The achievement of tissue engineering can be highly depending on the capability to generate complicated, cell seeded three dimensional (3D) micro/nano-structures. So, various fabrication techniques that can be used to precisely design the architecture and topography of scaffolding materials will signify a key aspect of multi-functional tissue engineering. Previous methods for obtaining scaffolds based on top-down are often not satisfactory to produce complex micro/nano-structures due to the lack of control on scaffold architecture, porosity, and cellular interactions. However, a bioprinting method can be used to design sophisticated 3D tissue scaffolds that can be engineered to mimic the tissue architecture using computer aided approach. Also, in recent, the method has been modified and optimized to fabricate scaffolds using various natural biopolymers (collagen, alginate, and chitosan etc.). Variation of the topological structure and polymer concentration allowed tailoring the physical and biological properties of the scaffolds. In this presentation, the 3D bioprinting supplemented with a newly designed plasma treatment for attaining highly bioactive and functional scaffolds for tissue engineering applications will be introduced. Moreover, various in vivo and in vitro results will show that the fabricated scaffolds can carry out their structural and biological functionality.
In multicellular organisms, including humans, understanding expression specificity at the tissue level is essential for interpreting protein function, such as tissue differentiation. We developed a prediction approach via generated sequence features from overrepresented patterns in housekeeping (HK) and tissue-specific (TS) genes to classify TS expression in humans. Using TS domains and transcriptional factor binding sites (TFBSs), sequence characteristics were used as indices of expressed tissues in a Random Forest algorithm by scoring exclusive patterns considering the biological intuition; TFBSs regulate gene expression, and the domains reflect the functional specificity of a TS gene. Our proposed approach displayed better performance than previous attempts and was validated using computational and experimental methods.
Tissue engineering and stem cells show potentials to restore lost or malfunctioning human tissues or organs. Another cell source for tissue engineering of cardiovascular tissues is stem cell. This study reports the development of cardiovascular tissues using tissue engineering and mesenchymal stem cells. The blood vessels and heart valves were fabricated by culturing mesenchymal stem cells on biodegradable synthetic or natural matrices. Bone marrow was isolated from dogs or rats and mesenchymal stem cells were cultured. The cells were seeded onto biodegradable synthetic or natural matrices and implanted in dogs. Histological and immunohistochemical analyses were performed to examine the regenerated cardiovascular tissues. Histological and immunohistochemical analyses showed the complete regeneration of blood vessels and heart valves. Fluorescent labeling of cells prior to implantation and fluorescence examination of the regenerated tissues revealed that the implanted cells reconstituted the cardiovascular tissues. This study demonstrates the potential of tissue engineering and mesenchymal stem cells for the regeneration of functional cardiovascular tissues or organs.
Emerging trends for cardiac tissue engineering are focused on increasing the biocompatibility and tissue regeneration ability of artificial heart tissue by incorporating various cell sources and bioactive molecules. Although primary cardiomyocytes can be successfully implanted, clinical applications are restricted due to their low survival rates and poor proliferation. To develop successful cardiovascular tissue regeneration systems, new technologies must be introduced to improve myocardial regeneration. Electrospinning is a simple, versatile technique for fabricating nanofibers. Here, we discuss various biodegradable polymers (natural, synthetic, and combinatorial polymers) that can be used for fiber fabrication. We also describe a series of fiber modification methods that can increase cell survival, proliferation, and migration and provide supporting mechanical properties by mimicking micro-environment structures, such as the extracellular matrix (ECM). In addition, the applications and types of nanofiber-based scaffolds for myocardial regeneration are described. Heart disease is one of the major life-threatening diseases with high mortality and incidence worldwide. Several model systems, such as primary cells and animals, have been used to understand heart diseases and establish appropriate treatments. However, they have limitations in accuracy and reproducibility in recapitulating disease pathophysiology and evaluating drug responses. In recent years, three-dimensional (3D) cardiac tissue models produced using tissue engineering technology and human cells have outperformed conventional models. In particular, the integration of cell reprogramming techniques with bioengineering platforms (e.g., microfluidics, scaffolds, bioprinting, and biophysical stimuli) has facilitated the development of heart-on-a-chip, cardiac spheroid/organoid, and engineered heart tissue (EHT) to recapitulate the structural and functional features of the native human heart. These cardiac models have improved heart disease modeling and toxicological evaluation. In this review, we summarize the cell types for the fabrication of cardiac tissue models, introduce diverse 3D human cardiac tissue models, and discuss the strategies to enhance their complexity and maturity. Finally, recent studies in the modeling of various heart diseases are reviewed.
The central strategy in tissue engineering involves a biomaterial scaffold as a delivery carrier of cells and a depot to deliver bioactive molecules. The ability of scaffolds to control cellular response to direct particular repair and regeneration processes is essential to obtain functional tissue engineering constructs. Therefore, many efforts have been made to understand local interactions of cells with their extracellular matrix (ECM) microenvironment and exploit these interactions for designing an ideal scaffold mimicking the chemical, physiological, and structural features of native ECM. ECM is composed of a number of biomacromolecules including proteins, glycosaminoglycans, and proteoglycans, which are assembled together to form complex 3-dimensional network. Electrospinning is a process to generate highly porous 3-dimensional fibrous structure with nano to micro scaled-diameter, which can closely mimic the structure of ECM. In this presentation, our approaches to develop biomimetic electrospun fibers for modulation of cell function will be discussed.
Scaffold fabrication for regenerating functional human tissues has an important role in tissue engineering, and there has been much progress in research on scaffold fabrication. However, current methods are limited by the mechanical properties of existing biodegradable materials and the irregular structures that they produce. Recently, Solid freeform fabrication (SFF) technology was remarked by fabricating 3D free-form micro-structures. Among SFF technologies, we tried to fabricate scaffolds using micro-stereolithography which contain the highest resolution of all SFF technologies and precision deposition system which can use various biomaterials. And we developed the CAD/CAM system to automate the process of scaffold fabrication and fabricate the patient customized scaffolds. These results showed the unlimited possibilities of our SFF technologies in tissue engineering.
Polymer brush is a soft material unit tethered covalently on the surface of scaffolds. It can induce functional and structural modification of a substrate's properties. Such surface coating approach has attracted special attentions in the fields of stem cell biology, tissue engineering, and regenerative medicine due to facile fabrication, usability of various polymers, extracellular matrix (ECM)-like structural features, and in vivo stability. Here, we summarized polymer brush-based grafting approaches comparing self-assembled monolayer (SAM)-based coating method, in addition to physico-chemical characterization techniques for surfaces such as wettability, stiffness/elasticity, roughness, and chemical composition that can affect cell adhesion, differentiation, and proliferation. We also reviewed recent advancements in cell biological applications of polymer brushes by focusing on stem cell differentiation and 3D supports/implants for tissue formation. Understanding cell behaviors on polymer brushes in the scale of nanometer length can contribute to systematic understandings of cellular responses at the interface of polymers and scaffolds and their simultaneous effects on cell behaviors for promising platform designs.
BACKGROUND: Bioprinting has recently appeared as a powerful tool for building complex tissue and organ structures. However, the application of bioprinting to regenerative medicine has limitations, due to the restricted choices of bio-ink for cytocompatible cell encapsulation and the integrity of the fabricated structures. METHODS: In this study, we developed hybrid bio-inks based on acrylated hyaluronic acid (HA) for immobilizing bio-active peptides and tyramine-conjugated hyaluronic acids for fast gelation. RESULTS: Conventional acrylated HA-based hydrogels have a gelation time of more than 30 min, whereas hybrid bio-ink has been rapidly gelated within 200 s. Fibroblast cells cultured in this hybrid bio-ink up to 7 days showed >90% viability. As a guidance cue for stem cell differentiation, we immobilized four different bio-active peptides: BMP-7-derived peptides (BMP-7D) and osteopontin for osteogenesis, and substance-P (SP) and Ac-SDKP (SDKP) for angiogenesis. Mesenchymal stem cells cultured in these hybrid bio-inks showed the highest angiogenic and osteogenic activity cultured in bio-ink immobilized with a SP or BMP-7D peptide. This bio-ink was loaded in a three-dimensional (3D) bioprinting device showing reproducible printing features. CONCLUSION: We have developed bio-inks that combine biochemical and mechanical cues. Biochemical cues were able to regulate differentiation of cells, and mechanical cues enabled printing structuring. This multi-functional bio-ink can be used for complex tissue engineering and regenerative medicine.
