Actin Dysfunction Induces Cell Cycle Delay at G2/M with Sustained ERK and RSK Activation in IMR-90 Normal Human Fibroblasts

  • The actin cytoskeleton plays a key role in the entry of mitosis as well as in cytokinesis. In a previous study, we showed that actin disruption delays mitotic entry at G2/M by sustained activation of extracellular signal-related kinase 1/2 (ERK1/2) in primary cells but not in transformed cancer cell lines. Here, we examined the mechanism of cell cycle delay at G2/M by actin dysfunction in IMR-90 normal human fibroblasts. We observed that de-polymerization of actin with cytochalasin D (CD) constitutively activated ribosomal S6 kinase (RSK) and induced inhibitory phosphorylation of Cdc2 (Tyr 15) in IMR-90 cells. In the presence of an actin defect in IMR-90 cells, activating phosphorylation of Wee1 kinase (Ser 642) and inhibitory phosphorylation of Cdc25C (Ser 216) was also maintained. However, when kinase-dead RSK (DN-RSK) was overexpressed, we observed sustained activation of ERK1/2, but no delay in the G2/M transition, demonstrating that RSK functions downstream of ERK in cell cycle delay by actin dysfunction. In DN-RSK overexpressing IMR-90 cells treated with CD, phosphorylation of Cdc25C (Ser 216) was blocked and phosphorylation of Cdc2 (Tyr 15) was decreased, but the phosphorylation of Wee1 (Ser 642) was maintained, demonstrating that RSK directly controls phosphorylation of Cdc25C (Ser 216), but not the activity of Wee1. These results strongly suggest that actin dysfunction in primary cells activates ERK1/2 to inhibit Cdc2, delaying the cell cycle at G2/M by activating downstream RSK, which phosphorylates and blocks Cdc25C, and by directly activating Wee1.

  • The advent of 5G mobile communications, which is expected in 2020, will provide many services such as Internet of Things (IoT) and vehicle-to-infra/vehicle/nomadic (V2X) communication. There are many requirements to realizing these services: reduced latency, high data rate and reliability, and real-time service. In particular, a high level of reliability and delay sensitivity with an increased data rate are very important for M2M, IoT, and Factory 4.0. Around the world, 5G standardization organizations have considered these services and grouped them to finally derive the technical requirements and service scenarios. The first scenario is broadcast services that use a high data rate for multiple cases of sporting events or emergencies. The second scenario is as support for e-Health, car reliability, etc.; the third scenario is related to VR games with delay sensitivity and real-time techniques. Recently, these groups have been forming agreements on the requirements for such scenarios and the target level. Various techniques are being studied to satisfy such requirements and are being discussed in the context of software-defined networking (SDN) as the next-generation network architecture. SDN is being used to standardize ONF and basically refers to a structure that separates signals for the control plane from the packets for the data plane. One of the best examples for low latency and high reliability is an intelligent traffic system (ITS) using V2X. Because a car passes a small cell of the 5G network very rapidly, the messages to be delivered in the event of an emergency have to be transported in a very short time. This is a typical example requiring high delay sensitivity. 5G has to support a high reliability and delay sensitivity requirements for V2X in the field of traffic control. For these reasons, V2X is a major application of critical delay. V2X (vehicle-to-infra/vehicle/nomadic) represents all types of communication methods applicable to road and vehicles. It refers to a connected or networked vehicle. V2X can be divided into three kinds of communications. First is the communication between a vehicle and infrastructure (vehicle-to-infrastructure; V2I). Second is the communication between a vehicle and another vehicle (vehicle-to-vehicle; V2V). Third is the communication between a vehicle and mobile equipment (vehicle-to-nomadic devices; V2N). This will be added in the future in various fields. Because the SDN structure is under consideration as the next-generation network architecture, the SDN architecture is significant. However, the centralized architecture of SDN can be considered as an unfavorable structure for delay-sensitive services because a centralized architecture is needed to communicate with many nodes and provide processing power. Therefore, in the case of emergency V2X communications, delay-related control functions require a tree supporting structure. For such a scenario, the architecture of the network processing the vehicle information is a major variable affecting delay. Because it is difficult to meet the desired level of delay sensitivity with a typical fully centralized SDN structure, research on the optimal size of an SDN for processing information is needed. This study examined the SDN architecture considering the V2X emergency delay requirements of a 5G network in the worst-case scenario and performed a system-level simulation on the speed of the car, radius, and cell tier to derive a range of cells for information transfer in SDN network. In the simulation, because 5G provides a sufficiently high data rate, the information for neighboring vehicle support to the car was assumed to be without errors. Furthermore, the 5G small cell was assumed to have a cell radius of 50-100 m, and the maximum speed of the vehicle was considered to be 30-200 km/h in order to examine the network architecture to minimize the delay.

  • Methods of Rock fragmentation are used rock of housing repair development at KU-SAN DONG area in seoul Youn-Pyong Ku. So, Theorical analyses of the effect of vibration and frequency on structural damage around old housed also discussed. The results can be summarized as follows: 1. A area(Rock area not more than 15m Ku-San Mention) Some Empirical equations were obtained $V=K\{{\frac{D}{W}}1/3\}^{-n}$ where the values for n and K are estimated to be -1.64 and 94 respectively, this values were obtained only theorical analyses. If we have 125g charge this area is impossible blasting operation, so this area must be worked by SRS(Super Rock Splitter) method. 2. B area(Rock area from 15m to 25m in a boundary line from Ku-San Mention) This area charge is about 125g in a delay time by some empirical equation s. So, this area can be blasting operations by small charge. 3. C area(Rock area from 25m to 35m in a boundary line from Ku-San Mention) This area charge is about 500g in delay time by some empirical equation s. So, this area can be blasting operations by middle charge. 4. D area(Rock area more then 35m in a boundary line from Ku-San Mention) This area charge is about 1000g in a delay time by some empirical equation s. So, this area can be blasting operations by middle charge.

Expediting Data through Erasure Coding in Networks with High Coefficient of Variation of Transfer Time (전송시간의 변화가 큰 네트워크에서 이레이저 코딩을 적용한 긴급 데이터 전송 방법 및 성능 분석)

  • In this paper, we focus on end-to-end transfer delay improvement by using erasure coding when delivering expediting message consisting of M packets in networks with high coefficient of variation of transfer time. In the scheme, M packets are divided into b groups with each having g packets. Each group is erasure coded with additional r packets and transmitted. Since the first arrived g packets among g+r packets completes the delivery of the group, the delivery time of the expediting message is reduced. For the scheme, we investigate the optimum group size and number of redundancy packets considering delivery delay reduction and additional transmission cost caused by using erasure coding. From the results of the investigation, we see that the proposed scheme is effective in networks having high variability of transfer time and would be very useful and practical especially for the case that expedited deliveries of messages are needed.

Transmission Performance of VoIP Traffic over MANETs (MANET에서 VoIP 트래픽의 전송성능)

  • In this paper, some performance characteristics of VoIP(Voice over Internet Protocol) for MANET(Mobile Ad-hoc Networks) with simulation is studied and appropriate condition for implementation of VoIP service is suggested. VoIP simulator is implemented with NS(Network Simulator)-2. VoIP traffic for simulation is generated with some codecs of G.711, G.723.1, G.726-32, G.729A, GSM.AMR and iLBC. As simulation results for traffic transmission under $670{\times}670m$ 50node MANET environment, performance data for MOS(Mean Opinion Score), network delay, packet loss rate and transmission bandwidth are measured. Normalized analysis about measured results shows that maximum VoIP connection satisfying VoIP service quality condition is 15.

  • The spatial variability of soil heat fluxes in a conifer forest was investigated by meteorological measurement. The maximum daily averages of R $s_{dn}$ and Rn were about 260 W $m^{-2}$ and 180 W $m^{-2}$ . The daily average of G was typically 10% of net radiation during mid-July to mid-August. The measured soil heat flux of $G_{6}$ was suitable to calculate G within 2% error during the study period. A time delay in the maximum nux at a depth of 0.1 m by heat storage was observed. About 10 to 15 W $m^{-2}$ of error can occur, if it is neglected.

Radio Propagation Characteristics of Different Frequency Bands in Multiple Paths According to Antenna Position in an Indoor Lobby Environment (실내 로비 환경에서 안테나 위치에 따른 다중 경로의 서로 다른 주파수 대역의 전파 특성)

  • The radio propagation characteristics of the 6, 10, and 17 GHz frequency bands in multiple paths in an indoor lobby environment were analyzed. The line-of-sight (LOS) and non-LOS (NLOS) paths were measured from a distance of 2-16 m (0.5 m intervals) from the transmitting to the receiving antenna positions. For basic transmission losses, three parameters were compared using the floating intercept path loss model corresponding to the path. For a root mean square delay spread, the measurement results were compared for cumulative probabilities of 10, 50, and 90%. Propagation loss and propagation delay occurred in all measured frequencies owing to the existence of pillars and an unusual lobby structure. Thus, a measurement scenario for an indoor lobby environment and the provision of standard measurement data was proposed. The results may facilitate research on the radio propagation characteristics of 5G and millimeter-wave bands in indoor lobby environments with various structures.

  • This study focuses on the hardware implementation of fast and low-system-complexity multiplier over GF(2$^{m}$ ). From the properties of an irreducible AOP of degree m. the modular reduction in GF(2$^{m}$ ) multiplicative operation can be simplified using cyclic shift operation. And then, GF(2$^{m}$ ) multiplicative operation can be established using the away structure of AND and XOR gates. The proposed multiplier is composed of m(m+1) 2-input AND gates and (m+1)$^2$ 2-input XOR gates. And the minimum critical path delay is Τ$_{A+}$〔lo $g_2$$^{m}$ 〕Τ$_{x}$ proposed multiplier obtained have low circuit complexity and delay time, and the interconnections of the circuit are regular, well-suited for VLSI realization.n.

Application of array comparative genomic hybridization in Korean children under 6 years old with global developmental delay

  • Purpose: Recent advancements in molecular techniques have greatly contributed to the discovery of genetic causes of unexplained developmental delay. Here, we describe the results of array comparative genomic hybridization (CGH) and the clinical features of 27 patients with global developmental delay. Methods: We included 27 children who fulfilled the following criteria: Korean children under 6 years with global developmental delay; children who had at least one or more physical or neurological problem other than global developmental delay; and patients in whom both array CGH and G-banded karyotyping tests were performed. Results: Fifteen male and 12 female patients with a mean age of $29.3{\pm}17.6months$ were included. The most common physical and neurological abnormalities were facial dysmorphism (n=16), epilepsy (n=7), and hypotonia (n=7). Pathogenic copy number variation results were observed in 4 patients (14.8%): 18.73 Mb dup(2)(p24.2p25.3) and 1.62 Mb del(20p13) (patient 1); 22.31 Mb dup(2) (p22.3p25.1) and 4.01 Mb dup(2)(p21p22.1) (patient 2); 12.08 Mb del(4)(q22.1q24) (patient 3); and 1.19 Mb del(1)(q21.1) (patient 4). One patient (3.7%) displayed a variant of uncertain significance. Four patients (14.8%) displayed discordance between G-banded karyotyping and array CGH results. Among patients with normal array CGH results, 4 (16%) revealed brain anomalies such as schizencephaly and hydranencephaly. One patient was diagnosed with Rett syndrome and one with $M{\ddot{o}}bius$ syndrome. Conclusion: As chromosomal microarray can elucidate the cause of previously unexplained developmental delay, it should be considered as a first-tier cytogenetic diagnostic test for children with unexplained developmental delay.

Bit-Parallel Systolic Divider in Finite Field GF(2m) (유한 필드 GF(2m)상의 비트-패러럴 시스톨릭 나눗셈기)

  • This paper presents a high-speed bit-parallel systolic divider for computing modular division A($\chi$)/B($\chi$) mod G($\chi$) in finite fields GF$(2^m)$. The presented divider is based on the binary GCD algorithm and verified through FPGA implementation. The proposed architecture produces division results at a rate of one every 1 clock cycles after an initial delay of 5m-2. Analysis shows that the proposed divider provides a significant reduction in both chip area and computational delay time compared to previously proposed systolic dividers with the same I/O format. In addition, since the proposed architecture does not restrict the choice of irreducible polynomials and has regularity and modularity, it provides a high flexibility and Scalability with respect to the field size m. Therefore, the proposed divider is well suited to VLSI implementation.
