In this study, the xylitol dehydrogenase (XYL2) gene was expressed in Saccharomyces cerevisiae as a host cell for ease of use in the degradation of lignocellulosic biomass (xylose). To select suitable expression systems for the S.XYL2 gene from S. cerevisiae and the P.XYL2 gene from Pichia stipitis, $pGMF{\alpha}-S.XYL2$, $pGMF{\alpha}-P.XYL2$, $pAMF{\alpha}-S.XYL2$ and $pAMF{\alpha}-P.XYL2$ plasmids with the GAL10 promoter and ADH1 promoter, respectively, were constructed. The mating factor ${\alpha}$ ($MF{\alpha}$) signal sequence was also connected to each promoter to allow secretion. Each plasmid was transformed into S. cerevisiae $SEY2102{\Delta}trp1$ strain and the xylitol dehydrogenase activity was investigated. The GAL10 promoter proved more suitable than the ADH1 promoter for expression of the XYL2 gene, and the xylitol dehydrogenase activity from P. stipitis was twice that from S. cerevisiae. The xylitol dehydrogenase showed $NAD^+$-dependent activity and about 77% of the recombinant xylitol dehydrogenase was secreted into the periplasmic space of the $SEY2102{\Delta}trp1/pGMF{\alpha}-P.XYL2$ strain. The xylitol dehydrogenase activity was increased by up to 41% when a glucose/xylose mixture was supplied as a carbon source, rather than glucose alone. The expression system and culture conditions optimized in this study resulted in large amounts of xylitol dehydrogenase using S. cerevisiae as the host strain, indicating the potential of this expression system for use in bioethanol production and industrial applications.

The physiological activities of cultivated Korean blue mussel (Mytilus edulis) and New Zealand green-lipped mussel (Perna canaliculus) were analyzed and compared. Both hot water extracts of blue mussel flesh (BMF) and green-lipped mussel flesh (GMF) showed increased activities of alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). BMF showed increased ADH and slightly decreased ALDH activities compared to GMF. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of BMF was higher than that of GMF at the same concentration. BMF and GMF showed similar inhibitory activity against angiotensin converting enzyme at a concentration of 30 mg/mL. These results suggest that cultivated Korean blue mussel has similar physiological activity with New Zealand green-lipped mussel.
The galactic magnetic field (GMF) and the intergalactic magnetic field (IGMF) affect the propagation of ultra-high energy cosmic rays (UHECRs) from the source to us. Here we examine the influences of the GMF/IGFM and the dependence of their sky distribution on galactic latitude, b. We analyze the correlation between the arrival direction (AD) of UHECRs observed by the Pierre Auger Observatory and the large-scale structure of the universe in regions of sky divided by b. Specifically, we compare the AD distribution of observed UHECRs to that of mock UHECRs generated from a source model constructed with active galactic nuclei. Our source model has the smearing angle as a free parameter that reflects the deflection angle of UHECRs from the source. The results show that larger smearing angles are required for the observed distribution of UHECRs in lower galactic latitude regions. We obtain, for instance, a $1{\sigma}$ credible interval for smearing angle of $0^{\circ}{\leq}{\theta}_s{\leq}72^{\circ}$ at high galactic latitudes, $60^{\circ}$ < $\left|{b}\right|{\leq}90^{\circ}$, and of $75^{\circ}{\leq}{\theta}_s{\leq}180^{\circ}$, $-30^{\circ}{\leq}b{\leq}30^{\circ}$, at low galactic latitudes, respectively. The results show that the influence of the GMF is stronger than that of the IGMF. In addition, we can estimate the strength of GMFs by these values; if we assume that UHECRs would have heavier nuclei, the estimated strengths of GMF are consistent with the observational value of a few ${\mu}G$. More data from the future experiments may make UHECR astronomy possible.
Xylitol is widely used in the food and medical industry. It is produced by the reduction of xylose (lignocellulosic biomass) in the Saccharomyces cerevisiae strain, which is considered genetically safe. In this study, the expression system of the GRE3 (YHR104W) gene that encodes xylose reductase was constructed to efficiently produce xylitol in the S. cerevisiae strain, and the secretory production of xylose reductase was investigated. To select a suitable promoter for the expression of the GRE3 gene, pGMF-GRE3 and pAMF-GRE3 plasmid with GAL10 promoter and ADH1 promoter, respectively, were constructed. The mating factor ${\alpha}$ ($MF{\alpha}$) signal sequence was also connected to each promoter for secretory production. Each plasmid was transformed into S. cerevisiae $SEY2102{\Delta}trp1$, and $SEY2102{\Delta}trp1$/pGMF- GRE3 and $SEY2102{\Delta}trp1$/pAMF-GRE3 transformants were selected. In the $SEY2102{\Delta}trp1$/pGMF-GRE3 strain, the total activity of xylose reductase reached 0.34 unit/mg-protein when NADPH was used as a cofactor; this activity was 1.5 fold higher than that in $SEY2102{\Delta}trp1$/pAMF-GRE3 with ADH1 as the promoter. The secretion efficiency was 91% in both strains, indicating that most of the recombinant xylose reductase was efficiently secreted in the extracellular fraction. In a baffled flask culture of the $SEY2102{\Delta}trp1$/pGMF-GRE3 strain, 12.1 g/l of xylitol was produced from 20 g/l of xylose, and ~83% of the consumed xylose was reduced to xylitol.
Business process modeling is to represent the business activities in a graphical notation that enterprises use to achieve their customer's requirements. Nowadays, lots of requirements are changed quickly and variously. Therefore, business process models should provide the means which can prepare for changes by analyzing (pointing) elements of business process that are likely to alter. In this paper, we propose a business process family model (BPFM) which represents the commonalities and the variabilities of a set of business processes. In addition, we propose the process which develops the tools for BPFM based on Eclipse Plug-In Graphical Modeling Framework (GMF). The variabilities of Business Process are modeled by using expanded model elements of UML2.0 activity diagram.
Sea surface wind is a fundamental element for understanding the oceanic phenomena and for analyzing changes of the Earth environment caused by global warming. Global research institutes have developed and operated scatterometers to accurately and continuously observe the sea surface wind, with the accuracy of approximately ${\pm}20^{\circ}$ for wind direction and ${\pm}2m\;s^{-1}$ for wind speed. Given that the spatial resolution of the scatterometer is 12.5-25.0 km, the applicability of the data to the coastal area is limited due to complicated coastal lines and many islands around the Korean Peninsula. In contrast, Synthetic Aperture Radar (SAR), one of microwave sensors, is an all-weather instrument, which enables us to retrieve sea surface wind with high resolution (<1 km) and compensate the sparse resolution of the scatterometer. In this study, we investigated the Geophysical Model Functions (GMF), which are the algorithms for retrieval of sea surface wind speed from the SAR data depending on each band such as C-, L-, or X-band radar. We reviewed in the simulation of the backscattering coefficients for relative wind direction, incidence angle, and wind speed by applying LMOD, CMOD, and XMOD model functions, and analyzed the characteristics of each GMF. We investigated previous studies about the validation of wind speed from the SAR data using these GMFs. The accuracy of sea surface wind from SAR data changed with respect to observation mode, GMF type, reference data for validation, preprocessing method, and the method for calculation of relative wind direction. It is expected that this study contributes to the potential users of SAR images who retrieve wind speeds from SAR data at the coastal region around the Korean Peninsula.
The synergism of active and passive microwave techniques for hurricane ocean wind remote sensing is explored. We performed the analysis of Windsat data for Atlantic hurricanes in 2003-2005. The polarimetric third Stokes parameter observations from the Windsat 10, 18 and 37 GHz channels were collocated with the ocean surface winds from the Holland wind model, the NOAA HWind wind vectors and the Global Data Assimilation System (GDAS) operated by the National Center for Environmental Prediction (NCEP). The collocated data were binned as a function of wind speed and wind direction, and were expanded by sinusoidal series of the relative azimuth angles between wind and observation directions. The coefficients of the sinusoidal series, corrected for atmospheric attenuation, have been used to develop an empirical geophysical model function (GMF). The Windsat GMF for extreme high wind compares very well with the aircraft radiometer and radar measurements.

The purpose of this study is to perform parameter selection and develop a method for the condition monitoring of an LCD conveyance robot. To determine the vibration characteristics of the driving part of the robot, the gear mesh frequency (GMF) of the speed-reducing gearbox is calculated and confirmed by frequency analysis. In order to ensure reproducibility of the measured data, an appropriate working pattern is selected and experiments are carried out. For condition monitoring of the robot, a wireless measurement system is constructed and used in parameter selection, with the GMF as the center. A method involving the use of the standard deviation of the measured data and another method involving the use of multiple value of amplitude are proposed.

Sea surface winds in the sea off the east coast of Korea were derived from L-band ALOS (Advanced Land Observing Satellite) PALSAR (Phased Array type L-band Synthetic Aperture Radar) data and their characteristics of errors were analyzed. We could retrieve high-resolution wind vectors off the east coast of Korea including the coastal region, which has been substantially unavailable from satellite scatterometers. Retrieved SAR-wind speeds showed a good agreement with in-situ buoy measurement by showing relatively small an root-mean-square (RMS) error of 0.67 m/s. Comparisons of the wind vectors from SAR and scatterometer presented RMS errors of 2.16 m/s and $19.24^{\circ}$, 3.62 m/s and $28.02^{\circ}$ for L-band GMF (Geophysical Model Function) algorithm 2009 and 2007, respectively, which tended to be somewhat higher than the expected limit of satellite scatterometer winds errors. L-band SAR-derived wind field exhibited the characteristic dependence on wind direction and incidence angle. The previous version (L-band GMF 2007) revealed large errors at small incidence angles of less than $21^{\circ}$. By contrast, the L-band GMF 2009, which improved the effect of incidence angle on the model function by considering a quadratic function instead of a linear relationship, greatly enhanced the quality of wind speed from 6.80 m/s to 1.14 m/s at small incident angles. This study addressed that the causes of wind retrieval errors should be intensively studied for diverse applications of L-band SAR-derived winds, especially in terms of the effects of wind direction and incidence angle, and other potential error sources.
