Osteoporosis develops with high prevalence in both postmenopausal women and hypogonadal men. Osteoporosis results in significant morbidity, but no cure has been established. Mesenchymal stem cells (MSCs) critically contribute to bone homeostasis and possess potent immunomodulatory/anti-inflammatory capability. Here, we investigated the therapeutic efficacy of using an infusion of MSCs to treat sex hormone-deficient bone loss and its underlying mechanisms. In particular, we compared the impacts of MSC cytotherapy in the two genders with the aim of examining potential gender differences. Using the gonadectomy (GNX) model, we confirmed that the osteoporotic phenotypes were substantially consistent between female and male mice. Importantly, systemic MSC transplantation (MSCT) not only rescued trabecular bone loss in GNX mice but also restored cortical bone mass and bone quality. Unexpectedly, no differences were detected between the genders. Furthermore, MSCT demonstrated an equal efficiency in rectifying the bone remodeling balance in both genders of GNX animals, as proven by the comparable recovery of bone formation and parallel normalization of bone resorption. Mechanistically, using green fluorescent protein (GFP)-based cell-tracing, we demonstrated rapid engraftment but poor inhabitation of donor MSCs in the GNX recipient bone marrow of each gender. Alternatively, MSCT uniformly reduced the $CD3^+T$-cell population and suppressed the serum levels of inflammatory cytokines in reversing female and male GNX osteoporosis, which was attributed to the ability of the MSC to induce T-cell apoptosis. Immunosuppression in the microenvironment eventually led to functional recovery of endogenous MSCs, which resulted in restored osteogenesis and normalized behavior to modulate osteoclastogenesis. Collectively, these data revealed recipient sexually monomorphic responses to MSC therapy in gonadal steroid deficiency-induced osteoporosis via immunosuppression/anti-inflammation and resident stem cell recovery.

Sarcopenia, a common clinical syndrome in older adults, is defined as decreased muscle mass, strength, and physical performance. Since sarcopenia is associated with the incidence of functional decline, falls, and even mortality in older adults, researchers and health care providers have been keen to accumulate clinical evidence to advocate the screening and prevention of sarcopenia progression in older adults. The factors that may accelerate the loss of muscle mass and function include chronic diseases, inactivity, and deficiency in appropriate nutritional support. Among these, nutritional support is considered an initial step to delay the progression of muscle wasting and improve physical performance in community-dwelling older adults. However, a nationwide study suggested that most Korean older adults do not consume sufficient dietary protein to maintain their muscle mass. Furthermore, considering age-associated anabolic resistance to dietary protein, higher protein intake should be emphasized in older adults than in younger people. To develop a dietary protein recommendation for older adults in Korea, we reviewed the relevant literature, including interventional studies from Korea. From these, we recommend that older adults consume at least 1.2 g of protein per kg of body weight per day (g/kg/day) to delay the progression of muscle wasting. The amount we recommend (1.2 g/kg/day) is 31.4% higher than the previously suggested recommended daily allowance (i.e., 0.91 g/kg/day) for the general population of Korea. Also, evidence to date suggests that the combination of exercise and nutritional support may enhance the beneficial effects of protein intake in older adults in Korea. We found that the current studies are insufficient to build population-based guidelines for older adults, and we call for further researches in Korea.
본 연구는 슈퍼홍미 미강 추출물이 폐경 후 갱년기 증상에 및 대사 개선 효과를 확인하고자 수행되었다. 무월경이 지속된 지 5년 미만의 여성을 대상으로 12주동안 진행되었으며 위약대조군 15명, 실험군 15명으로 배정하였고, 1일 2개의 캡슐을 섭취하도록 하였다(슈퍼홍미 미강추출물 700 mg/day). 시험 종료 후 슈퍼홍미 미강 추출물 섭취군의 체중과 BMI 그리고 중성지질과 및 총콜레스테롤은 수준이 유의하게 감소하였다. 혈중 HDL 콜레스테롤과 ApoA1 농도는 약 10% 증가하여 지질대사 개선에 효과를 기대할 수 있었다. 항당뇨과 관련된 지표 중 혈중 glucose와 인슐린이 유의하게 감소하여 인슐린 저항성 지표인 HOMA-IR이 감소한 것을 확인할 수 있었고, 아디포넥틴 수준이 유의하게 증가하였으며, $TNF-{\alpha}$는 슈퍼홍미 미강 추출물 섭취군에서 투여 전보다 유의하게 감소하여 폐경 이후 당질대사 개선에 도움을 줄 수 있을 것으로 보인다. 항산화활성 분석 결과 SOD1, GSH 그리고 TBARS 수준이 실험군에서 감소하였으며, AOPP 는 유의적인 차이를 보이지 않았다. 여성호르몬 중 $17{\beta}-estradiol$과 progesterone 농도는 위약대조군에서 유의하게 감소하였으나 실험군은 섭취기간동안 호로몬 농도를 유지한 것으로 나타나 대조군에 비해 높은 수준을 나타났다. 폐경을 겪은 여성들은 여성호르몬 저하로 인해 갱년기 증상 및 대사증후군 발병 가능성이 증가하는데 슈퍼홍미 미강과 같은 기능성 식품 섭취로 대사증후군 위험인자들에 대한 호전이 높을 것이라 보고 이러한 연구 결과는 갱년기 여성 건강에 시사하는 바가 크다고 생각된다.
노화에 따른 난소 기능 저하와 성호르몬 생성 감소는 호르몬 결핍을 유발하여 여성의 갱년기를 유발한다. 여성 갱년기 증상 개선을 목적으로 에스트로겐 유사 물질이 함유된 천연물을 연구하였다. 본 연구는 난소 적출술을 받은 쥐를 대상으로 Gastrodia elata Blume 추출물(TVB-1000)의 효능을 평가하였다. 난소 적출 수술을 한뒤 7일의 회복 기간 후 TVB-1000 처리군에게 16, 40, 100 mg/kg을 12주 동안 경구 투여하였다. OVX 양성 대조군에는 17β-estradiol을 10 ㎍/kg의 용량으로 쥐의 등 부위에 피하 주사하였다. 그 결과 TVB-1000 처리군에서 심혈관질환 지표인 TG와 LDL-C가 모든 농도에서 유의하게 감소했다. LDL-C/HDLC 비는 TVB-1000 투여군 중 Medium, High군에서 유의하게 감소하였다. 또한 TVB-1000 고농도 처리군에서 복부지방 무게가 통계적으로 유의하게 감소하였다. ER- α의 발현량은 저농도 및 중농도 투여군에서 통계적 유의성을 보이며 증가하였으나 ER-β는 유의한 변화를 보이지 않았다. 실험을 통해 TVB-1000은 에스트로겐 유사 효과로 인해 갱년기 증상 중 하나인 심혈관계 질환을 예방하는 기능성 식품 소재로써 유용할 수 있음을 보여준다.
생체 면역조직에서는 면역세포의 성장, 분화에 있어서 매우 중요한 역할을 수행하는 면역조직 기질세포가 존재하며, 이들은 서로 연결된 3차원적인 그물구조를 형성하면서 그 사이의 공간에 위치한 면역세포와의 상호작용을 통해 다양한 면역반응을 수행한다. 따라서 생체환경을 모사한 면역세포의 배양이 이루어지기 위해서는 면역세포들이 상호작용할 수 있는 3차원적 면역조직 기질세포 뼈대의 구축이 매우 중요한 의의를 지닌다. 특히 면역반응에서 핵심적인 기능을 수행하는 T세포의 생존, 성장 및 분화에 있어서 필수적인 역할을 하는 흉선상피세포에 대한 3차원적 배양은 T세포의 연구에 필수적으로 요구되지만, 아직 이에 관한 연구가 거의 이루어지지 않은 실정이다. 본 연구에서 흉선상피세포는 폴리도파민으로 코팅된 PCL 및 PCL/PLGA 지지체에서 비코팅군에 비해 부착 및 성장이 촉진되었다. 또한 폴리도파민으로 코팅된 지지체에서 흉선상피세포를 배양하였을 때 2차원 배양군에 비해 흉선세포형성촉진인자의 유전자 발현이 더 증가하였다. 따라서 본 연구는 면역조직 기질세포의 3차원 배양 기술의 개발에 크게 기여할 수 있을 것으로 사료된다.
Objective: p66Shc, a 66 kDa protein isoform encoded by the proto-oncogene SHC, is an essential intracellular redox homeostasis regulatory enzyme that is involved in the regulation of cellular oxidative stress, apoptosis induction and the occurrence of multiple age-related diseases. This study investigated the expression profile and functional characteristics of p66Shc during preimplantation embryo development in sheep. Methods: The expression pattern of p66Shc during preimplantation embryo development in sheep at the mRNA and protein levels were studied by quantitative real-time polymerase chain reaction (RT-qPCR) and immunofluorescence staining. The effect of p66Shc knockdown on the developmental potential were evaluated by cleavage rate, morula rate and blastocyst rate. The effect of p66Shc deficiency on reactive oxygen species (ROS) production, DNA oxidative damage and the expression of antioxidant enzymes (e.g., catalase and manganese superoxide dismutase [MnSOD]) were also investigated by immunofluorescence staining. Results: Our results showed that p66Shc mRNA and protein were expressed in all stages of sheep early embryos and that p66Shc mRNA was significantly downregulated in the 4-to 8-cell stage (p<0.05) and significantly upregulated in the morula and blastocyst stages after embryonic genome activation (EGA) (p<0.05). Immunofluorescence staining showed that the p66Shc protein was mainly located in the peripheral region of the blastomere cytoplasm at different stages of preimplantation embryonic development. Notably, serine (Ser36)-phosphorylated p66Shc localized only in the cytoplasm during the 2- to 8-cell stage prior to EGA, while phosphorylated (Ser36) p66Shc localized not only in the cytoplasm but also predominantly in the nucleus after EGA. RNAi-mediated silencing of p66Shc via microinjection of p66Shc siRNA into sheep zygotes resulted in significant decreases in p66Shc mRNA and protein levels (p<0.05). Knockdown of p66Shc resulted in significant declines in the levels of intracellular ROS (p<0.05) and the DNA damage marker 8-hydroxy2'-deoxyguanosine (p<0.05), markedly increased MnSOD levels (p<0.05) and resulted in a tendency to develop to the morula stage. Conclusion: These results indicate that p66Shc is involved in the metabolic regulation of ROS production and DNA oxidative damage during sheep early embryonic development.
연구배경 : 결핵의 방어면역에서 IFN-${\gamma}$는 매우 중요한 역할을 한다. IFN-${\gamma}$ 수용체의 완전결손이 있는 환자는 Mycobacterium 감염에 감수성이 높아서 매우 치명 적이다. 매우 드문 일부 Mycobacterium 감염에서 IFN-${\gamma}$ 수용체의 완전결손 외에 여러 종류의 부분결손들과 STAT1의 돌연변이들이 알려져 있다. 특히, IFN-${\gamma}$ 수용체의 부분결손이 있는 환자들의 경우 병리학적 소견파 임상적 경과가 일반적 결핵과 유사한 소견을 보이는 것으로 알려져 있다. 방 법 : 임상적 결핵환자에서 IFN-${\gamma}$ 수용체의 기능을 평가하여 대조군과 비교하였다. IFN-${\gamma}$ 수용체의 이상이 있을 가능성이 가장 높은 파종성 결핵환자의 DNA에서 앞서 발표된 증례들에서 확인된 IFN-${\gamma}$ 수용제와 STAT1의 유전적 이상을 확인하였다. 결 과 : 말초혈액 단핵구를 recombinant IFN-${\gamma}$ (100, 1000IU/ml)로 자극하였을 때 대조군과 환자군에 모두 HLA-DR과 CD64의 발현이 증가하였으나, 두 군간의 차이는 없었다. 대조군과 환자군에서 모두 LPS자극하였을 때 TNF-${\alpha}$의 생산이 증가하였으며 IFN-${\gamma}$로 전처치 후에 다시 LPS로 자극하였을 때 TNF-${\alpha}$의 생산은 더욱 더 증가하였다. 그러나, 두 군간의 통계적으로 유의한 차이는 없었다. 파종성 결핵 환자에서 기존에 알려진 IFNgR1과 STAT1의 돌연 변이를 확인하였을 때 특이한 이상소견을 발견할 수 없었다. 결 론 : 임상적 결핵에서 IFN-${\gamma}$ 수용체의 기능적 및 유전적 이상을 확인할 수 없었다. 기존의 연구에서 BCG와 NTM 감염과 관련 있는 것으로 알려진 IFN-${\gamma}$수용체의 이상 만으로는 임상적 결핵의 개체감수성을 설명할 수 없었다.
Objectives : Parkinson's disease is a well-known neurodegenerative disease characterized by dopaminergic cell death in the substantia nigra. The reactive gliosis by activated astrocytes and microglias is no more regarded as a simple sequel of neuronal cell death. Microglial activation takes place in a stereotypic pattern with graded morphologic and functional(resting, activated and phagocytic) changes. In Parkinson's disease animal model, the degree of microglial activation along the nigro-striatal dopaminergic tract has not been studied intensively. The purpose of this study was to elucidate the characteristics of microglial reaction and to grade its degree of activation at substantia nigra and corpus striatum using 6-hydroxydopamine induced rat model of Parkinson's disease. Methods : Using Sprague-Dawley rat, parkinsonian model was made by 6-hydroxydopamine(OHDA) induced destruction of medial and lateral substantia nigra(SN). The rat was sacrificed 3-, 5-, 7-, 14- and 21-day-after operation. For control group, we injected saline with same manner and sacrificed 3-day after operation. With immunohistochemistry, we examined dopaminergic neuronal cells and microglial expression using tyrosine hydroxylase (TH) and OX-42 antibodies, respectively. Also we performed in situ hybridization for osteopontin, a possible marker of subset in activated microglia. Results : 1) In lesioned side of substantia nigra and corpus striatum, the TH immunoreactivity was markedly decreased in whole experimental groups. 2) Using optical densitometry, microglia induced immunoreactivity of OX-42 was counted at SN and corpus striatum. At SN, it was increased significantly on the lesioned side in control and all time-dependent experimental groups. At striatum, it was increased significantly in post lesion 3-day group only(p <0.05). Compared to control group, immunoreactivity of OX-42 on lesioned side was increased in groups, except post lesion 21-day group, at SN. Only post lesion 3-day group showed significance at striatum(p <0.05). Compared to SN region, immunoreactivity of OX-42 was much weaker in striatum. 3) Microscopically, the microglias showed typically different activation pattern. At SN, numerous phagocytic microglias were found at pars compacta and reticularis of lesion side. At striatum, no phagocytic form was found and the intensity of staining was much weaker. 4) At SN, the immunoreactivity of osteopontin showed definite laterality and it was markedly increased at pars compacta of lesion side with relatively short duration time. At striatum, however, it was not detected by in situ hybridization technique. Conclusion : The nigral 6-OHDA induced rat model of Parkinson's disease revealed several characteristic patterns of microglial reaction. At SN, microglias was activated shortly after direct neuronal damage and maintained for about three weeks. In contrast, despite of sufficient dopaminergic insufficiency at striatum, activation of microglias was trivial, and distinguished 3 day later. Antegrade slow neuronal degeneration is major pathophysiology in striatal dopaminergic deficiency. So, the acuteness of neuronal damage and consequential degree of neuronal degeneration may be important factor for microglial activation in neurodegenerative diseases such as Parkinson's disease. Additionally, osteopontin may be a possible marker for several subsets of activated microglia, possibly the phagocytic form.
음주 후 숙취는 사람에 따라 증상과 정도가 상이 하지만 일반적으로 갈증, 피로감, 두통, 전신권태, 위장장애, 구토, 설사, 비타민 결핍 등이 나타난다. 이러한 숙취현상은 간세포와 체내에 축적된 알코올 및 알코올에 함유되어 있는 발효 부산물인 에틸아세테이트, 아세트알데히드 등의 전구물질들의 작용에 의해 발생한다. 이에 숙취현상을 해소하기 위한 연구로 연어 추출물 중 정액 또는 정소 추출물인 핵산-당-인산으로 이루어진 고분자이며 다당체인 Polydeoxyribonucleotide(PDRN)과 동일한 Origin, 동일 구조를 가지며 PDRN에 비해 가격이 저렴한 수용성 연어 추출물 분말을 사용하였다. 그리고 D-Glucuronic acid와 N-Acetyl glucosamine으로 구성된 고분자 다당체로 뛰어난 생체 적합성, 점탄성 및 보습력을 보유하고 있어서 숙취현상 중 나타나는 생체수분 감소량 및 피부수분 함유량저하에 대한 부분에 효과가 있어 항산화 및 피부보습효과를 제공하는 히알루론산(Hyaluronic acid)을 감마선으로 조사하여 사용하였다. 연어추출물 분말과 히알루론산 등을 이용하여 조성물을 제조한 후 체내 에탄올, 아세트알데히드 감소량, 혈중 아세트알데히드 농도와 초산 농도를 측정하여 알코올 분해효과를 평가하였고, 피부 수분 증발량과 피부 수분량을 조사하여 피부개선을 평가하고 항산화 및 피부보습효과를 제공하는지 여부를 판단하였다. 이에 에탄올은 연어추출물을 첨가하였을 때가 무첨가에 비해 5배에서 7배의 감소량을 보였고 아세트알데히드 3배에서 5배 이상의 감소량을 보였다. 또한 혈중 아세트알데히드 농도와 초산 농도의 변화에서는 무첨가 대조군에 비해 급격한 저하 변화를 보여 알코올 분해효과가 있음을 확인하여 숙취해소 효과가 있음을 확인할 수 있었다. 그리고 히알루론산 원료를 첨가하여 사용하였을 때 피부 수분함유량이 높았으며, 피부 수분 증발량이 감소됨이 확인되었다. 이에 고분자 다당체인 히알루론산은 뛰어난 점탄성 및 보습력을 보유하고 있어서 숙취현상 중 나타나는 생체수분 감소량 및 피부수분 함유량저하에 효과가 있어 항산화 및 피부보습효과를 제공하는 것으로 사료되어 연어추출물 분말과 히알루론산을 주원료로 하는 조성물이 음주 후 나타나는 숙취현상에 효과가 있다고 할 수 있다.

서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
