Mammary Gland-Specific Expression of Biologically Active Human Osteoprotegerin in Transgenic Mice

  • Osteoprotegerin (OPG) is a secreted glycoprotein that regulates bone resorption by inhibiting differentiation and activation of osteoclast, thereby potentially useful for the treatment of many bone diseases associated with increased bone loss. In this study, we designed a novel cDNA expression cassette by modifying the potent and mammary gland-specific goat ${\beta}$-casein/hGH hybrid gene construct and examined human OPG (hOPG) cDNA expression in transgenic mice. Six transgenic mice all successfully expressed hOPG in their milk at the level of 0.06-2,000 ${\mu}g/ml$. An estimated molecular weight of the milk hOPG was 55 kDa in SDS-PAGE, which is the same as a naturally glycosylated monomer. This hOPG expression was highly specific to the mammary glands of transgenic mice. hOPG mRNA was not detected in any organs analyzed except mammary gland. Functional integrity of milk hOPG was evaluated by TRAP (tartrate-resistant acid phosphatase) activity assay in bone marrow cell cultures. OPG ligand (OPG-L) treatment increased TRAP activity by two fold but it was completely abolished by co-treatment with transgenic milk containing hOPG. Taken together, our novel cDNA expression cassette could direct an efficient expression of biologically active hOPG, a potential candidate pharmaceutical for bone diseases, only in the mammary gland of transgenic mice.

2-히드록시에틸 메타크릴레이트와 기능성 사이레인 단량체와의 공중합에 관한 연구 (Copolymerization of 2-Hydroxyethyl Methacrylate with Functional Silane Monomers)

  • 2-Hydroxyethyl methacrylate(HEMA) was copolymerized with allyltrimethylsilane- (ATMS) and allylt.imethoxysilane(ATMOS) at 70。C in N, N'dimethylformamide- (DMV) using $\alpha$ , $\alpha$'-azobisisobutyronitrile(AIBN). The compositions of unreacted monomers were determined by gas chromatographic analysis. The monomer reactivity ratios were determined by Fireman-Ross, Helen-Tiidus and intersection methods. The average values are as follows : $$r_1(HEMA) : 6.62 $\pm$ 0.07, r_2(ATMS) : 0.07 $\pm$ 0.01 for HEMA-ATMS system.$$ $$r_l(HEMA) : 4.09 $\pm$ 0.14, r_2(ATMOS) : 0.06 $\pm$ 0.01 for HEMA-ATMOS toys tem.$$ The lower rl(HEMA) In the HEMA-ATMOS system as compared to HEMA ATMS system may be contributed to higher relative reactivity of ATMOS toward the poly(HEMA) radical. The compositions of synthesized copolymers were determined from silicon con tents estimated gravimetrically. The thermal stabilities of the copolymers were investigated by thermogravimetry(TG). The enthalpic changes associated with the endothermic transition were evaluated by differential scanning calorimetry(DSC). The swelling properties of the copolymers in water were also investigated.

Polymerization and Physical Property Assessment of Optical Lens Materials Containing Amide Group

  • The basic hydrogel lens with addition of N,N-dimethylacrylamide (DMA) and N-methyl-N-vinylacetamide (NMV) were manufactured. The optical and physical characteristics of ophthalmic lens were evaluated by measuring water content, oxygen permeability (Dk), refractive index and optical transmittance. The water content & oxygen permeability (Dk) of sample containing Ref., DMA group and NMV group was in the average of 34.48%, 35.54~49.19% & $13.003{\sim}18.468{\times}10^{-11}(cm^2/sec)$($mlO_2/ml{\times}mmHg$) and 36.28~44.95% & $12.270{\sim}16.883{\times}10^{-11}(cm^2/sec)$ ($mlO_2/ml{\times}mmHg$), respectively. And also, refractive index of the sample containing Ref., DMA group and NMV group was in the average of 1.4350, 1.4330~1.4131 and 1.4335~1.4195, respectively. Standard hydrogel monomer containing DMA and NMV was expected to be used usefully as a material for fabricating hydrophilic functional ophthalmic lens.

메틸메타크릴레이트의 환경친화적인 광중합 및 열분해특성 (A Characteristics of Environmental Fraternitive Photopolymerization and Thermal Degradation on Methyl Methacrylate)

  • Photopolymerization, the utilization of electromagnetic radiation(or light) as the energy source for polymerization of functional monomers, oligomers is the basis of important commercial processes with broad applicability, including photoimaging and RV curing of coatings and inks. The objective of this study is to investigate the characteristics of environmental fraternitive photopolymerization of methyl methacrylate(MMA). This work is the first step to continue further research about alkyl methacrylate. The experiment was done in aqueous solution under the influence of photo-initiator concentration(0.05-0.25mol/l), light intensity (5000-9000 ${\mu}J/cm^2$) and monomer concentration(2-6mol/l). Methyl methacrylate was polymerized to high conversion ratio using hydrogen peroxide($H_2O_2$) and the kinetics model we have obtained is as follows. $R_p=k_p[S]^{0.41}[M]^{0.62}[L]^{2.45} exp(53.64/RT$). The differential method of thermogravimetric analysis(Friedman method) was used to obtain value of activation energy on decomposition reaction. The average value of it res 45.4Kca1/mol.

Corona방전처리에 의한 PVC film의 대전방지가공 (Antistatic Finishing of PVC Film Treated with Corona Discharge)

  • Polyvinyl chloride (PVC) sheets were treated with corona discharge to produce peroxy radicals on the surfaces. The peroxy radicals formed on the PVC surfaces were subsequently used as initiators for the graft polymerization of acrylic acid or acrylamide in an aqueous solution. Introduction of acrylic acid and acrylamide on the PVC sheet could be confirmed by the observation of carbonyl and primary amine absorptions based on carboxylic acid and amide, respectively. The water contact angle$(90^\circ)$ of PVC sheet was constant, irrespective of time, while corona-treated and functional monomer-grafted PVCs were slowly increased with time, showing the rearrangement of surface polar groups in air condition. The water contact angle of PVC sheet$(90^\circ)$ was decreased by corona treatment$(78^\circ)$, and further decreased by the grafting of acrylic acid$(55^\circ)$ and acrylamide$(56^\circ)$ , indicating increased hydrophilicity of the modified surfaces. The half-life periods of surface voltage on acrylic acid- (62 sec) and acrylamide-grafted PVC (147sec) were significantly decreased when compared to those on PVC (3,115 sec) and corona-treated PVC (463sec). These results mean that acrylic acid- and acrylamide-grafted PVCs could be used as the antistatic sheets.

음이온 교환막 제조를 위한 염화비닐벤질 고분자가 그라프트된 ETFE 필름의 방사선 합성과 디아민류와의 4차 아민화에 관한 연구 (A Study on the Radiolytic Synthesis of PVBC-grafted ETFE Films and Their Quaternarization with Diamines for the Preparation of Anion Exchange Membranes)

  • In this study, several anion exchange membranes were successfully prepared by radiation grafting of vinylbenzyl chloride (VBC) monomer onto a polyethylene-co-tetrafluoroethylene (ETFE) film and subsequent quaternarization with diamins such as 1,4-diazbicyclo[2,2,2]octane, N,N,N',N'-tetramethyl-1,3-diaminopropane, N,N,N',N'-tetramethyl-1,6-diaminohexane, and trimethylamine. The anion exchange capacities of the prepared membranes were determined by using acid-base titration method. The cross-sectional morphology of the membranes was studied by using SEM instrument. The cross-sectional distribution patterns of the grafted polymers with anion exchange functional groups were also studied by EDX analysis.

Study on the Characteristic of Elastomer Composite Containing Tungsten Powder

  • In order to develop an ultra-high-density elastomeric material for substitution of steel dynamic dampers, a new curing system and technique for high-loading of the filler were examined in this study. Mechanochemical modification of chloroprene rubber (MAH-g-CR) using an internal mixer was carried out with maleic anhydride (MAH) as a reactive monomer. The optimum amount of MAH was 10 phr and the efficient grafting of MAH on CR could be achieved at a mixing temperature of 100℃. After preparing MAH-g-CR, 50 mol% epoxidized natural rubber (ENR 50) was blended with MAH-g-CR to develop a "self-curable rubber blend system" via reaction between the functional groups of the elastomeric matrices without the curing agent and additives. The content of ENR 50 was fixed at 30 wt.% throughout evaluation of the curing behavior of the MAH-g-CR/ENR blend. Tungsten powder was added to the MAH-g-CR/ENR matrix up to 60 vol.% to obtain ultra-high-density, and the maximum density obtained was 7.57 g/㎤. Stable ts2 (scorch time) and t90 (90% cure time) could be obtained even when tungsten powder was incorporated up to 60 vol.%. In addition, the tensile strength and damping properties of MAH-g-CR/ENR containing 60 vol.% of tungsten were better than those of CR containing 60 vol.% of tungsten.

수분산성 아크릴계 점착제 중합 조건에 관한 연구 (Study on Polymerization Condition of Water-based Acrylic Adhesion)

  • 본 연구는 주단량체로 2-에틸헥실아크릴레이트(2-EHA), n-부틸아크릴레이트(n-BA), 메틸메타크릴레이트(MMA) 그리고 기능성 단량체로 아크릴산(AAc)을 사용한 유화중합에서 아크릴계 점착제를 합성하기 위한 최적의 합성 조건을 구하고자 한다. 이를 위하여 계면활성제 농도의 영향, 계면활성제 Hydrophilic lipophilic Balance(HLB) 값, 중합개시제의 농도와 완충제가 아크릴계 점착제의 물성에 미치는 영향을 연구하였다. 그 결과 계면활성제는 단량체 사용량에 대하여 3~5 wt%를 사용하였을 때, HLB 값이 12.3일 때 가장 안정한 점착체를 얻을 수 있었으며, 중합 개시제의 농도가 1 wt% 이상 사용하였을 때는 중합 속도는 증가하였으나 안정성이 저하되었다. 완충제로 sodium bicarbonate를 사용할 경우 0.4 wt% 이상 사용하여야 충분한 완충 효과가 있었으며, 단량체의 종류에 따라 약간의 차이는 있으나 완충제의 농도가 0.6~0.8 wt%가 적당하여 4성분계 유화중합에서 최적의 중합 조건을 제시할 수 있었다.

수용성 streptavidin의 Escherichia coli 에서 기능적 발현 (Functional Expression of Soluble Streptavidin in Escherichia coli)

  • Streptmyces avidinii에서 발현되는 Streptavidin은 vitamin H인 d-biotin 4분자에 결합하며 해리상수(Kd)가 10−15 M를 나타내는 아주 강한 비공유결합이다. 이러한 streptavidin과 biotin 상호간의 강한 결합력은 수많은 생물체 분자들의 탐지 및 특징을 연구하는데 응용되어져 왔으므로 Escherichia coli에서 수용성 streptavidin의 기능적 발현에 대한 연구는 매우 유용하다. 즉 Escherichia coli에서 streptavidin을 발현시키기 위해 streptavidin유전자를 T7 RNA polymerase/T7 promoter를 이용하는 pET-22b 플라스미드로 클로닝하였다. 또한 N-말단에 pelB leader를 포함하여 발현된 streptavidin의 periplasmic space로 운반하여 수용성 단백질형태의 분비를 촉진하였으며 C-말단에는 6개의 polyhistidine tags를 두어 정제하는데 사용되었다. 정제된 streptavidin단백질은 10-20 mg/ml 의 높은 회수율을 나타내었으며 SDS-PAGE에서 가열하는 경우 변성되어 17 kD인 monomer형태를, 가열하지 않는 경우에는 68 kDa으로 원래의 tetramer형태를 나타내었다. 따라서 streptavidin의 tetramer 구조는 비공유결합에 의해 이루어짐을 알 수 있었다. Size-exclusion chromatography에 의한 streptavidin의 구조 역시 tetramer를 재확인할 수 있었다. 정제된 수용성 streptavidin은 Westernblot실험에서 biotinylation된 단백질을 탐지하였으며 이 결과는 정제된 streptavidin이 biotin에 결합하는 기능이 존재함을 나타내었다. 이상의 모든 결과를 종합해보면 본 연구에서 구축된 발현시스템을 통하여 발현된 streptavidin은 높은 회수율을 나타내어 대량생산이 가능하였으며 자연상태의 streptavidin과 동일한 homotetramer를 형성하고 biotin에 결합할 수 있는 기능을 나타내었다.

식품연포장용 라미네이트 수성 감압점착제의 친환경적 적용에 대한 연구 (Design and Properties of Laminating Waterborne PSA for Eco-friendly Flexible Food Packaging)

  • 본 연구는 식품연포장 적층 라미네이트에 주로 사용되는 유성접착제를 대체하기 위해 아크릴 수성화방법을 활용하여 친환경적인 수성 점착제를 설계하였다. 방법으로는 음이온/비이온 그리고 인산에스테르 계면활성제들의 조합과 분자량조절제(chain transfer agent, CTA)의 양을 달리하고, 기능성단량체(functional monomer)의 종류를 달리하였다. 합성된 점착제의 가교 정도와 분자량의 영향을 각각 실험군 8종으로 점착제를 설계 합성하고 조사하였다. 그리고 별도로 분자량이 서로 다른 styrene/alpha-methyl styrene/acrylic acid (SAA) semipolymer 분산제(Mw = 13,000과 Mw = 8,600)를 기존의 계면활성제를 대신하여 사용해 감압점착제(Pressure Sensitive Adhesive, PSA)의 물성을 조사하였다. 합성된 10종의 감압점착제가 유연포장용 기본 물성 규격에 적합한지를 DSC, 입자크기, adhesion, 분자량의 물성 분석으로 평가하였다. 실험 결과 계면활성제는 음이온/비이온 조합으로, CTA는 0.2%로 기능성 단량체는 HEA, GMA를 사용한 점착제 2개의 실험군에서 유연포장에 적용 가능한 분자량과 유연성, 낮은 접착력, 작은 입자의 물성을 확인하였다.
