명태 유통 중 Pseudomonas spp. 농도의 예측 모델링과 민감도 분석 (Dynamic Modeling and Sensitivity Analysis for Predicting the Pseudomonas spp. Concentration in Alaska Pollack along the Distribution Path)

  • Dynamic modeling was used to predict the Pseudomonas spp. concentration in Alaska pollack under dynamic temperature conditions in a programmable incubator using Euler's method. The model evaluation showed good agreement between the predicted and measured concentrations of Pseudomonas spp. In the simulation, three kinds of distribution path were assumed: consumers buying from a distribution center (A), manufacturer (B), or direct market (C). Each of these distribution paths consists of six phases: shipping, warehousing/shipment, warehousing/storing, processing, market exhibition, and sale/consumption. Sensitivity analysis of each phase was also implemented. The Pseudomonas concentrations and sensitivities ($S_k$) at the terminal phases of the three paths were estimated to be (A) 11.174 log CFU/g and 10.550 log $S_k$, (B) 10.948 log CFU/g and 10.738 log $S_k$, and (C) 8.758 log CFU/g and 9.602 log $S_k$, respectively. The sensitivities indicated that path A has the highest risk of failure in managing the relevant phases.

Effect of Heavy Metals on Embryonic Development in the Mussel, Mytilus galloprovincialis

  • The embryos of marine bivalves have been commonly used in bioassays for quality assessments of marine environments. Although several standard protocols for the developmental bioassay of bivalves have been proposed, only a few trials for application of these protocols in environmental assessments or for the development of a new protocol with Korean species have been conducted. As such, there is a strong need to establish standard bioassay protocols with bivalves commonly found in Korean waters. To determine the sensitivity of Mytilus galloprovincialis to establish a standard bioassay, their fertilized eggs were exposed to six metals (Ag, Cd, Cr, Cu, Ni, and Zn). The order of biological impact was Ag > Cu > Ni > Zn > Cr > Cd and their lowest observed effective concentration were 5, 16.4, 25.4, 142, 187 and 1,500${\mu}g/l$, respectively. The proportion of normal larvae appeared to decrease linearly with the logarithm of each toxicant concentration within the tested range. The average values of median effective concentrations $(EC_{50})$ from the triplicate experiments for Ag, Cd, Cr, Cu, Ni, and Zn were 6.8, 1,797, 786, 16.6, 68.1, and 139.2${\mu}g/l$, respectively. There was a more than 100-fold difference in $EC_{50}$ values of Cu and Cd. The value of $EC_{50}$ or median lethal concentration of Cu was within the range observed for other bivalve developmental bioassays. The overall sensitivity of M. galloprovincialis in the present developmental bioassay was also similar to that of other marine organisms commonly used in aquatic bioassays (e.g. sea urchins, oysters). Hence, the bioassay using the embryo of M. galloprovincialis is considered to be a useful tool to monitor and evaluate the quality of marine aquatic environments.

LED 광원에 대한 암순응시 볼락 (Sebastes inermis)의 시각 스펙트럼 민감도 (Visual spectral sensitivity of dark-adapted rockfish (Sebastes inermis) in LED light source)

  • The characteristics of aquatic light fields are generally reflected in the visual systems of fishes inhabiting them. Therefore, research on light sensitivity of fish is useful to explain the correlation between the visual function and habitat, behavior and distribution of fish. Rockfish is an important species in coastal ecology and also one of the main species for culturing in Korea. To make a contribution on the maintenance of the fish resources and understanding the ecology of the rockfish, the visual spectral sensitivities of the dark-adapted rockfishes were measured in the range of visible light (405~660 nm) using a light source of light-emitting diodes (LEDs). In order to assess electrophysiological response of the fish, the ERGs (electroretinograms) of the dark-adapted rockfishes were recorded on a data logger (12 bits) and a laptop computer. Juvenile (n=5; weight: $20.3{\pm}5.2g$; total length: $10.3{\pm}0.7cm$) and adult (n=5; weight: $87.8{\pm}21.8g$; total length: $18.1{\pm}1.3cm$) rockfishes were used in experiment. The visual threshold of juvenile and adult rockfish were 11.66 (log quanta/$cm^2/s$) and 11.81 (log quanta/$cm^2/s$) in 574 nm, respectively. The peak wavelength of the spectral sensitivity in the dark-adapted juvenile and adult rockfish was commonly 551 nm (series of green color). Collectively, these results demonstrate that the rockfish has suitable visual capabilities for inhabiting coastal water in Korea.

  • 지속적인 산업화와 도시화에 따른 에너지 수요의 급증으로 인해 원유와 석유제품의 해양수송이 점점 증가하고 있다. 우리나라의 경우 국내의 유류 수요를 맞추기 위해서는 매일 30만톤급의 대형 유조선이 입항해야 하기 때문에 대형 유류오염 사고에 노출되어 있다 해양 유류오염사고의 가능성을 줄이기 위해 부단한 노력에도 불구하고 회복하기 어려운 유출 사고는 인간의 실수, 악천후 또는 복합적인 이유로 일어나게 된다. 따라서 적절한 대응시스템으로 지역방제계획을 세워 최악의 경우에 대비하고 해양환경과 연안자원을 보호해야만 한다. 특히 보호의 우선순위를 명확히 하고 현장 방제전략을 세움으로써 유출과 방제의 노력이 환경에 미치는 영향을 줄이기 요구되는 방제정보지도에 초점을 맞추었다. 본 논문에서는 기름에 대한 민감도의 정의와 개발한 소프트웨어의 주요특짚과 함께 방제정보지도를 간략히 소개한다.

ATG5 knockout promotes paclitaxel sensitivity in drug-resistant cells via induction of necrotic cell death

  • Autophagy regulators are often effective as potential cancer therapeutic agents. Here, we investigated paclitaxel sensitivity in cells with knockout (KO) of ATG5 gene. The ATG5 KO in multidrug resistant v-Ha-ras-transformed NIH 3T3 cells (Ras-NIH 3T3/Mdr) was generated using the CRISPR/Cas9 technology. The qPCR and LC3 immunoblot confirmed knockout of the gene and protein of ATG5, respectively. The ATG5 KO restored the sensitivity of Ras-NIH 3T3/Mdr cells to paclitaxel. Interestingly, ATG5 overexpression restored autophagy function in ATG5 KO cells, but failed to rescue paclitaxel resistance. These results raise the possibility that low level of resistance to paclitaxel in ATG5 KO cells may be related to other roles of ATG5 independent of its function in autophagy. The ATG5 KO significantly induced a G2/M arrest in cell cycle progression. Additionally, ATG5 KO caused necrosis of a high proportion of cells after paclitaxel treatment. These data suggest that the difference in sensitivity to paclitaxel between ATG5 KO and their parental MDR cells may result from the disparity in the proportions of necrotic cells in both populations. Thus, our results demonstrate that the ATG5 KO in paclitaxel resistant cells leads to a marked G2/M arrest and sensitizes cells to paclitaxel-induced necrosis.

Effect of Autophagy-Related Beclin1 on Sensitivity of Cisplatin-Resistant Ovarian Cancer Cells to Chemotherapeutic Agents

  • The purpose of the study was to determine the effects of autophagy related gene Beclin1 at different levels of expression on the sensitivity of cisplatin-resistant ovarian cancer cells (SKOV3/DDP) to different chemotherapeutics. In pSUPER-Beclin1 transfected cells, real-time fluorescence quantitative RT-PCR and Western blot analysis showed that expression was significantly inhibited. Flow cytometry revealed that the mean fluorescence intensity (MDC), reflecting autophagy, and cells in the G0/G1 phase were markedly reduced. When compared with the blank control group, inhibition of Beclin1 expression in SKOV3/DDP cells not only increased the rate of apoptosis following treatment with chemotherapeutics, but also increased the sensitivity. These findings suggest that Beclin1 expression plays an important role in chemotherapeutic agent-induced death of SKOV3/DDP cells. Inhibition of autophagy related gene Beclin1 expression in SKOV3/DDP cells may increase the rate of apoptosis and elevate the sensitivity to chemotherapeutics.

소금민감성유전자와 비만 (Salt-sensitive genes and their relation to obesity)

  • Purpose: Although it is well known thatmortality and morbidity due to cardiovascular diseases are higher in salt-sensitive subjects than in salt-resistant subjects, their underlying mechanisms related to obesity remain unclear. Here, we focused on salt-sensitive gene variants unrelated to monogenic obesity that interacted with sodium intake in humans. Methods: This review was written based on the modified $3^rd$ step of Khans' systematic review. Instead of the literature, subject genes were based on candidate genes screened from our preliminary Genome-Wide Association Study (GWAS). Finally, literature related to five genes strongly associated with salt sensitivity were analyzed to elucidate the mechanism of obesity. Results: Salt sensitivity is a measure of how blood pressure responds to salt intake, and people are either salt-sensitive or salt-resistant. Otherwise, dietary sodium restriction may not be beneficial for everyone since salt sensitivity may be associated with inherited susceptibility. According to our previous GWAS studies, 10 candidate genes and 11 single nucleotide polymorphisms (SNPs) associated with salt sensitivity were suggested, including angiotensin converting enzyme (ACE), ${\alpha}$-adducin1 (ADD1), angiotensinogen (AGT), cytochrome P450 family 11-subfamily ${\beta}$-2 ($CYP11{\beta}$-2), epithelial sodium channel (ENaC), G-protein b3 subunit (GNB3), G protein-coupled receptor kinases type 4 (GRK4 A142V, GRK4 A486V), $11{\beta}$-hydroxysteroid dehydrogenase type-2 (HSD $11{\beta}$-2), neural precursor cell-expressed developmentally down regulated 4 like (NEDD4L),and solute carrier family 12(sodium/chloride transporters)-member 3 (SLC 12A3). We found that polymorphisms of salt-sensitive genes such as ACE, $CYP11{\beta}$-2, GRK4, SLC12A3, and GNB3 may be positively associated with human obesity. Conclusion: Despite gender, ethnic, and age differences in genetics studies, hypertensive obese children and adults who are carriers of specific salt-sensitive genes are recommended to reduce their sodium intake. We believe that our findings can contribute to the prevention of early-onset of chronic diseases in obese children by facilitating personalized diet-management of obesity from childhood to adulthood.

Cephalosporin C 내성과 7-Aminocephalosporanic Acid 감수성을 지닌 균주의 선발 및 특성 (Isolation and Characterization of a Cephalosporin C Resistant and 7-Aminocephalosporanic Acid Sensitive Strain)

  • A strain which showed cephalosporin C resistance and 7-aminocephalosporanic acid sensitivity was isolated from nature. Among the isolates, SS5 was sensitive to cephalosporin C, penicillin G, ampicillin, 7-aminocephalosporanic acid, 6-aminopenicillanic acid, and 7-aminodeacetoxy cephatosporanic acid at concentrations of 1,000 $\mu $g/ml, 2,000 $\mu $g/ml, 3,000 $\mu $g/ml, 30 $\mu $g/ml 100 $\mu $g/ml and 100 $\mu $g/ml, respectively. But SS5 was sensitive at very low concentration of chloramphenicol, kanamycin, neomycin, streptomycin and tetracycline. Since SS5 was sensitive to 7-ACA (30 $\mu $g/ml) and didn't have $\beta $-lactamase activity on the cephalosporin C, SS5 could be useful as an indicator strain for the production of 7-ACA, which is an important precursor for the synthesis of many semisynthetic cephalosporins.

Detection and Quantification of Toxin-Producing Microcystis aeruginosa Strain in Water by NanoGene Assay

  • We demonstrated the quantitative detection of a toxin-producing Microcystis aeruginosa (M. aeruginosa) strain with the laboratory protocol of the NanoGene assay. The NanoGene assay was selected because its laboratory protocol is in the process of being transplanted into a portable system. The mcyD gene of M. aeruginosa was targeted and, as expected, its corresponding fluorescence signal was linearly proportional to the mcyD gene copy number. The sensitivity of the NanoGene assay for this purpose was validated using both dsDNA mcyD gene amplicons and genomic DNAs (gDNA). The limit of detection was determined to be 38 mcyD gene copies per reaction and 9 algal cells/ml water. The specificity of the assay was also demonstrated by the addition of gDNA extracted from environmental algae into the hybridization reaction. Detection of M. aeruginosa was performed in the environmental samples with environmentally relevant sensitivity (${\sim}10^5$ algal cells/ml) and specificity. As expected, M. aeruginosa were not detected in nonspecific environmental algal gDNA over the range of $2{\times}10^0$ to $2{\times}10^7$ algal cells/ml.

무선/무전원 러브파 바이오센서 개발 (Development of wireless/battery-free Love wave biosensor)

  • This paper reports a novel wireless love-wave biosensor on $41^{\circ}$ YX $LiNbO_3$ piezoelectric substrate and $SiO_2$ guiding layer for Immunoglobulin G (IgG) detection by protein binding. Different from the traditional biosensors based on surface acoustic wave (SAW) oscillator structured by delay line/resonators, a 440MHz reflective delay line consists of SPUDTs and three reflectors placed on $41^{\circ}$ YX $LiNbO_3$ in a row was fabricated as the sensor element. Good linearity, reproducibility, and high sensitivity were observed in the IgG concentration range 1~65nM. Unique advantages as high sensitivity, passive and simple measurement system are present over currently available other biosensors.
