Food Safety of Functional Neoglycoproteins Prepared by Covalent Attachment of Galactomannan to Food Proteins

  • Hen egg-white lysozyme, ovalbumin, egg-yolk phosvitin, acid-precipitated soy protein and $\alpha$$_{sl}$ milk casein were covalently linked with galactomannan through a controlled dry-heating at 6$0^{\circ}C$ under 79% relative humidity without any chemical reagent. Neoglycosylation by the covalent binding of polysaccharide chains brought a significant improvement into the surface functionalities of food proteins. Excellent emulsifying properties and foaming properties were observed in all protein-galactomannan conjugates. Bacterial mutagenesis tests and animal dose test were done to evaluate the food safety of the protein-galactomannan conjugates. The neo-glycoproteins were negative for Ames test using Salmonella typhimurium TA100 (hisG46) and TA98 (hisD3052) strains, and rec-assay using Bacillus subtilis Hl7 (rec) and M45 (re $c^{+}$) strains. All substances were also nontoxic for oral administration to rats. L $D_{50}$ 's of these substances were all more than 7.5 g/kg body-weight of rat. No effect was also observed in the weight increases and the concentrations of total cholesterol, triglyceride and phospholipids in blood serum of the administrated rats with 7.5 g/kg conjugates. Thus, Maillard-type protein-polysaccharide conjugates prepared by covalent attachment of galactomannan to food proteins were proposed to be useful as a safe functional biopolymer in this study.y.

The Current Status and Prospect of Sericultural Byproduct Industry in China

  • Sericulture is a traditional agro-industry, which involves mulberry cultivation and silkworm rearing, has made great contributes to the human civilization. With the development of national economy and modem technology, mulberry and silkworm are being used to develop products with functionality besides the traditional cocoon production in China. In this paper, we brief the current developing situation of sericultural byproducts with functionality in the following aspects. (1) Functional products from silkworm larvae: silkworm powder, white muscardine silkworm, isolation and purification of anti-bacterial proteins from the larvae and production of medically valuable substances by Bombyx mori nuclear polyhedrosis virus (BmNPV) vector. (2) Utilization of silkworm feces: for pillow and for isolation of chlorophyll etc. (3) Production of valuable Chinese traditional medicine like Cordyceps sinensis with pupae, functional utilization of pupa protein and chitin. (4) Silk as additives to cosmetics, silk food and medical materials. (5) Functional utilization of mulberry: cultivation of edible fungus on mulberry shoots as medium, mulberry fruit drinks, mulberry tea, etc. The prospect of sericultural byproduct industry in China is also discussed.

건강기능식품 안전관리 연구방향 (Research Direction for Functional Foods Safety)

  • 건강효과나 보건효과를 표방한 다양한 건강기능식품이 시장에 유통되고 있다 이들 제품은 분명한 식품형태의 것으로부터 정체, 캡슐 상 까지여서 의약품과 오인 혼동될 우려가 높으며 전문적 지식이 없는 사람이 통상의 식품으로서 판매하고 있는 일이 많고 건강기능식품용 질병의 치료 목적으로 사용하는 경우도 적지 않다. 건강기능식품제도의 창설이나 법적 규제에 의해 혼란스런 건강기능식품의 상황을 개선하기 위한 노력이 계속되어 왔지만 아직 소비자에게는 제대로 전달되지 않고 있다. 따라서 건강기능식품이 질병의 치료에 이용되는 문제나 위법으로 의약품성분을 첨가한 제품이 유통되는 문제는 끊이지 않고 있고 이러한 문제는 인터넷 등의 정보 전달수단의 발달에 따라 더욱 확대되고 있다. 건강기능식품의 문제는 제품자체의 문제와 이용방법의 문제로 크게 나눌 수 있지만 어느 쪽이나 적절한 정보를 소비자에게 전달해서 그 실태가 틀림없이 인식되어야만 어느 정도 개선 헬 수 있다고 생각된다. 건강가능식품과 관련되어 야기 될 수 있는 문제들은 위법제품, 유해물질 혼입, 알레르기 병자에 대한 투여시 고려되어야 할 사항, 의약품과의 상호작용, 성분표시가 있어도 그 순도와 함량이 극히 애매해 검출되지 않을 수도 있는 문제, 대사활성화가 일어나는 제품, Alkaloid가 함유된 경우 및 비타민 미네랄의 과잉섭취와 부족에 의한 건강피해 리스크 등 많은 경우가 있다. 건강기능식품과 관련한 부작용 사례가 해마다 증가하고 있는 이유는 불량 건강기능식품 업체가 그 효과와 효능을 과장해서 선전하는 것도 중요한 부분을 차지하며 실제로 허위 과대 광고로 인해 적발된 사례가 연평균 821건에 달한다. 식품의약품안전청은 홈페이지를 통해 건강기능식품 문제의 개선과 홍보를 위해 정보를 제공하고 있지만 건강기능 식품의 적정사용과 의약품 건강기능식품 간 상호작용을 피하기 위해서는 개선의 여지가 있다. 건강기능식품문제의 개선을 위해서는 승인된 제품에 대한 정보 부작용 수집의 제도화 및 재평가 제도의 정착, 인체적용 전 시험과 인체적용 시험조건에 대한 표준화가 시급하다. 또한 적절한 정보제공 활동을 위해서는 건강기능식품 의약품 등 이종 데이터간의 유기적 연동을 위한 통합 DB시스템 및 사용자가 데이터를 편리하게 검색할 수 있는 사용자 인터페이스도 요망된다.

재식간격 및 재배 기간에 따른 고구마 수량 및 유용성분 함량 평가 (Yield of Tuber Roots and Functional Substances According to the Planting Interval and Cultivation Period in Sweetpotato (Ipomoea batatas L.))

  • 본 연구에서는 유용성분 함유 고구마 품종의 대량생산을 위한 재배법 개발을 위하여 재식간격 및 재배기간에 따른 괴근 수량 및 다양한 유용성분 함량 분석을 수행하였다. 이러한 결과는 고구마 수량 및 유용성분 대량생산을 위한 재배법 개선에 유용한 정보를 제공해줄 것으로 생각된다. 1. 두 품종 모두 재배기간이 길수록 총 괴근 수량과 300 g 이상 괴근 수량이 증가하는 경향을 나타내었다. 2. 주황미의 경우 재식간격별 수량 조사 결과, 70×20 cm 정식구에서 재배기간이 길어질수록 총 괴근 수량과 300 g이상 괴근 수량이 가장 높게 나타났으며, 신자미의 경우에는 재식간격에 따른 총 괴근 수량의 변화는 큰 차이가 없었으나 300 g이상 괴근 수량은 140일과 160일 재배구에서 70×30 cm와 70×35 cm 정식구의 수량이 70×20 cm과 70×25 cm 정식구에 비해 높게 나타났다 3. 주황미 괴근의 베타카로틴의 총 함량도 재배기간이 길수록 높은 수치를 나타냈으며 70×20 cm 재배 간격에서 베타카로틴의 함량이 다른 재배간격에서 베타카로틴의 함량에 비해 가장 높게 나타났다. 4. 신자미 괴근의 안토시아닌의 총 함량 역시 재배기간 120일 대비 140일과 160일에서 크게 증가하였다. 재식간격에 따른 총 안토시아닌 수량의 변화는 큰 차이가 없었으나 300 g이상 괴근 수량에 해당하는 안토시아닌 총 함량은 140일과 160일 재배구에서 70×30 cm와 70×35 cm 정식구의 수량이 70×20 cm과 70×25 cm 정식구에 비해 높게 나타났다. 5. 총 폴리페놀 및 플라보노이드 함량을 분석한 결과 두 함량 모두 주황미 보다 신자미가 현저히 높은 함량을 나타내었으며, 160일 재배구에서 가장 높게 나타났다. 특히 300 g이상 괴근의 두 유용성분의 함량은 70×30 cm 또는 70×35 cm에서 가장 높은 수치를 나타내었다. 6. 고구마 유용성분 대량 생산을 위한 재배방법으로 두 품종 모두 재배기간은 160일이 적합하였으나 재식밀도는 주황미의 경우 특히 70×20 cm 재배 간격에서 베타카로틴의 함량이 다른 재배간격에서 베타카로틴의 함량에 비해 가장 높게 나타났으며, 신자미의 경우는 70×30 cm 또는 70×35 cm에서 300 g이상 괴근의 유용성분 총 함량이 높은 경향을 나타내었다

헛개나무과병추출물 중 퀘르세틴 분석 (Analysis of quercetin in extract of Hovenia dulcis Thunb)

  • "건강기능식품에 관한 법률"에 따라 건강기능식품이란 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조한 식품을 말한다. 또한, 건강기능식품은 고시화한 원료와 영업자가 개별적으로 신청한 개별인정형 원료로 나누어진다. 그 중 개별인정형 원료의 시험법은 고시화를 위해 표준화가 필요하며 2가지 이상의 원료가 복합적으로 혼합되어 제조될 경우 제품에 대한 적용성 검토가 요구된다. 개별인 정형 원료 중 헛개나무과병추출물의 지표성분인 퀘르세틴을 대상으로 전처리방법을 표준화하였으며, 표준화한 퀘르세틴 시험법의 밸리데이션을 수행한 결과 회수율은 97.1~105.4%, 표준편차는 1.15~3.11%였다. 검량선의 직선성은 결정계수($R^2$)가 0.999 이상을 나타냈고 검출한계는 $0.12{\mu}g/mL$, 정량한계는 $0.36{\mu}g/mL$를 나타냈다. 표준화한 퀘르세틴 시험법의 적용성 검토를 위해 헛개나무과병추출물의 유통제품을 수거하여 분석한 결과, 퀘르세틴 표시 함량 대비 82.5~105.1%로 적합하였다.

발아 채소 추출물의 항산화 활성 비교 (The Comparison of Antioxidative Activities of Sprouts Extract)

  • This study was carried to develop the new material of functional food which antioxidant of natural substances of sprouts. We compared antioxdiative activity and antioxidant substances exist in sprouts. Antioxidaive activities of sprouts were measured by total polyphenolic acid contents, electron donating activity(EDA), SOD-liked activity and hydroxyl radical scavenging ability. Total polyphenolic acid content was very higher in Rape sprouts extract(Rap) than other sprout extracts. Also Rape extract was showed the most excellent antioxdiative activity in SOD liked activity at 86.94%. The EDA was ordered Rape sprouts extract(Rap)>Tatsoi sprouts extract(Tat)>Broccoli sprouts extract(Bro)>Alfalfa sprout extract(Alf). Hydroxy radical scavenging ability was the most effective in Rape sprouts extract(Rap). Therefore we could be certain that Rape sprouts extract(Rap) was the most effective in antioxidative activity from sprouts.

Saengmaeg-san as an ergogenic aid: improving exercise performance

  • Sports drinks help optimize and improve performance by delaying and eliminating the buildup of fatigue-causing substances in the body during exercise. Saengmaeg-san is a nature-friendly traditional beverage that has no side effects on the human body and can quench thirst. However, studies on the relationship between exercise ability and Saengmaeg-san are insufficient. The purpose of this study was to prescribe Saengmaeg-san during the summer training period of 4 weeks and to analyze the effect on body composition and exercise performance. Seventeen male participants were divided into 3 groups (Saengmaeg-san acid intake group [n=9], placebo group [n=8]), and body composition (height, weight, muscle mass, fat mass, BMI) and conducted exercise performance (total exercise time and HRmax). In our study, Saengmaeg-san intake had a positive effect on exercise performance, such as decreased body fat percentage, increased exercise time, and decreased HRmax. Therefore, Saengmaeg-san showed the potential as a sports drink. In the future, additional studies on fatigue-related substances, immune function-markers, and blood lipids are needed in order to clearly explain the change in exercise performance due to consumption of Saengmaeg-san.

물오리나무 유래 고함량 Oregonin 추출방법 및 생리활성 연구 (Extraction Method and Physiological Activity of High Content Oregonin Derived from Plant of Alnus sibirica Fisch. ex Turcz)

  • Oregonin is known as a representative active substance of plants of the Alnus species. In this study, we will show the results of establishing a method for extracting trace amounts of useful substances in plants. Additionally, a variety of physiological activity studies were conducted with extracts containing high content of useful substances. In this study, anti-oxidant, anti-inflammatory and anti-allergic activities of supercritical fluid residue 60% EtOH extracts of Alnus sibirica (ASCFR) were investigated in vitro. To explore anti-oxidant and anti-inflammatory characteristics of supercritical extract, DPPH radical scavenging, and ABTS radical scavenging activity, inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells and ${\beta}$-hexosaminidase release in IgE-stimulated RBL-2H3 cells were examined, respectively. Results showed a concentration-dependent increase in DPPH and ABTS radical scavenging activity. Cells stimulated with LPS produced more nitric oxide than normal control cells. However, cells treated with supercritical fluid residue 60% EtOH extract decreased production in a concentration-dependent manner. In anti-allergic activity studies, ASCFR showed concentration-dependentanti-allergic activity. Based on results and strong antioxidant activity, ASCFR has potential as basic research materials for development of therapeutic supplements-based medicines or functional cosmetics related to chronic inflammatory skin immunity diseases.

김치 소비 패턴 및 기능성 김치에 관한 소비자 인식 (A Study on Kimchi Consumption Pattern and Consumer's Perception about the Functional Kimchi in Seoul and Gyeonggi-do Areas)

  • This study is conducted to investigate to the consumption pattern of Kimchi and perception about the functional Kimchi of consumer. The survey was done between October 1 to October 15, 2011 among 294 male and female adults aged 19 and over in Seoul and Gyeonggi-do areas. The gender distribution of subjects was 33.3% males and 66.7% females. 64.3% of subjects prepared Kimchi by themselves, 23.5% of subjects received Kimchi from relatives and 12.2% of subjects purchased Kimchi from the market. In addition, the rate of preparing Kimchi at home is highest in those aged fifty or over. Only 41.8% of subjects knew how to make Kimchi. 72.1% of subjects responded that they ate Kimchi one or more a day. 46.6% of subjects have purchased commercial Chinese cabbage Kimchi. The amount of one-time purchase of commercial Kimchi were investigated; 45.2% of subjects have been buying 500-1 kg, 34.4% of the subjects bought less than 500 g, and 11.2% of subjects bought 1-3 kg. 28.2% of subjects buy Kimchi at the supermarket and warehouse market. With regard to the evaluation of Kimchi taste, most consumers were not satisfied with the sweetness of Kimchi. In this result, the perception about functional Kimchi was very low. Consumer's demands were as follows: nutrient enhancement, strengthening of biologically active substances, lactic acid bacteria enhancement in order. Small sales units were preferred by the consumers, and complementation of sweetness of kimchi was required. Various Kimchi including functional Kimchi must be developed to meet the needs of consumers.

Functional characterization of gibberellin signaling-related genes in Panax ginseng

  • Gibberellins (GAs) are essential phytohormones for plant growth that influence developmental processes and crop yields. Recent functional genomic analyses of model plants have yielded good characterizations of the canonical GA signaling pathways and related genes. Although Panax ginseng has long been considered to have economic and medicinal importance, functional genomic studies of the GA signaling pathways in this crucial perennial herb plant have been rarely conducted. Here, we identified and performed functional analysis of the GA signaling-related genes, including PgGID1s, PgSLY1s, and PgRGAs. We confirmed that the physiological role of GA signaling components in P. ginseng was evolutionarily conserved. In addition, the important functional domains and amino acid residues for protein interactions among active GA, GID1, SCFSLY1, and RGA were also functionally conserved. Prediction and comparison of crystallographic structural similarities between PgGID1s and AtGID1a supported their function as GA receptors. Moreover, the subcellular localization and GA-dependent promotion of DELLA degradation in P. ginseng was similar to the canonical GA signaling pathways in other plants. Finally, we found that overexpression of PgRGA2 and PgSLY1-1 was sufficient to complement the GA-related phenotypes of atgid1a/c double- and rga quintuple-mutants, respectively. This critical information for these GA signaling genes has the potential to facilitate future genetic engineering and breeding of P. ginseng for increased crop yield and production of useful substances.
