Non Destructive Fast Determination of Fatty Acid Composition by Near Infrared Reflectance Spectroscopy in Sesame

  • To investigate seed non destructive and fast determination technique utilizing near infrared reflectance spectroscopy (NIRs) for screening ultra high oleic (C18:1) and linoleic (C18:2) fatty acid content sesame varieties among genetic resources and lines of pedigree generations of cross and mutation breeding were carried out in National Institute of Crop Science (NICS). 150 among 378 landraces and introduced cultivars were released to analyse fatty acids by NIRs and gas chromatography (GC). Average content of each fatty acid was 9.64% in palmitic acid (C16:0), 4.73% in stearic acid (C18:0), 42.26% in oleic acid and 43.38% in linoleic acid by GC. The content range of each fatty acid was from 7.29 to 12.27% in palmitic, 6.49% from 2.39 to 8.88% in stearic, 12.59% of wider range compared to that of stearic and palmitic from 37.36 to 49.95% in oleic and of the widest from 30.60 to 47.40% in linoleic acid. Spectrums analyzed by NIRs were distributed from 400 to 2,500 nm wavelengths and varietal distribution of fatty acids were appeared as regular distribution. Varietal differences of oleic acid content good for food processing and human health by NIRs was 14.08% of which 1.49% wider range than that of GC from 38.31 to 52.39%. Varietal differences of linoleic acid content by NIRs was 16.41% of which 0.39% narrower range than that of GC from 30.60 to 47.01%. Varietal differences of oleic and linoleic acid content in NIRs analysis were appeared relatively similar inclination compared with those of GC. Partial least square regression (PLSR) among multiple variant regression (MVR) in NIRs calibration statistics was carried out in spectrum characteristics on the wavelength from 700 to 2,500 nm with oleic and linoleic acids. Correlation coefficient of root square (RSQ) in oleic acid content was 0.724 of which 72.4 percent of sample varieties among all distributed in the range of 0.570 percent of standard error when calibrated (SEC) which were considerably acceptable in statistic confidence significantly for analysis between NIRs and GC. Standard error of cross validation (SECV) of oleic acid was 0.725 of which distributed in the range of 0.725 percent standard error among the samples of mother population between analyzed value by NIRs analysis and analyzed value by GC. RSQ of linoleic acid content was 0.735 of which 73.5 percent of sample varieties among all distributed in the range of 0.643 percent of SEC. SECV of linoleic acid was 0.711 of which distributed in the range of 0.711 percent standard error among the samples of mother population between NIRs analysis and GC analysis. Consequently, adoption NIR analysis for fatty acids of oleic and linoleic instead that of GC was recognized statistically significant between NIRs and GC analysis through not only majority of samples distributed in the range of negligible SEC but also SECV. For enlarging and increasing statistic significance of NIRs analysis, wider range of fatty acids contented sesame germplasm should be kept on releasing additionally for increasing correlation coefficient of RSQ and reducing SEC and SECV in the future.

Comparison of the Cold-Pressed Peel Oil Composition between ]Korean and Japanese Satsuma Mandarin (Citrus unshiu Marcov. forma Miyagawa-wase) by GC, GC-MS and GC-O

  • The comparison of the volatile flavor components from Korean and Japanese Satsuma mandarin (C. unshiu Marcov. forma Miyagawa-wase) peel oils, isolated by cold-pressing, was performed by gas chromatography, mass-spectrometry and gas chromatography-olfactometry (GC-O). Eighty-five volatile components were identified in each oil by GC and GC-MS. Forty-three components were detected in each oil by GC-O. The total amount of monoterpene hydrocarbons was 95.88% (Korean mandarin) and 95.29% (Japanese mandarin). Limonene, ${\gamma}$-terpinene, myrcene and $\alpha$-pinene were the main components of the cold-pressed oils from the both samples. The volatile composition of the Japanese mandarin was characterized by a higher content of sesquiterpene hydrocarbons, especially bicyclogermacrene, $\alpha$-humullene and valencene. The volatile composition of two samples can easily be distinguished by the percentages of aldehydes, ketones and esters, which were found at higher levels in the Japanese mandarin. The sweet and fruity flavor was stronger in the Korean mandarin oil while herbaceous flavor was stronger in Japanese sample. From GC-O data it is suggested that the sweet and fruity flavor of the Korean mandarin resulted from terpinolene and linalool, and the herbaceous note of the Japanese mandarin from $\alpha$-humullene, nepal, ι-carvone and perill aldehyde.

Carcass Characteristics and Meat Quality of Swamp Buffaloes (Bubalus bubalis) Fattened at Different Feeding Intensities

  • Twenty-four male 1-year old swamp buffaloes (Bubalus bubalis) were randomly allocated to 4 groups. One group grazed on guinea grass (GG) and another on guinea grass and the legume Stylosanthes guianensis (GL). The other two groups were kept in pens and fed freshly cut guinea grass and concentrate at an amount of 1.5% (GC1.5) and 2.0% (GC2.0) of body weight, respectively. The effect of the different feeding intensities on carcass characteristics and meat quality were assessed. The mean body weight at slaughter was 398 (${\pm}16$) kg. Average daily gain was higher in concentrate-supplemented groups (570 and 540 g/d in GC1.5 and GC2.0, respectively) when compared to GG (316 g/d) and GL (354 g/d) (p<0.01). Likewise, the warm carcass weight was higher in GC1.5 and GC2.0 compared to GG and GL. Dressing percentage was 48.1% and 49.5% in GC1.5 and GC2.0 in comparison to 42.9% and 44.8% observed in GG and GL, respectively. Meat of Longissimus throracis from GC1.5 and GC2.0 was redder in color (p<0.01), while water holding capacity (drip and thawing loss) was improved in pasture-fed groups (p<0.05). Protein and fat content of Longissimus thoracis was higher in animals supplemented with concentrate (p<0.01), as was cholesterol content (p<0.05), whereas PUFA:SFA ratio was higher and n-6/n-3 ratio lower (p<0.01) in pasture-fed buffaloes. Results of the present study showed that the supplementation of pasture with concentrate enhances the growth and carcass characteristics of swamp buffaloes expressed in superior dressing percentage, better muscling, and redder meat with a higher content of protein and fat, whereas animals grazing only on pasture had a more favorable fatty acid profile and water holding capacity. In conclusion, the supplementation of concentrate at a rate of about 1.5% of body weight is recommended to improve the performance and carcass quality of buffaloes.

형개의 정유 성분인 Limonene, Menthone, Pulegone의 부위별 함량 연구 (Content of Limonene, Menthone, Pulegone from Different Parts of Schizonepeta tenuifolia Briquet)

  • Objectives : To compare limonene, menthone and pulegone content from different part of Schizonepeta tenuifolia Briquet. Methods : The quantitative content of essential oil was determined according to the method of Chinese Pharmacopoeia. GC analysis of essential oil was carried out using a flame ionization detector. The results obtained were confirmed by GC/MS. Results : The content of essential was high on the leaf part. The content of menthone was higher than other ingredients. When compare the content observed by harvesting time, the highest pick was shown on middle of August. Conclusion : This research can be used to modify or feedbackthe article in the Korean Pharmacopoeia.

Vitis vinifera 적포도주 휘발성분의 분리 및 동정 (Isolation and Identification of Volatile Compounds from Red Wine Manufactured with Vitis vinifera grapes)

  • Vitis vinifera 적포도주의 화학성분, 고급 알콜, esters 및 휘발성분 등을 측정하기 위하여 HPLC, GC및 GC-MS등의 기기를 이용하였다. 포도주의 저장 숙성중에는 산류의 변화가 많았는데, 특히 tartaric acid가 현저히 감소하였다. Total phenolics와 phenol flavonoid의 함량은 Malbec 품종으로 제조한 포도주가 각각 470mg/L과 245mg/L로서 Cabernet sauvignon (310mg/L, 135mg/L)과 Cabernet franc (425mg/L, 125mg/L) 포도주보다 많았고, nonfalvonoid의 함량은 Cabernet franc 포도주가 300mg/L로서 Malbec (225mg/L)과 Cabernet sauvignon (175mg/L) 포도주보다 많았다. Acetaldehyde 함량은 Cabernet sauvignon (26mg/L)과 Cabernet franc (28mg/L에 비하여 Malter(33mg/L) 포도주에서 많았다. Methyl alcohol, propanol 및 isoamyl alcohol은 다른 두 포도주에 비하여 Cabernet sauvignon 포도주에서 많았고, isobutanol은 Malbec 포도주가 64mg/L이었으며, ethylacetate는 Cabernet franc 포도주가 35mg/L로서 Malbec (28mg/L)과 Cabernet sauvignon (23mg/L) 보다 많았다. 세품종(Cabernet sauvignon, Cabernet franc, 및 Malbec)의 포도주 농축물로부터 GC를 이용하여 87~91여종의 휘발성분을 분리하였고, 그 중 terpine-4-ol을 비롯한 35종을 GC-MS를 사용하여 동정하였다.

쌀의 호응집성에 대한 QTLs 분석 (Analysis of Quantitative Trait Loci (QTL) Associated with the Gel Consistency in Rice)

  • 본 연구에서는 자포니카형인 '낙동'과 통일형인 '삼강' 조합의 DH 집단을 이용하여 식미를 결정하는 특성 중 하나인 호응집성과 미립의 이화학적 특성인 천립중, 장폭비, 아밀로즈 함량, 단백질 함량, 지질 함량 및 전분 함량 간 상관관계를 검정하였다. 또한 호응집성의 QTL을 분석하고, 각 QTL에 속하는 DNA marker와 DH 집단의 호응집성 및 품종별 호응집성 간 관계를 분석하였다. '삼강/낙동' DH 집단의 호응집성 범위는 35~94 mm로 비교적 넓은 범위의 변이를 나타내었고 양친의 범위를 벗어나는 초월분리현상을 나타내었다. '삼강/낙동' DH 집단에서 호응집성은 미립의 이화학적특성 중 아밀로즈 함량과는 연으로 분류된 계통에서만 부의 유의성 있는 상관관계를 나타내었고, 지질 함량과는 중 및 전체 계통에서는 부의 상관관계를 나타내었으나 연으로 분류된 계통과는 정의 상관관계를 나타내었다. 단백질 함량과는 연으로 분류된 계통과는 정의 상관관계, 중과 전체 계통에서는 부의 상관관계를 나타내었으며, 전분 함량에서는 연으로 분류된 계통에서만 호응집성과 부의 상관관계를 나타내었다. 호응집성과 연관된 QTL은 4번(qgc4)과 11번(qgc11) 염색체에서 탐색되었으며 LOD score는 각각 3.1, 2.9를 나타내었고 두 QTL의 설명 가능한 표현형 변이는 23%로 비교적 크게 작용하고 있었다. '삼강/낙동' DH 집단에서 gel의 길이가 긴 상위 10개 계통과 짧은 하위 10개 계통을 대상으로 QTL 연관 DNA marker와 호응집성간의 관계분석에서 S4026과 RM287은 gel의 길이와 연관성이 높은 것으로 나타났다.

KOH 첨가에 의한 페놀수지로 제조된 유리상탄소의 미세구조제어 (Effect of KOH Addition on Pore Structure of Glassy Carbon Prepared by Polymerization of Phenolic Resin)

  • 페놀수지 원료에 인산 경화제를 사용하여 유리상탄소를 제조할 때, 활성화제인 KOH를 첨가하여 미세구조를 제어하였다. KOH의 첨가량에 따라 수율 및 물성을 조사하였다. KOH를 첨가하지 않고 제조한 유리상탄소는 미세기공을 거의 포함하지 않지만, KOH의 첨가량이 증가될수록 미세구조가 발달하여 KOH/페놀수지 비가 0.9에서 비표면적율 870 $m^2/g$정도까지 증가시킨 다공질의 유리상탄소를 얻을 수 있었다. 탄화수율은 KOH의 첨가량이 증가될수록 감소하여 40% 수준에서 15%까지 떨어졌고, 전기비저항은 50${\times}$$10^{-4}$에서 60${\times}$$10^{-4}$$\Omega$.cm으로 약간 증가하였으며, 물 속에서 측정한 겉보기 밀도는 1.5에서 0.9g/$cm^3$까지 감소하였다.

선택이온 측정법에 의한 십자화과 채소중의 Sulforahane 함량 (Determination of sulforaphane in cruciferous vegetables by SIM)

  • 한국인이 흔히 섭취하는 십자화과 채소와 백합과 채소중의 S-methylsulfinylbutyl isothiocyanate, 즉, sulforaphane 함량을 GC/MS로 분석하여 가장 많이 함유된 채소를 선별하고, 십자화과 및 백합과 채소에 속하는 20여종의 채소 용매추출물을 GC/MS에 주입하여 sulforaphane 표준품의 질량분석 스펙트럼을 근거로 선택이온 측정법 GC/MS (SIM)에 의해 정량하였다. 그 결과, 브로콜리에 $80.2{\sim}631.4\;ppm$으로 가장 많이 함유되어 있었으며, 그 다음으로 순무, 무청, 자색 양배추, 무, 케일, 흰색 양배추, 콜리플라워, 배추, 청경채의 순이었으며, 갓, 돌산갓, 무순 및 부추 그리고 달래, 골파, 양파 및 마늘 등 백합과 채소에는 함유되어 있지 않았다. 브로콜리 품종 중에서 sulforaphane이 가장 많은 품종은 'Pilgrim'과 '1243'이었다. 브로콜리는 식용부위별로 품종 간에 sulforaphane 함량이 달랐는데, 꽃에 가장 많이 함유된 품종은 'Pilgrim' 이었고, 대에 가장 많이 함유된 품종은 '1243'으로 700 ppm 이상의 sulforaphane이 존재하였다.

Comparative Studies on Medicinal Constituents of Korean and Chinese Angelicae Dahuricae Radix

  • The medicinal constituents contained in Korean and Chinese Angelicae dahuricae Radix were compared by confirming their qualities. From the extracts of Angelicae dahuricae Radix, oxypeucedanin, imperatorin, and alloisoimperatorin etc. furanocoumarin derivatives were identified by GC/MS analysis. Through GC/FID analysis, the furanocoumarin derivatives content of Baizi cultivated in Korea was more than that cultivated in China, except for one cultivated at Ankuk province, and so confirmed to possess, on the whole, good quality medicinal constituents by content, as compared with the ones cultivated in China.

딸기의 RAPD를 위한 PCR의 최적조건 (Optimum Condition of Polymerase Chain Reaction Techniques for Randomly Amplified Polymorphic DNA of Strawberry)

  • 본 연구는 random primer를 이용하여 PCR을 수행하기 위한 딸기 DNA증폭의 최적조건을 구명하여 조직배양된 딸기 배양묘와 모본과의 유전적인 동일성의 여부 및 품종을 판별할 수 있는 marker를 개발하기 위하여 수행하였다. 추출한 딸기 커(\ulcorner)잣 DNA를 proteinase-K나 RNase-H를 처리하였을때 깨끗하고 순수한 DNA band를 확인할 수 있었으며, 50ng의 template DNA, 10pmol의 primer, 37oC annealing 온도로 45 cycle로서 PCR을 행하는 것이 가장 효율적이었다. 상기 실험결과로서 PCR적정조건을 확립한 후, UBC primer를 대상으로 딸기 여봉 DNA에서 PCR를 수행하여 RAPD의 양상을 조사한 결과 총 90개의 primer 중에서 딸기 genomic DNA에서 PCR product를 형성한 것은 46개였으며, 총 형성된 band의 수는 158개로 나타났다. Band를 형성한 primer와 band를 형성하지 않은 primer간의 GC content를 비교하면 band를 형성한 primer의 경우 GC content는 평균 67.4%이었다. 그러나 band를 형성하지 못한 primer의 경우에는GC함량이 평균58%이었다.
