The antigenicity of EPO (erythropoietin) was investigated in guinea pig, mice and rats. Antigenicity tests-active systemic anaphylaxis (ASA), passive cutaneous anaphylaxis (PCA) of this materials were performed according to the established Regulation of Korean National Institute of Safety Research (1996, 4, 16). The results were followed: 1. After sensitizaion with EPO emulsified with complete Freund's adjuvant (CFA), guinea pigs didn't show any anaphylatic shock symptom in the ASA test 2. After sensitization with antisera of EPO sensitized mice, blue spots were observed on the hypodermis of back of rats in the PCA test, but diameter of each spot was smaller than 5 mm. From the results of this investigation, the antigenicity of EPO was negative under the present experimental condition.

Background: The immunomodulatory effects of Korean mistletoe (Viscum album Coloratum) on the innate immune responses of eel (Anguilla japonica) were studied. Methods: Mistletoe, Freund’s complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control was injected into eel peritoneal cavities. Results: Nitroblue tetrazolium (NBT)-positive cells in the head kidney of eel were significantly augmented by the second day post-injection of mistletoe. Reactive oxygen intermediates (ROI) were more produced in mistletoe-injected fish kidney leucocytes than in FCA-injected ones. The level of lysozyme activity in the serum of fish 2 days after injection with mistletoe was also significantly higher than that in the serum of the control fish. The optimal concentration of mistletoe in inducing the highest serum lysozyme activity was revealed to 500${\mu}$g/200 g of fish. In phagocytic activity assay, mistletoe-sensitized eel kidney phagocytes captured more zymosan than did the control fish. Conclusion: Korean mistletoe appeared to be a good activator of the non-specific immune responses of eel.

Antigenicity of Recombinant Granulocyte macrophage colony stimulating factor(LBD-005), a newly developed drug for hematopoietic growth, was investigated in mice and guinea pigs. 1. Mice showed production of antibodies against LBD-005 (100mg/kg) with alumin]m hydroxide gel(alum) as an adjuvant, judged by the heterologous anaphylaxis(PCA) test using rats. On the other hand, antibodies against ovalbumin(OVA) inoculated with alum were definitely detected. 2. In the studies with guinea pigs, both the inoculation of LBD-005 only and of LBD-005 with complete Freund's adjuvant(CFA) as an adjuvant did not produce positive reactions in homologous passive cutaneous anaphylaxis (PCA).

엘라스타제를 기니픽에 감작시킨 후에 항체검출시험을 실시하였다. 감작은 경구투여 및 피하주사의 두가지 방법으로 하였다. 경구감작은 투여직전 엘라스타제를 생리식염수에 용해한 후 강제투여하였으며, 피하감작은 엘라스타제를 Freund's complete adjuvant 에 현탁시킨 후, 목 뒷부분에 주사하였다. 항체생성의 양성, 음성 검출은 경구감작군에 대해, 능동적전신성아나필랙시 (ASA번)과 면역확산번 (ID법)으로, 피하감작군에 대해 수신피부아나필랙시 (PCA법)으로 행하였다. 결과, 경구감작군은 임상용량 (180 Unit/kg)의 경우는 ASA법 및 ID법 모두에서 음성을 보였으나, 임상 10배용량의 경우는 ASA법에서는 양성을 보인 동물이 있었으나 통계학적 우의성은 없었으며 (p<0.1), ID법에서는 음성을 보였다. 피하감작군은 모든 투여군에서 PCA양성을 보였다. 결론적으로 엘라스타제는 항원성이 인정되나, 임상용량을 경구적으로 투여한 경우는 항원성이 나타나지 않았다.

본 연구는 랫드에서 Freund's complete adjuvant를 랫드의 후지에 투여하여 인의적으로 유발된 관절염을 진단하기 위한 방법으로 $^{99m}Technetium-MDP (^{99m}Tc_MDP$)을 이용한 골 스캔의 임상적 의의를 찾고자 하였다. 성숙 Sprague-Dawley 암컷 랫드를 대조군과 실험군으로 분리하고, 골의 스캔은 0.05mCi의 $^{99m}Tc_MDP$를 정맥 주사하여 3시간 후에 전신 영상을 얻었다. 골의 스캔과 방사선학적 평가를 비교한 겨과 관절염 유발 7일 후부터 관절염의 병변이 관찰되었지만, 방사선학적 소견에서는 14일 후부터 병변이 관찰되었다. 골의 스캔에서 관절염 유발 7일 후에 유의적으로 골의 이상적 섭취 증각가 관찰되었고(p<0.05), 21일 후에는 전지와 후지의 모든 부위에서 골의 이상적 섭취 증가가 관찰되었지만, 방사선학적 소견에서는 후지에서만 병변 부위가 관찰되었다. 이상의 결과에서 $^{99m}Tc_MDP$을 이용한 골의 스캔은 류마티스 관절염을 임상적으로 진단하는데 방사선학적 진단보다 효과적인 방법임을 알 수가 있었고, 골의 스캔은 류마티스 관절염을 초기에 진단하는데 유효한 진단방법임을 확인하였다.

Antigenic potential of a novel cephalosporin, IDC7181 was examined a active systemic anaphylaxis (ASA) and passive cutaneous anaphylaxis (PCA) test, in guinea pig and mouse-rat models, respectively. In ASA test, IDC7181 induced the signs of restlessness in a few animals immunized with a high dose (100 mg/kg) of IDC7181 alone or in combination with Freund's complete adjuvant (FCA). In PCA test, only one of ten sera from the animals immunized with a high dose (100 mg/kg) of IDC7181 in the absence or presence of FCA showed positive reaction. The positive reaction, induced by IDC7181, which may be due to $\beta$-lactam ring, in ASA and PCA tests were negligible in comparison with those of traditional cephalosporins. Taken together, it is suggested that IDC7181 do not cause immunological problems in clinical dosage.

Antigenicity of DA-3285, human recombinant erythropoietin which was produced from mammalian cells, was examined in guinea pigs and mice. In active systemic anaphylactic test, mild anaphylactic signs were observed in guinea pigs sensitized subcutaneously with DA-3285 or DA-3285 incorporated with complete Freund's adjuvant(CFA) when challenged with high dose(1000 IU/Kg) of DA-3285. Other groups showed negative responses. In mouse-rat passive cutaneous anaphylaxis test, 20% sera of mice immunized with DA-3285 or DA-3285 mixed with aluminum hydroxide(alum) showed mild positive responses. In the case of indirect haemagglutination reaction(IHA) test, when sheep red blood cells coated with DA-3285 was incubated with mouse serum, all the serum samples were showed negative responses. These results suggest that DA-3285 has a very weak antigenic potential and probably would not induce systemic allergic reactions in clinical uses.

Aqueous extreact of fresh ginseng (AFG) was examined for the antigenicity in Hartley guinea pigs in comparision with ovalbumin (OVA). When guinea pigs were sensitized with AFG emulsified with complete Freund's adjuvant (CFA), these animals showed negative reactions in active systemic anaphylaxis (ASA), active cutaneous anaphylaxis (ACA) and passive cutaneous anaphylaxis (PCA) tests and passive hemagglutination (PHA) reaction. On the contrary, when guinea pigs were sensitized with OVA emulsified with CFA as positive controls, these animals disclosed positive reactions in ASA, ACA and PCA tests and PHA reaction. From these results, AFG was considered not to possess antigenic properties in guinea pigs. In addition, the dose levels of AFG empolyed in the present experiment were confirmed not to suppress immune reactions.

The studies on the anti-inflammatory and analgesic activities were carried out to develop the domestic natural sources of the underground parts of Caragana koreana Nakai (Leguminosae), which have been used for the treatment of neuralgia, arthritis, and gout etc. in folk remedies. The methanol extract of the root was shown significant antiinflammatory effect in the paw edema induced by carrageenin, and also indicated remarkable preventive and therapeutic effects to chronic arthritis model induced by Freund's complete adjuvant in rats. It also has inhibited significantly the increase in vascular permeability and the number of writhings induced by acetic acid in mice.

To investigate effects of Paeonia Radix Rubra aqua-acupuncture on Adjuvant Athritis in rats, the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum were measured and histological test of the muscular tissue were done in the arthritis part. 1. After arthritis of Sprague dawley(SD) rats was induced by injecting Freund's complete adjuvant for 2 weeks, normal saline was injected for the Control group and Paeonia Radix Rubra acua-acupuncture was injected for the Exp. group during 30 days. Selected point was $D{\acute{u}}bf(ST_{35})$ in both groups. And then the inframmator volume of plantar were checked. The volume of the plantar was $0.84{\pm}0.14mm$ in Control group and $0.57{\pm}0.09mm$ in Exp. group, the swelling of plantar was restricted significanily in Exp, group(P<0.05). 2. The number of WBC was $10.34{\pm}0.14(10^3/ml)$ in normal group and $37.47{\pm}5.46(10^3/ml)$ in Control group. It was $25.21{\pm}3.45(10^3/ml)$ in Exp. group. This fact showed that Exp group. with Paeonia Radix Rubra acua-acupuncture was more effective than Control group in the treatment of arthritis(P<0.05). 3. The content of total protein in the blood serum were $6.14{\pm}0.43g/d{\ell}$ in normal group, $7.95{\pm}0.94g/d{\ell}$ in Control group, and $6.82{\pm}1.23g/d{\ell}$ in Exp. group. There was no significance in total protein between Exp. group and Control group from the statistical analysis. 4. The contents of albumin in the blood serum were $2.94{\pm}0.13g/d{\ell}$ in normal group, $2.01{\pm}0.48g/d{\ell}$ in Control group and $2.75{\pm}0.14g/d{\ell}$ in Exp, group. This fact showed that Exp. group had significant increase in the serum albumin from the statistical analysis, in comparison with Control group(P<0.05). 5. The contents of globulin in the blood serum were $3.19{\pm}0.48g/d{\ell}$ in normal group, $4.70{\pm}1.26g/d{\ell}$ in Control group and $3.58{\pm}0.73g/d{\ell}$ in Exp. group. There was no significance in the serum globulin between Exp. group and Control group from the statistical analysis. 6. In histological examinadon, because inflammatory reaction was active, remarkably irregular tissue and large amount of inflammatory cells were found in Control. group. But Exp. group showed that tissues were in some degree recovered, the number of inflamed cells was decreased and inflammation was restrained. From these results, it is shown Paeonia Radix Rubra aqua-acupuncture restrained inflammatory reaction and muscular tissue necrosis in SD rat's plantar was restrained by Freund's complete adjuvant.
