A unicellular algae, Chlorella vulgaris(CV), was used as a biological response modifier. The effect of CV on forced swimming test and blood biochemical parameters related to fatigue was investigated. Blood urea nitrogen(BUN); creatine kinase(CK); lactic dehydrogenase(LDH); glucose(Glc); total protein(TP); and albumin were determined. CV was orally administered to mice in the range of 0.05 to 0.15 g/kg/day. A forced swimming test results on 3 and 7 day after administration of CV, showed that immobility time was decreased in the CV-administered group(0.15 g/kg). In addition, the contents of BUN in the blood serum were decreased in CV-fed group. The contents of CK and LDH were tended to decrease, but not statistically significant. The plasma Glc level was increased in CV-fed groups(0.05 and 0.1 g/kg) compared to control group. It had no effect on the elevation of TP and albumin level. The results indicate that CV could improve physical stamina.
T-2 toxin is one of mycotoxins produced by fungi such as Fusarium spp. and possesses a potent cytotoxicity to eukaryotic cell. The contamination of mycotoxins in cereals and feedstuffs is one of the great concerns in health authorities. Therefore, the development of the specific, sensitive and simplified analysis method for T -2 toxin is required. During more than ten years, several chemical and biological analysis methods were proposed and applied for the detection and quantification of T-2 toxin. TLC, GLC-FID and GC-MS are widely employed, but these methods required numerous clean-up procedures before analysis, and the detection limit for T-2 toxin is more than 10 ppb. Biological analysis methods with dermal tissues and cultured cells are not specific to T-2 toxin, since T-2 toxin and other related derivatives possess a similar toxicological activity although their relative activity is different each otber. Based on tbe specific reaction between antibody and antigen, the authors tried to introduce the immunochemical methods for determination of T-2 toxin. The enzyme-linked immunosorbent assay method using monoclonal antibody for T-2 toxin was applied to analyse T-2 toxin. The detection limit of T-2 toxin by ELISA method was 0.1 ppb. The correlation between ELISA and GC-MS method on these samples was very high. ELISA method developed for the detection and quantification of T -2 toxin in this paper possesses simplicity, high sensitivity and specific for T-2 toxin. Furthermore, the ELISA method with T-2 toxin monoclonal antibody was an excellent tool for the screening of Fusarium spp. which was suspected to produce T-2 toxin.

The present study was conducted to elucidate the triglyceride composition of walnut oil. The triglycerides were separated by thin layer chromatography (TLC) and fractionated on the basis of partition numbers by reverse phase high performance liquid chromatography (HPLC) on a column packed with $\mu$-bondapak $C_{18}$ using methanol-chloroform mixture as a solvent system. Each of these collected fractions was fractionated again on the basis of acyl carbon number of triglyceride by gas liquid chromatography (GLC). The fatty acid composition of triglycerides for each partition numbered group was also analyzed by GLC. From the results, it was found that walnut oil consists of ten kinds of triglycerides, and the patterns of major ones in walnut oil were as follows: 53.3% of $(C_{18:2},\;C_{18:2},\;C_{18:2})$, 10.1% of $(C_{18:1},\;C_{18:2},\;C_{18:2})$, 5.4% of $(C_{18:1},\;C_{18:1},\;C_{18:2})$, 4.3% of $(C_{18:1},\;C_{18:2},\;C_{18:3})$, 3.9% of $(C_{18:0},\;C_{18:2},\;C_{18:2})$, 2.0% of $(C_{18:0},\;C_{18:1},\;C_{18:2})$, 1.8% of $(C_{18:0},\;C_{18:2},\;C_{18:2})$.
An analytical method was developed to determine residues of haloxyfop-R and its methyl ester in soils and soybeans using gas-liquid chromatography (GLC) with electron capture detector (ECD). Soil or soybean sample was acidified and extracted with acetone. The extract was then subjected to ion-associated partition to individually separate haloxyfop-R and the neutral methyl ester. One phase containing haloxyfop-R was methylated with $BF_3$/methanol, partitioned to n-hexane and analyzed by GLC/ECD. The other phase containing the methyl ester was further purified by Florisil column chromatography prior to GLC determination. No cross contamination was found between two phases containing each of the acid and methyl ester, thus two compounds can be separately determined as the identical haloxyfop-R-methyl. Overall recoveries of haloxyfop-R from fortified samples averaged 88.2${\pm}$3.9% (n=12) and 88.3${\pm}$4.0% (n=6) for soils and soybeans respectively, and those of haloxyfop-R-methyl showed mean values of 89.2${\pm}$4.0% (n=12) and 85.6${\pm}$5.6% (n=6). Detection limits of both haloxyfop-R and its methyl esterwere 0.005㎎/㎏ and 0.01㎎/㎏ for soil and soybean samples respectively.
To get further information on the behavior of phorate(0,0-diethyl S-ethylthiomethyl phosphorodithioate) in soil under the subtropical conditions, a field experiment has been conducted. Phorate granule (10%) was applied to silt loam soil at the rate of 40kg a.i./ha and incorporated to 10cm soil depth. Residues of phorate and its metabolites in soil were determined with GLC and confirmed qualitatively with TLC. Phorate was rapidly oxidized to its sulfoxide and sulfone. Therefore, main metabolic pathway of phorate in soil was the oxidation of phorate to phorate sulfoxide and sulfone. Phorate sulfoxide and sulfone were relatively more persistent than phorate itself. Phoratoxon was detected at low level only up to 30 days after treatment and its sulfoxide and sulfone were not detected during the whole experimental period. Toluene-acetonitrile-nitromethane(40 : 30 : 30, v/v/v) solvent system separated satisfactorily phorate and its five metabolites. Most of the residues was found in the initial incorporation depth $(0{\sim}10cm)$. Consequently, insecticides showed a little downward movement.
Chitin, a $\beta$-1,4 polymer of N-acetyl-D-glucosamine, is one of the most abundant organic compounds in nature. Chitinase (EC 3.2.1.14) is an enzyme that degrades chitin to chito-oligosaccharides, diacetyl rhitobiose and N-acetyl-D-glucosamine. An extracellular chitinase-producing bacterial strain was isolated from soil and named to as Bacillus subtilis JK-56. Optimum culture condition of B. subtilis JK-56 for the production of chitinase was 1% chitin, 0.5% polypepton, 0.1% KCl, 0.05% MnS $O_4$.4$H_2O$, 37$^{\circ}C$, initial pH 7.0 and 40 hour culture time. When B. subtilis JK-56 was grown in the optimum medium, one major active band and two minor active bands were detected by native-PAGE and active staining of the gel. Among them, the major band was purified from the culture supernatant by 70% ammonium sulfate precipitation and native-PAGE with BIO-RAD Model 491 Prep-Cell and named as Chi-56A. Its molecular weight was estimated to be 53kDa monomer and the isoelectric point (pI) was pH 4.3. The pH and temperature for the optimum activity of Chi-56A were pH 6.0 and $65^{\circ}C$, respectively. Chi-56A was stable up to $65^{\circ}C$ and in alkaline region. Its $K_{m}$ value for colloidal chitin was 17.33g/L. HPLC analysis of the reaction products confirmed that Chi-56A was an exo type chitinase.e.
