한약재를 이용한 음료의 개발 및 혈관개선에 미치는 기능성 평가 (Development of Drink from Composition with Medicinal Plants and Evaluation of its Physiological Function in Aorta Relaxation)

  • The purpose of this study was to investigate the possibility of medicinal plants application as an edible functional food resources. We carried out to develop a traditional functional beverage by using hot-water extraction of 4 medicinal plants(Polygonatun sibiricum, Ophiopogonis radix, Lycii fructus, Schizandriae fructus) and we examined the effects of drink on physiological function in aorta relaxation. Thus, the effect of developed beverage on phenylephrine induced contraction of isolated rat thoracic aorta. Contractile force was measured with force displacement transducer under 1.5g loading tension. Brix, pH and titratable acidity of developed drink were 9.5%, 3.3 and 0.22%. The approximate nutritional composition of beverage was carbohydrate, 5.98%, crude protein, 0.70%, crude fat, 0.20% and crude ash, 0.20%. Developed beverage contained K(4.00 ㎎%), Na(3.68 ㎎%), Ca(2.54 ㎎%), Mg(1.60 ㎎%) and Fe(0.29㎎%). The contraction forces by injection of phenylephrine in isolated thoracic aorta were significantly low in each experimental groups compared with control groups, These results that developed drink with medicinal plants can be used as a functional material to decrease aorta contraction.

국내산 키위의 색상별 항산화능 및 아질산염 소거능 비교 (Comparison of Antioxidant and Nitrite Scavenging Activities of Different Colored Kiwis Cultivated in Korea)

  • This study was to conducted to investigate the antioxidative activity of 70% ethanol extracts of kiwi of three different colors (gold, green and red) by measuring DPPH, ABTS radical scavenging activity, tyrosinase inhibitory effect, metal chelating effect, reducing power, and nitrite scavenging activity. Total polyphenol contents were: gold kiwi, 3.09 mg GAE/g, green kiwi 2.71 mg GAE/g, and red kiwi 4.59 mg GAE/g, respectively. Red kiwi showed higher antioxdidant activity than gold and green kiwi. DPPH and ABTS radical scavenging activity, and nitrite scavenging activity of red kiwi exhibited 94.83, 99.57, and 97.88%, respectively, at a concentration of 20 mg/mL, which were equal to those of ascorbic acid (positive control). Metal chelating effect of red kiwi was superior to that of ascorbic acid. Therefore, the availability of red kiwi will be increased in the field of functional material for food additives and value added products.

Fermented Kochujang Supplement Shows Anti-obesity Effects by Controlling Lipid Metabolism in C57BL/6J Mice Fed High Fat Diet

  • The aim of the present study was to assess the anti-obesity effects of fermented kochujang supplement in C57BL/6J mice. Thirty mice were divided into 3 groups; normal diet control group (ND), high fat diet control group (HD), and high fat diet plus kochujang supplemented group (HDK). Results were as follows: 1. Fennented kochujang supplement in high fat diet decreased body weight and epidydimal and back fat weight compared to non-supplement in HD group. 2. Lipid content and blood glucose level were lower in HDK group than HD group. 3. Fermented kochujang supplement increased mRNA level of lipolytic genes such as acyl-CoA synthetase (ACS), carnitine palmitoyltransferase-1 (CPT-1), and uncoupling proteins-1 (UCP-1) expression, whereas decreased mRNA level of adipogenic genes such as acetyl CoA carboxylase (ACC) expression. These findings suggest that fermented kochujang supplement in high fat diet normalized body weight, epididymal and back fat weight, lipid content, and blood glucose levels through controlling lipid metabolism and provides basic information on the control of obesity.

스피루리나 첨가 쌀엿강정의 항산화 특성 (Antioxidant Activities of Riceyeotgangjung with Added Spirulina Powder)

  • The objective of this study was to develop high quality Riceyeotgangjung (RYG) with high antioxidant activity and a longer shelf-life. Spirulina (1, 2, and 3%) with added RYG was prepared, and antioxidant activities were examined under storage at $60^{\circ}C$ for 15 days. The total phenol content remaining in the spirulina with added RYG at 15 days of storage was much higher than that of fresh RYG without spirulina. The IC50 values for DPPH and hydroxyl radical scavenging activities decreased with increasing amounts of spirulina, whereas these values increased with storage time. TBA and peroxide values decreased with increasing amounts of spirulina. From these results, adding spirulina to RYG delayed lipid rancidity and prolonged shelf-life. Adding spirulina powder to RYG, which is a healthy functional food material, improved antioxidant activity and delayed deterioration during storage.

병풀(Centella asiatica) 착즙액과 추출물의 Asiaticoside 분석법 검증 및 항산화 활성 (Validation of Asiaticoside as Marker Compound of Centella asiatica Juice and Extract, and Its Antioxidant Activity)

  • Centella asiatica (C. asiatica) has been widely used in food, cosmetics, and pharmaceutical industry as a functional material. In a previous study, we have investigated not only pharmacological effects such as antioxidative and anti-inflammatory effects, but also analyzed various functional ingredients. In this study, triterpenoids were analyzed using HPLC-DAD to determine marker compounds among functional ingredients. When triterpenoids were analyzed, asiaticoside from C. asiatica was determined as an optimal marker compound. Next, specificity, linearity, limited of detection (LOD), limited of quantification (LOQ), precision, accuracy, and range were evaluated using HPLC-DAD to determine asiaticoside contents in C. asiatica juice and extracts. The specificity was elucidated by chromatogram and retention time using an established analytical method. The coefficient of correlation obtained was 0.9996. LOD was 4.99 ㎍/mL and LOQ was 15.12 ㎍/mL. Intra- and inter-day precision of asiaticoside were determined to be 0.48~1.68% and 0.08~1.09%, respectively. Furthermore, the recovery rate of asiaticoside was 98.88% and the analytical range of Field-70E was determined to be 0.625~10 mg/mL. As a results of evaluating ABTS, DPPH, and FRAP antioxidative effect, Field-70E showed potent antioxidant activities. Results of this study could be used as basic data for quality standardization of C. astiatica juice and extracts.

Fluorescence-labelling for analysis of protein in starch using asymmetrical flow field-flow fractionation (AF4)

  • Starch is a mixture of amylose (AMY) and amylopectin (AMP) which are different in physical properties such as molar mass (M), rms radius ($R_g$) and hydrodynamic diameter ($d_H$). The rheological and functional properties of starch are influenced by various factors including the molecular size, molar mass distribution (MD) and the concentration ratio of AMY and AMP. It is also important to analyze proteinaceous material in starch as they affect the flavor and texture of food to which starch is added. In this study, asymmetrical flow field-flow fractionation (AF4) was employed for separation and quantitation of AMY and AMP in starches (Amaranth, potato, taros and quinoa). AF4 was coupled with a multi-angle light scattering (MALS) and a refractive index (RI) detector for determination of the absolute M, MD and molecular structure. It was found that AMP has the M and $R_g$ ranging $3.7{\times}10^7{\sim}6.5{\times}10^8g/mol$ and 84 ~ 250 nm, respectively. Also the existence of branch was confirmed in higher M. In addition, proteinaceous material in starch was analyzed by AF4 coupled with a fluorescence detector (FS) after fluorescence-labeling. AF4-FS with fluorescence-labelling showed a potential for investigation on existence of proteinaceous material and the interaction between proteinaceous material and polysaccharide in starch.

기능성 골판지 상자로 포장한 '신고' 배의 신선도 유지효과 (Freshness Preserving of 'Shingo' Pear using Corrugated Paperboard Box Laminated with Functional MA Film)

  • 골판지상자에 기능성 필름을 첨합한 포장상자(FC)의 배 저장 중 품질유지 상태를 조사하고자 대조구로 기존에 유통중인 골판지 상자와 0.02, 0.04mm의 LDPE필름과, 0.02와 0.04mm의 기능성MA필름(CE)에 배를 포장하여 5 C에 60일간 저장하면서 신선도 변화를 비교 고찰하였다. 중량변화는 대조구는 2.9% 감소했으며, FC, LDPE와 CE 포장구들은 1.0-0.5% 감소했다. 경도는 각 포장방법간에 차이가 없었으며 비타민C 함량도 필름포장구들과 FC 포장구에서 대조구보다 더 높게 유지되고 있었다. 산도와 당도는 대조구가 필름포장구들과 FC 포장구 보다 변화가 심하게 나타났고 전체적인 외관상의 기호도 FC 포장구와 필름포장구에서 양호한 것으로 나타났다. 이상의 결과에서 FC 포장재를 신선도 유지에 활용할 수 있을 것으로 판단되었다.

땅콩나물 추출물의 항산화 효과 (Antioxidative Effects of Peanut Sprout Extracts)

  • 땅콩나물을 기능성식품 소재로 활용하기 위하여 충북, 경북, 전북, 중국 및 베트남산 땅콩 및 이들을 9일 동안 발아시킨 땅콩나물 메탄올 추출물의 폴리페놀 함량과 항산화 활성을 측정하였다. 땅콩나물 추출물은 땅콩 추출물에 비하여 폴리페놀 함량이 높았으며, 특히 경북 땅콩나물 추출물에서 20.4 mg/g으로 가장 높게 나타났다. 9일간 발아시킨 땅콩나물 추출물은 모두 땅콩 추출물에 비하여 그 활성이 높게 나타났으며, 특히 경북 땅콩나물 추출물의 활성이 $200\;{\mu}L/mL$ 농도에서 37.68%로 가장 높게 나타났다. 또한 환원력의 경우도 이와 유사한 경향이었다. 경북 땅콩나물에서 잎, 줄기 및 뿌리의 부위별 추출물에 대한 수소공여능을 측정한 결과 잎 추출물의 활성이 가장 높았으며, 그 활성은 $200\;{\mu}L/mL$ 농도에서 90%로 높게 나타났다. 또한 ABTS radical 및 $\beta$-carotene bleaching 활성 모두 줄기나 뿌리 추출물에서의 활성보다 잎 추출물에서 우수하였다. 경북 땅콩 및 땅콩나물의 resveratrol 함량의 분석한 결과 땅콩나물에서의 그 함량이 $15.05\;{\mu}g/g$로서 땅콩에서의 $1.42\;{\mu}g/g$에 비하여 그 함량이 높게 나타났다. 따라서 본 연구결과를 고려해볼 때 땅콩나물을 항산화 기능을 함유한 기능성식품 소재로 활용할 수 있을 것으로 생각된다.

해조류 가공식품 및 부산물을 이용한 제품 개발 (Development of Value-Added Products Using Seaweeds)

  • 국내 해조류 자원의 이용성 증대를 위해서 해조류의 기능성, 유산균 생육 특성, 가공식품에의 이용, 생분해성 포장재의 소재, 그리고 부산물을 이용한 동물사료화 및 퇴비화 가능성의 연구 보문을 조사하였다. 기능성물질 연구에서는 유용색소 (Fucoxanthin)의 추출과 부위별 함량 및 분광학적인 특성이 있으며, 해조류로부터 alginate의 함량 조사와 최적추출조건 확립에 대한 연구를 수행하여 AASA (Acid alkali soluble alginate) 추출방법에서 3% $Na_2CO_3$ 농도와, $H_2SO_4$의 농도 0.4 N에서 가장 높은 수율을 얻었다. 톳으로부터 산알칼리 (AASA) 방법으로 추출한 알긴산에 인위적으로 sulfate를 흡착 Lactobacillus acidophilus를 접종한 후 배양하면서 유산균의 성장에 대한 영향을 살펴 본 결과 모든 농도에서 이들 배지에 S-alginate를 첨가한 것이 유산균의 성장을 증가시켰다. 미역귀 추출물로 잼을 제조하였고, 김, 미역, 다시마의 물 추출물로 물성이 우수하고 관능성적도 양호한 젤리를 제조하였다. 해조류 젤리를 상품화하는데 있어서 해조류가 갖는 해조취와 젤리색소의 안정화가 상품화의 요소로 대두되어 이를 개선할 방법도 제시되었다. 해조 간장과 된장 및 두부에서는 다당류나 무기질 등 영양적인 기능성과 관능적 기호성을 향상시킬 수 있었다. 알긴산 필름의 물성 증진을 위하여 $CaCl_2$를 필름용액에 직접 첨가 또는 필름을 $CaCl_2$용액에 침지하는 두 가지 방법에서 수분 저항성이 강한 필름을 제조할 수 있었다. 미이용 해조분말이나 해조가공 부산물로 얻어지는 해조분말을 이용하여 새로운 생분해성 포장소재로 사용할 수 있는 가능성이 있다. 해조류의 영양성분 및 아미노산 함량을 분석한 결과 사료첨가제로서 충분한 가치가 있었으나 다량의 염분 함량으로 인하여 그 가치가 평가 절하되었다. 따라서 해조류의 사료적 가치를 증진시키기 위하여 발효처리를 하였으며, 그 결과 기능성 영양소 (불포화 지방산)가 증가되는 효과가 있었다.
