Diurnal and Seasonal Variation of Chlorophyll Fluorescence from Korean Fir Plants on Mt. Halla (한라산 구상나무 잎의 엽록소형광의 일변화와 계절적 변화)

  • Chlorophyll fluorescence of needles of Korean fir (Abies koreana) plants and environmental factors of their natural habitat were investigated in order to obtain the information for environmental adaptation and conservation of Korean fir plants. The photochemical efficiency of photosystem II, Fv/Fm, of Korean fir needles was significantly low (0.19-0.36) in the winter, whereas it was high (0.8-0.86) in the summer. The Fv/Fm value of the winter was slightly higher at mid-day than at dawn, suggesting that mid-day environmental conditions of the winter were favorable on needles of Korean fir plants. In contrast, the mid-day Fv/Fm value of the summer maintained high (around 0.8). It indicates that mid-day environmental conditions of the summer did not induce photodamage, although it caused a slight decrease in the Fv/Fm values. The non-photochemical fluorescence quenching (NPQ) of Korean fir needles was very low (0-0.01) all through the day in the winter. However, it was high (0.76) at mid-day in the summer. These results suggest that Korean fir plants have a system for the protection of PS II from mid-day environmental stresses of the summer. In the winter, the Fv/Fm values were positively correlated with temperature, light intensity and relative humidity, although NPQ values showed no correlation with any of them. In the summer, the Fv/Fm values were positively correlated with relative humidity but negatively correlated with temperature and light intensity. These results indicate that increase of tempera-ture, light intensity and relative humidity lead to promotion of the photochemical efficiency in the winter and high temperature and light intensity may cause photoinhibition in the summer.

Effect of different water levels on the photosynthetic pigments of crops

  • An excess soil water condition is one of the major problems for the field crops growing in paddy fields because of their poor drainage and less availability for oxygen uptake which leads to adversely affect the photosynthesis. Therefore, the current study was undertaken with aim to investigate the effects groundwater level on the photosynthetic response of soy bean (Urum), red bean (Arari), sesame (Geonbaek), perilla (Dayu) after the transplanting to the lysimeter to investigate the plant-water relation and their effect on photosynthesis. The chlorophyll content of the crops according to the humid conditions of the soy bean, sesame and the perilla was found to be 5%, 6.89 % and 13.7% higher than that of the groundwater treated at 40cm, respectively. On the other hand, the chlorophyll content of adzuki bean decreased 6.6% from the groundwater level of 40cm, and the sorghum decreased by 5.7%. As a result of investigating the Fv / Fm value of groundwater, the adzuki bean at 20cm above groundwater was lower than that of groundwater by 40cm immediately before flowering. The Fv / Fm value of soy bean and sesame at 40cm above groundwater were lowered by flowering under groundwater 20 cm and Fv / Fm value of sorghum is increased at 40 cm treatment immediately before flowering while the Fv / Fm values of the perilla had no significant difference in comparison to those at 20 cm and 40 cm of groundwater. In the case of chlorophyll fluorescence reaction, it is known that the when the absolute value is closer to 0.82, the stress is considered less. As a result of comparing the numerical values of the crops, it was found that the sorghum was the most stressed followed by adzuki bean and sesame, while the soy beans and perilla was found on the average, as they received less stress.

Effect of Irrigation Starting Point of Soil on Chlorophyll Fluorescence, Stem Sap Flux Relative Rate and Leaf Temperature of Cucumber in Greenhouse (시설 토양 오이재배에서 관수개시점 처리가 광합성 형광반응, 줄기수액흐름 및 엽온에 미치는 영향)

  • This experiment was conducted to investigate the effect on chlorophyll fluorescence, stem sap flux relative rate (SFRR) and leaf temperature of cucumber when irrigation is controlled using a soil moisture tensiometer. Cucumber (Cucumis sativus L.) 'Chungchun' was irrigated of 10-10-20 kPa and 20-10-10 kPa by soil starting point of irrigation at each growth stage. At the 66 days after treatment (DAT) of 736 to 854 W·m-2 and above 32℃, chlorophyll fluorescence variables (Fo, Fm, Fv/Fm) values showed significantly different between treatments. The Fo and Fv/Fm value in the daytime (10:30 am to 6:00 pm) at 66 DAT was higher in 20-10-10 kPa treatment than in 10-10-20 kPa treatment. The Fv/Fm value decreased when the leaf temperature was increased. There was no difference in leaf growth (length, width and area) at 28 and 66 DAT, but the chlorophyll content (SPAD value) was significantly higher in 20-10-10kPa treatment. SFRR and leaf temperature increased with light intensity and temperature increased. In both treatments, the SFRR started to increase sharply between 8 am and 9 am when the solar radiation is 170 W·m-2 or higher. The soil temperature of the treatments decreased after irrigation, that showed 31.0℃ at 10-10-20kPa and 28.5℃ at 20-10-10kPa on July 5 (820W·m-2 at 1 pm). However, there was no difference in SFRR, leaf temperature, temperature difference (leaf temperature - air temperature) and VPD between treatments. SFRR was significantly positive correlate with the leaf temperature (p < 0.01, r = 0.770). The SFRR and leaf temperature showed positive significant correlation with solar radiation, temperature, soil temperature, soil moisture content and VPD. There was a negative significant correlation with relative humidity and temperature difference.

The Effects of Acidic Electrolytic Water on the Development of Barley Chloroplast (산성 전해수가 보리(Hordeum vulgae L.) 엽록체의 발달에 미치는 영향)

  • To investigate the effects of strong acidic electrolytic water on the chloroplast, barley leaves were treated with strong acidic electrolytic water(pH 2.5). And to investigate the effects of weak acidic electrolytic water on the chloroplast development, etiolated barley leaves were treated with weak acidic electrolytic water(pH 6.5) during greening period. Chl contents, Fo, Fv, and Chl fluorescence quenching coefficient in barley leaves were measured during and after treatment of acidic electrolytic water. The following results were obtained. Chl a, b, and carotenoid were decreased with treatment of strong acidic electrolytic water. Chl contents were significantly decreased than that of the control after 5 min. These results provide evidence that the strong acidic electrolytic water dissimilate the Chl and so that the value of Fo was slightly increased. The strong acidic electrolytic water damaged PS II because Fo was increased and Fv, Fm, and Fv/Fm ratio were decreased. qP, qNP and qE were decreased. On the other hand qI was increased than that of the control. But Chl content and Chl fluorescence patterns were a little changed as the pH increase over 4.0 Chl a, b, and carotenoid were increased with treatment of weak acidic electrolytic water during greening period. Chl contents were significantly increased than that of control after 12 hours greening. These results provide evidence that the weak acidic electrolytic water accelerated the chlorophyll synthesis. And the weak acidic electrolytic water accelerated PS II development because Fv, Fm, qP and Fv/Fm ratio were increased than that of the control.

The Effect of NaCl on the Greening of Etiolated Leaves of Barely (Hordeum vulgare L.) Seedings (NaCl이 황백화된 보리(Hordeum vulgare L.) 잎의 녹화에 미치는 영향)

  • The effects on photosynthesis of NaCl(0, 0.2, 0.4, 0.6, 0.8 or 1.0 M) were examined in etiolated barley seedlings. Chlorophyll(Chl) a, Chl b and carotenoid contents, Chl a fluorescence and quenching coefficients of Chl fluorescence have been determined in the primary leaves of etiolated barley seedlings cultivated under low light(60 $\mu$$m^{-2}\;s^{-1}$). Chl a, b, and carotenoid contents were decreased remarkably in comparison with the control at 0.4 M NaCl. However, the value of Fo and Fv were decreased at 0.6 M NaCl and the ratio of Fv/Fm were deceased at 1.0 M NaCl. Chlorophyll synthesis was seriously inhibited from 0.4 M NaCl, and the photosynthetic electron transport system was inhibited from 0.6 M NaCl. Quantum of photosystem II reaction center was inhibited at 1.0 M NaCl. The effects of NaCl on the Chl content were raised in a 6 hrs, but the effects of NaCl on the value of Fo, Fv and Fv/Fm were raised in 30 hrs. The value of qP was decreased in comparison with the control at all concentrations, but there was a small change in the value qE. These results provide evidence that NaCl inhibited effects of various concentration of NaCl were inhibited quinone redox, however, proton gradient between thylakoid membranes was little damaged.

Effects of Petroleum Spray Oil on Photosynthesis Characteristics in Citrus Leaves (Petroleum Spray Oil 살포가 감귤 잎의 광합성관련 특성에 미치는 영향)

  • Recently, petroleum spray oil(PSO) has been used to control key pests in integrated pest management (IPM) of citrus and other orchards in Australia and USA. In order to clarify the influences of a newly developed PSO (D-C Tron $Plus^{(R)}$) on citrus leaves, 0.33% or 1.0% of PSO were sprayed to potted 4-year-old citrus trees under some kinds of condition, and then the changes of photosynthesis, transpiration, stomatal conductance and chlorophyll fluorescence(Fv/Fm) were determined. When sprayed with 1.0% PSO, the photosynthetic rate, transpiration and stomatal conductance of citrus leaves were decreased by 20%, and then recovered in 20 days after treatment (DAT), while there were little influences by the spray of 0.33% PSO. The value of Fv/Fm decreased more under the $34/24^{\circ}C$ temperature condition than that of under the $30/20^{\circ}C$ and $28/16^{\circ}C$ condition. The high temperature ($50^{\circ}C$ for 10 hours)-treated trees sprayed with PSO 1.0% or PSO 1.0% plus dithianon 1/2000 dilution showed not only the increase of rate in dropped leaf but also the reduced photosynthesis and Fv/Fm compared with $30/20^{\circ}C$ temperature-treated ones. From the results of this study, the spray of 1.0% PSO can inhibit the physiological activities in citrus leaf, particularly under high temperature condition after spray or the mixing-spray with a fungicide (dithianon WP, 75%).

Initial Growth Responses of Four Woody Plants for Indoor Landscaping according to Irrigation Frequency (관수주기에 따른 실내녹화용 목본식물 4종의 초기 생육반응)

  • This study was carried out to investigate growth characteristics of woody plants that are widely used indoors in accordance with irrigation frequency and to find the optimum irrigation conditions for plants that help to improve the indoor environment. Four woody plants used in this study included Ardisia pusilla, Clusia rosea, Fatsia japonica, and Ficus elastica. They were planted in pots with a diameter of 10cm and cultivated in three different irrigation frequencies: two times per week, one time per week, and one time per two weeks. After 120 days, they were measured by plant height, fresh weight, dry weight, SPAD value, leaf color, leaf water potential, chlorophyll fluorescence (Fv/Fm), and photosynthetic rate. The average soil moisture content was $48.8{\pm}2.1%$ in two times per week, $25.2{\pm}4.4%$ in one time per week, and $10.3{\pm}2.4%$ in one time per two weeks. For A. pusilla, leaf water potential was higher, and Fv/Fm value was 0.731 in two times per week irrigation, showing more wetness. For A. pusilla, F. japonica and F. elastica photosynthetic rate was significantly lower in one time per two weeks irrigation, appearing to be more sensitive to drying than C. rosea. When irrigated one time per week, with the soil's volume average moisture content of 25%, all four woody plants used in this experiment proved to grow smooth. Thus, it was determined to be good for use in indoor landscaping.

Effect of Silicon on Growth and Temperature Stress Tolerance of Nephrolepis exaltata 'Corditas'

  • Effect of silicon (Si) nutrition on growth and temperature stress tolerance of Nephrolepis exaltata 'Corditas' grown in a soilless substrate was examined. In vitro-grown acclimatized plantlets were transplanted into the pots containing a coir-based substrate. A nutrient solution containing 0, 50, or $100mg{\cdot}L^{-1}$ Si was supplied through a drip-irrigation system. After 5 months of cultiv ation, S i-treated and -untreated p lants were grown at 10, 25, or $40{\pm}1^{\circ}C$ under a 12 h photoperiod with $530{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD and 60% RH. After 7 days, chlorophyll content and chlorophyll fluorescence parameters were measured. Silicon nutrition had a negative effect on growth characteristics of N. exaltata 'Corditas'. However, Si-treated plants had more tolerance to temperature stress than the control plants. The Fv/Fm value was not significantly different when the plants were exposed to $25^{\circ}C$. However, significant difference in Fv/Fm was recorded when plants were exposed to 10 or $40^{\circ}C$. Thus, Fv/Fm could be used as an indicator of low and high temperature tolerance in ferns. The present study also suggests that application of Si may be used to enhance temperature tolerance of ferns.

Effect of Light Intensity on the Growth Responses of Three Woody Plants for Indoor Landscaping (실내녹화용 목본식물 3종의 초기 생육반응에 미치는 광량의 영향)

  • The purpose of this study was to investigate the effects of light intensity on the initial growth response of three woody plants for indoor landscaping; Ardisia pusilla, Clusia rosea and Fatsia japonica. The plants were planted in 10cm pots, the light intensities used were of four levels-15, 30, 60, $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD-and light irradiation time was set to 12/12 (day/night). Growth responses including plant height, leaf length, leaf width, chlorophyll fluorescence (Fv/Fm), SPAD and Hunter values were measured at 4-week intervals, and shoot weight and root weight of fresh and dry plants were measured after completion of the experiment. Fatsia japonica tended to show greater leaf length and leaf width as light intensity became greater, while other plants did not show any significant differences at different light intensities. The Fv/Fm value of the Ardisia pusilla was found to be stressed at 60 and $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, while the Fv/Fm values were within normal range with other plants or at other light intensity levels to show no stress. Only Clusia rosea showed significantly different SPAD values at $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and there was no significant SPAD value difference found with other plants or at other light intensity levels. While Hunter values of the Ardisia pusilla did not show any significant differences at any light intensity levels, Clusia rosea and Fatsia japonica showed specificity in L, a and b values at 60 and $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. Ardisia pusilla showed a big stem growth at $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and Clusia rosea showed a steady growth at 60 and $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$.

The Effect of 4Ca^{2+}$ on $Cd^{2+}$ -induced Physiological Toxicity in Commelina communis L.

  • 3-weeks old Commelina was transferred to and grown in Hoagland solution ($\pm$ 100 $\mu{M}$ $Cd^{2+}$, 100 $\mu{M}$ $Cd^{2+}$ + 100$\mu{M}$ $Ca^{2+}$, 100 $\mu{M}$ $Cd^{2+}$ + 200 $\mu{M}$ EGTA) for two weeks and then a number of physiological activities was investigated. $Cd^{2+}$ reduced total chlorophyll content up to 29% at a week and 75% at two weeks. In the treatment of $Cd^{2+}$$Ca^{2+}$, the total chlorophyll content was reduced to 29% at a week and 80% at two weeks. $Cd^{2+}$ reduced 24% of Fv/Fm after two weeks. In case of $Cd^{2+}$$Ca^{2+}$, Fv/Fm was reduced 55% at a week, but after two weeks, the plants were almost dead and Fv/Fm could not be measured. When EGTA was treated with $Cd^{2+}$, the value of Fv/Fm was not affected. There were no differences of water potential between the control and the treatment of $Cd^{2+}$+EGTA toy a week, but in other treatments. water potential was reduced. $Cd^{2+}$ reduced about 21% of water potential and $Cd^{2+}$$Ca^{2+}$ reduced 43% of water potential after two weeks. $Cd^{2+}$ inhibited 21% of photosynthetic activity at a week and 32% at two weeks. In case of photosynthetic activity, $Cd^{2+}$$Ca^{2+}$ inhibited 58% at a week and 73% at two weeks. $Cd^{2+}$+EGTA inhibited 15% of photosynthetic activity at a week and 21% at two weeks. Similar results were found in stomatal conductance. From the above results, it was observed that the treatment of $Ca^{2+}$ with $Cd^{2+}$ induced more reduction of a series of physiological responses than those of the treatment of $Cd^{2+}$ alone. Therefore, it could be concluded that $Ca^{2+}$ did not reduce the toxicity of $Cd^{2+}$, but enhanced $Cd^{2+}$ -induced physiological toxicities, but EGTA induced an decrease of $Cd^{2+}$ -induced physiological toxicities.
