Studies on the Ligninolytic Enzyme Activities During Biological Bleaching of Kraft Pulp with Newly Isolated Lignin-Degrading Fungi

  • A screening has been performed to find hyper-ligninolytic fungi, which degtrade beech and pine lignin extensively in order to broaden the understanding of the ligninolytic enzymes elaborated by various white-rot fungi. One hundred and twenty two ligninolytic strains were selected from decayed woods with a selective medium for screening ligninolytic wood-rotting fungi. Two of them, Phanerochaete sordida YK-624 and YK-472, showed much higher ligninolytic activity and selectivity in beech-wood degradation than typical lignin-degrading fungi, phanerochaete chrysosporium and Coriolus versicolor. They also degraded birch dioxane lignin and residual lignin in unbleached kraft pulp(UKP) much more extensively than P. chrysosporium and C. versicolor. During fungal treatment of beech wood-powder, the fungus strain P. sordida YK-624 showed higher activity of extracellular manganese peroxidase (MnP) in the medium than P. chrysosporium. It also showed MnP activity, which would not be lignin peroxidast during treatment of oxygen-bleached kraft pulp(OKP) and under enzyme-inducing conditin.

  • Anaerobic chytridiomycete fungi are now well recognized as one of the major components of rumen microflora. Since the discovery of anaerobic fungi, the knowledge upon their morphology and physiology has been accumulated. It is certain that they gave roles in ruminal fiber digestion, although their quantitative contribution to rumen digestion is still unclear. Their role in fiber digestion is complicated by the dietary factors and the interaction with other microorganisms. We aim at reviewing such information in this article. Considerable attention gas been paid to the polysaccharidase of these fungi. Analysis on the fungal genes encoding these enzymes has been performed in several laboratories. This article also covers the genetical analysis of fungal polysaccharidases.

Mycotoxin분비 수종균주들의 배양여과액으로 처리된 HeLa세포의 전자현미경적 관찰 (Electron Microscopec Observations on the HeLa Cells treated with culture Filtrates of Mycotoxin-Producing Fungi)

  • The fine structure of HeLa cells treated with several mycotoxin-producing fungi (Aspergillus flavus ATCC 15517, Aspergillus parastiticus RIB 1037, Penicillium toxicarium RIB 4002, Penicillium cirinum SWU)238, Penicillium islandicum IFO 5235, Penicillium tadum IFO 5787 and Pencillium brunneum RIB 1172) has been examined and some details have been descried. The normal HeLa cell have numerous microvilli, large ovoid nucleus, pleomorphic mitochondria, electron-dense body, Golgi complex, mid-body and endoplasmic reticulum etc. Certain specific structural changes induced by culture filtrates of several mycotoxin-producing fungi have been noted. These alterations induced disappearance of Golgi complex, rER vacuolization, nucleolus attachment to the nuclear envelope nad appearance of certain vacuoles. There were not any changes by the treatment of culture filtrates of non-toxic fungi and only cell debris of some specimens can be observed by the injury of culture filtrates. The experimental animals treated with mycotoxin-producing fungi (Aspergillus flavus ATCC 15517, Aspergillus parasilicus RIB 1037, Penicillum citrinum SWU 238, Penicillium toxicarium RIB 4002, and Penicillium islandicum IFO 5235) were mal cells treated with culture filtrates.

Effects of Interspecific Interactions of Arbuscular Mycorrhizal Fungi on Growth of Soybean and Corn

  • Growth responses of Zea mays and Glycine max to colonization by mixture of combination of three species of arbuscular mycorrhizal (AM) fungi, two species of Glomus and a species of Scutellospora were compared. In Zea mays, plants inoculated with single species of AM fungi showed significantly higher in dry weight than non-mycorrhizal plant for all three AM fungal species. Also, growth of plants inoculated with spores of two species of AM fungi was significantly higher than nonmycorrhizal control except for plants inoculated with two Glomus species. When three species of AM fungi were inoculated, the plants showed the highest growth. In Glycine max, plants with single AM fungal species inoculation were not significantly different in plant growth from nonmycorrhizal plants. When the plants were inoculated with combination of two or more AM fungal species, their growth significantly increased compared to nonmycorrhizal plants. In both plant species, mycorrhizal root colonization by Scutellospora species was significantly lower than by Glomus species.

  • damages of cotton cloth and characteristics of fabroid degradation were studied by Chaetomium globosum and Aspergillus niger which presupposed as powerful erosive fungi to cellulose fiber by means of tensile strength. The results obtained are as follows: 1. the growth(rate) of fungi in malt extract agar was superior to potato agar for two weeks. 2. Chaetomium globosum showed mostly severe damage t the cotton cloth in malt extract agar media at pH 4.5. 3. Tensile strength was reduced with time by Aspergillus niger-coenzyme and Chaetomium globosum-coenzyme reaction. In comparison with Chaetomium globosum and Aspergillus niger, the former weaken tensile strength about 15.8% and the latter enfeebled 10.0% after 124 hours. 4. after 30 days the breeding of fungi in pH 4.5 malt extract agar media, critical damage of cotton cloth was observe, I. e., 92.4% damage by chaetomium globosum and 74.9% lose by aspergillus nige respectively.

침엽수의 잎에서 분리한 국내 미기록 내생균 3종에 대한 보고 (First Report of Three Species of Endophytic Fungi Isolated from Needle Leaves of Conifers in Korea)

  • 경기도와 강원도에 위치한 화악산, 명지산 그리고 설악산에서 침엽수를 채집하여 균 분리를 수행하였다. 특히, 눈측백과 분비나무의 내생균에 대한 연구가 최초로 수행되었으며, 본 연구를 통해 이들로부터 분리한 균에 대한 형태적, 유전적 동정을 수행하였다. 그 결과 Cryptosporiopsis actinidiae, Pestalotiopsis mangiferae, Pyrenochaeta cava가 확인되었으며, 이에 대해 최초로 국내에서 눈측백과 분비나무에 분포함을 보고하였다.

  • 곤충이 곰팡이병에 감염되는 것을 흔히 볼 수 있고, 대부분은 이들에 대해 감수성을 나타내고 있다. 최근 살충제 저항성이 문제가 되면서 곤충병원성 곰팡이를 이용한 모기 방제에 대한 연구가 더욱 주목을 받고 있고, Beauveria bassiana, Metarhizium anisopliae, Lagenidium giganteum와 같은 병원성 곰팡이를 모기 방제에 사용하기 위해 노력 중이다. 이 총설에서는 말라리아, 뎅기열, 사상충, 황열병 등을 옮기는 모기들의 방제에 사용되는 곤충병원성 곰팡이를 정리하고 이들의 활용방안을 논하였다.

  • 1996년 4월1일부터 1998년 3월 31일까지 지리산의 국립공원에서 많은 고등균류를 채집하였다. 그것들을 생태적 자원면에서 분류한 결과는 다음과 같다. 희귀 균류는 진균문에서는 줄솔밭버섯(Omphalina ericetorum)을 포함하여 15종이고 변형균문에서는 아재비털먼지(Trichia affinis), 청황색 자루먼지(Physarum viride var. aurantium)을 2종을 확인하였다. 그리고 식용가능한 균류는 135종, 재배 가능한 버섯은 16종, 독버섯은 35종, 항암버섯(약용포함)은 68종, 균근형성균은 51종, 목재부후균(낙엽분해균포함)은 177종이었다.

Diversity of Endophytic Fungi Isolated from Pinus densiflora and Juniperus rigida Distributed in Mt. Baekryeonsan and Mt. Johangsan, Korea

  • We investigated the biodiversity of endophytic fungi in Pinus densiflora and Junipreus rigida from Mt. Baekryeonsan and Mt. Johangsan in Korea. In total, 59 isolates were discovered from 26 host plants and identified using sequences from the rDNA internal transcribed spacer (ITS) region. Twenty taxa of endophytic fungi were identified in this study; 62% of isolates belonged to Leotiomycetes, 34% belonged to Sordariomyetes, and 4% belonged to Agaricomycetes. Results confirmed three previously unreported endophytic fungi in Korea: Phanerochaete concrescens, Creosphaeria sassafras, and Scopuloides rimosa. Community analysis showed that the fungal diversity patterns of the samples from Mt Baekryeonsan and Mt. Johangsan are similar to those from previous studies and varied depending upon host and locality.

New Record of three Penicillium species isolated from the Freshwater Environment in Korea

  • In this study, we aimed to observe the diversity of aquatic fungi by collecting deposits of soil, plants, and plant litter from streams and rivers. Three Penicillium strains were isolated from soil in streams. Based on the morphological characteristics and phylogenetic analysis using DNA sequences of the internal transcribed spacer, β-tubulin and calmodulin genes, the isolates were identified as Penicillium guanacastense, P. saturniforme, and P. scabrosum. These three fungi have not yet been reported in Korea.
