  • An ESR study has been made on free radicals produced in frozen aqueous solutions (ices) of glycine, DL-${\alpha}$-alanine, DL-serine, L-cysteine, DL-leucine and DL-isoleucine by gamma-irradiation at dry ice temperature. All free radicals induced were decayed concomitant to the successive annealing but the radical species which is believed to be dominant seems to be stable even near the melting point of the ice. These dominant species were found to be identical to those resulted from direct action of radiation in the solid at room temperature. Small but significant changes in the spectra of glycine and DL-${\alpha}$-alanine were observed by varying the microwave power. These results seem to support the view that the spectra obtained were composite consisting of more than two different resonances having different power saturation characteristics. The relative contribution of unidentified resonances to the composite spectra was greater for solutions of low concentration. These resonances are assumed to be induced by indirect effects, mainly hydrogen abstraction by radiation produced hydroxyl radicals and also C-N bond cleavage by hydrated electrons.

Numerical Analysis on Freezing in the Ship Voyaging in Polar Regions

  • For vessels operating in the cold climate regions, the ballast water inside or hopper tanks above the waterline may be frozen, starting at the top of the tank and at the side walls. Therefore, countermeasures against freeze-up of the ballast tank such as air-bubbling system, hot steam injecting system, heating coil system and water circulating system are taken to prevent freeze-up phenomenon; however, there are no rigorous investigations of anti-freezing to examine the effectiveness and validity of systems against freeze-up of the ballast tank, in which the temperatures are about -$25^{\circ}C$ (ambient air temperature) and $0^{\circ}C$ (sea water), respectively. In this paper, to ensure reasonable specifications for cold regions if the measures from the above-mentioned systems against freeze-up are effective, the phenomenon of ballast tank freeze-up is simulated and discussed in low temperature conditions. With the results using the commercial CFD code, CFX 14, the most cost-effective solution is conducted to prevent being frozen along the outer surface.

Diluents and Cryoprotectants for Cryopreservation of Filefish Thamnaconus modestus Sperm

  • The present study aimed to find the best diluent and cryoprotectant for sperm cryopreservation of filefish Thamnaconus modestus. Two kinds of artificial seminal plasma(ASP1, ASP2), 0.3 M glucose and marine fish Ringer's solution(MFRS) were employed as diluent. Dimethyl sulfoxide(DMSO) and methanol as cryoprotectant were selected for sperm cryopreservation. Sperm was diluted at the ratio of 1:3 with diluents containing cryoprotectants and adjusted for final concentration at 5%, 10%, 15% and 20%. Mixed milt was frozen at liquid nitrogen vapor after equilibration for 5 min. The highest motility($40.5{\pm}2.8%$) and swimming speed($81.5{\pm}4.1{\mu}m/s$) of frozen/thawed sperm were observed in ASP1 diluent containing 10% DMSO and in ASP2 containing 15% DMSO, respectively. Results showed that cryopreservation with ASP as diluent and DMSO as cryoprotectant could be adopted for long term storage of filefish sperm.

Detection of Wound-inducible Trans-Cinnamic Acid-4-Hydroxylase in Avocado, Persea americana, Roots

  • Trans-cinnamic acid-4-hydroxylase(tC4H) is the first cytochrome P450-dependent monooxygenase of the phenylpropanoid pathway. The roots of avocado seedlings were wounded and examined to determine whether the tC4H would be activated in response to wounding and/or whether tC4H activity be modulated by the application of exogenous p-coumarate. At the specified length of times, the wounded and treated roots were either frozen in liquid nitrogen or used immediately to extract microsomal proteins. The microsomal proteins were subjected to immunoblot analysis using polyclonal antibodies against CYP73 of tC4H gene. In this study, tC4H was induced in wounded roots sealed in bags within 6 hours, and in low level({TEX}$10^{-8}${/TEX}M) of p-coumarate solution within 24 hours, whereas the olution without p-coumarate and high levels of p-coumarate solution repressed tC4H induction in wounded roots. These results indicate that tC4H is induced by wounding in the root of avocado, and is inhibited by the application of exogenous p-coumarate.

Enhancement in the Viability and Biosensing activity of Freeze-Dried Recombinant Bioluminescent Bacteria

  • The genetically-engineered Escherichia coli strain, DPD2540, which contains a fabA:::luxCDAbefusion gene, gives a bioluminescent output when membrane fatty acid synthesis is needed. For more pactical application of this strain in the filed as biosensor, freezedrying was adopted. A 12% surcrose solution with Luria-Bertani (LB) broth, as determined by the viability after freeze-drying, was found to be most most effective composition for lyophilization solution among various compositions testitons tested. Rapid freezing with liquid nitrogen also gave the best viability after freeze-drying as compared to samples frozen at-7$0^{\circ}C$ and -2$0^{\circ}C$. The biosensing activities of the cells showed a greater sensitivity when the cells from the expontial phase were freeze-dried. Finally, the optimum temperature for use of the freeze-dried cells in the biodencor field was determined.

Photocycloaddition Reaction of 8-Methoxypsoralen and 5,7-Dimethoxycoumarin with Maleimide

  • $C_4$-Photocycloaddition of 8-methoxypsoralen (8-MOP) and 5,7-dimethoxycoumarin (DMC) to maleimide was studied in order to elucidate the mechanism of the photobiological activities of these molecules. The photoreaction was carried out in chloroform solution and frozen aqueous solution state. The major product was isolated and characterized by spectroscopic methods. The photoadduct between 8-MOP and maleimide was shown to be an 1:1 $C_4$-cycloadduct through the photocycloaddition of 4',5'-furyl double bond of 8-MOP to maleimide. The stereochemistry of cyclobutane ring of this photoadduct is consistent with the anti configuration. The photoadduct between DMC and maleimide was shown to be an 1:1 $C_4$-cycloadduct through the photocycloaddition of 3,4-pyrone double bond of DMC to maleimide.

솔비톨 첨가에 의한 냉동 해산물의 레토르트 후 품질 향상 (Quality improvement of retorted frozen seafood by adding sorbitol)

  • 고온에서 가열 살균한 냉동 해산물의 텍스처 경화 개선 효과를 당알코올을 이용한 얼음결정 억제 측면에서 연구, 고찰하여 상업적으로 적용 가능한 전처리 방법을 확립하였다. 레토르트 낙지, 오징어 및 소라의 텍스처 경화 현상 개선 실험 결과 2, 4%(w/w) 솔비톨 용액을 혼합 후 급속냉동, 해동 후 고온 가열한 경우 무처리구 대비 텍스처의 기계적 측정 지표인 경도값이 9-36% 내외 감소하여 텍스처 연화 효과가 발생하는 것으로 나타났다. 냉동 해산물의 냉동 과정에서 발생하는 얼음결정 형성에 따른 단백 조직의 수축에 의한 질긴 식감과 세포 조직 손상이 솔비톨 첨가에 따른 미세 얼음결정 형성과 보수력 향상으로 최소화되어 고온 가열 후에도 무처리구 대비 수분 용출이 감소하여 텍스처 연화가 발생함을 알 수 있었다(1,3,13-15). 또한 솔비톨 처리 시 단백 조직의 보수력 향상으로 중량 변화에 근거한 수율도 2-5% 내외 상승하였다. 오징어살과 소라살에서 5점 척도법 분석형 관능검사 결과 전처리 실험구의 탄력성 및 식감 기호도가 무처리구 대비 통계적 유의성 있게 0.2-0.4점 우수한 것으로 나타나 텍스처 기기 측정치와 동일하게 관능 분석에서도 텍스처 연화에 따른 식감 향상 효과가 검증 되었다. 이상과 같이 레토르트 냉동 해산물 원료의 냉동변성 현상을 억제하기 위하여 솔비톨과 염을 이용하여 급속 냉동시키는 얼음결정 억제 전처리 공정을 적용한 결과 오징어살과 소라살에서서 냉동변성 방지 효과가 확인되었으며, 오징어살의 최적 처리조건은 2% (w/w) 솔비톨 용액, 소라살의 최적 처리조건은 4% (w/w) 솔비톨 용액이었다.

유전자재조합 감자의 검정을 위한 DNA분리 및 PCR검출의 최적조건 탐색 (Optimized Condition of Genomic DNA Extraction and PCR Methods for GMO Detection in Potato)

  • 국내에서 시판되는 감자와 수입 감자스낵류로부터 상용 DNA 추출 kit 및 CTAB-phenol/chloroform 추출법등을 이용하여 시료특성에 따른 genomic DNA를 추출방법을 선정하고 PCR 정성검사를 실시하였다. 생감자의 경우 STE 용액으로 과량의 전분을 제거한 다음 DNA를 추출한 경우 순도 높은 DNA를 추출할 수 있었으며 상용 추출 kit를 이용한 경우 lysis buffer와 함께 ${\alpha}-/{\beta}$-amylase를 각각 또는 혼합으로 처리하거나 추출된 DNA 용액에 마지막 단계에서 효소를 처리한 시료군에서 고순도의 DNA를 추출할 수 있었으며, 효소 처리군에서는 ${\alpha}-/{\beta}$-amylase를 혼합으로 처리한 경우에 DNA 추출수율이 높았다. 냉동가공감자의 경우 silica-coated bead법을 이용하여 효소를 처리한 경우와 CTAB-페놀 클로로포름 처리군에서 DNA가 추출되었다. 또한, 각 방법으로 추출한 DNA에 대하여 감자의 내인성 유전자인 Pss 프라이머를 사용하여 PCR을 한 결과 모든 시료에서 추출된 DNA에 대하여 내부표준유전자 증폭산물이 검출되었다. 고도의 가공처리를 거친 수입 감자스낵(fabricated potato chips)과 냉동가공 감자(frozen fried potato) 등은 계면활성제인 CTAB(cetyl trimethyl ammonium bromide)과 페놀-클로로포름 혼합액을 이용하여 추출하고 이를 template로 하여 PCR 증폭을 실시하였다. 그 결과, Fig. 8에 제시한 바대로 감자의 내인성 유전자인 Pss 특이적 산물인 216bp의 산물이 냉동감자가공품과 감자칩에서 검출되었으며 재조합유전자인 New leaf plus 유래의 증폭산물(234bp)와 New lear Y유래의 증폭산물(225bp)는 검출되지 않았다. 본 실험의 결과 시료의 가공특성과 적용한 추출 kit 및 방법에 따라 genomic DNA 순도 및 추출수율이 크게 차이가 났으며 이것이 결국 PCR 결과에 의음성 혹은 의양성 등에 영향을 미치게 될 것으로 판단된다. 또한, 동일한 DNA추출방법에 의해서도 DNA가 추출되지 않은 경우가 있어서 동일한 시료에서 2회 반복 추출하는 것이 의음성결과를 피할 수 있는 방법으로 판단된다.

Heparin이 소 정자의 수정능획득반응에 미치는 영향에 관한 연구 (Study on Effect of Heparin on Bovine Sperm Capacitation)

  • To investigate the effect of heparin on sperm capacitation. The acrosome reaction of bovine sperm which were incubated in mTALP containing heparin and the in vitro development of bovine follicular oocytes which were cocultured with heparin-treated sperm were evaluated, and the resutls were as follows : 1. When bovine fresh sperm were incubated in mTALP solution containing 0, 5, 10 and 25$\mu\textrm{g}$/ml heparin for 15 to 840 minutes, there was no significant difference between motilies of heparin-treated sperm and untreated sperm, but the acrosome-reaction rate of heparin-treated sperm was significantly higher than that of untreated sperm. Moreover the acrosome reaction rate of was sperm treated with heparin was significantly increased after incubating for 15 minutes. 2. When fresh sperm were incubated in mTALP solution containing 10$\mu\textrm{g}$/ml heparn for 840 minutes, the motility of sper incubated for 840 minutes was lower than those of sperm incubated for 0, 15, 60 and 120 minutes, but the acrosome-reaction rate of sperm incubated for 840 minutes was higher than those of the others. 3. When frozen-sperm were incubated in mTALP solution containing 10$\mu\textrm{g}$/ml heparin for 120 minutes, the acrosome reaction rate of sperm was significantly increased after incubating for 15 minutes. 4. When fresh sperm treated with heparin were cocultured with bovine follicular oocytes, 16.7 to 23.7% of the oocytes were developed to 2-8 cell stage.

초급속동결에 있어서 Vitrification Solution 개발과 FDA 생사판정이 수정란의 배양과 이식후 착상에 미치는 영향 II. Vitrification Solution내의 비수과성 물질(Ficoll, sucrose)과 평형시각이 초급속동결 융해후 Mouse Morulae의 생존율에 미치는 영향 (Effects of the Improvement of Vitrification Solution and FDA-test on the Embryo Survival and Conception Rate by Ultrarapid Freezing II. Effects of the Addition Level of Non-permeable Cryoprotectants (Ficoll, sucrose) in Vitrification Solution and Equilibration Time on the Survival of Vitrofied Mouse Embryos)

  • This study was carried out to study effects of the addition level of acetamide and non-permeable cryoprotectants(Ficoll, sucrose) in VS(20% glycerol+10% ethyleneglycol) and equilibration time on the survival of vitrified mouse morulae. The results are summarized as follows: 1. When 10, 15 and 20% of acetamide were added to the new vitrification solution(20G 10E), FDA-scores of embryos were 4.4(control), 4.4(10%), 3.6(15, 20%), respectively. The addition of acetamide did not affect the survival of forzen-thawed morulae(P<0.05). 2. The survival rate betwen 5 min(3.5) and 10 min(4.6), 10 min(4.6) and 20 min(3.2) of equilibration in 10% sucrose, and 20 min(3.2) and 5 min(4.0), or 10 min(4.3) in 20% sucrose were significantly different(P<0.05). The highest survival(4.6) rate was obtained in mouse morulae equilibrated in VS(20G 10E) containing 10% sucrose for 10 minutes. 3. FDA-score of morulae frozen in the new vitrification solution containing 0, 10, 20 and 30% Ficoll was 4.5, 4.2, 4.4 and 4.6, respectively and had no significant effect among concentrations of Ficoll(P>0.05). The development rate after culture(24h) was 89%(20% Ficoll) and 93%(30% Ficoll), respectively.
