Modified Drum Priming and Exogenous Application of 24-Epibrassinolide (24-EBL) for Enhancing Germination under High Temperature Condition in Lettuce Seeds

  • This study was conducted to investigate the effects of modified drum priming and 24-Epibrassinolide (24-EBL) treatment to improve the seed quality for export. 40, 50 and 60% seed moisture content (SMC) of hydrated seeds were incubated for 16 and 24 h in a container with a relative humidity of 99% at 26 rpm for a modified drum priming treatment. The treated seeds were sown at $20^{\circ}C$ and $30^{\circ}C$ (12/12h, light/dark) with four replications of 25 seeds on pleated paper. The seeds were hydrated with water or 24-EBL solutions of $10^{-7}$, $10^{-8}$ and $10^{-9}M$, respectively. The germination of the modified drum primed seeds (24 h incubation after 60% SMC hydration) improved to 1.6 days mean germination time (MGT) and $46%{\cdot}day^{-1}$ germination rate (GR), while the untreated seeds showed 2.1 days MGT and $28%{\cdot}day^{-1}$ GR. The modified drum priming (60% SMC and 24 h incubation with $10^{-9}M$ 24-EBL) showed improved results in MGT (1.8 days) and GR (55%) at $20^{\circ}C$, whereas untreated seeds showed 2.3 days MGT and 44% GR. Under $30^{\circ}C$, germination of modified drum primed seeds was significantly improved in GP (80%), GR ($31%{\cdot}day^{-1}$), HS (55%) and MGT (3.3 days), however, untreated seeds showed decreased GP (27%), GR ($22%{\cdot}day^{-1}$), HS (55%) and MGT (4.8 days). This study showed that the germination of lettuce seeds is enhanced by 24 h drum incubation with 24-EBL and this method can be used effectively to achieve the benefits of early germination and uniform seedling development. In addition, these treatments circumvent thermo-dormancy of lettuce seed and have a possibility of high-quality and environment-friendly seed processing.

A Study on the Airborne Pollen Calender in Mt. Kwanak (冠岳山에 있어서 空中에 飛散하는 花紛層에 關한 硏究)

  • The airborne pollens were investigated by Durhan's pollen trap in Mt. Kwanak from March 1, 1984 to February 28, 1985. The observed airborn pollens were identified 22 families and 49 genera, which were divided into 27 tree pollens. Most airborne pollens were found from late-March to late-September and a few from early-November to late-February, 1985. The first observed pollen of the year was Alnus on March 5 and the last was Artemisia on Decomber 19, 1984. The maximum amount of daily airborne pollens was 823 grains/cm2 on May 16. Peak day and the maximum amount of major airborne pollens were as follows; 166 grains/cm2/day of Alnus pollen on April 4, 38 gr. of Populus on April. 13, 22 gr. of Quercus on May 10, 791 gr. of Pinus on May 26, 139 gr. on May 26 and 104 gr. of Graminae on September 6, 24 gr. of Ambrosia on Sep. 10 and 236 gr. on May 26 and 104 gr. of Graminae on September 6, 24 gr. of Ambrosia airborne pollen for one year was 12,733 grains/cm2 which were composed of 61% tree pollens and 39% herb ones. Decreasing order of pollen counts was Pinus, Alnus, Quercus and Populus in trees, and Graiminae, Artemisia and Ambrosia in herbs. Most of the airborn pollens were deposited in the day-time and a few at night, the maximum at 2∼4p.m.and the mininum at 8∼10 p.m.

A Study on Immoblization of Pepsin (��프신의 고정화(固定化)에 관한 연구(硏究))

  • Several enzyme immobilization methods has been compared for immobilization of pepsin. Carboxymethyl cellulose and diethylaminoethyl cellulose were activated with Hcl and with NaOH, and were used for immobilization of pepsin. Sepharose-4B was activated cyanogen bromide, and was used for immobilization of pepsin. Porous glass beads were derivatized with 3-aminopropyitrlethoxysilane and with succinicanhydride, and were used for immobilization of pepsin. The results abtained were summarized as follow, 1. 10 mg/gr. dry bead and 15mg/gr. dry bead of pepsin were absorbed to CM-cellulose and DEAE-cellulose, 20 mg/gr. dry bead and 27 mg/gr. dry bead were coupled to CM-cellulose and DEAE-cellulose with glutaraldehyde respectively. Enzyme yields were 22% and 24% of soluble pepsin. 2. 16 mg/gr. dry bead of pepsin was attached to cyanogen bromide activated sepharose-4B, 19mg/gr. dry bead was cross linked to the activated bead with glutaraldehyde. Immobilized enzyme activity was 23% of soluble pepsin. 3. 40 mg/gr. dry bead of pepsin was conjugated to the derivatized glass beads. Immobilized enzyme activity was 45% of soluble pepsin.

Effects of Ginseng Radix on the Cell Cycle Regulation in Human Fetal Osteoblast (인삼이 사람태아골모세포의 세포주기조절에 미치는 영향)

  • Ginseng Radix(GR) had been used widely from oriental medicine and the effects of it have been investigated by many researchers. The purpose of present study was to investigate the effects of GR on the cell cycle progression and its molecular mechanism in human fetal osteoblast. The results were as follows. Increased cell proliferation was observed in cells exposed to 100 ng/ml, 10 ng/ml of GR-1 at 12 hours and 24 hours, 1 ${\mu}g$/ml of GR-1 at 48 hours, and 100 ${\mu}g$/ml, 10 ${\mu}g$/ml of GK-2 at 12 hours, all treatment groups of GR-2 at 24 hours(p<0.05). S phase and G1 phase was increased in the group of treated with 100 ng/ml of GR-1, with 10 ${\mu}g$/ml and 1 ${\mu}g$/ml of GR-2, with 100 ${\mu}g$/ ml and 10 ${\mu}g$/ml of GR-3 in the cell cycle analysis. The cell cycle regulation protein levels of Cyclin D1, Cyclin E, CDK 2. CDK 4 and CDK 6 were increased in the group of treated with 1 ${\mu}g$/ml and 100 ng/ml of GR-1, with 10 ${\mu}g$/ml and 1 ${\mu}g$/ml of GR-2, with 100 ${\mu}g$/ ml and 10 ${\mu}g$/ml of GR-3. On the other hand, p21 was decreased in the treatment group with 1 ${\mu}g$/ml and 100 ng/ml of GR-1, with 10 ${\mu}g$/ml and 1 ${\mu}g$/ml of GR-2, 10 ${\mu}g$/ml of GR-3, and p53 and p16 was decreased in the treatment group with 100 ng/ml of GR-1, 100 ${\mu}g$/ml and GR-3 and pRb was decreased in the all treatment groups except 1 ${\mu}g$/ml of GR-1. These results suggested that GR increases the cell proliferation and the cell cycle progression in human fetal osteoblast, which is linked to increased cell cycle regulation protein levels of Cyclin D1 , Cyclin E, CDK 2, CDK 4, CDK 6 and decreased cell cycle regulation protein levels of p21, pRb.

Study on swelling of starch granules using gravitational field-flow fractionation (GrFFF) (중력 장-흐름 분획법을 이용한 전분 입자의 swelling에 관한 연구)

  • Swelling of starch granules by water-sorption causes a progressive or sometimes abrupt change in sorption behavior as a result of structural alterations and the possible exposure of new sites with high affinity for water. It is thus of interest to examine the time-dependent change in the size or shape of the starch granules. Gravitational field-flow fractionation (GrFFF) utilizes the earth's gravity as the external field, and is useful for separation of micron-sized particles with larger particles eluting earlier than smaller ones. In this study, GrFFF was used to monitor the swelling of two starch granules, potato starch and sweet potato starch during contact time of 11-12 days at room temperature in water. Results from GrFFF were compared with those obtained from optical microscope (OM). For both starch granules, the mean sizes were increased with time spent in water.

Determination of Optimal Harvest Time of Chuchung Variety Green Rice® (Oryza sativa L.) with High Contents of GABA, γ-Oryzanol, and α-Tocopherol

  • In our previous study, an early-maturing variety of rice (Oryza sativa L.), Jinbu can have feature with unique green color, various phytochemicals as well as nutritive components by the optimal early harvesting, called Green Rice$^{(R)}$ (GR). The aims of the present field experiments were to evaluate the changes in the weight of 1,000 kernels, yield, and contents of proximate and bioactive compounds in Chuchung, a mid-late maturing variety, during the pre-harvest maturation of rough rice and to research the appropriate harvest time and potent bioactivity of Chuchung GR. The weights of 1,000 kernels of Chuchung GR dramatically increased until 27 days after heading (DAH). The yields of Chuchung GR declined after 27 DAH and significantly declined to 0.0% after 45 DAH. The caloric value and total mineral contents were higher in the GR than in the full ripe stage, the brown rice (BR). In the GR, the contents of bioactive compounds, such as ${\gamma}$-aminobutyric acid, ${\gamma}$-oryzanol, and ${\alpha}$-tocopherol, were much higher (P<0.05) than those in the BR, specifically during 24~27 DAH. Therefore, bioactive Chuchung GR can be produced with a reasonable yield at 24~27 DAH and it could be useful for applications in various nutritive and functional food products.

AN EXPERIMENTAL STUDY ON PENETRATION OF DYE IN FILLING MATERIALS (수종(數種) 수복물(修復物)의 색소침투(色素浸透)에 관(關)한 실험적(實驗的) 연구(硏究))

  • The purpose of this study was to measure penetration of dye stuff(5% Methylene blue, Hematoxylin, Crystal violet and Safranin-O) on silicate cement, Adaptic, Hi-pol and unfilled resin. Each filling material was mixed on the mixing pad and the mixed material was inserted with condensation force of 500gr, 1000gr and 2000gr and without condensation force into preformed glass tube (10mm in diameter and 10mm in height). The specimen was stored in the air for 24 hours, then specimen was immersed in various dye solution (5% methylene blue, hematoxin, crystal violet and safranin-O) for different period of time (l hour and 24 hours). These dye-treated specimen was cut horizontally at the middle portion and the dye penetration in cut surface was measured. Following results were obtained. 1. Pentration of various dye was excessive in silicate cement with and without Condensation force. 2. There has been no evidence of dye penetration in unfilled resin. 3. Dye penetration occurced with in 1 hour period and the extending time didn't affect the dye penetration.

Simultaneous Voltammetric Determination of Mefenamic Acid and Paracetamol using Graphene Nanosheets/Nickel Oxide Nanoparticles Modified Carbon Paste Electrode

  • A new modified carbon paste electrode (CPE) was constructed based on nickel oxide nanoparticles (NiONPs) and graphene nanosheets (Gr) for simultaneous determination of paracetamol (PCM) and mefenamic acid (MFA) in aqueous media and pharmaceutical dosage forms. NiONPs were synthesized via a simple and inexpensive technique and characterized using X-ray diffraction method. Scanning electron microscopy was used for the characterization of the morphology of modified carbon paste electrode (NiONPs/Gr/CPE). Voltammetric studies suggest that the NiONPs and Gr provide a synergistic augmentation that can increase current responses by improvement of electron transfers of these compounds on the NiONPs/Gr/CPE surface. Using cyclic voltammetry, the NiONPs/Gr/CPE showed good sensitivity and selectivity for the determination of PCM and MFA in individually or mixture standard samples in the linear range of $0.1-30{\mu}g\;mL^{-1}$. The resulted limit of detection and limit of quantification were 20 and $60ng\;mL^{-1}$ for PCM, 24 and $72ng\;mL^{-1}$ for MFA, respectively. The analytical performance of the NiONPs/Gr/CPE was evaluated for the determination of PCM and MFA in pharmaceutical dosage forms with satisfactory results.

Neural Mechanisms Underlying Antidepressant-Like Effects of Glycyrrhizae Radix in Rats

  • Glycyrrhizae radix (GR) is an herbal medicine commonly used in East Asia for treating a variety of diseases, including stomach disorders. In this study, the antidepressant-like activity of GR was investigated using the forced swimming test (FST) in rats. After the FST, the expression of c-Fos and corticotrophin releasing factor (CRF) was assessed by immunochemistry of brain samples from the paraventricular nucleus of the hypothalamus (PVN). The results of the FST showed that a high dose (400 mg/kg) of extract was very effective in reducing immobility(P<0.01), and increased climbing. In addition, treatment with GR (400 mg/kg) significantly decreased the expression of c-Fos and CRF in the PVN, compared to controls. In conclusion, the findings of this study demonstrated that GR effectively reduced behavioral and physiological depression responses in an animal model of depression, suggesting that GR might be useful in the treatment of clinical depression.
