PURPOSES : The objective of this study is to evaluate the environmental resistance of bio-polymer concrete for use of pavement materials developed for reducing the carbon-dioxide. METHODS : The compression, tension, and bending strength tests were conducted on the bio-polymer concrete specimens with and without environmental conditioning. The specimens were conditioned using the freezing-thaw and accelerated weathering process for long period of time. To assess the resistance against chloride, the chloride ion penetration resistance tests were carried out on the bio-polymer concrete specimens. RESULTS : Test results show that the maximum difference in strength between specimens with and without conditioning is about 2.6MPa indicating that the effect of environmental conditioning on specimen strength is negligible. Based on the chloride ion penetration resistance test, the penetration quantity of electric charge of the specimens is zero and there is no ion penetration within the bio-polymer concrete. CONCLUSIONS : It is found from this study that there is slight change in strength of bio-polymer concretes before and after environmental conditioning process and no chloride ion penetration observed in these specimens. Therefore, the developed bio-polymer concretes can be applied effectively as pavement materials due to the small change of physical properties with environment change.
To evaluate the practical application of oyster shells as construction materials, an experimental study was performed. More specifically, the long-term mechanical properties and durability of concrete blended with oyster shells were investigated. Test results indicate that long-term strength of concrete blended with 10% oyster shells is almost identical to that of normal concrete. However, the long-term strength of concrete blended with 20% oyster shells Is appreciably lower than that of normal concrete. 1'hereby, concrete with higher oyster shell has the possibility giving a bad influence on the concrete long-term strength. Elastic modulus of concrete blended with crushed oyster shells decreases as the blending mixture ratio increases. Namely, the modulus is reduced by approximately 10∼15% when oyster shells are blended up to 20% replacing the fine aggregate. The drying shrinkage strain increases as the blending ratio increases. In addition, the existing model code of drying shrinkage does not coincide with the test results of this study. An adequate prediction equation needs to be developed. The utilization of oyster shells as the fine aggregate in concrete has an insignificant effect on freezing and thawing resistance, carbonation and sulfuric acid attack of concrete recycling. However, water permeability is considerably improved.
Present studies were conducted to investigate the developmental stage and the location of embryos in the reproductive tract at various times after ovulation, the morphologically normal after thawing of embryos preserved in liquid nitrogen, and the survival after transferring frozen-thawed embryos. The results obtained were as follows: 1. Embryo stage and location in the reproductive tract after hCG administration. For the investigation of embryo stage and location in the reproductive tract after ovulation, rabbits were laparotomized at 24, 40, 48, 72 and 120 hrs post hCG injection, simultaneously with mating. the oviducts and uteri were flushed out with PBS medium containing 50% rabbit serum, respectively. 1) Most of embryos was remained in the oviduct within 48 hrs, with the lapse of time, embryos were started to move to uterus and shifted in uterus at 72 hrs after hCG injection. 2) The representatives of embryos stage collected at 24, 40, 48, 72 and 120 hrs were 1-cell(60.4%), 8-cell to early morula (52.3, 39.3%), late blastocyst (95.5%) stages, respectively. 2. Morphological normality and survival of the frozen-thawed embryos. For the evalution of the quality and viability on the frozen-thawed embryos, immediately after thawing, embryos were assessed by morphologically normal under a dissecting microscope, and a further test of frozen-thawed embryos was made by transferring the morphologically normal embryos to the uteri of recipient rabbit induced pseudopregnancy by the injection of hCG at the time of hCG injection in donor rabbits. 1) The propotions of embryos which a, pp.ared morphologically normal was higher when 8-cell (85.7%) and morula(90.5%) were used for freezing than when 4-cell (66.7%) and blastocyst (75.8%) were used. 2) Preganacies were observed at Day 15 after transfer of frozen-thawed 8-cell (7/13), morula (19/42) and blastocyst (3/19) but not after transfer of embryos at 4-cell stage.

The purpose of this study was to evaluate the effect of addition of ethylene glycol, glycerol and sucrose to TCG (Tris, Citric Acid, Glucose, Egg Yolk) and DMSO Frozen. The extender containing Egg yolk concentration (10%, 20%) affects viability and acrosome morphology of rabbit sperm. Sperm viability was then assessed for the freezing extenders TCGD (Tris + Citricacid + Glucose + DMSO), TCGED (Tris + Citricacid + Glucose + Egg yolk + DMSO), TCGGD (Tris + Citricacid + Glucose + Glycerol + DMSO) and TCGSD Tris + Citricacid + Glucose + Sucrose + DMSO) during thawing at 38℃. for 20 seconds, respectively. TCG + 10% egg yolk (viability: 77.0 ± 0.8, NAI: 73.3 ± 0.9) was significantly (sperm viability and normal acrosome interaction (NAI)) higher than TCG + 20% egg yolk (70.7 ± 1.1, 70.0 ± 0.9) in the sperm normalcy analysis according to the yolk concentration. TCGGD (53.4 ± 0.1, 62.3 ± 0.4), TCGSD (61.3 ± 0.0, 67.1 ± 0.1) sperm viability and normal acrosome interaction (NAI) in frozen spermatozoa are TCGD (46.4 ± 2.8 and 56.3 ± 1. 4) and TCGED (23.0 ± 1.1 and 54.6 ± 1.4) extenders was thawed at 38℃ for 20 seconds. According to the results from each frozen bulking agent, sperm membrane integrity by hypotonic swelling test (HOST) analysis in TCGGD (59.8 ± 0.7), TCGSD (59.3 ± 0.5) was significantly high compared to other experimental groups (p < 0.05). In conclusion, these results suggested that TCGGD and TCGSD extenders enhance survivability of rabbit sperm after frozen-thawing.
This study was attempted to develop the physicochemical ad morphological stability test methods for the cream and lotion formulations among the cosmetic foundations and to provide the guidance for the stability methods with respect to basic emulsions and creams. With these developed stability test methods, we can evaluate the expired date or life time of the available basic cosmetics, especially basic lotions ad creams. Also, the stability test methods established in this study can be used as a guideline to test physical and morphological stability of cosmetics in the future. Thus, we selected two types of basic cosmetics such as lotions and creams made by four different cosmetic companies ad applied them to the stability test methods depending on the temperature changes such as temperature cycling and freezing-thawing cycling test. After the temperature changes, the conductivity, turbidity, particle size, creaming ratio and pH changes of the creams and lotions were evaluated and morphological changes such as crystal formation, odor, color and feeling of the creams and lotions were also tested. As the results of the stability tests, all the tested creams and lotions except for one lotion were stable. Therefore, it may be concluded that these short-term accelerated stability tests as physical stability test depending on the temperature change study were suitable for the stability testing methods for the basic cosmetics and may be useful for the establishment of the guideline for the stability test of cosmetics.
This study was to test whether Hanwoo in vitro matured oocytes can be successfully cryopreserved by a new vitrification procedure using MVC method. For the vitrification, oocytes were pretreated in 10% ethylene glycol (EG10) for 5-10 min, exposed in EG30 for 30 sec, each oocytes were individually put on the inner wall of 0.25 $m\ell$ straw, and then straws were directly plunged into L$N_2$. Thawing was taken by 4-step procedures [1.0 Msucrose (MS), 0.5 MS, 0.25 MS, and 0.125 MS] at 37$^{\circ}C$. In vitro developmental capacity (survival, cleavage ($\geq$2-cell) and blastocyst rates) in vitrified group was no significant difference compared to that in other treatment groups (exposed; 100.0, 74.4, 32.3% and control; 100.0, 78.3, 36.3%): high mean percentage of oocytes (91.2%) was survived, 69.4% of them were cleaved and 27.9% of cleaved embryos were developed to blastocyst. Especially, after transfer of in vitro developed embryos in vitrified group, four of six recipient animals were found to pregnant and three of them were ongoing pregnant by manual palpation at 250 days after transfer. However, among them, two healthy female calves (23 and 25kg) were born. This result demonstrates that MVC method is very appropriate freezing method for the Hanwoo in vitro matured oocytes and that ovum bank can be maintained efficiently by MVC cryopreservation method.
현재 건축구조물에 사용되는 단열재의 단열성능은 초기성능을 기준으로 설계 시에만 반영하고 있으며, 장기 내구성 저하에 따른 단열성능 감소는 반영되고 있지 않다. 본 연구에서는 가속내구성 시험을 통해 단열재의 내구성 저하에 따른 단열성능 저하를 검토하였다. 연구결과, 발포폴리스티렌 단열재 비드법의 경우 표준환경 조건 및 실험실 가속 시험 조건에서는 경시변화에 따른 성능저하가 나타나지 않았으나, 동결 융해 시험 조건에는 성능이 저하하는 것으로 나타났다. 반면, 압출법의 경우 동결 융해 시험 조건에서는 단열성능 저하가 작았지만, 경시변화 초기에 내부 가스의 방출로 급격한 성능저하를 나타내었다. 또한, 경질 폴리우레탄 폼 단열재의 경우 다른 단열재에 비해 초기 단열성능이 매우 뛰어났으나, 실험실 가속 시험조건 및 동결 융해 시험 조건에서는 성능이 다소 저하하는 것으로 나타났다.
PURPOSES: The purpose of this study is to evaluate physical properties, durability, fatigue resistance, and long-term performance of poly-urethane concrete (PU) which can be possible application of thin layer on long-span orthotropic steel bridge and to check structural stability of bridge structure. METHODS : Various tests of physical properties, such as flexural strength, tensile strength, bond strength and coefficient of thermal expansion tests were conducted for physical property evaluation using two types of poly urethane concrete which have different curing time. Freezing and thawing test, accelerated weathering test and chloride ion penetration test were performed to evaluate the effect of exposed to marine environment. Beam fatigue test and small scale accelerated pavement test were performed to assess the resistance of PU against fatigue damage and long-term performance. Structural analysis were conducted to figure out structural stability of bridge structure and thin bridge deck pavement system. RESULTS: The property tests results showed that similar results were observed overall however the flexural strength of PUa was higher than those of PUb. It was also found that PU materials showed durability at marine environment. Beam fatigue test results showed that the resistances of the PUa against fatigue damage were two times higher than those of the PUb. It was found form small scale accelerated pavement test to evaluate long-term performance that there is no distress observed after 800,000 load applications. Structural analysis to figure out structural stability of bridge structure and thin bridge deck pavement system indicated that bridge structures were needed to increase thickness of steel deck plate or to improve longitudinal rib shape. CONCLUSIONS: It has been known that the use of PU can be positively considered to thin layer on long-span orthotropic steel bridge in terms of properties considered marine environment, resistance of fatigue damage and long-term performance.
In this study, Field test was performed using high-quality additions and accelerators to obtain the improvement of the strength on domestic shotcrete and quality test based on EFNARC was performed. In addition, Deterioration test that combined the Freezing-thawing and Carbonation was also performed in order to investigate a long-term durability of high-strength shotcrete. As a result of field test, a promotion ratio of early strength is $90\sim97%$ in case of using alkali-free accelerators. And a compressive strength of shotcrete using Micro-silica fume was $45.2\sim55.8MPa$ and the flexible strength was $5.01\sim6.66MPa$, so a promotion ratio of strength was $37\sim79%$, $17\sim61%$ respectively. It was showed that increment effect of strength by the Micro-silica fume replacement of $7.5\sim10%$ for cement mass was remarkable. It was also realized that application of Micro-silica fume to shotcrete reduced deterioration and improved a long-term durability of shotcrete.
The purpose of this study was to investigate the effects of developmental stage and equilibration time on survival of rabbit embryos following freezing by vitrification. Adult New Zealand White female rabbits were superovulated with PMSG and hCG. The 8-cell stage embryos were collected from 40 to 45 hours after hCG injection by flushing oviducts with Dulbecco's phosphated buffered saline and in vitro cultured in TCM-199 containing 10% fetal calf serum(FCS). Each embryos developed in vitro to 16-cell, compact morula and blastocyst was cryopreserved and cultured following thawing to examine their developmental potential to expanded blastocyst stage in vitro. The frozen-thawed-cultured embryos were stained with Hoechst 33342, and their nuclei were counted using a fluorescence microscope. On the toxicity test of EFS solution as cryopreservation, the survival rates of 8-cell stage embryos was decreased in reverse to increasing of exposure time over 5 minutes. The post-thaw survival rates of embryos on equilibration times was significantly(P<0.05) higher for 2 min. than for 5 or 10 minutes. From morula to blastocyst of rabbit embryos was more suitable than 8-cell stage for cryopreservation by vitrification. The higher post-thaw survival rate of embryos can be achieved by keeping the cryoprotectant at $4^{\circ}C$ than at $20^{\circ}C$. The mean number of nuclei per embryo following freezing by vitrification and in vitro culture to expanded blastocyst at compacted morula and blastcyst was not significantly differ from fresh blastocyst.
