Effect of Exogenous Application of Sodium Nitroprusside on Alleviation of Low Temperature Stress in Kimchi Cabbage (Brassica rapa ssp. pekinensis) (Sodium Nitroprusside 처리가 배추의 저온 스트레스 경감에 미치는 영향)

  • The effects of exogenous sodium nitroprusside (SNP, nitric oxide donor) on the growth, yield, photosynthetic characteristics, and antioxidant enzyme activity of kimchi cabbage (Brassica rapa L. subsp. pekinensis (Lour.) Hanelt) was studied under the low temperature conditions. Kimchi cabbages were treated with SNP of three concentrations (7.5, 15, 30 mg·L-1) for three times at four-day intervals and exposed to low temperature (16/7℃) stress for seven days. SNP treatment induced increases of net photosynthetic rate (Pn), stomatal conductance (Gs), intracellular CO2 concentration (Ci) and transpiration rate (Tr) under the stress condition with the highest level after the third treatment. The contents of malondialdehyde (MDA) and H2O2 were significantly lower in the treatment of SNP compared to the non-treated control. The activity of ascorbate peroxidase (APX), catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD), increased in treated plants by up to 38, 187, 24 and 175%, respectively compared to the non-treated control. SNP-treated and untreated plants had similar growth characteristics. Compared to the control group, SNP-treatment increased fresh weight and leaf area by 5%. Overall, our findings suggest that the application of sodium nitroprusside to the leaves contributes to reducing physiological damage and enhancing the activities of antioxidant enzymes, thereby improving low temperature stress tolerance in kimchi cabbage.

  • This study compared the production systems and packaging methods as pre- and post-harvest factors for Korean head cabbage, which is minimally processed at the industry locus. It examined how different crop production systems, specifically winter and spring cultivations, affect the characteristics of the Korean head cabbage after harvest. The quality of changes in cabbage characteristics was evaluated over 4 weeks at 10℃. After harvesting, soluble solids content (SSC), hardness, and DPPH (2,2-diphenyl-1-picrylhydrazyl) levels exhibited variations depending on the crop production system, with winter kimchi cabbage maintaining higher levels. These attributes were also influenced by the crop systems and packaging methods. Significant differences were observed in the changes in fresh weight of the Korean head cabbage depending on the packaging method; the extent of these changes was reduced with polypropylene (PP) film packaging. Although the freshness of Korean head cabbage decreased during storage, the appearance was somewhat preserved through film packaging after harvest. The results indicated that the properties of stored Korean head cabbages experienced different effects on pre- and post-harvest quality and characteristics, depending on the measurement items.

Selection of Plant Species for Phytoremediation of Arsenic Contaminated Sandy Soil in a Pine Forest at Janghang, Korea (장항 송림 비소오염토양의 식물재배정화를 위한 식물종 선정)

  • A series of experiments were performed to select suitable plant species for phytoremediation of arsenic (As) contaminated sandy pine forest soil in Janghang. Native plant species could uptake and remove As and sustain growth under the allelopathy of the pine forest and half-shade environmental conditions. We transplanted a total of 11 species into the pine forest in a pot, cultured them in a greenhouse for 3 months, and then harvested these plants to measure As accumulation, fresh weight, bioconcentration factor (BCF), and the amount of As removal per plant. The BCF of Lampranthus spectabilis was 3.52 and the amount of As taken up in Pennisetum alopecuroides shoots was 111.95 mg/kg. Higher biomass plants Lampranthus spectabilis and Lonicera japonica took up 8.49 mg/kg and 2.87 mg/kg of As in the above-ground parts, respectively. We applied oxalaic acid of 10, 20, and 40 mmol/kg-soil in total (divided into 15-20 splits) to each pot over a period of one month to enhance As uptake. Results showed no significant changes in plant growth or soil dehydrogenase activity. However, a statistically significant increase (p<0.05) in As uptake in Pennisetum alopecuroides was observed when a higher amount of oxalic acid (40 mmol/kg-soil) was applied.

Establishment of an Efficient Screening Methods for Resistance of Chinese Cabbage to Clubroot Disease (배추 뿌리혹병의 효율적인 저항성 검정법 확립)

  • Clubroot caused by Plasmodiophora brassicae is an important disease of crucifer crops worldwide. This study aimed to establish an efficient screening method to determine resistant cultivars of Chinese cabbage against P. brassicae. To do this, we investigated the virulence of seven P. brassicae isolates using seedlings of susceptible Chinese cabbage cultivar. The isolates exhibited different virulence in the plants and were divided into three groups based on their virulence. When we explored the disease occurrence in Chinese cabbage seedlings according to photoperiod after inoculation of P. brassicae and incubation temperature, the plants with all-day light showed higher disease severity than seedlings cultivated under 14 hr of light a day. The occurrence of clubroot disease was most severe at 25℃, followed by 20℃ and 18℃, but the fresh weight of clubroot of the seedlings cultivated at 20℃ was the highest, followed by plants grown at 25℃ and 18℃. When the seedlings of two commercial resistant cultivars were inoculated with the mixed spore suspensions of two different pathotype isolates of P. brassicae, disease severity increased as the spore concentration of the susceptible P. brassicae isolate among the two strains increased, suggesting that the clubroot development by different pathotype isolates was independent and not influenced by each other. Taken together, our results provide a faster and more accurate screening methods to determine the resistance of Chinese cabbage against P. brassicae.

Effect of Air Temperature on Growth and Phytochemical Content of Beet and Ssamchoo (온도처리가 비트와 쌈추의 생육과 생리활성 물질 함량에 미치는 영향)

  • The consumption of leaf vegetables has been steadily increasing in Korea. Leaf vegetables are used for "Ssam (vegetable wrap-up), eaf vegetables has been steadily increasing in Korea. Leaf vegetables are used for asoned condiments inside several layers of young vegetable leaves. This study investigated the effect of air temperature on the growth and phytochemical contents of beet (Beta vulgaris L.) and Ssamchoo (Brassica lee L. ssp. namai) grown in a closed-type plant factory system where fluorescent lamps were used as an artificial light source. Seeds of beet and Ssamchoo were sown in a peat-lite germination mix. The roots of 20-day-old seedlings were washed, and the seedlings were planted on a styrofoam board and grown in hydroponic beds for 25 days under fluorescent light. Plants were exposed to one of three different air temperature regimes (20, 25 and $30^{\circ}C$ during the day combined with $18^{\circ}C$ during the night), which were monitored with a sensor at 30 cm above the plant canopy. Increased plant height and leaf area were observed in beet at $25^{\circ}C$ and $30^{\circ}C$ compared to $20^{\circ}C$. For Ssamchoo, the greatest plant height, leaf area, fresh weight and dry weight were obtained at $20^{\circ}C$. Ascorbic acid content of beet and Ssamchoo leaves were highest at $30^{\circ}C$. In beet, total polyphenol and flavonoid contents were higher at $20^{\circ}C$ (42.4, $197.0mg{\cdot}g^{-1}DW$) and $25^{\circ}C$ (46.9, $217.0mg{\cdot}g^{-1}DW$) than $30^{\circ}C$ (22.4, $88.0mg{\cdot}g^{-1}DW$). In Ssamchoo, total polyphenol and flavonoid contents were also higher at $20^{\circ}C$ (79.2, $268.2mg{\cdot}g^{-1}DW$) and $25^{\circ}C$ (66.3, $258.3mg{\cdot}g^{-1}DW$), respectively, than $30^{\circ}C$ (53.7, $134.7mg{\cdot}g^{-1}DW$). Hence, the optimum temperature appears to be $20^{\circ}C$ for growing both beet and Ssamchoo in a closed-type plant factory system with fluorescent light.

Effect of Shading, Light Quality, and Chemical Elicitation on Growth and Bioactive Compound Content of Potentilla kleiniana Wight et Arnott (가락지나물의 생장과 생리활성물질 함량에 미치는 차광, 광질 및 화학적 엘리시테이션)

  • Potentilla kleiniana is a perennial herb beloning to Rosaceae family. Herein we investigated the effect of light intensity, light quality and chemical elicitor on plant growth and the accumulation of bioactive compounds in P. kleiniana. After 60 days of cultivation under different shading level [0% (200 μmol·m−2·s−1), 35% (95 μmol·m−2·s−1), 55% (65 μmol·m−2·s−1), 75% (40 μmol·m−2·s−1)] in the greenhouse, chlorophyll and carotenoid content were the highest under 35% treatment, however, plant height, leaf number and biomass were the highest under non-shading. As a result of cultivation among strong light condition as a control, florescence and three mixture light sources [red:white:blue (RWB) = 8:1:1, red:blue (RB) = 8:2, red:green:blue (RGB) = 8:1:1] as treatments in plant growth chamber (25 ± 2℃, 185 ± 3 μmol·m−2·s−1), growth, biomass, chlorophyll content low difference between total phenolic compouds and flavonoid content were higher under RWB treatment. DPPH radical elimination ability was the highest under all treatments especially florescence and RGB treatment except control. As a result of treating chemical elicitor [salicylic acid (SA), methyl jasmonate (MeJA)] concentration (0, 50, 100, 200 μM) respectively, plant height, petiole diameter and biomass were higher under non-treatment, MeJA 50 μM. It was investigated that fresh weight and dry weight under MeJA 50 μM treatment were especially a little high. Total phenolic compounds and flavonoid content of SA 50 μM treatment was the highest but DPPH radical elimination ability was significantly the highest under MeJA 200 μM (88.65%) and MeJA 50 μM (87.84%) treatment. Thus, this study suggested that we determined optimal shading and light quality in the greenhouse and plant growth chamber also confirmed bioactive compound content, antioxidant ratio increase according to different chemical elicitation concentration.

Effect of Light Intensity on the Growth Responses of Three Woody Plants for Indoor Landscaping (실내녹화용 목본식물 3종의 초기 생육반응에 미치는 광량의 영향)

  • The purpose of this study was to investigate the effects of light intensity on the initial growth response of three woody plants for indoor landscaping; Ardisia pusilla, Clusia rosea and Fatsia japonica. The plants were planted in 10cm pots, the light intensities used were of four levels-15, 30, 60, $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD-and light irradiation time was set to 12/12 (day/night). Growth responses including plant height, leaf length, leaf width, chlorophyll fluorescence (Fv/Fm), SPAD and Hunter values were measured at 4-week intervals, and shoot weight and root weight of fresh and dry plants were measured after completion of the experiment. Fatsia japonica tended to show greater leaf length and leaf width as light intensity became greater, while other plants did not show any significant differences at different light intensities. The Fv/Fm value of the Ardisia pusilla was found to be stressed at 60 and $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, while the Fv/Fm values were within normal range with other plants or at other light intensity levels to show no stress. Only Clusia rosea showed significantly different SPAD values at $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and there was no significant SPAD value difference found with other plants or at other light intensity levels. While Hunter values of the Ardisia pusilla did not show any significant differences at any light intensity levels, Clusia rosea and Fatsia japonica showed specificity in L, a and b values at 60 and $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. Ardisia pusilla showed a big stem growth at $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and Clusia rosea showed a steady growth at 60 and $120{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$.

Effect of Dietary Supplementation of Fresh Water Algae Euglena on the Performance and Fatty Acid Composition of Breast Muscle of Broiler Chickens (육계에서 담수녹조류 Euglena 첨가사료가 생산성 및 흉근의 지방산 조성에 미치는 영향)

  • A feeding trial was conducted with Euglena strains grown under different media. The effect of supplementation of Euglena on the performance, nutrient availability and fatty acid composition of breast muscle was studied. In experiment I, two hundred ten hatched broiler chicks (Ross) were assigned to seven dietary treatments for 5 weeks. Each treatment consisted of 3 replications with 10 birds each. Control diet was formulated to have $22\%$ CP and 3,150 kcal ME/kg for starter diet, $19\%$ CP and 3,200 kcal ME/kg for finisher diet. Euglena gracilis Z. (EG) was added to control diet at the plevel of 0.25, 0.5, $1.0\%$ and Euglena gracilis Z. bleached and DHA enriched (EGBD; a strain mutated by streptomycin and cultivated in DHA enriched medium) at the level of 0.5, 1.0, $2.0\%$ in the diet. In experiment 2, two hundred fifty hatched broiler chicks (Ross) were assigned to five dietary treatments: T1; Control, T2; T1 + Euglena gracilis Z. DHA enriched (EGD; cultivated in DHA enriched medium) $0.5\%$, T3; T1 + EGD $1.0\%$, T4; T1 + EGBD $0.5\%$, T5; T1 + EGBD $1.0\%$. The weight gain and feed consumption were measured weekly. Fatty acid composition of breast muscle was determined. In experiments I and 2, Euglena supplementation had no significant effects on weight gain, feed intake and feed conversion ratio. In experiment 1, EGBD treatments significantly increased DHA concentration but decreased concentration of linoleic acid and arachidonic acid in breast muscle. EGBD 2% treatment showed the highest DHA concentration (14.27%) which is 3.9 times of that of the control ($3.66\%$). In experiment 2, $1.0\%$ EGBD treatment showed highest EPA, lignoceric acid and DHA level in breast muscle (P<0.05). Also, EGD treatments significantly increased DHA and EPA concentration. It was concluded that EGBD and EGD can be supplemented to broiler diet to produce DHA enriched broiler meat.

  • Crisphead lettuce(Lactuca sativa L.) was grown in NFT to investigate the effects of NO$_3$-N and NH$_4$-N ratio in nutrient solution and elevated $CO_2$ treatment in the crisphead lettuce growth. This experiment has been conducted under three different ratios of NO$_3$-N:NH$_4$-N(100:0, 75:25, 50:50) with two $CO_2$ concentration (control, 1500ppm ). The results are as follows; 1. In the case of not controlling pH and EC in nutrient solution, pH was gradually increased in NO$_3$-N:NH$_4$-N=100:0 treatment but rapidly decreased in the nutrient solution 2. Daily changes of NO$_3$-N and NH$_4$-N were observed without controlling the nutrient solution. In the treatments of NO$_3$-N:NH$_4$-N ratios were 75:25 and 50:50, NO$_3$-N absorption rates were 27.7% and 26.1%, while NH$_4$-N absorption rates were 87.9% and 71.2%, respectively. 3. There was little differences in total nitrogen of leaves. However phosphorus, potassium, calcium and magnesium contents were highly shown in the treatment of $CO_2$ 1500ppm and 100:0 ratio of NO$_3$-N:NH$_4$-N. 4. Higher $CO_2$ assimilation rate was shown in plants grown under $CO_2$ 1500ppm and 100:0 ratio of NO$_3$-N:NH$_4$-N. It dropped significantly with the increase of NH$_4$- N rates in nutrient solution. 5. Fresh weight, leaf number, root length and root weight of crisphead lettuce were far better in the treatment of $CO_2$ 1500ppm and 100:0 ratio of NO$_3$-N:NH$_4$-N. Growth differences by $CO_2$ elevation were not shown in other NO$_3$-N:NH$_4$-N treatments. 6. The highest nitrate contents of leaves were shown in NO$_3$-N single treatment but shown the lowest vitamin C contents. Nitrate contents of leaves were decreased by $CO_2$ but the effect was slight treatment.

Effects or $H_2O$ Fraction or Dioscorea japonica Thunb with selenium on plasma Glucose and Lipid Metabolism in Streptozotocin Induced Diabetic Rats (당뇨 흰쥐에서의 참마 $H_2O$ 분획물과 Selenium 보충시 혈당 및 지질대사에 미치는 영향)

  • The purpose of this study was to investigate the effect of H$_2$O fraction of Dioscorea japonica Thunb(DJT) with selenium(Se) treatment on blood glucose and lipid metabolism in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats(180-220g) weighing were divided into five groups, that is one normal group and four diabetic experimental groups : the STZ-control group, the DJT group, the DJT-Se group and the Se group. Diabetes mellitus was induced in the male rats by injection of STZ into the tail vein at a dose of 45mg/kg B.W. The H,0 fraction of DJT(500mg/kg) given orally were administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na$_2$SeO$_3$(2mg/kg diet), which was prepared fresh daily. The body weight and food intake was monitored daily and plasma levels of glucose, insulin, hematocrit and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglyceride and fire fatty acid were measured. The activities of aminotrans ferase were analysed. The body weight gain was shown to be significantly higher in the normal group than all diabetic groups. The blood glucose levels of the DIT-Se group was significantly lower compared to those of the experimental groups. The administration of H$_2$O fraction of DJT and selenium showed an increase in plasma protein concentrations. The plasma cholesterol levels of all STZ-groups were not significantly different and HDL-cholesterol levels were increased in the diabetic experimental groups fed on H$_2$O fraction of DJT or Se supplementation. Plasma triglyceride levels were lower in the DJT-Se and Se group than in the STZ-control group. free fatty acid levels were not significantly differ among STZ-control groups. STZ treatment increased aminotransferase activity and that of DJT group was highest. In conclusion, the data from the present experiments indicate that the treatment of the H$_2$O fraction of DJT with selenium showed a synergistic effect and the two can have an influence on hyperg1ycemia and lipid metabolites when administered together.
