6$ and PSA<10 ng/mL showing bone metastasis. Conclusions: In our study, there were still small percentage of patients with bone metastasis even when low values of PSA (PSA<10 ng/mL) and GS ($GS{\leq}6$) were applied.

Characteristics of Separation of Water/Bitumen Emulsion by Chemical Demulsifier (화학적 항유화제에 의한 물/비튜멘 에멀젼의 분리특성)

  • In this study, the separation of water/bitumen emulsion was investigated by chemical demulsification method. Motor oils (GS Caltex Deluxe Gold V 7.5W/30, Hyundai gear oil 85W/140) and asphalt (AP-5, KS M 2201, Dongnam Petrochemical MFG. Co.) were used as model oils in the preliminary experiments to effectively remove water from water/bitumen emulsion. The bitumen extracted from Canadian oilsands was used in this study. The water/oil emulsion was not separated without demulsifiers, and Hyundai motor oil showed higher efficiency of water separation at a low concentration of demulsifier compared with that for GS Caltex motor oil. However, as the concentration increased, the efficiency did not rapidly increase compared with that of GS Caltex motor oil. It was highly speculated that the water phase of Hyundai motor oil was not dispersed well compared with that of GS Caltex motor oil because the viscosity of Hyundai motor oil was much higher than that of GS Caltex motor oil. The demulsifier of higher HLB (hydrophilic - lipophilic balance) value had high separation efficiencies in water/oil emulsion. The TWEEN 20 (polyoxyethylene sorbitan monolaurate solution) showed better separation efficiency than other demulsifiers.

The VLAN implementation scheme on IBM-NP4GS3 based Packet Forwarding Engine (IBM NP4GS3 기반 패킷 포워딩 엔진에서의 VLAN 구현 방안)

  • 기존 라우터시스템에서의 패킷 포워딩 엔진은 최적화된 룩업 알고리즘을 통한 소프트웨어 방식으로 구현되었으나, 처리해야 할 데이터가 폭주하면서 전용화된 룩업칩을 이용한 포워딩 엔진으로 대체되어 가고 있다. 그러나, 이러한 룩업칩 기반 패킷 포워딩 엔진은 가격이 비싼 것은 물론. 급변하는 통신 프로토콜에 적응하기 힘든 단점을 가지게 되었으며, 이로 인해서 최근에는 Network Processor 기반의 패킷 포워딩 엔진에 대한 개발이 활발해지고 있다. 본 연구소에서도 초고속광가입자망 사업에 적용되는 포워딩 엔진을 IBM의 2.5G Network Processor 인 NP4GS3 칩을 이용하고 있으며, GbE/POS/E-PON 등의 인터페이스를 지원하고 있다. 또한 본 사업에서는 가입자망에서의 Layer2 broadcasting시의 트래픽 폭주와 가입자간의 보안 기능 등을 지원하기 위해 VLAN 서비스를 적용할 예정이다. 본 논문에서는 일반적인 VLAN 기능에 대해서 알아보고, IBM NP4GS3에서 지원되는 VLAN 기능을 분석하였으며, 이를 토대로 NP4GS3 기반 패킷 포워딩 엔진에서 VLAN 구현 방안에 대하여 기술하였다.

Effect of Ginseng Saponin on the Analgesic Effect and Tolerance Development of Clonidine

  • The antagonism against clonidine-induced analgesia by ginseng saponin (GS) and the inhibitory effect of GS on the development of clonidine-induced tolerance were evaluated in mice. GS, when administered systemically, intracerebrally and intrathecally, antagonized significantly the analgesic effect of clonidine. GS, when injected intraperitoneally not only inhibited the development of clonidine-induced analgesic tolerance, but also enhanced the analgesic effect of clonidine on the 2nd and 5th day. Naloxone did not antagonize the analgesic effect of clonidine and had no influence on the deveolpment of tolerance of both acute and delayed types. These results indicate that the antagonism against clonidine-induced analgesia and the inhibition of the deveolpment of clonidine-induced tolerance by GS are not mediated by the opioid mechanism.

Ginsenosides Promote Proliferation of Cultured Ovarian Germ Cells Involving Protein Kinase C-mediated System in Embryonic Chickens

  • The effect of ginsenosides (GS) on germ cell proliferation was evaluated with a chicken ovarian germ-somatic cell coculture model and the mechanism involving protein kinase C (PKC) pathway was investigated. Ovarian cells were cultured in serum-free McCoy's 5A medium and challenged with GS alone or in combinations with PKC activator (phorbol 12-myristate 13-acetate, PMA) or inhibitor ($H_7$) for 48 h. The number of germ cells was counted and the proliferating cells were identified by immunocytochemistry of proliferating cell nuclear antigen (PCNA). Results showed that GS significantly increased germ cell proliferation and this stimulating effect was further increased by PMA, but inhibited by H7, in a dose-dependent manner. Moreover, GS-elevated PCNA expression and the PCNA -labeling index of germ cells displayed similar changes with the increased numbers of germ cells. These results indicated that GS stimulated proliferation of ovarian germ cells with involvement of the PKC-mediated system.

Isolation and identification of Bacillus sp, GS Producing Xylanase (Xylanase생산성 Bacillus sp. GS의 분리 및 동정)

  • To utilize hemicellulosic biomass efficiently, the microorganism producing xylanase was isolated from fermented sawdust. It was a Gram positive, aerobic and rod shape bacterium. It had endospore and secreted strong hydrolases, such as amylase and protease. Through morphological, cultural and physiological tests, it was identified as Bacillus sp. GS. To increase the productivity of xylanase from Bacillus sp. GS, the enzyme production medium was optimized. The composition of the medium and incubation conditions were like follows xylan 1.25%, yeast extract 0.1%, NaN030.2%, K2HP04 0.1%, MgSO4 0.02%, mineral salt 0.005%, pH 6.5, incubation temperature 37$^{\circ}C$.
