Analysis of Fra-X Gene Using Hair Root DNA

  • Extract of DNA for analysis of fragile X syndrome is usually performed by blood, the researches using hair root as specimen have been gradually spread. In this study, analyze fra X gene of the patients in mentally retarded children facilities was conducted using hair root DNA with molecular biologic test (PCR). The number of total subjects was 24, boys were 12, the average age was $17({\pm}3)$, and girls were 12, the average age was $18({\pm}2)$. In girls, normal size of band of 222 bp appeared in all lanes. Also, in all lanes except control in 517 bp, micro band appeared. Moreover, with appearance of band of 1198bp in lanes 2, 3, 4, 5, it is estimated that it is the band of full mutation whose CGG repeated sequences are more than 200. But it showed the peculiarity that it appeared with normal band in all the same lanes, thus it is not reasonable to judge it is the band of full mutation and further studies are needed. These results appeared in 50%, 6 of 12 mentally retarded girls. As the result of mentally retarded boys, normal band appeared in about 222 bp in control, however in experiment group, normal band did not appear. In 43%, 7 out of 12 boys, band did not either appeared in 1198bp, which showed different patterns from that of girls.

Evaluation of Failure Behavior of a Pipe Containing Circumferential Notch-Type Wall Thinning (원주방향 노치형 감육부를 가진 배관의 손상거동 평가)

  • In order to evaluate a failure behavior of pipe with notch-type wall thinning, the present study performed full-scale pipe tests using the 102mm, Schedule 80 pipe specimen simulated notch- and circular-type thinning defects. The pipe tests were conducted under the conditions of both monotonic and cyclic bending moment at a constant internal pressure of 10 MPa. From the results. of experiment the failure mode, load carrying capacity, deformation ability, and fatigue life of a notch-type wall thinned pipe were investigated, and they were compared with those of a circular-type wall thinned pipe. The failure mode of notched pipe was similar to that of circular-type thinned pipe under the monotonic bending load. Under the cyclic bending load, however, the mode was clearly distinguished with variation in the shape of wall thinning. The load carrying capacity of a pipe containing notch-type wall thinning was about the same or slightly lower than that of a pipe containing circular-type wall thinning when the thinning area was subjected to tensile stress, whereas it was higher than that of a pipe containing circular-type thinning defect when the thinning area was subjected to compressive stress. On the other hand, the deformation ability and fatigue life of a notch-type wall thinned pipe was lower than those of a circular-type wall thinned pipe.

An Estimation of Deformation for Composites by DIC (DIC에 의한 복합재료 변형측정)

  • The estimation of deformation and strain for the twill-weave carbon fiber reinforced plastic composite(CFRP) during the test with a digital image correlation system were implemented experimentally. The carbon fiber reinforced plastic composites have been developed as the edge technology materials. The plain, twill and satin weave types are commonly used for the CFRP composites. Thus, it is essential to find the deformation characteristics for those types of CFRP more easily. Especially the DIC method can express the visual strain distributions at the full range of the interested areas in the structures. In this study, the mechanical properties of twill-weave CFRP composite and the variation of strains in a full field of the specimen were estimated. The experiments were performed under a tensile loading and 3-point bending test with strain gages. Futhermore the DIC deformation results were estimated for the comparison. The results showed the deformation and strain contours visually well in all region of the interested areas and so usefulness for the safety control of the structures.

Axial compression performance of basalt-fiber-reinforced recycled-concrete-filled square steel tubular stub column

  • This study aimed to inspect the axial compression mechanical performance of basalt-fiber-reinforced recycled - concrete (BFRRC)-filled square steel tubular stub column. The replacement ratio of recycled coarse aggregate (RCA) and the basalt fiber (BF) dosage were used as variation parameters, and the axial compression performance tests of 15 BFRRC-filled square steel tubular stub column specimens were conducted. The failure mode and the load-displacement/strain curve of the specimen were measured. The working process of the BFRRC-filled square steel tubular stub column was divided into three stages, namely, elastic-elastoplasticity, sudden drawdown, and plasticity. The influence of the design parameters on the peak bearing capacity, energy dissipation performance, and other axial compression performance indexes was discussed. A mathematical model of segmental stiffness degradation was proposed on the basis of the degradation law of combined secant-stiffness under axial compression. The full-process curve equation of axial compressive stress-strain was proposed by introducing the influencing factors, including the RCA replacement ratio and the BF dosage, and the calculated curve agreed well with the test-measured curve.

An Experimental Study on the Structural Performance of Horizontally Curved Precast PSC Girder (프리캐스트 곡선 PSC 거더의 구조 성능에 관한 실험연구)

  • The main purpose of this study is to investigate the static behavior of a horizontally curved prestressed concrete (PSC) girder. A 30m long full-scale curved PSC girder with 80.0m radius is fabricated by a portable curved form system. Deflections and concrete strains at the middle of span were measured. The obtained experimental results have been compared to those from F.E.A. analysis. When a initial crack developed, the applied load was 1.3 times the service design load and the vertical deflection at the middle of span satisfied the requirement for a live load state according to the Korea Bridge Design Specifications (2010). Also, the ductility of the full scale specimen satisfied the limit in the Specifications (2010). To verify the experimental results, a numerical F.E. analysis was carried and confirmed that the data were similar with results from the test above. The horizontally curved PSC girder fabricated on site was found to have enough strength for safety under and after construction.

  • The evaluation of shear stiffness of shear connection in composite bridges with CIP concrete deck is analysed. Shear stiffness of shear connection in full-depth precast concrete deck bridges is obtained from experiments. 3-dimensional finite element analyses of push-out specimen are carried out to investigate the effects of characteristics of filling material strength in shear connection on shear stiffness and local stress distribution. The load-slip relations obtained from the analyses are compared with those of experiments. The equation of initial shear stiffness of shear connection in precast concrete deck bridge is proposed. Linear analyses are performed to evaluate the effects of the shank diameter of shear connector and the strength of mortar on the characteristics of deterioration and failure load obtained by the failure criterions of each material. The failure loads are estimated and compared with test results.

Full-scale testing on the flexural behavior of an innovative dovetail UHPC joint of composite bridges

  • This paper presents a full-scale experimental test to investigate the flexural behavior of an innovative dovetail ultra-high performance concrete (UHPC) joint designed for the 5th Nanjing Yangtze River Bridge. The test specimen had a dimension of 3600 × 1600 × 170 mm, in accordance with the real bridge. The failure mode, crack pattern and structural response were presented. The ductility and stiffness degradation of the tested specimens were explicitly discussed. Test results indicated that different from conventional reinforced concrete slabs, well-distributed cracks with small spacing were observed for UHPC joint slabs at failure. The average nominal flexural cracking strength of the test specimens was 7.7 MPa, signifying good crack resistance of the proposed dovetail UHPC joint. It is recommended that high grade reinforcement be cooperatively used to take full advantage of the superior mechanical property of UHPC. A new ductility index, expressed by dividing the ultimate deflection by flexural cracking deflection, was introduced to evaluate the post-cracking ductility capacity. Finally, a strut-and-tie (STM) model was developed to predict the ultimate strength of the proposed UHPC joint.

Estimation of Moment Resisting Property for Pin Connection Using Shear Strength of Small Glulam Specimens (집성재 소시험편의 전단강도에 의한 핀접합부의 모멘트 저항성능 예측)

  • Most connections for the glulam structural members consisted of connector and fastener. The mechanical behaviour of the connection can be occurred by the dowel bearing resistance and wood shear by the fastener. This study aims at the examination of the shear properties for the small specimen with lamination components and for the full-sized pin connection and the moment resisting property for the double shear full-sized pin connection using structural column and beam members. Small specimens including glue line shows greater density and shear strength by the lamination effect than other specimens. It is needed that estimations of double shear property and moment resistance for the pin connections should be adjusted in some degree. For the better and safe estimation of moment resistance strength for the column-beam pin connection, however, the shear strength of small specimens should be deducted by 10%.

Is "Initial Size of the Graft the Real Culprit behind Primary Contraction of Full-Thickness Skin Graft"?-A Cross-Sectional Study

  • Background Primary contraction of full-thickness graft has been traditionally quoted to be 40%. There are lacunae in literature to elaborate on the factors influencing it ever since. Methods About 75 subjects who underwent full-thickness grafting procedures to resurface small defects were included in the study. The initial and final graft dimensions after primary contraction were traced on X-ray templates and the percentage of contraction was evaluated using the graphical method. This was further correlated with age, collagen, elastic matrix metalloproteinases-1 (MMP-1) and -2 content along with dermal thickness of the skin specimen sent from the graft. Results The primary contraction of the graft had a very significant correlation only with the initial size of graft harvested with a linear regression of 33.3% and a Spearman's correlation of 0.587 significant at a p-value of 0.001. Conclusion This study though preliminary tries to highlight an important factor that primary contraction of grafts is a physical phenomenon independent of its contents like collagen, elastin, or MMP-1 and -2 or age and dependent on its initial size of harvest instead.

A Study for Vibration Characteristics of RC Slab with Hybrid Beams in Large Span Educational Facilities (대공간 교육시설에 사용되는 합성보 및 콘크리트 슬래브의 진동평가에 대한 연구)

  • In this study, vibration characteristics of reinforced concrete slab in large span educational facilities were evaluated. A 21.75m X 14.4m full scale reinforced concrete slab specimen was constructed with pre-flex hybrid beams. Vibrations were generated by three different methods such as free falling method of a 6kg sand bag, a 70kg person walking method and impact method by impulse hammer. Vibrations were generated more than 3 times at single location. Vibration characteristic data were collected by SA390 signal analyzer machine at 5 different locations.
