Preparation of Glass-like High-density Carbon by Polymerization of Thermosetting Resin (열경화성 수지의 축중합에 의한 고밀도 유리상 탄소의 제조)

  • Due to its low density, good mechanical properties and chemical inertness, glassy carbon(GC) has been studied for appications in several fields. A raw thermosetting resin of furanic resin was polymerized with a curing agent of p-toluenesulfonic acid monohydrate. The maximum yield of GC was obtained at the curing agent content of 1.0 wt% in furanic resin. In order to make thick GC, the affect of graphite filler addition to the furanic resin was investigated. The density and electrical resitivity of GC after graphitization were 1.45 $g/cm^{3}$ and 47 ${\times}10^{-4}$ ${\Omega}$ ${\cdot}$ cm respectively and the amorphous structure of GC was confirmed by XRD profiles with very broad peaks comparable to those of graphite at $206^{\circ}$ and $45^{\circ}$.

Changes in Volatile Constituents of Zingiber officinale Roscoe Rhizomes During Storage (생강 저장 중의 향기성분 변화)

  • This study was carried out to investigate the changes in volatile constituents concerning with the flavor of the green ginger (Zingiber officinale Roscoe) during storage in underground pit ($15^{\circ}C$, RH 95%). And the constituents of essential oil of etiolated shoots formed on the mother rhizomes during the five months storage in the dark under same conditions were compared with those of mother rhizomes. The essential oils of Korean domestic ginger (Bong-dong cultivar) were isolated by simultaneous steam distillation and extraction method (SDE). Then the compositions of the essential oils were analysed by GC and GC-MS spectrometry. The major compounds of essential oil from the fresh rhizomes were zingiberene, $citronellol+{\beta}-sesquiphellandrene$, ${\beta}-phellandrene$, camphene, geranial, ${\gamma}-bisabolene$, ar-curcumene+geranyl acetate, ${\alpha}-pinene$, ${\beta}-gurjunene$, limonene and neral. The content of monoterpene hydrocarbons increased with a concomitant lowering in the amounts of sesquiterpene hrdrocarbons and oxygenated sesquiterpenes during storage of rhizomes although contents of the oxygenated monoterpens changed little or slightly during the storage. During the storage the content of such monoterpenes as camphene, ${\beta}-phellandrene$ and citral (neral and geranial) increased whereas the content of such sesquiterpenes as zingiberene and $citronellol+{\beta}-sesquiphellandrene$ decreased. The composition of shoot oil differed from that of mother rhizome oil in having higher content of terpene hydrocarbons and also in the higher content of bornyl acetate, ${\beta}-gurjunene$ and ar-curcumene+geranyl acetate and lower in citral (neral and geranial).

Diminished Vascular Guanylyl Cyclase Activity in Deoxycorticosterone Acetate-Salt Hypertension

  • Pathophysiological implications of the vascular nitric oxide (NO)/cGMP pathway in hypertension were investigated. Sprague-Dawley rats were made deoxycorticosterone acetate (DOCA)-salt hypertensive for six weeks. The protein expression of endothelial constitutive NO synthase (ecNOS) and the tissue content of NO were determined in the thoracic aorta. The protein expression and catalytic activity of soluble guanylyl cyclase (GC) were also determined. Systolic blood pressure measured on the day of experiment was significantly higher in the experimental group than in the control. The hypertension was associated with decreases in the vascular tissue content of NO metabolites, concomitantly with the expression of ecNOS proteins. The protein expression of GC was not affected, while its catalytic activity was significantly decreased in hypertension. These results indicate that the high blood pressure is associated with a decreased activity of vascular NO/cGMP pathway in DOCA-salt hypertension.

  • The prescription of Gal Gun Tang, which has been used for treatment of cold, fever, and muscular pain in Chinese herb medicine, is produced in the form of decoction However, the storage problem for this dosage form remains unsettled. Using HPLC and GC, we examined quantitative change of major constituents caused by time-progress and temperature-change. Nine major constituents, such as cinnamaldehyde, cinnamic acid, ephedrine, puerarin, paeoniflorin, daidzin, benzoic acid, glycyrrhizin and liquiritin, were selected as references. The content of cinnamaldehyde significantle decreased by the increase of temperature. While that of cinnamic acid increased. Benzoic acid showed the most significant change of the content in three months at $40^{\circ}C$. It is suggested that most of constituents are considerably stable when kept frozen.

A Study on the Flavor Compounds of Dongchimi (동치미의 맛 성분에 관한 연구)

  • To study the most optimum condition of Dongchimi, the various Dongchimi distinct from the amount of salt, the temperature of fermentation and the ratio of radish to water were examed by sensory evaluation. Also, the content of volatile organic acids and nonvolatile organic acids were investigated by GC, the content of sulfur compounds by GC/MS. Volatile organic acids were identified with those butyl esters and nonvolatile organic acids were done with those TMS derivatives. Sulfur compounds and those decomposed products were extracted by steam distillation In results, the most optimum conditions ware salt 2.4% fermented temperature $4^{\circ}C$, ratio 1:1.5. Volatile organic acids detected were formic, acetic acid, and the amounts were effected by saltness. Nonvolatile organic acids detected were lactio, malio, fumario, tartario acid. Lactic acid was thought to effect overall eating quality. Sulfur compounds were almost the isothiocyanate groups in raw radish, which little in fermented Dongchimi.

Isolation and Identification of Biologically Active Components from Korean Valerian Roots (한국산 쥐오줌풀로부터 생리활성 성분의 분리 및 동정)

  • The contents of valepotriates, valerenic acid and their derivatives as mild sedative and antispamodic principles in two Korean valerian roots (Valeriana officinalis var. latifolia Miq. and V. fauriei var. dasycarpa Hara) were investigated and were compared with those in European valerian roots(Valeriana officinalis L.) by BPLC method. Among valepotriate compounds, valtrate was detected as a small amount in two Korean valerian roots, and iso-valtrate and dihydrovaltrate were detected as a trace amount. Among valerenic acid and their derivatives, valerenic acid were contained from 0.9∼3.46 mg/g base on dry weight. Over the vegetation periods investigated, the content of valepotriates remained more or less constant, hut the content of valerenic acid reached its maximum in 3-year old age. The compound isolated from Korean V. officinalis var. latifolia was elucidated as valerenic acid by spectroscopic data such as GC-MS, $^1$H-NMR and $^{13}$ C-NMR, and comparison of GC retention time with that of authentic compound.

  • An easy, sensitive and rapid method for the analysis of phytol, a strong ACAT (Acyl-CoA: Cholesterol Acyltransferase) inhibitory diterpenoid, was established. The pre-treatment of sample for GC analysis of phytol includes aq. MeOH extraction, partition of the extracts between EtOAc and water, and filtration of organic phase through Supelclean LC-Si SPE tube. The GC analysis of phytol for a variety of Lactuca sativa was carried out on DB-5 capillary column with flame ionization detector (FID). The consistency in phytol contents according to each variety was not shown. Especially, Jeoktoma, as RS-2 type, showed the highest phytol content.

Classification of Capsicum annuum Germplasm Using Random Amplified Polymorphic DNA (RAPD를 이용한 고추(Capsicum annuum) 유전자원의 분류)

  • This study was initiated to evaluate genetic relationship among various domestic and exotic pepper accessions using random amplified polymorphic DNA(RAPD) markers. The results suggested that the optimum conditions for PCR with random primers in Capsicum spp. could be obtained with 3mM of $MgCl_2$, 1.5U of Taq. DNA polymerase, 10ng of template DNA, $200{\mu}M$ of dNTPs, 200nM of random primer, and $42^{\circ}C$ of annealing temperature. Sixteen random primers showing high band intensity and reproducibility were selected from 80 random primers. Primers having 70% GC content were more effective in DNA amplification than primers having 60% GC content. The total 93 DNA bands including 71 polymorphic bands and 22 monomorphic bands were obtained with selected 16 random primers for 31 pepper cultivars and lines. About 4.4 polymorphic bands per primer were produced. Similarity coefficients were calculated by using 71 polymorphic bands and dendrogram based on the similarity coefficient showed clear classification of 31 peppers into three Capsicum species of Capsicum annuum, Capsicum chinense and Capsicum chacoense.

STELLAR CONTENTS AND GLOBULAR CLUSTER CANDIDATES IN THE SCULPTOR GROUP GALAXY NGC 300

  • We present UBVI CCD photometry of the stellar contents and globular cluster(GC) candidates in the spiral galaxy NGC 300 in the Sculptor group. Color-magnitude diagrams for 18 OB associations having more than 30 member stars are presented. The slope of the initial mass function for the bright stars in NGC 300 is estimated to be ${\Gamma}= -2.6{\pm} 0.3$. Assuming the distance to NGC 300 of (m - M)o = 26.53 $\pm$ 0.07, the mean absolute magnitude of three brightest blue stars is obtained to be < $M_v^{BSG}$ (3) > = -8.95 mag. We have performed search for GCs in NGC 300 and have found 17 GC candidates in this galaxy. Some characteristics of these GC candidates are discussed.

Quantification of Trans Fatty Acids in Processed Foods by Soxhlet Extraction Method (Soxhlet 추출법에 의한 가공 식품의 Trans 지방산 정량)

  • This study was designed to determine total trans fatty acids (tFAs) content of processed foods commonly consumed in the Korean diet and to prepare the database for the estimation of tFAs intake in Koreans. Total fat and tFAs content was determined by Soxhlet extraction method and attenuated total reflection infrared spectroscopy (ATR-IR), respectively. The tFAs positional isomers were analyzed using GC/MS spectrometer. In margarines, the content of tFAs ranged from 8.27% to 28.53%. Shortenings contained higher levels of tFAs (1.98~11.33%) than lards (1.83~1.96%). The content of tFAs in grilled confectioneries was wide (7.37~26.54%). Instant popcorns contained the highest amount of tFAs. Fried chickens had 0.44 to 14.85% of tFAs and french fries had 5.18 to 27.01% of tFAs. In fried snacks (crispy) and chocolates, tFAs were not detected. The amount of tFAs per serving size was the highest in instant popcorn, followed by french fries, fried snack (tortilla), doughnuts, and grilled confectioneries. TFAs isomer of margarines was mostly C18:1${\Delta}$9t. In shortenings and lards, the most abundant positional isomer of tFAs was C18:1${\Delta}$9t and C18:2${\Delta}$9t ${\Delta}$12t. Correlation coefficient of tFAs content between IR and GC/MS method was $r^2$=0.91.
