Comparison of Fruit Quality among Fruits Set on Various Position within Cluster in 'Niitaka' Pears (화총내 착과위치에 따른 '신고' 배 (Pyrus pyrifolia Nakai) 과실의 품질비교)

  • In order to prepare the technical fundamental in production of premium quality fruits of 'Niitaka' pears, we compared the quality parameters and storability of the fruits that had various fruit set position in a cluster. The fruits set on $4^{th}$ from basal part in fruit cluster had the largest fruit size. The weight of fruits set on the higher position than $4^{th}$ from basal part in fruit cluster showed decreasing tendency. The production rates of marketable fruits which had 500g to 899g in weight were the high in $3-5^{th}$ from basal part in cluster than those in other position. The differences of fruit height and diameter which represent balanced fruit shape also showed less difference in the fruits set on $4-5^{th}$ from basal part in cluster than the fruits set closely on basal part in cluster. The fruits set on $3-4^{th}$ from basal part in cluster showed lower rate in incidence of core breakdown and pithiness disorder during 4 months of cold storage than those on the closer position to basal part in cluster.

Studies for the Effective Diagnosis and Treatment of Bovine Piroplasmosis (젖소의 파이로프라스마증(症)의 효과적인 집단검색과 치료방법에 관한 연구)

  • In the endemic area of bovine piroplasmosis in Chonbuk Province, total of 486 heads of Holstein cow selected from 14 farms randomly were examined the infection status of piroplasma and their blood values. The results obtained were summarized as follows: 1. The etiological agent was only Theileria sergenti and none of the Babesia sp. infection. 2. All of the 486 heads of examined cow were infected with Theileria sergenti (100%). 3. In the initial examination of 11 farms in early summer, the means of blood values were as follows; RBC:$526{\pm}84(10^4/mm^3$), Ht:$27.9{\pm}2.8%$, SP:$7.5{\pm}0.6g/dl$, WBC: $11586{\pm}354/mm^3$, and Fibrinogen: $578.5{\pm}164.1mg/dl$. 4. In the second examinations in slimmer season, the blood values of examined cows had little varieties. But in the winter season, cows housed in the barn, the blood values were greatly increased; RBC: $601{\pm}77(10^4/mm^3)$, (F-value: 37.186**), Ht: $30.5{\pm}1.3%$(F-value: 15.626**) Hb: $12.2{\pm}1.3g/dl$(F-value: 5.899**), SP: $7.4{\pm}0.6g/dl$(F-value: 5.05**). 5. To determine the piroplasma infection in the many herds of cow, the measurement of Ht, Hb, and SP values etc. were more effective and helpful to find the weak cow because the examined process are not only to be easily carry out many samples but also to be done very simple and rapidly. 6. The indications of therapeutic standard values were stablished as follows; the less than 20% in the Ht value, below the 10g/dl in the Hb value, and less than 6g/dl in SP value. When the one among the therapeutic standard values, Ht, Hb, and SP was below the therapeutic standard value, the cow was treated with nutriments. When more than two among them were below the therapeutic standard, the cow was treated with Berenil, at once. In this way, there was no one head which represented clinical signs of piroplasmosis among 486 heads of cow at 14 farms during the year of 1986. 7. In a Korean native cattle infected with Thieleria sergenti seriously, the recovery of blood values was very prolonged by the treatment of Berenil only. But by the transfusion of 300ml blood collected from normal cow, the blood values were recovered rapidly.

A Study of Relation Between Dietary Vitamin A Intake and Serum Vitamin A Levels and Cancer Risk in Korea (한국인의 식이섭취와 암 유발의 관계에 관한 연구 -제 1 보 특히 ${\beta}-Carotene$ 섭취량과 혈청내 수준을 중심으로 -)

  • Even though the anticarcinogenic effect of dietary factors especially beta - carotene has been reported by various investigators, the mechanism of the action of ${\beta}-carotene$ has not yet been identified. We carried out the present study to determine the possibilities of relative cancer risk related to dietary intake of vitamin A ( both ${\beta}-carotene$ and retinol ) and blood levels of vitamin A among Koreans. The subjects were divided into two groups; cancer patients and controls. Blood levels for ${\beta}-carotene$ and retinol were analyzed by alumina column chromatography and colorimetry. Dietary intake was examined by food profile and convenient method for evaluating nutritional status through recalling 10 years of food habits. The results obtained are as follows : 1 ) Calorie, protein, fat, and carbohydrate intakes of cancer patient were lower than those of control. Calorie and carbohydrate intakes showed no significant difference but protein and fat intakes were significantly lower in cancer patients. According to cancer sites, in stomach cancer only fat intake was significantly lower than that of control. In lung and larynx cancer calorie, protein, fat and carbohydrate intakes showed similar trend as in control. 2 ) Vitamin A intake of cancer patient was significantly lower than that of control. It was estimated that 83.6% of total Vitamin A intake were provided by ${\beta}-carotene$ for control and cancer patient respectively. 3 ) The mean intake of dietary ${\beta}-carotene$ in cancer patient was significantly lower than that in control ( $7002\;\mu}g/day$ versus $10326\;{\mu}g/day$ ) According to cancer sites in mean intake of dietary ${\beta}-carotene$ was significantly lower in all but stomach cancer compared with that of control. Lung and larynx cancer showed lowest ${\beta}-carotene$ intake with mean value of $5855{\mu}g/day$ and $5492{\mu}g/day$ respectively. 4 ) The mean intake of dietary retinol in cancer patient was significantly lower than that in control ( $245{\mu}g/day$ versus $338{\mu}g/day$ ), but the difference was not significant. 5 ) The relative risk of all cancers in the first (lowest) to the forth quartile level of ${\beta}-carotene$ consumption such as $0-5999{\mu}g/day$. $6000-8999{\mu}g/day$, $9000-11999{\mu}g/day$/ day and $12000-20000{\mu}g/day$ was 85 : 1.7 : 20 : 1.0. The relative risk of all cancers in the first (lowest) to the forth quartile level of retinol consumption, such as $0-299{\mu}g/day$, $300-599{\mu}g/day$, 600-899${\mu}g/day$, and $900-1200{\mu}g/day$, was 1.14 : 067 : 0.21 : 1.0. 6 ) The various food group consumption of cancer patient were significantly lower than those of control in green leafy vegetables, fruits, sea weeds, milk and cheese and eggs. But the Kimchie consumption in cancer patient was three fold higher than that of control ( $1840\;{\mu}g/day$ versus $562\;{\mu}g/day$ ) and in the stomach cancer, Kimchie consumption was the highest, ( $1890\;{\mu}g/day$) According to cancer sites, the consumption of green leafy vegetables was significantly lower in all but stomach cancer compared to control and other vegetables showed no difference between two. In lung and larynx cancer, green leafy vegetables consumption was lowest ( $6094{\mu}g/day$ $5921{\mu}g/day$) and milk and cheese consumption was also( $5\;{\mu}g/day$ and $11{\mu}g/day$) 7 ) The recovery of ${\beta}-carotene$ from human serum by alumina column chromatography was $94.4{\pm}2.3%$. 8 ) Cancer patients showed significantly lower serum retinol ($56.4{\pm}18.1\:{\mu}g/100ml$ versus $72.2{\pm}21.8\:{\mu}g/100ml$) and ${\beta}-carotene$ ($48.9{\pm}33.8\:{\mu}g/100ml$ versus $72.2{\pm}42.6\:{\mu}g/100ml$) concentrations than in controls. 9 ) But breast cancer patients were not significantly different from controls in their serum retinol and ${\beta}-carotene$ concentrations.

Effects of a Diet Containing Green Tea Powder on the Physicochemical Properties of Eggs (산란계에 녹차 첨가 사료가 계란의 이화학적 성질에 미치는 영향)

  • The effects of green tea on the physicochemical properties of eggs were investigated. One-hundred-and-fifty Isa Brown hens (27 weeks of age) were studied over 10 weeks. Green tea was added to the diet at levels of 4% and 8% w/w. The contents of Ca, K, P, Fe and Mg in eggs increased upon dietary supplementation with green tea powder, and were higher in the yolk than in the white, with the exception of Mg. The increase was greatest for Ca, from 41.0 mg% to 119.8 mg%, in egg white. For Fe, an increase from 3.7 mg% to 12.6 mg% was apparent in egg yolk. When hens consumed the green tea-supplemented diets, average egg weight decreased from 68.8 g to 64.4 g, but the total cholesterol content of egg yolk did not significantly change (control value: 1,899.1 mg% test value: 2,011.3 mg%). In sensory evaluation tests, egg white was similar in terms of astringency and grayness regardless of diet, and egg yolk was yellow-to-orange in color, and rated as fishy or slightly fishy, when green tea diets were administered. Such diets reduced pH values in all of the white, yolk, and white/yolk combination groups. The major fatty acids of egg yolk, constituting more than 90% of total fatty acids, were palmitic, stearic, oleic, and linoleic acid. Green tea diets reduced the saturated fatty acid level from 32.0% to 27.4% of total fatty acids, and unsaturated fatty acid levels increased from 68.0% to 72.6% in egg yolk.

Isolation and Characterization of Lectin from Aloe vera (Aloe vera 중의 렉틴의 분리 및 특성)

  • The lectins from mucilaginous jelly and green epidermis of Aloe vera were isolated by gel and affinity chromatography. The molecular weights of the lectins were determined by SDS-PAGE. The molecular weights of the lectins from mucilaginous jelly isolated by Sephadex G-100 were 58.7 kD and 33.3 kD, and that isolated by acid-treated Sepharose 4B was 176.4 kD. The molecular weights of the lectins from epidermis isolated by Sephadex G-100 were 221.1, 54.0 and 32.5 kD respectively. And that isolated by acid-treated Sepharose 4B was 222.0 and 158.0 kD. The agglutinating activity of lectin from jelly was inhibited by D-galactose, lactose and D-galactosamine, but that from epidermis was not inhibited by lactose. The activity was stable at the pH range of $7.0{\sim}9.0$ and at the temperature $0{\sim}60^{\circ}C$.

Establishment of a Nondestructive Analysis Method for Lignan Content in Sesame using Near Infrared Reflectance Spectroscopy (근적외선분광(NIRS)을 이용한 참깨의 lignan 함량 비파괴 분석 방법 확립)

  • Sesamin and sesamolin are major lignan components with a wide range of potential biological activities of sesame seeds. Near infrared reflectance spectroscopy (NIRS) is a rapid and non-destructive analysis method widely used for the quantitative determination of major components in many agricultural products. This study was conducted to develop a screening method to determine the lignan contents for sesame breeding. Sesamin and sesamolin contents of 482 sesame samples ranged from 0.03-14.40 mg/g and 0.10-3.79 mg/g with an average of 4.93 mg/g and 1.74 mg/g, respectively. Each sample was scanned using NIRS and calculated for the calibration and validation equations. The optimal performance calibration model was obtained from the original spectra using partial least squares (PLS). The coefficient of determination in calibration (R2) and standard error of calibration (SEC) were 0.963 and 0.861 for sesamin and 0.875 and 0.292 for sesamolin, respectively. Cross-validation results of the NIRS equation showed an R2 of 0.889 in the prediction for sesamin and 0.781 for sesamolin and a standard error of cross-validation (SECV) of 1.163 for sesamin and 0.417 for sesamolin. The results showed that the NIRS equation for sesamin and sesamolin could be effective in selecting high lignan sesame lines in early generations of sesame breeding.

Effect of Storage Temperature on the Quality of Tomato (저장 온도에 따른 토마토의 품질 변화)

  • Tomato were stored at different temperatures($10^{\circ}C$, $20^{\circ}C$, room temperature and $30^{\circ}C$) for 5 days. During the storage period, Brix, pH, color, texture, vitamin C, lycopene were analyzed. Brix and pH had a little change. Texture force of tomato decreased with storage time and we could see a softening for tomato stored at $30^{\circ}C$ for 1 day. Addtional, the $L^*$(lightness) and $b^*$(yellowness) decreased and $a^*$(redness) increased with storage time. Addtionally, content of vitamin C increased up to 9.08 mg/100 g~17.82 mg/100 g after 5 days storage according storage temperature, whereas content of lycopene increased up to 3.81 mg/kg~34.56 mg/kg after 5 days storage according storage temperature. Optimal mature temperature for tomato was room temperature.

Effect of Rice Fermented using Poria cocos (a Wood-decay Fungus) Mycelium on Fermentation of Doenjang (Soybean Paste) (복령(Poria cocos) 균사체 발효쌀의 첨가가 된장의 품질에 미치는 영향)

  • We investigated changes in doenjang quality upon addition of rice fermented with Poria cocos mycelium. Sensory evaluation showed that rice addition to 15% (w/w) was optimal. The content of free amino acids was 1,899.17 mg% in doenjang fermented for 90 days with P. cocos mycelium. The rice contained seven essential amino acids: leucine, valine, phenylalanine, isoleucine, lysine, threonine, and methionine. The levels of essential amino acids increased after fermentation with P. cocos mycelium. Vitamin A (7.47 RE/100 g), Vitamin D (0.45 mg/100 g), and Vitamin E (5.73 mg/100 g) were detected in the experimental preparation but not in the control. In terms of electron-donating ability, the highest scores were 73.8% (in a water extract) and 76% (in an ethanolic extract) of doenjang fermented with P. cocos mycelium. Nitrite-scavenging ability was higher in the ethanolic extract than in the water extract of rice fermented with P. cocos mycelium.

Characteristics of Manure and Estimation of Nutrient and Pollutant of Holstein Dairy Cattle (홀스타인 젖소 분뇨의 특성과 비료성분 및 오염물질 부하량 추정)

  • This study was conducted to determine fertilizer nutrient and pollutant production of Holstein dairy cattle by estimating manure characteristics. The moisture content of feces was 83.9% and 95.1% for urine. The pH of feces and urine were in the ranges of 7.0~7.4 and 7.5~7.8, respectively. The average BOD5, COD, SS, T-N, T-P concentrations of the dairy feces were 18,294, 52,765, 102,889, 2,575, and 457mg/ℓ, respectively. Dairy urine showed lower levels of BOD5(5,455mg/ℓ), COD(8,089mg/ℓ), SS(593mg/ℓ), T-N(3,401mg/l), and T-P(13mg/ℓ) than feces. The total daily produced pollutant amounts of a dairy cow were 924.1g(Milking cow), 538.8g(Dry cow), 284.4g(Heifer) of BOD5, 2,336.5g (Milking cow), 1,651.8g(Dry cow), 734.1g(Heifer) of COD and 4,210.1g(Milking cow), 2,417.1g(Dry cow), 1,629.1g(Heifer) of SS and 194.8g(Milking cow), 96.4g(Dry cow), 58.3g(Heifer) of T-N and 24.0g(Milking cow), 10.2g(Dry cow), 6.1g(Heifer) of T-P. The calculated amount of pollutants produced by a 450kg dairy cow for one year were 181.3kg of BOD5, 492.5kg of COD, 899.9kg of SS, 36.0kg of T-N and 4.1kg of T-P. The total yearly estimated pollutant production from all head(497,261) of dairy cattle in Korea is 90,149 tons of BOD5, 244,890 tons of COD, 447,491 tons of SS, 17,898 tons of T-N and 2,008 tons of T-P. The fertilizer nutrient concentrations of dairy feces was 0.26% N, 0.1% P2O5 and 0.14% K2O. Urine was found to contain 0.34% N, 0.003% of P2O5 and 0.31% K2O. The total daily fertilizer nutrients produced by dairy cattle were 197.4g (Milking cow), 97.4g(Dry cow), and 57.9g(Heifer) of Nitrogen, 54.2g(Milking cow), 22.2g(Dry cow), and 14.2g(Heifer) of P2O5 and 110.8g(Milking cow), 80.4g (Dry cow), and 39.5g(Heifer) of K2O. The total yearly estimated fertilizer nutrient produced by a 450kg dairy animal is 36.2kg of N, 8.8kg of P2O5, 24.6kg of K2O. The estimated yearly fertilizer nutrient production from all dairy cattle in Korea is 18,000 tons of N, 4,397 tons of P2O5, 12,206 tons of K2O. Dairy manure contains useful trace minerals for crops, such as CaO and MgO, which are contained in similar levels to commercial compost being sold in the domestic market. Concentrations of harmful trace minerals, such as As, Cd, Hg, Pb, Cr, Cu, Ni, Zn, met the Korea compost standard regulations, with some of these minerals being in undetected amounts.

Characteristics and Action Pattern of Protease from Bacillus Subtilis Globigii CCKS-118 in Korean Traditional Soy Sauce (한국재래간장으로 부터 분리한 Bacillus subtilis globigii CCKS-118이 생성하는 pretense의 특성 및 작용양상)

  • The production of bacterial protease and its characteristics were investigated with Bacillus subtilis globigii CCKS-118 which was isolated from Korean traditional soy sauce. The optimum culture condition of the strain for the production of alkaline protease was as follow : 2% soluble starch, 0.2% yeast extract, 0.1% $(NH_4)_2SO_4$, 0.2% $MgSO_4$, pH 7.5, $35^{\circ}C$ and 20h rs. The optimum pH and temperature for the enzyme action of alkaline protease producing Bacillus subtilis globigii CCKS-118 were pH 9.0 and $50^{\circ}C$, respectively. The enzyme was relatively stable at $pH\;6.0{\sim}9.0$ and at temperature below $40^{\circ}C$. The activity of the enzyme was inhibited by $Hg^{2+}$ whereas $Cu^{2+}$ gave rather activating effects on the enzyme activity. The enzyme was inhibited by phenylmethane-sulfonyl fluoride indicating serine pretense metal ion group are required for the enzyme activity. Km value was $1.242{\times}10^{-4}M$, $V_{max}$ value was $25.99\;{\mu}g/min$. This enzyme hydrolyzed casein more rapidly than the hemoglobin.
