Hybridization and Use Of Grapes as an Oviposition Substrate Improves the Adaptation of Olive Fly Bactrocera oleae (Rossi) (Diptera: Tephritidae) to Artificial Rearing Conditions

  • The olive fly Bactrocera oleae (Rossi) is the key pest for olive cultivation worldwide. Substantial effort has been invested in the development of the sterile insect technique (SIT) to control this pest. One of the limitations to develop SIT technology for olive fruit fly is the low ability of wild females to lay eggs in other medium than olive fruits, and their slow adaptation to oviposition in artificial substrates. In the present study, fruit grapes were used as an alternative egg collection medium to harvest eggs and young larvae from freshly colonized wild strains originating from France, Italy, Spain and Croatia. The larvae were allowed to develop into the fruits until the second instar, before they were extracted out and further reared on a standard artificial diet. Furthermore, F1 to F4 female flies were alternatively offered wax bottles to oviposit. Finally, the performance of hybrid strains created from crosses between wild and long colonised flies was assessed. The results showed that females of all 4 wild strains readily oviposited eggs in grapes and from the F2 generation onward, females from all strains were adapted to laying eggs in wax bottles. No difference was observed in eggs and pupae production among all strains tested. The findings are discussed for their implications on SIT application against olive fruit fly.

Superoxide Dismutase유사활성을 지닌 식물체가 Oxidative Stress를 받고 있는 초파리의 수명에 미치는 영향 (Effect of Plant Extracts with Superoxide Dismutase-like Activity on Survival of Fruit Flies under Oxidative Stress)

  • 초파리를 이용하여 superoxide dismutase (SOD) 유사활성이 높다고 알려진 식물체 12 종의 효능을 평가하였다. 평가방법으로는 초파리 체내에서 superoxide를 발생시켜 초파리를 죽게 하는 paraquat (Pq)를 초파리 식이에 공급하고 상기 식물체 추출물을 투여하였을 때 초파리의 배양시간에 따른 생존율을 비교하여 분석하였다. Pq와 함께 마늘, 상치, 키위, 팽이버섯 추출물을 동시에 공급하였을 때 이들은 산소상해를 받고 있는 초파리에 비해 효과를 나타내지 못하였다. 그러나, 이들 식물체를 함유한 배지에 10일간 미리 적응시킨 후 식물체를 계속 공급하면서 60 mM 농도로 Pq를 함께 공급하였을 때 배양 1일 후 Pq 처리구의 생존율은 약 11%였으나 식물체 첨가구의 생존율은 $35{\sim}63%$로 높게 나타났다. 즉, 미리 적응시키면 SOD 유사활성이 있는 식물체는 초파리가 활성산소에 노출되더라도 산소독성에 예방효과를 나타낼 수 있음을 시사하고 있다. 한편, 초파리를 20 mM의 Pq가 첨가된 배지에 노출시켜 산소상해를 입히고 생존한 초파리만을 모아 60 mM의 Pq와 식물계 추출물이 함께 첨가된 배지로 옳기고 2일 후 생존율을 측정하였을 때 Pq첨가구 초파리의 생존율은 32%였으나 상치, 키위, 팽이버섯, 양파, 감, 고사리, 폴리플라워 첨가구 초파리의 생존율은 $64{\sim}82%$로 나타났다. 결과적으로 이들 식물체는 산소상해에 대하여 일종의 치유효과를 나타내는 것으로 판단되었다. 조사한 식물체 중 미나리, 표고버섯, 느타리 버섯, 브로콜리는 치유효과를 발휘하지 못하였다.

Terpinyl acetate-단백질먹이 유인제를 이용한 호박과실파리류 연중발생 모니터링 기술 및 살포용 방제 제형 개발 (Monitoring Technique of Pumpkin Fruit Flies Using Terpinyl Acetate-Protein Diet Lure and Development of Its Spraying Formulation for The Fly Control)

  • 국내에 호박을 가해하는 두 과실파리는 Zeugodacus 속으로 분류된다. 호박꽃과실파리(Z. scutellata) 수컷은 큐루어(Cuelure)에 유인되어 포장에서 이 과실파리의 발생을 모니터링 하는 데 이용된다. 이에 반해, 호박과실파리(Z. depressa) 수컷을 유인하는 물질은 아직 알려지지 않았다. 단백질 먹이 유인제가 대부분의 과실파리를 유인하게 되는데 여기에 terpinyl acetate (TA)를 추가하여 호박과실파리의 유인력을 높였다. 본 연구는 TA-단백질먹이 유인제를 이용하여 호박 재배지에서 호박과실파리의 연중 발생을 모니터링 하는 데 목적을 두었다. TA-단백질먹이 유인제의 효율성을 증명하기 위해 호박꽃과실파리를 대상으로 큐루어와 TA-단백질먹이 유인제를 이용하여 2019년 한 해 동안 발생을 모니터링하고 이 두 유인제로 얻어진 연중 모니터링 자료를 비교하였다. 두 유인제를 이용한 호박꽃과실파리의 연중 발생 패턴은 10월 하순을 제외하면 유사하였다. TA-단백질먹이 유인제는 10월 하순 시기에도 지속적인 호박꽃과실파리의 포획을 보였는데 이는 9월 하순 이후 호박꽃과실파리의 성충 휴면 유기에 따른 유인 행동 차이로 이해되고 있다. TA-단백질먹이 유인제를 이용한 호박과실파리의 연중 모니터링은 크게 두 개의 발생 최성기를 보여 하나는 7월 중순이고 다른 하나는 8~9월에 나타나며 포획된 개체들의 80% 이상이 암컷이었다. 이러한 TA-단백질먹이 유인제의 두 과실파리류에 대한 유인 효과를 바탕으로 스피노사드(spinosad) 살충제를 가미한 수화제를 제조하고 호박 재배지에 살포하였다. 처리 7 일경과 후 약 70% 이상의 방제 효과를 기록하였다. 그러나 이 방제 효과는 처리 이후 기간이 경과하면서 낮아지는 경향을 나타냈다. 이상의 결과는 TA-단백질먹이 유인제를 이용하여 야외 호박 재배지에서 호박과실파리의 연중 모니터링이 가능하다는 것을 보여 주었다. 또한 스피노사드를 가미한 TA-단백질먹이 유살제가 이들 호박과실파리류 방제에 응용될 수 있다는 것을 제시하고 있다.

Overexpressed Drosophila DNA Methyltransferase 2 Isoform C Interacts with Hsp70 in Vivo

  • Shen and colleagues (Lin et al., 2004) have recently shown that overexpression of the Drosophila DNA methyltransferase 2 isoform C, dDnmt2c, extended life span of fruit flies, probably due to increased expression of small heat shock proteins such as Hsp22 or Hsp26. Here, I demonstrate with immunoprecipitations that overexpressed dDnmt2c interacts with endogenous Hsp70 protein in vivo in S2 cells. However, its C-terminal half, dDnmt2c(178-345) forms approximately 10-fold more Hsp70-containing protein complexe than wild-type dDnmt2c. Overexpressed dDnmt2c(178-345) but not the full length dDnmt2c is able to increase endogenous mRNA levels of the small heat shock proteins, Hsp26 and Hsp22. I provide evidence that dDnmt2c(178-345) increases Hsp26 promoter activity via two heat shock elements, HSE6 and HSE7. Simultaneously overexpressed Hsp40 or a dominant negative form of heat shock factor abrogates the dDnmt2c(178-345)-dependent increase in Hsp26 transcription. The data support a model in which the activation of heat shock factor normally found as an inactive monomer bound to chaperones is linked to the overexpressed C-terminus of dDnmt2c. Despite the differences observed in flies and S2 cells, these findings provide a possible explanation for the extended lifespan in dDnmt2c-overexpressing flies with increased levels of small heat shock proteins.

Multi-host Pathogenesis by Pseudomonas aeruginosa and Use of Drosophila melanogaster as a New Model Host

  • Fruit fly, Drosophila melanogaster has developed efficient immune mechanisms to prevent microbial infection, which are consisted of cellular and humoral responses. During the systemic or local infection, two distinct pathways (Toll and Imd) play major roles in antimicrobial peptide synthesis. The Toll pathway is required to defend Gram-positive bacterial and fungal infections, whereas the Imd pathway is important in Gram-negative bacterial infection. We have shown that the infection of the opportunistic Gram-negative bacterium, Pseudomonas aeruginosa strain PA14 (PA14) into fly dorsal thorax can kill the flies within 48 h ($100\%$ mortality) in our optimized infection condition, suggesting that the PA14 strain can cause disease progress in fly model system. We found that flies carrying a constitutively activated mutant form of the Toll receptor $(Tl^{10b})$ showed increased resistance to P. aeruginosa infection and that flies carrying mutations in the Toll signaling pathway as well as in the Imd signaling pathway was more susceptible to PA14 infection. All these results imply that the Toll pathway might be important in the resistance to this pathogenic Gram-negative bacterial infection.

Artificial rearing of the olive fruit fly Bactrocera oleae (Rossi) (Diptera: Tephritidae) for use in the Sterile Insect Technique: improvements of the egg collection system

  • One major constraint in the development and implementation of a successful and cost-effective area-wide integrated pest management (AW-IPM) programme with a SIT component for Bactrocera oleae (Diptera: Tephritidae) is the ability to produce a large number of high quality mass-reared individuals. The aim of this study was to develop a more efficient and practical egg collection system in an attempt to improve the mass-rearing of this species. The following basic parameters were examined: egg production per female, egg hatch, pupal recovery, pupal weight, adult emergence and percentage of fliers. Three different strains (Israel wild-type, France wild-type, and Greece laboratory) were tested and each strain was evaluated for six generations. Female flies of the Israel strain produced significantly more eggs per female than the other two strains, but egg hatch was significantly lower. Egg hatch of the France wild type and the Greece laboratory strain was similar. For all other parameters, there was no significant difference between strains; however, there was a significant generational effect for all parameters observed. As a result of this study, a protocol was developed for the mass-rearing of this species that included the use of large adult holding cages that could house up to 96,000 flies per cage. The newly developed method of egg collection using a flat wax panel as one of the sides of an adult holding cage proved to be cost-effective, efficient, making colony growth easier for industrial mass-rearing.

Two combined amino acids promote sleep activity in caffeine-induced sleepless model systems

  • BACKGROUND/OBJECTIVES: The aim of this study was to evaluate the biological and sleep-promoting effects of combined ${\gamma}$-aminobutyric acid (GABA) and 5-hydroxytryptophan (5-HTP) using caffeine-induced sleepless fruit flies, ICR mice, and Sprague-Dawley rats. MATERIALS/METHODS: Video-tracking analysis was applied to investigate behavioral changes of Drosophila melanogaster. Pentobarbital-induced sleep test and electroencephalogram (EEG) patterns were used for analysis of sleep latency, duration, and quantity and quality of sleep in vertebrate models. RESULTS: Administration of combined GABA/5-HTP could significantly reverse the caffeine induced total distance of flies (P < 0.001). Also, individually administered and combined GABA/5-HTP significantly increased the total sleeping time in the caffeine-induced sleepless ICR mice (P < 0.001). In the caffeine-induced sleepless SD-rats, combined GABA/5-HTP showed significant differences in sleep quality between individual amino acid administrations (P < 0.05). CONCLUSIONS: Taken together, we identified inhibitory effects of combined GABA/5-HTP in locomotor activity, sleep quantity and quality in caffeine-induced sleepless models, indicating that combined GABA/5-HTP may be effective in patients with insomnia by providing sufficient sleep.

Impaired Taste Associative Memory and Memory Enhancement by Feeding Omija in Parkinson's Disease Fly Model

  • Neurodegeneration can result in memory loss in the central nervous system (CNS) and impairment of taste and smell in the peripheral nervous system (PNS). The neurodegeneration seen in Parkinson's disease (PD) is characterized by functional loss of dopaminergic neurons. Recent studies have also found a role for dopaminergic neurons in regulating taste memory rewards in insects. To investigate how taste memories and sugar sensitivity can be affected in PD, we utilized the $DJ-1{\beta}$ mutant fruit fly, $DJ-1{\beta}^{ex54}$, as a PD model. We performed binary choice feeding assays, electrophysiology and taste-mediated memory tests to explore the function of the $DJ-1{\beta}$ gene in terms of sugar sensitivity as well as associative taste memory. We found that PD flies exhibited an impaired ability to discriminate sucrose across a range of sugar concentrations, with normal responses at only very high concentrations of sugar. They also showed an impairment in associative taste memory. We highlight that the taste impairment and memory defect in $DJ-1{\beta}^{ex54}$ can be recovered by the expression of wild-type $DJ-1{\beta}$ gene in the dopaminergic neurons. We also emphasized the role of dopaminergic neurons in restoring taste memory function. This impaired memory property of $DJ-1{\beta}^{ex54}$ flies also allows them to be used as a model system for finding supplementary dietary foods that can improve memory function. Here we provide evidence that the associative taste memory of both control and $DJ-1{\beta}^{ex54}$ flies can be enhanced with dietary supplementation of the medicinal plant, omija.

Structure and Function of the Developmental Signaling Molecule Hedgehog

  • Hh proteins represent a new signaling paradigm in metazoan development. In species ranging from fruit flies to humans, Hh proteins mediate multiple processes vital to appropriate pattern formation in the developing embryo. Hh proteins undergo an autoprocessing event in which the full-length protein is cleaved into N-terminal and C-terminal domains (Hh-N and Hh-C, respectively), and a cholesterol moiety becomes covalently attached to Hh-N. All known signaling activities of Hh proteins are mediated by Hh-N while both the cleavage and cholesterol transfer reactions are mediated by Hh-C. The cholesterol attached to Hh-N is required to retrict the range of Hh signaling and may be involved in ensuring appropriate reception of the Hh signal in target tissues. Disruptions of Hh signaling pathways lead to severe developmental defects in newborns and cancers in adults. While studies of Hh proteins have yielded a wealth of new insight into the molecular mechanisms of metazoan development, many outstanding questions concerning Hh signaling mechanisms ensure that unraveling the secrets of this molecule will keep scientists well entertained for the foreseeable future.

오리엔탈과실파리 예찰용 왁스형방출기 제작 및 분자진단기술 (Formulation of Wax Type Dispenser Monitoring the Oriental Fruit Fly, Bactrocera dorsalis, and Its Molecular Diagnostic Technology)

  • 오리엔탈과실파리(Bactrocera dorsalis)는 주요 검역 대상으로 지정된 해충이다. 이 해충에 대한 고감도 모니터링과 진단 기술이 초기 방역 처리에 요구된다. 본 연구는 오리엔탈과실파리를 유인하여 치사시킬 목적으로 메틸유제놀과 생물농약을 혼합한 왁스형 방출기를 제작하였다. 본 연구는 또한 5종의 주요 검역 과실파리(오리엔탈과실파리, 오이과실파리, 퀸즐랜드과실파리, 말레이시아과실파리, 지중해과실파리)에 대한 PCR 진단프라이머를 개발하였다. 이상의 모니터링용 왁스방출기과 분자진단기술을 말레이시아 코타키나발루 지역에서 실증 시험하였다.
