Isolation frequency of resistant isolates of Phytophthora capsici to metalaxyl was reported to be 38.9% through the resistance monitoring for metalaxyl in P. capsici causing pepper Phytophthora blight. Metalaxyl was very effective to mycelium growth, while not to zoosporangium germination and zoospore release. $EC_{50}$ values of metalaxyl in the inhibition of mycelium growth were 0.204, 0.151, 0.379, and $0.215\;{\mu}g\;mL^{-1}$ against each isolate sensitive to the fungicide as P. capsici 06-119, 06-143, P08-7, and P08-31, respectively, whilst those were 5.242, 5.724, 6.621, and $5.377\;{\mu}g\;mL^{-1}$ in P. capsici 06-125, 06-155, P08-50, and P08-60. For the field fitness, several factors, which were mycelium growth, zoosporangium germination, zoospore release, virulence to pepper plants, and the zoosporangium and the oospore production, were investigated with 4 sensitive isolates and 4 resistant isolates. Between 2 groups differentiated by the sensitivity of metalaxyl, there was no significance in mycelium growth, zoosporangium germination, zoospore release, and virulence to pepper plants. However, the zoosporangium and the oospore production in each resistant isolate, which were related to survival of P. capsici in fields, were superior to those of sensitive isolates. Based on results of this study, it was suggested that the increase of the percentage of resistant isolates to metalaxyl resulted from the high capacities of the zoosporangium and the oospore production.

Among 34 isolates of Colletotrichum spp., 20 from red pepper and 14 from apple, only one isolate 2001-45 was identified as the isolate resistant to sterol biosynthesis inhibiting fungicides. EC$_{50}$ values of the isolate 2001-45 were 153.5, 42.7, 34.0, and 17.1 $\mu\textrm{g}$/ml for myclobutanil, tebuconazole, hexaconazole and nuarimol, respectively, The resistance factor of the isolate 2001-45 against the other isolate 2001-44 to 4 above fungicides was ranged from 17 to 57. However, EC$_{50}$ value of the 2001-45 for prochloraz was 0.07 $\mu\textrm{g}$/ml, which was lower than those of the 2001-44 and the isolate JC24. For the fitness test of the 2001-45, mycelial growth, sporulation on PDA and pathogenicity on fruits were investigated. No difference in mycelial growth was found between 2001-45 and 2001-44, but great difference in sporulation. No symptom was developed by 2001-45 even by wound inoculation of pepper fruit. Therefore, this study indicated that the isolate 2001-45 was inferior to the other isolates in the fitness, although the isolate 2001-45 was highly resistant to sterol biosynthesis inhibiting fungicides.
Pepper plants were flooded at 0, 5, 10 and 15cm at seedling stage under the condition of greenhouse. Treatment of flooding times are 6, 12, 24, 48 and 120 hours. The results obtained are summarized as follows. Plant height, number of leaves, shoot and root fresh weight were not recovered the flooding damages regardless of flooding time and depth. Pepper plant died in flooding depth of 5cm or more for over 48 hours. Plants in fallen leaves were found at more than 5cm depth and 6 hours of floodings. Photosynthesis and respiration rate decreased in the 5cm flooding depth or more for 24 hours. Chlorophyll content and root activity decreased for 12 hours or more at all the flooding depth. Also, diffusion resistance of stomata cell increased as increased flooding time and depth. Diseases occurred remarkably in proportion to the depth and hours of flooding treatment. It was not possible to control the desease by fungicide, also then was no effects of foliar spray of urea. Weight of fruit per plant not decrease by the 12 hours of 0cm and the 6 hours of 5cm flooding but decreased at deeper and longer flooding. Average weight of a fruit increased. The yield could not expected in the depth of 5cm or more for over 48 hours, There was significant positive correlation between all the investigated characteristics of growth and yield. There was, however, negative correlation between number of leaf and diffusion resistance of stomata.
Effects of fungicides on the mycelial growth of Botrytis cinerea isolated from ginseng leaves were investigated by an agar dilution method. By using a agar dilution method, it was investigated the effect of fungicides, procymidone, carbendazim and the mixture with both of carbendazim and diethofencarb, on the mycelial growth of Botrytis cinerea isolates, which were isolated from infected leaves of ginseng in 2005 and 2006. With MIC (minimum inhibiton concentration) of procymidone against B. cinerea, pathogens were divided into two groups. While one showed the low MIC between 0.8 and $4.0{\mu}g/ml$, the other showed higher MIC above $20{\mu}g/ml$. In terms of the inhibition ratio of mycelial growth at the indicated concentration of procymidone, isolates of B. cinerea were divided into three groups; the sensitive, the intermediate resistant, and the resistant group. Each group was differentiated by $EC_{50}$; the sensitive group showed below $2.0{\mu}g/ml$, the intermediate resistant group between 2.0 to $5.0{\mu}g/ml$, and resistant group above $5.0{\mu}g/ml$. Compared with the ratio of resistant isolates of B. cinerea in 2005, the ratio in 2006 increased from 19.3% to 27.5%. Furthermore, the average $EC_{50}$ value of them increased from $10.0{\mu}g/ml$ in 2005 to $237.3{\mu}g/ml$ in 2006. The ratio of isolates showing the multiple resistance between procymidone and carbendazim was 40.2%, whereas the ratio was 4.0% showing the multiple resistance in the mixture.
Dollar spot, caused by Sclerotinia homoeocarpa, is the major disease in cool-season turfgrasses. Understanding the distribution of this pathogen in soil and thatch is important to developing disease control strategies. In this study, toothpicks were used to detect S. homoeocarpa in the turfgrass canopy, thatch, and soil at different distances from dollar spot infection centers. The effect of penetrant and contact fungicide applications with different water volumes on distribution of S. homoeocarpa was also investigated. S. homoeocarpa was detected in 100% of samples taken from the leaf canopy, 83.3% in thatch area, and 0% in the soil from within the infection center. S. homoeocarpa was isolated in 100% of samples taken from the edge of the infection center, but was only detected in 13% of the samples taken at 1.5 cm away from the infection center edge. S. homoeocarpa was isolated at a higher frequency in the propiconazole treated plots than those treated with chlorothalonil and was not detected in leaf canopy samples when either fungicides was applied with 6.78 L of water. In conclusion, the toothpick-aided detection technique has improved our understanding of S. homoeocarpa epidemiology and could be used as a diagnostic tool to detect for fungicide resistance on golf courses.
Fusarium head blight is an important disease of small grains. It is mainly caused by members of the Fusarium graminearum species complex (FGSC). Barley and wheat growers spray fungicides, especially demethylation-inhibitor fungicides, to suppress the disease. The objective of this study was to examine the changes in the sensitivity of the FGSC population to the triazole fungicide, propiconazole. A total of 124 and 350 isolates of FGSC were obtained from barley and wheat in Jeolla Province during 2010-2016 and 2020-2021, respectively. The species identity and trichothecene chemotypes of the FGSC isolates were determined based on polymerase chain reaction assays targeting translation elongation factor 1-alpha and TRI12 genes, respectively. Sensitivity to propiconazole was determined based on the effective concentration that reduced 50% of the mycelial growth (EC50) using the agar dilution method. Of all isolates, F. asiaticum with the nivalenol chemotype was the most common (83.9% in 2010-2016 and 96.0% in 2020-2021), followed by F. asiaticum with the 3-acetyl deoxynivalenol chemotype (12.1% in 2010-2016 and 2.9% in 2020-2021). The EC50 values of the isolates collected in 2010-2016 and 2020-2021 ranged from 0.0180 to 11.0166 ㎍/mL and 1.3104 to 17.9587 ㎍/mL, respectively. The mean EC50 value of the isolates increased from 3.8648 ㎍/mL in 2010-2016 to 5.9635 ㎍/mL in 2020-2021. The baseline resistance to propiconazole was determined to be 7 ㎍/mL, based on the EC50 value of isolates collected in 2010-2016, and the ratio of resistant isolates increased from 9.7% in 2010-2016 to 28.6% in 2020-2021.
Isotianil is a novel fungicide which induces systemic acquired resistance in plants. It has excellent preventive effects as low dosages against rice blast which is one of the most serious diseases in rice. The Maximum Residue Limit (MRL) of Isotianil in republic of korea was set to 0.1 mg/kg in rice, so it is necessary to determine levels of Isotianil residues in agricultural commodities for controlling food safety. Therefore, The purpose of this study was to develop analytical method for the determination of isotiical residues in agricultural commodities using GC-NPD/MSD. Isotianil was extracted with acetonitrile from apple, chinese cabbage, hulled rice, mandarin, pepper, and soybean. The extract was diluted with saline water, and then dichloromethane partition was followed to recover this fungcide from the aqueous phase. A solid phase extraction with Florisil cartridge was additionally employed for final clean up. Isotianil was analyzed and quantitated by GC-NPD and confirmed by GC-MSD. Average recovery of Isotianil ranged from 70.0 to 103.9% in six representative agricultural commodities with relative standard deviations less than 10%, and limit of quantification (LOQ) was 0.05 mg/kg.

Baseline sensitivity to benthiavalicarb, iprovalicarb and dimethomorph included into carboxylic acid amide (CAA) group was evaluated in 180 isolates of Phytophthora capsici over 4 years from 2005 to 2008. $EC_{50}$ (effective concentration inhibiting mycelial growth by 50%) value of benthiavalicarb ranged from $0.015{\mu}g\;mL^{-1}$ to $0.049{\mu}g\;mL^{-1}$ with a mean of $0.033{\mu}g\;mL^{-1}$. The mean values of $EC_{50}$ of iprovalicarb and dimethomorph were 0.411 (0.197 - 0.556) ${\mu}g\;mL^{-1}$ and 0.271 (0.101 - 0.798) ${\mu}g\;mL^{-1}$, respectively. Although there was no increasing tendency in $EC_{50}$of benthiavalicarb and iprovalicarb during 4 years, $EC_{50}$ of dimethomorph was increased gradually by laps of time. There was no cross-resistance between each fungicide used in this study and metalaxyl. Among fungicides included into CAA group, there was a positive correlation between benthiavalicarb and iprovalicarb, and between dimethomorp and mandipropamid.

Growth-inhibitory effects of citrus oils and agricultural fungicides, which were on the market, on several molds isolated from putrefied citrus fruits were investigated. When fungicidal activities of 11 kinds of synthetic agrochemicals against 6 species of molds, Alternaria alternata, Rhizopus sp., Botrytis cinerea, Monilia candida, Penicillium italicum and Penicillium digitatum, were investigated, agrochemicals containing mancozeb or fluazinam as an effective component had the broadest fungicidal spectrum. Agrochemicals containing iprodione, benomyl, azoxystrobin or thiophanate were less effective on the molds and those containing conazole derivative were intermediately effective. Resistance of the molds to the agrochemicals were species- and agrochemical-dependent. Among those molds tested, Penicillium italicum and Alternaria alternata showed relatively higher level of survival in the presence of synthetic fungicides. On the other hand, when the molds were exposed to citrus oils by direct contact, no cell could survive regardless of the species. The fungicidal activity of citrus oils was also confirmed by paper disk method and microscopic observation. These results suggested that citrus oils had broad killing activity against molds. Therefore, it would be necessary to design method for the application of citrus oils in order to improve post-harvest storage of citrus fruits.
Pythium blight caused by Pythium spp. is one of major diseases in putting green of golf course. In this study, microorganisms which are anatgonistic to Pythium aphanidermatum, a pathogen of pythium blight, were selected primary through in vitro tests, dual culture method and triple layer agar diffusion method. In vivo test against pythium blight were conducted to select the best candidate biocontrol microorganism by pot experiment in a plastic house. Bacillus subtilis GB-0365 was finally selected as a biocontrol agent against pythium blight. Relative Performance Indies(RPI) was used as a criterion of selecting potential biocontrol agent. B. subtilis GB-0365 showed resistance to major synthetic agrochemicals used in golf course. Alternative application of synthetic agrochemicals and B. subtilis GB-0365 was most effective to successfully contol pythium blight. B. subtilis GB-0365 suppressed the development of pythium bight of bentgrass by 56.4% as compared to non-treated control and its disease control efficacy was 60.9% of a synthetic fungicide Oxapro(WP) efficacy. B. subtilis GB-0365 has a potential to be a biocontrol agent for control of pythium blight.
