Effects of Postharvest Fruit Coating and Ethylene Scrubber in Polyethylene Film Wrapping on the Marketability of 'Fuji' Apples during Storage and Marketing (과실 피막제 및 Ethylene 제거제의 수확후 처리가 '후지'사과의 저장력과 상온유통에서의 상품성에 미치는 영향)

  • Postharvest treatments such as polyethylene film wrapping, ethylene scrubber, and fruit coatings were compared to find a potential effect on the fruit quality in 'Fuji' apples during storage and simulated marketing. Polyethylene film wrapping appeared to be excellent in preventing weight loss during storage but fruits lost weight at a high rate when exposed to ambient temperature during simulated marketing. Also, surface cracking disorder was observed in parts of film-wrapped fruit under marketing condition after storage of 21 weeks. Ethylene scrubber effectively removed ethylene in polyetheylene bags but this effect reduced along with the increase of storage time. Although Prolong-treated fruits were good in appearance at 12-week storage, no beneficial effect was found on the inhibition of weight loss during storage as well as marketing. In addition, wax-like substances were accumulated on the surface after storage of 21 weeks, resulting in the reduction of marketability. Water soluble wax seemed highly effective on the inhibition of weight loss during storage and marketing. However, parts of fruit were severely injured probably due to the high concentration of wax. Undamaged fruit showed excellent appearance in shape and flesh quality even after 21 weeks of storage. Further study is required to find the proper concentration of wax.

Effects of Medicinal Herb Extracts and Heat and Coating Treatments on the Browning Degree of Fresh-Cut Apples (한약재 추출물, 가열 및 코팅 처리가 신선절단 사과의 갈변도에 미치는 영향)

  • The effects of dip treatments of chemicals (ascorbic acid, cystein, sodium chloride, magnesium chloride, calcium chloride, histidine: 1% solution) and medicinal herbs (Pueraria thunbergiana, Angelica gigas, Dioscorea japonica, Lindera strychifolia, Cnidium officinale, Astragalus membranaceus: 1% extracts), heat shock ($40-60^{\circ}C$ water), and edible coating (albumin, dextrin, sucrose poly ester, whole soy flour: 0.5-4% solution) on the browning degree of fresh-cut 'Fuji' apples were studied. Among the each treatment methods, the surface browning of fresh-cut apples was best retarded by calcium chloride dip, Astragalus membranaceus dip, $45-50^{\circ}C$ heat shock, coating with 3% dextrin or 1% whole soy flour. The results suggest that the treatments of medicinal herbs extracts, heat shock, and edible coating can be used as alternative for the use of chemical agents for the antibrowning of fresh-cut apples.

Antioxidant and tyrosinase inhibitory activities of immature fruits of Malus pumila cv. Fuji (미성숙 사과의 항산화 및 tyrosinase 저해 활성 평가)

  • In this study, the antioxidant and tyrosinase inhibitory activities of aqueous ethanolic extract from the immature fruits of Malus pumila cv. Fuji were evaluated. The antioxidant capacities of the extract was investigated employing radical scavenging assays using DPPH and $ABTS^+$ radicals. The skin-whitening effect of M. pumila cv. Fuji extract was tested using mushroom tyrosinase assay. In addition, the total phenolic content was measured by a spectrophotometric analysis. All tested samples showed a dose-dependent radical scavenging and tyrosinase inhibitory activities. Among the tested samples, the ethyl acetate (EtOAc)-soluble portion from the immature fruits of M. pumila cv. Fuji was showed the significant DPPH and $ABTS^+$ radicals scavenging activities. Also, the tyrosinase inhibitory activity of the ethyl acetate (EtOAc)-soluble portion from immature apples was higher than other solvent-soluble portion. These results suggest that unripe fruits of M. pumila cv. Fuji could be considered as a new valuable source of natural antioxidant and skin-whitening agents. Systematic investigation of immature fruits of Malus pumila cv. Fuji will be performed for the further development of its biological properties.

Influence of Thermal Treatments on Germination and Internal Compositions of 'Hongro' and 'Fuji' Apple Trees during Endodormancy (내재휴면기 온도처리가 사과 '홍로' 와 '후지'의 발아와 내부물질 변화에 미치는 영향)

  • In this study, we investigated the limit temperature range which is effective for dormancy release of 'Hongro' and 'Fuji' apples during the endodormancy period. The germination rate was 50% or more in all treatments except of 'Hongro' $-5^{\circ}C$ treatment. The germination rate of 'Fuji' was 86.3% at the temperature of $-5^{\circ}C$. The concentrations of carbohydrate and mineral components were significantly different between treatments but did not show any tendency or specific change. However, the sorbitol contents of 'Hongro' $-5^{\circ}C$ treatment were higher at 29.62 mg/g than those of 'Fuji', which was not affected for dormancy release. The results of hormone analysis showed that ABA(abscisic acid) and JA(jasmonic acid) in 'Hongro' $-5^{\circ}C$ treatment were respectively 176.48 and 15.72 ng/g. ABA, JA and SA(salicylic acid) contents in 'Fuji' apple tree were significantly lower than those of 'Hongro'. As a result of this study, the limit temperature range effective for dormancy release was different according to the varieties, and 'Hongro' did not affect with 30.3% of germination rate at $-5^{\circ}C$. However, 'Fuji' is 86.3% even at $-5^{\circ}C$ it was suggested that 'Fuji' had a chilling accumulation for dormancy release.

Pollination Effect and Nesting Behavior of Osmia cornifrons on 'Hongro', Early and 'Fuji', Late-season Apple Cultivars (중생종 '홍로'와 만생종 '후지'사과에서 머리뿔가위벌(Osmia cornifrons)의 영소활동 특성과 화분매개효과)

  • In order to use Osmia cornifrons more effectively in apples, we investigated the pollination effect and nesting activity of O. cornifrons on 'Hongro', the middle-season cultivar, and 'Fuji', the late-season cultivar. There was a significant difference in the nesting activity and pollination effect of O. cornifrons depending on the apple cultivar. The nesting activity, rate of trap nesting, and reproduction in 'Fuji' were 2.5, 1.5, and 3.8 times greater than in 'Hongro', respectively. The pollination effect according to cultivar 'Fuji' was 1.6 times greater than that of 'Hongro' in the central fruit set. In terms of the quality of fruit, the asymmetry index of 'Fuji' was 2.5 times lower than that of 'Hongro', and the number of apple seeds of 'Fuji' was 1.9 times greater than that of 'Hongro'. The main reason for this result was the air temperature. The activity of O. cornifrons was most affected by air temperature (R2 = 0.578). It is expected that the nesting activity and pollination effect are great in 'Fuji' (17.4-24.1℃) when the temperature during the blooming period is higher than that of 'Hongro' (12.5-20.2℃). Our study provides important information on stable apple production for apple cultivators, and can be used as an evidence for changes in flora and insect fauna caused by climate change.

Molecular Characterization of an Apple cDNA Encoding Cinnamyl Alcohol Dehydrogenase

  • The study of lignin, a major component of secondary cell wall, has been partly focused on its removal from the woody part in the kraft pulping industry. Cinnamyl alcohol dehydrogenase (CAD; EC 1.1.l95) catalyzes the synthesis of cinnamyl alcohols from corresponding cinnamaldehydes. A cDNA clone, MdCADl, encoding putative CAD from apples (Malus domestica Borkh. cv Fuji) was characterized in this study. The clone contains an open reading frame of 325 amino acid residues, which shows a greater than 80% identity with Eucalyptus CADl. MdCADl mRNA was detectable in vegetative tissues and was strongly expressed in the fruit. The expression pattern of MdCADl mRNA in the fruit peel after light exposure was also examined. The mRNA was rapidly increased until 1 day after light exposure and remained stable thereafter, suggesting that MdCADl is light inducible. The inducibility of the MdCADl gene was examined using several environmental stresses. Mechanical wounding of leaves increased the MdCADl mRNA level and the induction was further increased by salicylic acid. Southern blot hybridization showed that there is either one or a few copies of CAD genes in apples. To our knowledge, it is believed that MdCADl is the first CAD clone expressed predominantly in fruit.

Detection and Distribution of Apple scar skin viroid-Korean Strain (ASSVd-K) from Apples Cultivated in Korea

  • Apple scar skin viroid (ASSVd) has been one of the most destructive diseases in Korean apple orchards. Symptoms of the scar skin viroid disease were detected in various apple cultivars, namely, Sansa, Fuji, Chukwang, Miki-Life, Hongro, and Songbongeum cultivated in the southern part of Korea. The RNA molecules were extracted from the apples bearing dapple apple symptoms with the application of CF-11 RNA extraction method. The purified RNAs were used for the synthesis of cDNA with RT-PCR. The PCR products were cloned and sequenced. The viroid RNA molecules from the six different cultivars bearing the dapple symptos showed the same nucleotide sequences as that of the Korean strain of ASSVd(ASSVd-K). ASSVd-K was detected from apple orchards in Kunwi, Sangju, Uiseong, Yeong-yang, Andong, and Youngduk in Gyeongbuk Province in 2001, and in Muju in Jeonbuk Province in 2002. As the viroid disease could be propagated vegetatively, it can be widely transmitted gradually in Korea.

Classification of Apple Coloration Using Image Processing System (영상처리(映像處理) 장치(裝置)를 이용(利用)한 사과의 색택(色澤) 판정(判定))

  • The aims of this study were to investigate the feasivility of analyzing a few sorting factors such as size, coloration and defect of apples with a monochrome image processing system and to find apparent properties which could be effectively used for apple sorting. The results are summarized as follows. 1. A computer program was made to analyze the projection area, coloration and defect of an apple with a monochrome image processing system. 2. The algorithm developed to compute the projedtion area of an apple was between on the proportional relation between a given reference area and the corresponding number of pixels, and the computing time was 0.74 to 0.82 second depending on the size of apple. 3. The coloration of an apple was expressed as the ratio of the gray value of a reference color to that of a given bounded area of the stem end surface (defined as coloration index), and the computing time was about 3.0 seconds with this algorithm. 4. Defect of an apple could be isolated by lowpass filtering and image subtraction but it took about 20 seconds in computing time. 5. The coloration of the Fuji apple could be classified into 3 to 4 groups by the coloration index and also, it was found that the correlation coefficient between the indices and sugar contents was 0.74. 6. The coloration index obtained from a given bounded area of the stem end side of the Fuji apple could represent the coloration of total surface with a correlation coefficient of 0.922.

First Report of an Apple Ring Rot Fungus Botryosphaeria kuwatsukai in Korea

  • Three fungal strains designated as KNUF-23-MG32, KNUF-23-YC8, and KNUF-23-MJ82 were isolated from the abnormal symptomatic apple trees during screening fungal pathogens collected in Jeollanam-do and Gyeongsangbuk-do, Korea. These fungal strains were found to have similar cultural and morphological characteristics close to the genus Botryosphaeria. Morphological characteristics were matched with B. kuwatsukai CBS 135219 but different with B. dothidea KACC 45481 and B. sinensis HMAS 246714T. Pathogenicity tests of strain KNUF-23-MG32 showed that this strain causes rot in Fuji apples. Molecular phylogenetic analysis based on the sequences of the internal transcribed spacer (ITS) regions, translation elongation factor 1-alpha (TEF1α), and partial β-tubulin (TUB2) showed that these strains are B. kuwatsukai. To the best of our knowledge, this is the first report of an apple ring rot fungus Botryosphaeria kuwatsukai in Korea.

Applications of Artificial Pollination, Spraying Gibberellin A4+7 Plus Benzyladenine for Production of Uniform Fruits in 'Fuji' Apples (인공수분(人工受粉) 및 Promalin 처리에 의한 사과 '후지' 품종(品種)의 과형(果形) 개선(改善))

  • Artificial pollination (AP) and gibberellin A4+7 plus benzyladenine (promalin) were applied alone and together. AP was applied at 10% flowering time with 'Senshu' pollen (Malus domestica cv. Senshu). 12mg/L promalin was applied at 0, 10 and 20 days after falling of central flowers, respectively. In promalin treatment with or without AP application methods, fruit length, weight and length/diameter (L/D) were higher than those of control and AP. However, in AP and AP+ promalin application, the number of seeds and seed weight were higher than those of control and promalin. In AP+ promalin treatment, 78.6% fruits showed their uniform fruit shape and so significantly enhanced fruit uniformity compared to other treatments. Also cortex and core thickness of fruits were greater at the apex than that of other treatment. Fruit with L/D ratio over 0.87 were highly produced by applications of AP+promalin and promalin than control and AP only. 'Fuji' apples in good shape can be produced by using of AP + promalin together.
