Mycobiota including Alternaria alternata, Fusarium verticillioides and Aspergillus pulverulents were tested for their ability to uptake radiocobalt(Co-60) and radiocesium(Cs-137) from radionuclide containing medium. A. alternata was the most efficient fungal species for uptake of radioisotopes, followed by A. pulverulents, whereas F. verticilliodies came in the last rank. The conditions of radioisotope uptake were optimized such as the form of the fungal organism either spores or mycelium, inoculum age and pH of growing medium. Furthermore the total pigments of the tested fungi were extracted and tested for their ability to bind with radioisotope, where melanin of A. alternata produced about 60% for radioisotope uptake out of total added radioisotope radioactivity. Moreover, transmission electron microscopic examination of radioisotope exposed spores showed high precipitation of melanin granules in the spore wall and within the cell as comparing to untreated spores.
The vibration of fungal cultures was evaluated to determine its potential effect on the dispersal of airborne fungal microorganisms suspected of being pathogens. An artificial vibration system, which simulates the actual environmental vibration of fungal structures, was designed and constructed for this purpose. Experiments featured the use of low-frequency vibrations similar to those induced by earthquakes. Within the range of conditions tested, the vibration of fungal cultures was found to affect the airflow-driven generation of bioaerosols.
An imperfect fungus Fusiococcum species was isolated from Quercus dentata. A naturally infected Daimyo oak tree was collected and showed elongate wounds on the stem. The fungal cultures were initially white and cottony, and later turned dark gray. Numerous solitary pycnidia were developed on the medium surface, and typically spherical. Yellowish conidial masses were exuded from pycnidia on the culture plates. Conidial masses were swollen and measured as approximately 100 to $300{\mu}m$ in length. It appeared that conidia were usually held together in globose to oval drops. Conidia were hyaline, single-celled (nonseptate), ellipsoid to fusoid, and measured as approximately $8.0{\times}2.7{\mu}m$. Based on these cultural and morphological characteristics, the fungal isolate was identified as a species of Fusicoccum Corda. To preserve and examine fungal spores exuded from pycnidia on the medium surface, a vapor fixation procedure for scanning electron microscopy was employed in this study. The specimens were exposed to the vapor of 2% (v/v) glutaraldehyde and 2% (w/v) osmium tetroxide each for 2 h. With the vapor fixation we obtained excellent retention of conidial masses in this study. The simple and versatile procedure for demonstrating fungal spores and their exudation from fruiting bodies would facilitate characterization of diverse pathological and environmental isolates as they are in native environments.

Nosema ceranae is an obligate intracellular fungal parasite that causes mortality in honey bees and enhances the susceptibility of honey bees to other pathogens. Efficient purification of Nosema spores from the midgut of infected honey bees is very important because Nosema is non-culturable and only seasonably available. To achieve a higher yield of spores from honey bees, in this study, we considered that the initial release of spores from the midgut tissues was the most critical step. The use of 2 mm beads along with enzymatic treatment with collagenase and trypsin enhanced the homogenization of tissues and the yield of released spores by approximately 2.95 times compared with the use of common 3 mm beads alone. The optimal time for the enzyme treatment was determined to be 1 hr as measured by the yield and viability of the spores. A one-step filtration using a filter paper with an $8-11{\mu}m$ pore size was sufficient for removing cell debris. This method may be useful to purify not only N. ceranae spores but also other Nosema spp. spores.
As the first step to develop efficient means to control fungal spore bioaerosols, we designed, manufactured, and evaluated a fungal spore aerosol generator. We studied the physical and biological properties of the fungal spore bioaerosols on two common fungal species. The results demonstrated that the fungal spore bioaerosol generator effectively produces fungal spore bioaerosols.
청미래덩굴의 근권 토양을 온실에서 배양하여 Glomeromycota에 속하는 균류의 포자를 분리하였다. 분리된 포자는 18S partial rDNA 영역의 DNA 염기서열 분석과, 더 정확한 동정을 위해 추가적으로 ITS 영역과 28S rDNA 영역까지 포함하는 $Kr{\ddot{u}}ger$ fragment 지역의 염기서열을 분석하여 동정하였다. 동정 과정에서 2종의 국내 미기록종 Glomeromycota 균류의 포자를 확인하였으며, 확인된 종은 Diversispora eburnea, Paraglomus laccatum이다. 확인된 미기록종 포자의 형태적 특성 및 계통적 분석의 결과에 대해 기술하였다.
Conidial development of Cordyceps militaris was observed from germinating ascospores and vegetative hyphae through light and scanning electron microscopy (SEM). Ascospores were discharged from fresh specimens of C. militaris in sterile water as well as Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates. We observed ascospore germination and conidial formation periodically. Under submerged condition in sterile water, most part-spores germinated unidirectionally and conidia were developed directly from the tips of germinating hyphae of part-spores within 36 h after ascospore discharge, showing microcyclic conidiation. First-formed conidia were cylindrical or clavate followed by globose and ellipsoidal ones. Germination of ascospores and conidial development were observed on SDAY agar by SEM. Slimy heads of conidia on variously arranged phialides, from solitary to whorl, developed 5 days after ascospore discharge. Besides, two distinct types of conidia, elongated pyriform or cylindrical and globose, were observed in the same slimy heads by SEM. Conidia were shown to be uninucleate with 4,6-diamidino-2-phenylindole staining. Conidiogenous cells were more slender than vegetative hyphae, having attenuated tips. Microcyclic conidiation, undifferentiated conidiogenous hyphae (phialides), polymorphic conidia and solitary, opposite to whorled type of phialidic arrangement are reported here as the characteristic features of asexual stage of C. militaris, which can be distinguished from other Cordyceps species.
Soil-borne fungal pathogens such as Verticillium and Rhizoctonia can colonize in the stem tissue of plant through root and lead to wilting symptoms of plant by blocking. water transportation. During the colonization of Rhizoctonia solani in the vascular tissue of tomato stems, particularly, phenylalanine ammonia-lyase (PAL) gene induction pattern was cyclized showing peak induction at two different time points (10 and 80 h) after fungal spores inoculation in vivo. In leaves or roots, however, no such cycling pattern was observed. The first induction peak may be due to an initial sporulation events leading to a second induction peak by a proliferation of fungal spores to the upper stems or other tissues from an initial spore trapping sites. Tomato PAL gene was also dramatically induced by wounding, light illumination and mercury chloride treatment but was not cyclized. Mercury chloride showed the earliest induction with all tissues even at half an hour after treatment.
The indoor environmental condition was assessed in houses with allergy (asthma and atopy) patients by use of a fungal detector. The fungal index was calculated from the growth rate of the sensor fungi in a fungal detector encapsulating the spores, Alternaria alternata S-78, Eurotium herbariorum J-183 and Aspergillus penicillioides K-712. Fungal indices were higher in asthma patient's houses than in control houses and Eurotium herbariorum showed the highest growth response among the sensor fungi. Dust mites allergen, Der f1, was also significantly high in allergy patient's houses where fungal indices above 10 were detected. A correlation was observed between the fungal indices and dust mite allergen proliferations in examined houses. Therefore, the fungal index can be a useful tool as an indirect indication for detecting chronic dampness that brings both contaminations by fungi and dust mite.
우포 습지에 서식하는 식물의 근권 토양에서 수지상균근균(AMF) 포자를 추출하였다. 추출한 포자는 형태적 특성과 18S rDNA 염기서열의 분자적 분석을 이용해 동정하였다. 그 결과 6속 9종의 AMF 포자가 분리된 것을 확인하였으며, 종 다양성 지수를 계산하여 분석한 결과 습지에서 AMF 포자의 종 다양성은 식물에 의해 영향을 받는 것을 확인하였다. 연구 과정에서 국내 미기록종 1종을 확인하였으며, 확인된 종은 Diversispora epigaea이다. 미기록종 AMF 포자의 형태적 특성 및 분자적 분석의 결과에 대해 서술하였다.
