김치 유산균 배양물의 부상용 배합사료내 첨가시 넙치(Paralichthys olivaceus) 치어의 성장, 체조성 및 면역성 반응에 미치는 효과 (Effects of the Inclusion of Kimchi Lactic Acid Bacterial Culture in Extruded Pellets on the Growth, Body Composition and Immune Response of Juvenile Olive Flounder Paralichthys olivaceus)

  • This study was performed to determine effects of the supplementation of Kimchi lactic acid bacterial culture in extruded pellets (EP) on the growth, body composition, blood chemistry and immune response of olive flounder Paralichthys olivaceus. Four hundred eighty individuals averaging 16.1 g were randomly distributed into 12, 180 L flow-through tanks (forty fish per tank). Four concentrations of Kimchi lactic acid bacterial culture (KL) were prepared: Control (0%), 0.1%, 0.2% and 0.5%. Three concentrations (0.1%, 0.2% and 0.5%) of Kimchi lactic acid bacterial culture were each diluted to 10% of EP weight and then fully absorbed by EP for 10 minutes. Each diet was fed to triplicate groups of fish. Fish were hand-fed to apparent satiation twice a day for 8 weeks. At the end of the 8-week feeding trial, the plasma lysozyme and bacterial activities of fish were determined. In addition, the cumulative mortality of fish was monitored for 8 days after their artificial infection with Edwardsiella tarda. The weight gain, specific growth rate, feed efficiency ratio, protein efficiency ratio, protein retention, hepatosomatic index and condition factor of fish were not affected by dietary supplementation with KL. None of the proximate composition, plasma parameters, lysozyme or bactericidal activities of fish was affected by dietary supplementation with KL. However, the cumulative mortalities of fish fed EP containing 0.1% and 0.5% Kimchi lactic acid bacterial culture were relatively low compared to that of fish fed the control diet. In conclusion, dietary supplementation with KL did not effectively improve growth, feed utilization, body composition, plasma chemistry, lysozyme, bactericidal activities or immune response of olive flounder after E. tarda infection under these experimental conditions.

화피(樺皮)의 항염(抗炎) 효과(效果)에 관(關)한 연구(硏究) (Anti-inflammatory Effects of an Ethanolic Extract form Betula Platyphylla)

  • Objectives : Betula Platyphylla(BP) is a traditional analgesic, anti-fungal, anti-inflammatory herb used in Chinese 1medicine. However, no information is available to explain its action. In this study. we investigated the anti-inflammatory 1effects of BP to elutidate the molecular pharmacological activity in the ethanol extract of BP(BPE). Methods : We performed WTS assay in lipopolysaccharide (LPS) stimulated RAW264.7 macrophages with BPE. Nitrite was measured by Griess assay, prostaglandin E2 (PGE2) by enzyme-linked immunosorbent assay (ELISA) in LPS induced RAW264.7 macrophages with BPE. Inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX-2) were determined by Western blot. Activation of nuclear factor-kappaB (NF-kB) was measured by electrophoretic mobility shift assay (EMSA). Results : BPE significantly suppressed production of nitric oxide (NO) and PGE2 in LPS-induced RAW264.7 macrophages in a dose-dependent manner. The maximal inhibition rate of NO and PGE2 production by BPE was ca. 88.8% and 93% at the concentration of $100{\mu}g/ml$ (non-cytotoxic concentration), respectively. BPE also decreased iNOS protein and COX-2 protein in LPS-induced RAW264.7 macrophages. EMSA demonstrated that BPE inhibited the DNA binding activity of the NF-kB. Conclusions : These results suggest that BPE inhibits NF-${\kappa}B$-mediated gene expression and downregulates inflammatory mediator production in RAW264.7 macrophages.

  • Components of extracellular matrix and the matrix-degrading enzymes are some of the key regulators of tumor metastasis and angiogenesis. Hyaluronic acid (HA), a matrix glycosaminoglycan, is known to promote tumor adhesion and migration, and its small fragments are angiogenic. Until now, we have compared levels of hyaluronidase, an enzyme that degrade HA, in normal adult prostate, benign prostate hyperplasia and prostate cancer tissues and in conditioned media from epithelial explant cultures, using a substrate (HA)-gel assay and ELISA-like assay (Kim et al., unpublished results). The present review described an overall characterization of hyaluronidases and its application to human diseases. The hyaluronidases are a family of enzymes that have, until recently, deed thorough explication. The substrate for these enzymes, hyaluronan, is becoming increasingly important, recognized now as a major participant in basic processes such as cell motility, wound healing, embryogenesis, and implicated in cancer progression. And in those lower life forms that torment human beings, hyaluronidase is associated with mechanisms of entry and spread, e.g. as a virulence factor for bacteria, for tissue dissection in gas gangrene, as a means of treponema spread in syphilis, and for penetration of skin and gut by nematode parasites. Hyaluronidase also comprises a component of the venom of a wide variety of organisms, including bees, wasps, hornets, spiders, scorpions, sh, snakes and lizards. Of particular interest is the homology between some of these venom hyaluronidases and the enzyme found in the plasma membrane of mammalian spermatozoa, attesting to the ancient nature of the conserved sequence, a 36% identity in a 300 amino acid stretch of the enzyme protein. Clearly, hyaluronidase is of biological interest, being involved in the pathophysiology of so many important' human disorders. Greater effort should be made in studying this family of enzymes that have, until recently, been overlooked. Also, oriental medical application of the hyaluronidase will be discussed with respect to inhibition and suppression of inflammation and malignacy.

Wisconsin의 Pinus resinosa 조림지(造林地) 생태계(生態系) 내(內)의 양료분포(養料分布) 및 순환(循環) (Distribution and Cycling of Elements in a Pinus resinosa Plantation-Ecosystem, Wisconsin)

  • Typic Udipsamment 토양(土壤)에 생장하고 있는 red pine (Pinus radiata) 조림지의 biogeochemistry가 Wisconsin의 central sand plains에서 조사(調査)되었다. Biomass와 net primary production은 각각 106Mg/ha 및 110Mg/ha/yr이었다. 식생(植生) 내(內)의 양료 농도는 2-3년생(年生) 침엽(針葉)에서 가장 많았고 그 다음은 당년생(當年生) 침엽(針葉), 당년생(當年生) 초지(梢枝), 수피, 생지(生枝), 고사지(枯死枝), 수간생성물, 수간 순(順)이었다. 조사된 수체(樹體) 성분에 있어서 당년생(當年生) 초지(梢枝)를 제외하고는 양료의 농도가 상부 3분지 1 부위(部位)에서 가장 높았고, 하부(下部) 3분지 1 부위(部位)에서 가장 낮았다. 식위(植位) 내(內)의 전체 양료는 N($222kg\;ha^{-1}$), Ca(217), K(85), Mg(38), S(31), P 순(順)이었다. 생태계 내(內)의 토양은 가용(可溶) P.K, Ca, Mg, S의 60-80%를 함유하고 있었다. N과 S의 약간의 급원(給源)이 강수(降水)인 반면에 Ca, Mg, P, K의 대부분은 토양풍화(土壤風化)로서 공급되었다. 조사양료의 용탈량(溶脫量)은 7 kg ha/yr미만이지만 S의 경우는 12kg ha/yr 이었다. Throughfal에 의(依)해서 N, K, S는 대부분 임상(林床)으로 순환되지만 Ca, Mg, P의 대부분은 litterfall에 의(依)해 돌아오는 것으로 사료된다. 엽(葉)의 양분용탈 때문에 throughfall의 양료의 함량은 강수의 함량보다 많았다. 강수(降水) 내(內)의 ammonium량(量)은 세근(細根)에 위한 흡수(吸水)때문에 토양상부(土壤上部) 7.5cm내(內)에서 3 가지 요인에 의하여 감소되었다.

휴대폰 디자인에 있어서 지배성 변화 측정 (Measuring Changes of Dominance in Designs of Mobile Phones)

  • 본 연구는 디자인요소의 변화과정을 통해 제품디자인요소(product design factors)의 지배력 정도(dominant degree)를 실증적으로 분석하고 있다. 선행연구에서는 지배적 디자인에 대한 측정방법은 구체적으로 제시되었지만 장기간에 걸쳐 데이터를 확보하지 못한 한계점으로 시간의 흐름에 따른 지배적 디자인(dominant design)의 실증연구가 제시되지 않았다. 따라서 본 연구에서는 연도별 디자인 변화 데이터를 확보한 후 휴대폰의 디자인 지배력(dominance)과 지배구조(structure change)가 어떻게 변했는지를 측정해 보았다. 측정결과, 휴대폰 디자인요소 중 바디형태(body form)와 모서리(edge)는 지배력(dominance)이 점점 증가하여 각각 직사각형으로 라운드 모서리로 집중화(convergence)되는 경향인 지배적 디자인요소를 가지고 있었고, 버튼방식(button type), 개폐방식(open type), DMB여부, 영상통화여부, 컬러조합, 바디컬러는 지배력이 높았다가 감소되면서 디자인수준(design levels)이 분산화(divergence) 되는 경향을 가지고 있었다. 이렇게 제품디자인요소의 지배력 정도를 측정함으로써 마케팅 관리자나 디자이너들에게 급변하는 시장 상황에서 제품전략을 수립하는데 도움이 될 것이다. 본 연구는 지배적 디자인에 대한 실증연구를 통해 신제품 개발시 지배적 디자인 등장에 대비한 성공적인 제품 개발의 가이드라인을 제공하는데 도움이 될 수 있다.

Water relations of plants under environmental stresses: role of aquaporins

  • Effects of low temperature ($8^{\circ}C$) on the hydraulic conductivity of young roots of a chilling-sensitive (cucumber; Cucumis sativus L.) and a chilling-resistant (figleaf gourd; Cucurbita ficifolia Bouche) crop have been measured at the levels of whole root systems (root hydraulic conductivity, $Lp_r$) and of individual cortical cells (cell hydraulic conductivity, Lp). In figleaf gourd, there was a reduction only in hydrostatic $Lp_r$ but not in osmotic $Lp_r$ suggesting that the activity of water channels was not much affected by low root temperature (LRT)treatment in this species. Changes in cell Lp in response to chilling and recovery were similar asroot level, although they were more intense at the root level. Roots of figleaf gourd recovered better from LRT treatment than those of cucumber. In figleaf gourd, recovery (both at the root and cell level) often resulted in Lp and $Lp_r$ values which were even bigger than the original, i.e. there was an overshoot in hydraulic conductivity. These effects were larger forosmotic (representing the cell-to-cell passage of water) than for hydrostatic $Lp_r$. After a short term (1 d) exposure to $8\;^{\circ}C$ followed by 1 d at $20\;^{\circ}C$, hydrostatic $Lp_r$ of cucumber nearly recovered and that of figleaf gourd still remained higher due to the overshoot. On the contrary, osmotic $Lp_r$ and cell Lp in both species remained high by a factor of 3 as compared to the control, possibly due to an increased activity of water channels. After pre-conditioning of roots at LRT, increased hydraulic conductivitywas completely inhibited by $HgCl_2$ at both the root and cell levels. Different from figleaf gourd, recovery from chilling was not complete in cucumber after longer exposure to LRT. It is concluded that at LRT, both changes in the activity of aquaporins and alterations of root anatomy determine the water uptake in both species. To better understand the aquaporin function in plants under various stress conditions, we examined the transgenic Arabidopsisand tobacco plants that constitutively overexpress ArabidopsisPIP1;4 or PIP2;5 under various abiotic stress conditions. No significant differences in growth rates were found between the transgenic and wild-type plants under favorable growth conditions. By contrast, overexpression of PIP1;4 or PIP2;5 had a negative effect on seed germination and seedling growth under drought stress, whereas it had a positive effect under cold stress and no effect under salt stress. Measurement of water transport by cell pressure probe revealed that these observed phenotypes under different stress conditions were closely correlated with the ability of water transport by each aquaporin in the transgenic plants. Together, our results demonstrate that PIP-type aquaporins play roles in seed germination, seedling growth, and stress response of Arabidopsis and tobacco plants under various stress conditions, and emphasize the importance of a single aquaporin-mediated water transport in these cellular processes.

동아시아의 30°N부근에서 여름철 태풍 강도변화 (Variations of the Summertime Tropical Cyclone Intensity near 30°N in East Asia)

  • In this paper, changes in the intensity (e.g., central pressure and maximum sustained wind speed) of Tropical Cyclone (TC) in summer in the regions located at $30^{\circ}N$ in East Asia from 1988 to 1991 were found. The intensity of TC from 1991 to 2007 was much higher than that of TC from 1965 to 1988. The reason for this was that the frequency of TCs passing China from 1991 to 2007 was much lower than that of TCs from 1965-1988 because a northeasterly wind caused by high-pressure circulation in East Asia got severer along the East Asian coast. Instead, TCs moved from the eastern region of the Tropical West Pacific to Korea and Japan mainly after passing the East China Sea due to the low-pressure circulation strengthened in the subtropical waters of East Asia. In addition, low Vertical Wind Shear (VWS) was created along the mid-latitude regions of East Asia and the main path of TCs from 1991 to 2007. Most of the regions in the Northwestern Pacific showed higher Sea Surface Temperature (SST) from 1991 to 2007, and had a good environment where TCs were able to maintain a higher intensity on the mid-latitude. In particular, a low sensible heat flux occurred due to high snow depth in East Asia in the spring of 1991 to 2007. Accordingly, the lower layer of East Asia showed high-pressure circulation, and the sea surrounding East Asia showed low-pressure circulation. Thus, the typical west-high, east-low pattern of winter atmospheric pressure was shown. The possibility of snowfall in East Asia in spring to be used as a factor for predicting the summer intensity of TC in the mid-latitude regions of East Asia was insinuated. The characteristics of TC in a low-latitude region were the same in Korea. The latest intensity of TCs got higher, and the landing location of TCs gradually changed from the west coast to the south coast.

Estimation of Pork Quality Traits Using Exsanguination Blood and Postmortem Muscle Metabolites

  • The current study was designed to estimate the pork quality traits using metabolites from exsanguination blood and postmortem muscle simultaneously under the Korean standard pre- and post-slaughter conditions. A total of 111 Yorkshire (pure breed and castrated male) pigs were evaluated under the Korean standard conditions. Measurements were taken of the levels of blood glucose and lactate at exsanguination, and muscle glycogen and lactate content at 45 min and 24 h postmortem. Certain pork quality traits were also evaluated. Correlation analysis and multiple regression analysis including stepwise regression were performed. Exsanguination blood glucose and lactate levels were positively correlated with each other, negatively related to postmortem muscle glycogen content and positively associated with postmortem muscle lactate content. A rapid and extended postmortem glycolysis was associated with high levels of blood glucose and lactate, with high muscle lactate content, and with low muscle glycogen content during postmortem. In addition, these were also correlated with paler meat color and reduced water holding capacity. The results of multiple regression analyses also showed that metabolites in exsanguination blood and postmortem muscle explained variations in pork quality traits. Especially, levels of blood glucose and lactate and content of muscle glycogen at early postmortem were significantly associated with an elevated early glycolytic rate. Furthermore, muscle lactate content at 24 h postmortem alone accounted for a considerable portion of the variation in pork quality traits. Based on these results, the current study confirmed that the main factor influencing pork quality traits is the ultimate lactate content in muscle via postmortem glycolysis, and that levels of blood glucose and lactate at exsanguination and contents of muscle glycogen and lactate at postmortem can explain a large portion of the variation in pork quality even under the standard slaughter conditions.

무안경식 Interactive 3D Display: 시청거리, 시청방위, 협응동작이 시각피로에 미치는 영향 (Glasses-free Interactive 3D Display: The Effects of Viewing Distance, Orientation and Manual Interaction on Visual Fatigue)

  • 본 연구에서는 interactive 3D(i3d) 시스템을 사용하는 시청 환경에서의 시각 피로를 측정하고, 시각 피로에 영향을 주는 중요한 변인이 무엇인지를 연구하였다. i3D는 무안경식 디스플레이로 사용자가 화면에 제시되는 자극에 대해 손을 사용한 간단한 상호작용을 지원한다. 실험 1에서는 i3D 환경에서 시각피로를 측정할 수 있는 설문문항을 개발하기 위해, 개방형 설문을 실시한 후 사용자의 응답을 다시 설문문항으로 사용하여 요인분석을 통해 문항의 타당성을 검증하였다. 실험 2에서는 실험 1에서 도출한 피로측정 설문지를 사용하여 다양한 시청환경에서 피로를 측정하였다. 협응동작(유/무), 시청거리(1/2/4m), 시청방위($0/28/56^{\circ}$)의 세 변인을 사용하여 피험자내 요인설계를 통해 각 조건에서의 시각피로를 측정하였다. 그 결과, 시청거리가 멀어질수록 시각피로가 줄어드는 결과를 획득하였으며, 시청방위와 협응동작은 시각피로에 영향을 미치지 않는 것을 확인하였다. 실험조건 중 극단적인 시청환경 (예를 들어, 시청거리 1m, 시청방위 $56^{\circ}$)인 조건에서는 피로감이 줄어드는 결과를 획득하였으나 이는 무안경식 디스플레이의 한계로 인해 입체감이 사라져 참가자들이 이중상을 경험한 결과인 것으로 해석하였다. 본 연구의 제한점과 후속 연구에 대한 제안을 종합논의에 제시하였다.

시계열 강수량 공간화를 위한 SCEM-UA 기반의 PRISM 매개변수 최적화 (Optimization of PRISM parameters using the SCEM-UA algorithm for gridded daily time series precipitation)

  • 상세한 수문기상자료 구축은 수자원 활용 계획을 수립하고 대응하는 데 있어 필수적인 요소로 인식되고 있다. 기후, 수문, 지리 및 환경 등의 다양한 영역에서 신뢰할 수 있는 공간적 강우량의 요구가 증가하고 있다. 지형의 약 70%가 산악 지형인 우리나라의 경우 기존의 면적가중 및 수치내삽 방법은 고도가 높은 지역의 기상인자를 추정하는 데 한계가 있는 것으로 평가 되고 있다. PRISM 기법은 일반적인 공간보간 방법에 부족한 지형적 특성을 반영한 격자형태의 기상인자를 생산할 수 있는 유용한 모형으로서 본 연구에서는 SCEM-UA 기법을 기반으로 일단위 시계열에서의 PRISM 모형을 최적화 수행하였으며, 그 결과 최소영향반경은 9.10 km, 최대는 34.99 km로 산정되었으며, 해양가중치에서 고도기준은 681.03 m, 최소 및 최대거리는 각각 9.85 km, 38.05 km가 추정되었다. 거리가중치계수는 약 0.87로 산정되어 PRISM 모의 결과가 거리에 매우 민감하다는 것을 확인하였다. 또한, 다양한 통계적 검정을 통해 생산된 강수 시계열이 관측시계열과 비교하여 유사한 특성을 갖는 것을 확인하였다.
