Studies on Screening of Paraquat Toxicity Reducing Agent and its Inhibition Mechanism (Paraquat 독성 경감제 검색 및 그 억제 기전에 관한 연구)

  • In this study, we intended to evaluate the modulatory effects of natural products, ${\beta}-carotene$, aloesin and semiessential amino acid, taurine on the toxicitiy of paraquat. In the taurine treated groups, serem glutamic oxaloacetic transaminase (s-GOT), serem glutamic pyruvic transaminase (s-GPT). blood urea nitrogen (BUN), creatinine, malondialdehyde (MDA), alkaline phosphatase (ALP) activity in serum and MDA, ALP activity, collagen in lung tissue were decreased to the normal values. In the aloesin treated groups, s-GPT, BUN, creatinine, MDA level in serum were decreased to the normal values significantly. In the ${\beta}-carotene$ treated group, only s-GPTactivity was reduced to the normal values. In the lung tissue of taurine treated groups, MDA value, G-6-phosphatase activity and collagen synthesis were recovered to the normal valuse and ALP activity was increase about 40%. From these results, we concluded that taurine is an effective agent to inhibit the pulmonary and internal organs toxicities induced by paraquat and the inhibition effects of taurine are due to remove free radicals directly.

Clinical Observation on Effects and Adverse Effects of Choweseuncheng-tang on Obesity Patients (비만 환자에 대한 조위승청탕의 효과 및 부작용에 관한 임상적 고찰)

  • Objective : This study was designed to investigate the effects and adverse effects of Choweseuncheng-tang on obesity patients Methods : Choweseuncheng-tang was given to 20 obesity patients who have no abnormality of GOT, GPT and past medical history. Additionally, electroacupuncture was performed on Fuchao(腑巢) with 60Hz. We compared body weight, body fat and laboratoy test(GOT, GPT, BUN, Creatinine) between before and after treatment. Blood pressure, heart rate and clinical symptoms were checked to investigate the side effects. Decrease of body weight was checked in accordance with Sasang constitutional type, appetite type, and BMI. Results : 6 patients(BMI 23-24.9), 10 patiets(BMI 25-29.9), and 4 patients(BMI over 30) were showed decrease of body weight. We found body fat was decerased $3.16{\pm}1.25%$. BUN, Creatinine, GOT and GPT were increased within normal range. There were adverse event of autonomic hyperactivity(11 patients), gastrointestinal symptoms(7 patients) and others(3 patients). Conclusions : According to above results, Choweseuncheng-tang could be used effectively, but should be taken cautiously on obese patients.

Effects of Time Elapse of Serum Separation on the Examination of Bovine Blood Chemical Values (혈청분해시간의 경과가 소 혈액화학치 검사에 미치는 영향)

  • To obtain the basic data for blood chemistry, the effects of the time elapse of serum separation on bovine blood chemical values were investigated. The results obtained are summerized as follows : 1. The constant fluctuation tendency and significance was not detected in GOT, GPT, BUN, Creatinine, ALP, CPX, Cholesterol, Ca, Mg and Pi. 2. Glucose showed the decrease tendency according to the time elapse of serum separation. As the values at 12 and 24 hours showed significant lower values than those at 1 and 2 hours (p<0.01) it was thougth that separation time of serum should be focussed for the glucose determination.

Inhibitory Effect of Schizandrin on Toxicity of Paraquat (Paraquat 독성에 대한 Schizandrin의 억제효과)

  • Paraquat is a useful nonselective herbicide widely used throughout the world. However, accidental or intentional ingestion of the paraquat cause fetal pulmonary injuring. But there is not suitable antidote of paraquat intoxication and therapeutic agents now be used are not effective. So, in this study we intended to evaluate the inhibitory effects of DDB(dimethyl-4,4'dimethoxy-5,6,5',6'-dimethylene dioxyphenyl-2,2'-dicarboxylate) on paraquat toxicity. DDB (100mg/kg) was administered orally to SD rats lhr after paraquat(50mg/kg) injection. After 24 hours, the biochemical parameters of blood and tissues were examined. In paraquat treated groups sGPT, sGOT, BUN, creatinine, MDA and alkaline phosphatase levels in blood and MDA, glucose-6-phosphatase activity in tissues were elevated by 2 to 5 times of normal values. However in schizandrin treated groups, sGPT, sGOT, MDA and alkaline phosphatase activity in blood and MDA and glucose-6-phosphatase activity were significantly decreased to notmal levels but not in biochemical parameters of nephrotoxicity, BUN and creatinine levels. Therefore, we concluded that schizandrin can be used as an antidote of pulmono, hepatotoxicity of paraquat.

  • In order to inspect safety and function regarding the reduction of cadmium poisoning by polyphenol compounds prepared from persimmon leaves (Diospyros kaki folium), animal test was done. Rats, group P-1 and P-2, were treated with cadmium and polyphenols of persimmon leaves. A control was just treated with cadmium. GOT activities in P-1 and in P-2 were lower than that of the control without considerable difference, whereas GPT activities decreased in P-1 and P-2. The SOD activity of liver tissue increased in P-l and P-2 compared with that of the control. GST activity was higher than that of the control without considerable difference in P-1; however, it increased in P-2. Lipid peroxide value considerably increased in both P-1 and P-2.

  • In order to investigate the inter-relation with the growth of the rice-root and its transaminase-activity, by measuring the growth of its root and transaminase-activity supplying this root with various nitrogen Compounds($NO_3$-N, $NH_4$-N and Amino acid.). The obtained results are summarized as follows; 1. Growth of rice-root supplied with $NO_3$-N is generally increased in length and weight, compared with that of the root fertilized by $NH_4$-N. 2. The above-metnioned root with $NH_4$-N is not only decreased in its weight and length but also is apt to inhibited its growth as the nitrogen concentruration is increased, in compared with the root provided with $NO_3$-N. 3. The activity of G.O.T. and G.P.T. for the root fertilized by $NH_4$-N, the badly grown root is generally increased, while of the root supplied with $NO_3$-N is decreased. 4. The activity of G.O.T. and G.P.T. for the root provided with amino acid known as the considerable growth inhibiting compound is generally decreased, while that of the badly-grown root is increased. 5. The activity of G.O.T. and G.P.T. in the supernatant fraction of the rice-root is for the most part, high and low in the mitochondrial fraction.

A Basic Study on User Experience Evaluation Based on User Experience Hierarchy Using ChatGPT 4.0 (챗지피티 4.0을 활용한 사용자 경험 계층 기반 사용자 경험 평가에 관한 기초적 연구)

  • With the rapid advancement of generative artificial intelligence technology, there is growing interest in how to utilize it in practical applications. Additionally, the importance of prompt engineering to generate results that meet user demands is being newly highlighted. Exploring the new possibilities of generative AI can hold significant value. This study aims to utilize ChatGPT 4.0, a leading generative AI, to propose an effective method for evaluating user experience through the analysis of online customer review data. The user experience evaluation method was based on the six-layer elements of user experience: 'functionality', 'reliability', 'usability', 'convenience', 'emotion', and 'significance'. For this study, a literature review was conducted to enhance the understanding of prompt engineering and to grasp the clear concept of the user experience hierarchy. Based on this, prompts were crafted, and experiments for the user experience evaluation method were carried out using the analysis of collected online customer review data. In this study, we reveal that when provided with accurate definitions and descriptions of the classification processes for user experience factors, ChatGPT demonstrated excellent performance in evaluating user experience. However, it was also found that due to time constraints, there were limitations in analyzing large volumes of data. By introducing and proposing a method to utilize ChatGPT 4.0 for user experience evaluation, we expect to contribute to the advancement of the UX field.

Effect of Jujube Methanol Extract on the Hepatotoxicity in $CCl_4$-Treated Rats (대추 메탄을 추출물이 사염화탄소투여에 의한 흰쥐의 간 세포독성에 미치는 영향)

  • To investigate effects of Jujube methanol extract on the carbon tetrachloride-induced liver damage in rats, experimental animals were divided into 4 groups; control group(CON), Jujube methanol extracttreated group(JME), $CCl_4$- treated groups(CCl), and Jujube methanol extract and $CCl_4$-treated group (JMC). Each group was sacrificed after 2 or 4week feeding and determined the activities of serum transaminase(GOT, GPT) and hepatic xanthine oxidase, superoxide dismutase(SOD), catalase and glutathione peroxidase(GSH-Px), and hepatic contents of thiobarbituric acid-reactants(TBARS) and glutathione in liver. The activities of sGOT and sGPT, and the hepatic content of TBARS after $CCl_4$-treatment were markedly increased, compared to CON, but those levels were significantly decreased by the pretreatment of Jujube methanol extract, especially in sGOT after 2 and 4 week and TBARS after 4week respectively. Xanthine oxidase activity was increased by $CCl_4$- treatment as compared to CON, but it was also inhibited by the pretreatment of Jujube methanol extract for 2 and 4 week. The activities of SOD, catalase and GSH-Px were elevated by $CCl_4$-treatment, compared to CON, but those elevated activities were showed significant decreasing effect by pretreatment of Jujube methanol extract after 2 and 4week as compared to CON, however, hepatic catalase activity was not affected significantly. These results suggest that Jujube methanol extract is believed to be a possible protective effect for the carbon tetrachloride-induced hepatotoxicity in rats.

Effects of Sachunghwan Extract on Liver Damage in Thioacetamide Induced Rats (사청환(瀉靑丸)이 thiaocetamide에 의한 백서(白鼠) 간손상(肝損傷)에 미치는 영향)

  • Objectives The purpose of this study is to examine of the effect of Sachunghwan on the liver disease. Methods It was induced liver damage rats by thioacetamide and dosed the extract orally and measured the activity rate of GOT, GPT, ALP and LDH. Results 1. GOT activity was more decreased in the experimental goup than in the control goup. Group I showed continuous significance after 72 hour, Group II was significance after 96 hours. 2. GPT activity was more decreased in the experimental group than in the control group. Group I was effective after 72 hours, but group II was effective after 96 hours. 3. The significance of ALP activity in the Group I and group II was revealed after 72 hours. 4. The significance of LDH activity in the Group I and group II was revealed after 96 hours. Conclusions It was showded that Sachunghwan extract was effective on liver disease.

  • The effects of foliar spray of Kinetin (6-Furfurylaminopurine) on the growth of mulberry tree and the metabolic activities in the leaves were measured. The results of this experimented are summarized as follows: 1. Foliar application of Kinetin was effective in increasing of leaf area, the fresh and dry weight of the leaves, the length and diameter of shoots. 2. Chlorophyll content was generally increased in all treated leaves. 3. All the treated leaves were higher in GOT activity than the control leaves but were lower in GPT activities. 4. The activities of the carbohydrate metabolism in the mulberry leaves were higher than that of the control in all treated leaves.
