차밭에 설치된 차광망의 동해경감 효과 (Effect of Shade Net on Reduction of Freezing Damage at a Tea Garden)

  • 월동기간 동안의 차나무에 대한 차광망 색상과 차광율에 따른 동해경감에 미치는 영향을 조사한 결과, 차광율 변화에 대한 효과는 차광율이 높아질수록 차나무생육이 불량하고 동해 피해율이 높았으며, 무처리구 대비 55% 차광율에서 신초 및 생엽수확량이 좋게 나타났다. 차광망 색상별로 보면, 투명망(차광망 55% 수준) 처리구와 녹색 차광망 처리구가 무처리구와 검정 차광망 처리구에 비해 생엽수확량과 피해면적이 감소하였다. 투명망 처리구가 무처리구에 비해 동해 피해율이 50%이상 감소하고, 생엽수확량은 무처리구보다 투명망 처리구에서 단위면적당(10a) 68kg 더 많았다. 차광망의 색상별 동해경감은 검정<녹색<투명 색상 순으로 동해 경감률이 다소 높게 나타났다. 처리구간의 미기상변화를 보면 처리구가 무처리구에 비해 평균기온은 $0.7^{\circ}C$ 낮았으며, 평균상대습도는 14.9% 높게 관측되었고, 지중온도는 $0.6^{\circ}C$ 낮은 반면에 토양수분은 4.6% 높게 관측 되었다. 또한 차광망 피복물 설치에 의한 평균풍속은 무처리구 대비 0.7m/s 감소하여, 바람에 의한 과잉 증발산과 토양의 건조를 줄여주는 효과가 있었으며, 동해경감의 방안으로 활용이 가능한 것으로 판단되었다.

인간 다-전핵기 (>2PN) 수정란의 초급속 동결에 관한 연구 (Cryopreservation of Human Multi-Pronuclear (PN) Zygote by Ultra-Rapid Freezing)

  • 본 연구는 인간 전핵기 수정란의 동결시 election microscope grid론 사용하는 초급속 동결방법이 유용하는지 여부를 조사하고자 실시하였다. 본 실험에서는 인간 IVF 시술시 생성되는 다-전핵기 (>2PN) 수정란을 정상 2PN 수정란 대신 사용하였으며, 또한 이들은 3PN과 $\geq4PN$ 수정란으로 나누어 전핵의 수에 따른 냉해와 융해 후 체외 배발달율을 조사하였다. 동해제는 30% ethylene glycol, 18% ficoll, 0.5 M sucrose와 10% FBS 등이 D-PBS에 첨가되어 제작된 EFS30을 사용하였다. 본 연구에서 얻어진 결과는 다음과 같다. 초급속동결-융해 후, 인간 다-전핵기 수정란의 생존율은 85.5%였다. 대조군과 동결군의 난할율을 진핵의 수에 따가 니누어 비교하였을 때, 각 군간에는 유의한 차이를 나타내지 않았다 (3PN; 81.3%와 85.4%, $\geq4PN$; 90.0%와 95.7%). 또한, 융해 후 체외발달율을 조사하였던바, $\geq4PN$수정란에서는 동결군 (4.5%)의 체외발달이 대조군 (44.4%)보다 유의하게 낮게 나타났던 반면, 3PN 수정란에서는 동결군 (22.0%)의 결과가 대조군 (38.5%)에 비해 유의한 차이를 나타내지 않았다. (p<0.05). 따라서 인간 다-전핵기 수정란은 EM grid와 EFS30 동결액을 이용한 초급속 동결로 배발달을 유도할 수 있다는 것을 알 수 있었다.

Effects of Discontinuous Percoll Gradient Containing Alpha-linolenic Acid on Characteristics of Frozen-thawed Boar Spermatozoa

  • This present study was conducted to investigate protective effect of discontinuous Percoll gradient containing alpha-linolenic acid (ALA) before freezing process on viability, acrosome damage, mitochondrial activity, and oxidative stress of frozen-thawed boar spermatozoa. The separation of spermatozoa by discontinuous Percoll gradient was performed by different concentration of Percoll solution (45/90%) containing ALA combined with bovine serum albumin (BSA), and collected sperm in each Percoll layer was cryopreserved. To evaluate viability, acrosome damage, mitochondrial activity, and reactive oxygen species (ROS) level of frozen-thawed sperm, flow cytometry was used. Morphological abnormalities were observed under light microscope. In results, viability of sperm from 90% Percoll layer was higher than control and 45% Percoll group (p < 0.05). Separated sperm in 90% Percoll layer had lower acrosome damage and morphological abnormalities than control as well as viability, whereas 45% Percoll group was higher (p < 0.05). Similar with acrosome damage and abnormalities, mitochondrial activity was slightly enhanced and the population of live sperm with high ROS level was decreased by 90% Percoll separation, however, there was no significant difference. Supplementation of 3 ng/mL ALA into Percoll solution increased sperm viability and decreased population of live sperm with high ROS compared to control (p < 0.05). In conclusion, discontinuous Percoll gradient before freezing process could improve efficiency of cryopreservation of boar sperm through selection of sperm with high freezing resistance, and supplement of ALA during Percoll gradient might contribute suppression of ROS generation via stabilizing of plasma membrane during cryopreservation.

점안용 우황약침액의 안점막자극실험 (Eye Irritation Test of Bovis Calculus Pharmacopuncture Solutions for Eye Drop)

  • Objective This study was done to investigate the safety of Bovis Calculus pharmacopuncture solution manufactured with freezing dryness method to use eye drop. Methods The eye irritation test of this material was performed according to the Regulation of Korea Food & Drug Administration (2005. 10. 21, KFDA 2005-60). After Bovis Calculus pharmacopuncture solution was medicated in the left eye of the rabbits, the auther observed eye irritation of the cornea, iris, conjunctiva at 1, 2, 3, 4 & 7day. Results 1. After Bovis Calculus pharmacopuncture solution was medicated in the left eye of the rabbits, there wasn't physical problem at 9 rabbits. 2. After Bovis Calculus pharmacopuncture solutionwas medicated in the left eye of the rabbits, there wasn't eye irritation of the cornea, iris, conjunctiva at 1, 2, 3, 4 & 7day. Conclusions I suggested that Bovis Calculus pharmacopuncture solution didn't induced eye irritation in rabbits.

굴절률을 이용한 수용액의 빙충전율 측정 (Measurement of the Ice Packing Factor of an Aqueous Solution Using the Index of Refraction)

  • In the present paper, a new method is proposed for the measurement of the ice packing factor (IPF) of an ice slurry using the index of refraction. The purpose of the new method is to improve the resolution of the measurement and to increase its resistance to electric noise as compared to the standard IPF measurement technique that relies on measurement of the freezing temperature. These two methods are similar in that they both obtain a concentration of aqueous solution from measured physical quantities and calculate the IPF using a relation between concentration and IPF. We experimented and compared the two methods, whose results were also compared with results from the calorimeter method obtaining the IPF directly They are in good agreement (within $5\%$), which demonstrates the validity of the newly proposed method.

  • The objective of this study is to examine an effective cryopreservation method and various vitrification containers on the survival vates of embroys. For the vitrification, in vitro produced embryos at blastocyst stage were exposed to ethylene glycol 5.5 M freezing solution (EG5.5) for 20 sec, loaded on containers such as grid, straw and paper and then immediately plunged into - 196$^{\circ}C$ L$N_2$. The blastocysts were thawed serially in 0.5, 0.25 and 0.125%; P < 0.05). Therefore, this study suggests that bovine embryos can be easily, effectively and successfully cryopreserved by grid, straw, and paper in the presence of freezing solution. Furthermore, vitrification using paper may be used as a no M sucrose in CR1aa, each for 1 min, and cultured in CR1 an medium supplemented with 10% FBS. After thawing, there were not significant differences in recovery rates of EM grid, straw and paper as 84.6, 88.3, and 93.7%, respectively (Table 1). However, survival rates of EM grid (78.1%) and paper (77.1%) showed significantly higher than straw (52. 1w method for bovine embryos.

  • A study on the heat and mass transfer for the drying time in spray freezing drying process was experimentally presented in this paper. A spray freezing dryer for the production of an inhalable powder medicine is developed and an albumin solution as protein specimen is adopted. The freeze-drying for the albumin solution is tested in three different height of the particles piled up in a tray such as 2 mm, 5 mm, 7 mm, and the drying time is estimated as 10 h, 15 h, 22 h. From this results, the correlation is suggested and the drying time with various height of the particles piled up in a tray can be estimated. The assessment on the drying time is conducted thought a microbalance and the rate of water content is measured. The results based on temperature and weight are compared. The difference of the results happens due to the heat transfer from the lateral side of the vial. It is thought that the better estimation in drying time can be predicted by considering the heat transfer effect.

  • This study investigates anti-icing design by conjugate heat transfer analysis in lab-scale printed circuit heat exchanger (PCHE) for supply of cryogenic high pressure liquid hydrogen. The conjugate heat transfer analysis by using computational dynamics (CFD) provided various temperature distributions at important locations in PCHE heat exchanger and predicted the possibility of freezing in hot channel. And, the effect of inlet temperature of glycol water was analyzed in order to resolve the freezing problem in PCHE.

생쥐 2-세포기 수정란의 초급속동결 (Ultrarapid Freezing of Mouse 2-Cell Embryos)

  • This study was carried out in order to investigate effects of cryoprotectant concentration and equilibration time on survival of ultrarapidly frozen 2-cell mouse embryos. Mouse 2-cell embryos, following dehydration by exposure to DMSO and sucrose, were directly immersed into liquid nitrogen and thawed in 37$^{\circ}C$ water. Viability was defined by development rate to the blastocyst stage after in vitro culture for 72 hours. The results are summarized as follows ; 1. When 0.25M of sucrose was added into the freezing medium at various concentrations of DMSO and dilution medium, higher development rate of embryo was obtained in 3.0M DMSO concentrations (82.6%). However, when sucrose concentraitons of 0.25 and 0.5 M were added to the freezing medium with 3.0 M DMSO and dilution medium, development rate of embryos were 81.7% and 24.1%, respectively. 2. In the equilibration time at room temperature, higher development rate was attained after short period of time (2.5min) in 3.0 M DMSO+0.25 M sucrose (85.9%). 3. The development rate of embryos at in vitro 2-cell, in vitro 2-cell, solution control and untreated control was 84.6%, 90.9%, 89.9%,, and 89.7%, respectively.

태양광 에너지를 이용한 발열논슬립의 성능분석 (Performance Analysis of Heating Nonslip using Solar Power Energy)

  • This study aims to analyze the performance of Heated-nonslip using renewable energy to prevent nonslip freezing during winter. For this purpose, power generation system and congratulatory devices using solar energy are designed, and it is designed to provide regular electricity to heat up nonslip through Electrical storage system(ESS). In this study, It is intended to analyze the level of electrical energy suitable for nonslip using 24V or 48V, and to measure the temperature changes and temperature distribution according to the location of the test object. As a result of the experiment, nonslip's frame temperature was measured at $-7.5{\sim}-5^{\circ}C$ on average, and $-1{\sim}-2^{\circ}C$ on the heating cable during the supply of 24V and this could not be the solution for defrosting freezing nonslip in the winter. As a result of heating nonslip by supplying 48V with an electrical power of 8W, the temperature of the nonslip was shown to be between $5^{\circ}C$ and $11^{\circ}C$ to $13^{\circ}C$. Even if the power supply was switched on and off every minute, the temperature did not drop below $4^{\circ}C$ and the frozen ice melted on the nonslip without freezing.
