Efficacy Test of Commercial Digestives Containing Antacids, Digestive Enzyme and Herbal Drug(I): In vitro and In vivo Evaluation (제산제, 소화효소제 및 생약제를 함유한 시판 복합소화효소제의 효력시험(I) : in vitro 및 초 vivo 제산력 시험)

  • The reaction rates, duration times and neutralizing capacities of the antacids which are frequently used in Korean market and three different commercial combination products were evaluated in vitro by Fuchs method and Johnson-duncan method, respectively. In vivo tests of combination products were determined in the fasted state of rat by Aspiration method. Comparing the result of in vitro test with that of in vivo test, the maximal pH was lowered by 2-3 value and the durational time increased by two folds in vivo test. Each antacid composition and combination products from three phamaceutical companies (A, B, and C) were studied, respectively. The duration times measured by Fuchs method were double compared to those by Johnson-Duncan method. A and C preparation maintained the pH range from 3 to 7 for 60 min by Fuchs method. In vovo test, maximum pH of A, B and C preparation was 6.50, 3.65, 2.65 and duration time of those was 200, 500, 0 min, respectively.

  • To solve the interaction of incident monochromatic waves with a bottom-fixed vertical circular cylinder, a numerical analysis by boundary element method is developed using three-dimensional linear potential theory. A numerical analysis by boundary element method is based on Green's theorem and introduce to an integral equation for the fluid velocity potential around the vertical circular cylinder. These numerical results are compared with those of ManCamy and Fuchs(1954) and Williams and Mansour(2002), and it has shown good relationship with their results. This numerical analysis developed by boundary element method will be applied for various offshore structures to be constructed in coastal zones in the future.

  • Conventional method for designing and installing anechoic chambers is to utilize porous wedges for the sound absorbers. As cutoff frequency lowers down such as 63Hz or 50Hz, the corresponding long wedges diminish the free field area of the chamber. In this study, a new broadband compact absorber(BCA) is introduced which absorbs acoustic energy down to 50Hz. Most prominent is that it measures only 250mm thick. A freely vibrating panel between the non-fibrous absorbers allows tuned absorption at the low frequency region in addition to the high frequency absorption resulted from the conventional absorber installed at the front. Standing waves at low frequency range are suppressed as the BCA modules which are tuned to the corresponding modes absorb sound energy effectively, resulting in anechoic condition. Not only the low frequency performances, but the high frequency absorption is measured to meet adequate conditions for the anechoic chamber. Realized BCA chambers are presented.

  • The combined products of antacid and anti-ulcer agent were prepared with antacid composed of aluminium hydroxide dried gel, magnesium hydroxide and simethicone with a ratio of 1:1:0.1 (M) and anti-ulcer agent, aceglutamide aluminium (AGA). The efficacy of antacid was evaluated in vitro with Fuchs, Johnson-Duncan and Rosset-Rice methods and in vivo using an aspiration method in rat. The addition of anti-ulcer agent did not affect the neutralizing capacity of M significantly. The combined products with the M/AGA ratios of 2.3:1 and 3.4:1 produced the maximum pH of $4.0{\sim}5.8$ and the duration time of $64{\sim}137$ min in vitro test. The in vivo neutralizing test in rats showed the rapid increase of gastric pH up to 3.5 within 30 min and the gastric pH of $4{\sim}6$ was kept for 5 hr.

  • An efficient method for the calculation of wave forces on a tension leg platform(TLP) is presented in this paper. It is based on the Morison's equation with two corrective terms. One is the reduction of the inertia forces on the vertical columns in order to include the wave diffraction effect particularly for small wave conditions. The other is the inclusion of the hydrodynamic forces acting at the bottoms of the columns. Numerical studies are carried out for a TLP in 1000 ft water with two different wave heading angles($0^{\circ}$ and $45^{\circ}$). The reponse amplitude operators(RAO's) for the TLP motions and top tether tension variations are obtained by the present method and the theoretically more accurate method based on the diffraction theory. A comparison has been made between the results obtained by two methods.

  • At an early stage of genomic investigations, a small sample of microarrays is used in gene expression experiments to identify small subsets of candidate genes for a further accurate investigation. Unlike the statistical analysis methods for a large sample of microarrays, an appropriate statistical method for identifying small subsets is a randomization test that provides exact P values. These exact P values from a randomization test for a small sample of microarrays are discrete. The possible existence of differentially expressed genes in the sample of a full set of genes can be tested for the null hypothesis of a uniform distribution. Subsets of smaller P values are of prime interest for a further accurate investigation and identifying these outlier cells from a multinomial distribution of P values is possible by M test of Fuchs et al. (1980). Above all, the genome-wide gene expressions in microarrays are correlated, but the majority of statistical analysis methods in the microarray analysis are based on an independence assumption of genes and ignore the possibly correlated expression levels. We investigated with simulation studies the effect that correlated gene expression levels could have on the randomization test results and M test results, and found that the effects are often not ignorable.

Differential Responses to TGF Alpha in between Invasive Squamous Cell Carcinoma Cell Line and Noninvasive One (침투성 상피암세포주와 비침투성 상피암세포주의 TGF alpha에 대한 반응의 차이)

  • Both SCC 12 and SCC 13 cell lines were derived from squamous cell carcinoma (SCC) of the skin (Wu and Rheinwald, 1981). In the present study, we compared the inherent invasive activity in their raft cultures where most in vivo characteristics of epidermis can be reproduced by cell culture method. The raft culture of SCC 12 cell line produced many invading colonies within the collagen lattice and basal-like cells in the middle of differentiating cell layers, but no invasive activity was observed in the SCC 13 raft culture. We investigated which factors are implicated in inherent invasive activity of SCC 12 cell line by examining basal levels of type I collagenase, EGF receptor, fibronectin, and its receptor in two cell lines. Among them, only type I collagenase was significantly higher in invasive SCC 12 cells than in non-invasive SCC 13 cells. Furthermore, we tried to investigate mechanisms underlying between SCC 12 cell's inherent invasive activity and its high basal level of type I collagenase. As one of them, discrepancy in TGF alpha mediated responses between two cell lines was observed. In SCC 13 cells, TGF alpha initially stimulated type I collagenase at 12 h after TGF alpha treatment and then its down regulation was followed from 24 h even though TGF alpha was continuously present in the medium. However in SCC 12 cells, TGF alpha continuously stimulated type I collegenase up to 48 h. We propose that defect in EGF receptor's down-regulation may be involved in lack of type I collagenase's down-regulation and its possible connection to invasive activity of SCC 12 cell line.
