Purification of \alpha-Cyclodextrin Glucanotransferase Excreted from Themophilic Geobacillus thermosac-chalytycus and Characterization of Transglycosylation Reaction of Glucosides. (호열성 Geobacillus thermosacchalytycus가 생산하는 \alpha-Cyclodextrin Glucanotransferase의 분리정제와 당전이 반응 특성)

  • $\alpha$-Cyclodextrin glucanotransferase excreted from a newly isolated Geobacillus thermosacchalytycus was purified through the ultrafiltraion, hydrophobic Sepharose CD-4B affinity chromatography, and gel filtration on Sephadex G-75, respectively. The molecular weight of the purified CGTase was 69 kDa and its N-terminal amino acid sequence was determined to be Asn-Leu-Asn-Lys-Val-Asn-Phe-Val-Ser-Asp-Val-Val-Val-Gln-Ile. The optimum pH and temperature were pH 6.0 and$ 60^{\circ}C$, respectively, and stably at the pH range of 6.0-8.0 and $60^{\circ}C$ in the presence of $Ca^{++}$. The excreted CGTase from the thermophilic G. thermosacchalytycus was $\alpha$-type showing a high coupling activity for the transglycosylation on various glucosides. The coupling reaction was carried out according to the random ternary complex mechanism.m.

  • In this study the mechanical properties of ferritic ductile cast iron in as-cast are discussed by metallographic considering the effect of phosphorus content(about 0 - 0.8wt.%). In ferritic S.G. cast iron containing about 4.2wt.% Si, 0.5wt.% Ni, 0.02wt.%B, these samples are investigated for castability, microstructure, machinability, wear resistance, mechanical and thermal properties. The main results are summarized as follows. 1) The chill depth increased greatly with P addition up to about 0.6wt.% but the tensile strength and the elongation are decreased smoothly. 2) The size of S.G. and the amount of steadite increase with increasing P content. This means the tendency of increasing the number of S.G. 3) The maximum value of wear by mechanical fracture was appeared at the abrasion speed of 1.14m/sec and the plastic Flow layers are stretched longly on the wear surface with decreasing P content. 4) Although the coefficient of thermal expansion increased with P addition, the cast iron growth was slowing down extremely at 0.6w.% P or more.

  • The effect of biomass concentration of Saccharomyces cerevisiae ATCC 24858 on specific growth rates, biomass yields, ethanol yields and productivity in the batch fermentation of rotary shaker was investigated. The specific growth rate decreased according to the increase in the biomass density and finally became zero at a biomass concentration, 55g/L. The ethanol yield $Y_{p/s}$ represented a constant value, 0.43, regardless of the change of biomass concentrations. However, the biomass yield $Y_{x/s}$ showed a trend to diminish in values with augmentation of biomass density and ultimately to reach zero at 55g/L of biomass concentration. The ethanol productivity increased linearly with biomass concentration so that, in case of initial sugar concentration, 170g/L, the productivity for 55g/L of biomass density rose up to 30g/L$\cdot$hr for all the batch fermentations. And also the ethanol concentration inhibiting completely the growth was verified 95g/L by applying experimental data to Luong's equation.

  • 24 samples which were incoulated with Aspergillus flavus and Bcillus subtilis and cultured on the steamed soybean media under various conditions-pH, moisture and temperature were-investigated on the production of aflatoxin by the interaction of these two microorganisms. 1) The amount of aflatoxin produced by mixed cultures of Aspergillus flavus and Bacillus subtilis was decreased significantly rather than that of single cultures of Aspergillus flavus. 2) Maximum production of crude aflatoxin was 2,560 ppm $(B_1,\;0.908\;ppm;\;B_2,\;0.261\;ppm;\;G_1,\;1.162\;ppm;\;G_2,\;0.229\;ppm)$ at 30% moisture, pH 5.0 and $20^{\circ}C$, whereas minimum production was 1.107 ppm $(B_1,\;0.341\;ppm;\;B_2,\;0.104\;ppm;\;G_1,\;532\;ppm;\;G_2,\;0.130\;ppm)$ at 63% moisture, pH 9.0 and $40^{\circ}C$.

  • This study aimed to evaluate urea excretion, nitrogen balance and microbial protein synthesis in lactating goats fed with diets containing different protein sources in the concentrate (soybean meal, cottonseed meal, aerial part of cassava hay and leucaena hay). Four Alpine goats whose mean body weight was $42.6{\pm}6.1kg$ at the beginning of the experiment, a mean lactation period of $94.0{\pm}9.0days$ and a production of $1.7{\pm}0.4kg$ of milk were distributed in a $4{\times}4$ Latin square with four periods of 15 days. Diets were formulated to be isonitrogenous, containing 103.0 g/kg of CP, 400 g/kg of Tifton 85 hay and 600 g/kg of concentrate. Diet containing cottonseed meal provided (p<0.05) increased excretion of urea and urea nitrogen in the urine (g/d and mg/kg of BW) when compared with leucaena hay. The diets affected the concentrations of urea nitrogen in plasma (p<0.05) and excretion of urea nitrogen in milk, being that soybean meal and cottonseed meal showed (p<0.05) higher than the average aerial part of the cassava hay. The use of diets with cottonseed meal as protein source in the concentrate in feeding of lactating goats provides greater nitrogen excretion in urine and negative nitrogen balance, while the concentrate with leucaena hay as a source of protein, provides greater ruminal microbial protein synthesis.

  • Alcaligenes eutrophus was successfully recovered from high cell density broth by pre-treatment with Fe-based coagulants. An inorganic coagulant, Fe$_2$(SO$_4$)$_3$, and a polymerized coagulant, Ferix-3, were used. Good coagulation was observed in broad pH range of 3 to 13, the floe size was increased with increasing pH of culture broth. The optimum pH of fermentation broth for cell recovery was 10 to 13. The optimum coagulant dosages to recover cells with 95% cell recovery were increased with increasing cell concentration. Optimal coagulant dosage was lower when the polymerized coagulant was used rather than the inorganic coagulant. The coexistence of NH$_4$$\^$+/ was increased coagulant requirement, and the coagulant requirement was 0.066g Fe$_3$$\^$+//g NH$_4$$\^$+/.

Effects of Benzo〔a〕pyrene on Growth and Photosynthesis of Phytoplankton (식물플랑크톤의 성장과 광합성에 대한 benzo〔a〕pyrene의 영향)

  • We examined the impacts of anthyopogenic pollutant (benzo〔a〕pyrene) on the growth and photosynthesis of five marine phytoplankton species (Skeletonema costatum, Heterosigma akashiwo, Prorocentrum dentatum, P. minimum, Aknshiwo sanguinea), which are dominant in Korean coastal water. After the 72 h exposure to benzo〔a〕pyrene, the dramatic decrease in cell numbers was observed in the range of 1 to 10 $\mu\textrm{g}$ L$^{-1}$ for S. costatum, P. minimum, P. dentatum, whereas for A. sanguinea and H. akashiwo at the low concentrations 0.1 to 1 $\mu\textrm{g}$ L$^{-1}$ . Among the 5 phytoplankton species, the highest growth inhibition concentration ($IC_{50}$/) was 6.20 $\mu\textrm{g}$ L$^{-1}$ for P. minimum, followed by 2.14 $\mu\textrm{g}$ L$^{-1}$ for P. dentatum, 1.68 $\mu\textrm{g}$ L$^{-1}$ for S. costatum, 0.74 $\mu\textrm{g}$ L$^{-1}$ for H. akashiwo, 0.10 $\mu\textrm{g}$ L$^{-1}$ for A. sanguinea. The five species exposed to the low concentration of 1 $\mu\textrm{g}$ L$^{-1}$ were recovered after transferring to new media, but the species exposed to the high concentrations of 10 and 100 $\mu\textrm{g}$ L$^{-1}$ were not recovered, with the exception of P. minimum. Those results indicate that the thecate dinoflagellate P. minimum is most tolerant to the chemical and the athecate dinoflagellate A. sanguinea is not. Geneyally, the cell-specific photosynthetic capacity of H. akashiwo exposed to the low concentrations of 0.1 and 1 $\mu\textrm{g}$ L$^{-1}$ was higher than that of the cells in the control, whereas the cells exposed to the high concentrations of 5 and 10 $\mu\textrm{g}$ L$^{-1}$ showed the negligible photosynthetic level by the first few days of the experiment. In the case of the cells exposed to the concentration of 5 $\mu\textrm{g}$ L$^{-1}$ , after 12 days of the experiment the photosynthetic capacity was increased toward the end of the experiment. This indicates that H. akashiwo may utilize the benzo〔a〕pyrene as a carton source for its growth when exposed to low concentrations. Results suggest that anthropogenic pollutants such as benzo〔a〕pyrene may have significant influence on the succession of phytoplankton species composition and the primary production in coastal marine environments.
