Effects of Pharbitidis Semen on 3T3-L1 Cells and Obesity-induced Rats (흑축이 3T3-L1 세포와 비만 유도 흰 쥐에 미치는 영향)

  • Objectives: This study was designed to evaluate the efficacy of Pharbitidis Semen (PS) on obesity using 3T3-L1 cells and high-fat-diet rats. Methods: After the administration of PS at a concentration of 0.125, 0.25, and 0.50 mg/ml in 3T3-L1 cells, the degree of differentiation of adipocytes was measured by Oil Red O staining. The experimental animals were divided into five groups in vivo. Normal group: diet-fed normal group (Normal), Control group (Control): high-fat-diet (HFD)-fed, Positive control group (PC): HFD+Contrave 200 mg/kg, PS_Low group: HFD+PS 250 mg/kg group, PS_High group: HFD+PS 500 mg/kg. Results: The total cholesterol, low density lipoprotein-cholesterol (LDL-cholesterol), and triglycerides (TG) content of the PS medication groups were significantly lower than those of the control group. In addition, high density lipoprotein cholesterol (HDL-cholesterol) tended to increase, and free fatty acid, total lipid, and phospholipid tended to decrease, but the decrease was not statistically significant. The administration of PS inhibited the differentiation of 3T3-L1 adipocytes, and a statistically significant difference was found in the group administered with 0.125 mg/ml. The mRNA expression of $PPAR-{\gamma}$ and leptin, which are involved in the differentiation of adipocytes, was observed using reverse transcription polymerase chain reaction (RT-PCR). The expressions of $PPAR-{\gamma}$ and leptin significantly decreased in comparison with the control and the positive control.

Nutritional Components and Physicochemical Properties of Hulled and Naked Oat Flours according to Particle Sizes (겉귀리 및 쌀귀리 분말의 입자크기별 영양성분 및 이화학적 특성)

  • Hulled oat (Chohan) and naked oat (Choyang) flours were analyzed according to particle size to investigate nutritional components and physicochemical properties. Particle size of naked oat flours was larger than that of hulled oat flours. As the mesh of the crushed ones decreased, particle sizes increased in value. The content of total ${\beta}$-glucan was highest in hulled oat flour (4.23%) with 60 mesh and in naked oat flour (4.26%) with 100 mesh. Most total ${\beta}$-glucan was soluble ${\beta}$-glucan in both flours (over 76%). Total starch of hulled oat flours (63.64~69.82%) was higher than that of naked oat flours (52.45~63.71%). Whereas amylose contents showed a negative correlation with total starch contents. Contents of free amino acids increased according to particle size, and each component was dependent on each type of amino acid. Moreover, while fatty acid composition was not significant, the content of most naked oats was higher than that of hulled oats. Besides, the ratio of unsaturated fatty acids (oleic acid and linoleate) in both types of oat flours was relatively higher than that of other grains. The pasting properties of peak viscosity, trough, and breakdown showed the highest value in both oats with 100 mesh. Further, the values of final viscosity and the setback were higher with relatively larger particle size. In addition, peak time exhibited a lower value with larger particle size, whereas it showed the opposite value for pasting temperature. Starch digestibility did not show any specific trend according to particle size, whereas expected glycemic index of hulled oats was lower than that of naked oats. Moreover, lower values were investigated at larger particle sizes. The results of this study provide basic useful information for processing of oat products to improve consumption of oats grown in Korea.

Food Components Characteristic of Oysters Produced in Korea (국내 산지별 굴의 성분 특성)

  • The detailed proximate, fatty/amino acid, mineral composition, texture, color, chemical and taste compounds of six oysters (four kinds of cultured oysters and two kinds of wild oysters) in Korea were investigated. Length and weight of the shell removed cultured and wild oysters were 4.7~5.1 and 3.0~4.2 cm, and 5.9~9.1 and 2.6~5.5 g, respectively. The proximate compositions were not significantly different between cultured and wild oysters. Amino nitrogen and volatile basic nitrogen content of these ones were 232.8~258.2 and 160.5~213.9 mg/100 g, 9.5~12.0 and 7.8~9.5 mg/100 g, respectively. As a texture characteristic of muscle, shearing force were 95~114 and 105~132 g. Amounts of total amino acids of cultured and wild oysters were 9,004~10,198 and 8,165~8,942 mg/100 g, respectively. Major amino acids and inorganic ions were aspartic acid, glutamic acid, proline, alanine, leucine, phenylalanine, lysine, arginine and K, Na, Ca, Fe, S, P, Zn. Major fatty acids of these ones were 16:0, 18:0, 16:1n-9, 18:1n-9, 22:1n-9, 16:4n-3, 20:5n-3 and 22:6n-3, and there was little difference between the two groups. Amounts of free amino acids of cultured and wild oysters extracts were 1,444~1,620 and 1,017~1,277 mg/100 g, respectively, and major ones were taurine, glutamic acid, glycine, alanine, tryptophan, ornithine and lysine. There is a little difference in glycine, tryptophan, ornithine and arginine contents, but TMAO and TMA contents were low in amount, and were not significantly different between the two groups.

  • For the purpose of obtaining basic data which can be applied to processing of retort pouched shellfishes, retort pouched seasoned ark shell, Anadara broughtonii, was prepared. The frozen ark shell was thawed and seasoned with a mixed seasoning powder prepared with $10.0\%$ of sorbitol, $2.0\%$ of table salt and $0.5\%$ of monosodium glutamate at $5^{\circ}C$ for 10 hours, and then dried at $45^{\circ}C$ for 4 hours. The dried seasoned ark shell was coated with $1.0\%$ sodium alginate solution, dried with cola air blast for 2 hours and then vacuum-packed in the laminated plastic film bag (polyester/casted polypropylene= $12{\mu}m/70{\mu}m,\;15{\times}16cm$), and finally sterilized up to Fo=6.0 in hot water circulating retort at $121^{\circ}C$ for 10 minutes. The major fatty acids of raw ark shell and retort pouched seasoned ark shell products were 16:0, 20:5, 22:6, 18:0 and 18:3, and predominant free amino acids of those were lysine, arginine, glycine, alanine, glutamic acid and leucine. In nucleotides and its related compounds of raw ark shell and retort pouched seasoned ark shell products, the most abundant one was AMP, and total extract-N of those was chiefly consisted of free amino acids, betaine and nucleotide and its related compounds. During the processing procedure such as drying and sterilization, unsaturated fatty acids slightly decreased while saturated fatty acids increased, and total extract-N content decreased about a half. From the results of chemical and microbial experiments during storage, it was concluded that the products could be preserved in a good condition for 100 days at room temperature, and their duality could be improved by the coating treatment of sodium alginate solution.

Quality Characteristics and Physiological Activities of Fermented Soybean by Lactic Acid Bacteria (유산발효대두의 품질특성 및 생리활성)

  • The quality characteristics and functional properties of fermented soybean by lactic acid bacteria (FSB) were investigated and compared with those of soybean (control) and commercial cheonggukjang Powder (CGP). The crude protein, lipid, and fiber contents of FSB were similar to those in CGP. The vitamin $B_2$ content in FSB (1.4 mg%) was similar to the control (1.3 mg%) whereas it was remarkably low in the CGP (0.2 mg%). The bright yellow color of FSB determined by Hunter's colormeter was quite different from the dark reddish brown color of the CGP. The pH of FSB was the lowest and thee amino-nitrogen was 517.2 mg%, which was higher than that in CGP (468.1 mg%). Glutamic acid, aspartic acid and leucine were the most abundant amino acids. In particular, the increase in the glutamic acid level was noticeable in FSB. The fatty acid compositions of FSB and CGP were similar to the control. However, the sucrose and fructose levels were lower after fermentation but the glucose level was higher. The results of isoflavone analysis by HPLC showed that the levels of daidzein and genistein in FSB were as much as 48.33 and 52.82 mg%, respectively, which is higher than that found in CGP. The DPPH free radical scavenging effects of FSB and CGP were 1.8 times higher an those of the control. The fibrinolytic activity determined by the diameter of the lysed area on the fibrin plate was the most effective in FSB. In conclusion, it is believed that FSB has a similar or higher quality than CGP. Therefore, FSB is expected to be good functional food material.

Quality Characteristics of Doenjang by Aging Period (전통 된장의 숙성 기간에 따른 감각·화학적 품질특성)

  • In order to characterize the quality of Doenjang, fermented Korean soybean paste, subjected to long-term aging, this study performed physico-chemical analyses and sensory evaluation according to aging period (from 1 to 9 years). Regarding the proximate composition of Doenjang according to aging period, moisture, crude protein, crude lipid, crude ash, and salt contents showed little differences among Doenjang samples. Amino-type nitrogen content was 1,046.7 mg% in the 1 year-aged sample, 990.9~996.9 mg% in the 2~5 year-aged samples, and 1,214.1~1,304.8 mg% in the samples fermented more than 5 years. ${\Delta}$E value, reflecting total color differences between the samples, increased according to aging period. Ratios of linoleic and linolenic acids, which are essential fatty acids in soybeans, constituted 55% of total fatty acids, which was the most abundant among all fatty acids. The major free sugar in Doenjang was fructose at a content of 1.6~2.2% in 1~9 year-aged Doenjang. Glycoside form of isoflavones in Meju constituted 77.1%, and the aglycon form constituted 22.9%. However, the glycoside type of isoflavones in soybeans was converted to aglycon type in Doenjang through fermentation and aging. In the sensory evaluation of Doenjang samples, brown color, salt smell, soy sauce flavor, and viscosity all increased according to aging period, whereas sweet flavor, roast smell, beany flavor, salty taste, and acrid taste showed no significant differences. In cluster analysis of the sensory attributes of Doenjang according to aging period, 1 year-aged Doenjang was significantly different between 2 year- and 3~5 year-aged Doenjang.

Changes in Nutraceutical Lipid Constituents of Pre- and Post-Geminated Brown Rice Oil (발아 전후 현미유에서의 기능성 지질성분 변화)

  • This study investigated the changes in the nutraceutical lipid components of brown rice oil after germination. Four different high-yielding cultivars (Dasan1, Segyejinmi, Hanareum1 and Hanareum2) of Korean brown rice were selected and brown rice oil was extracted from each cultivar before and after germination. Free fatty acid, squalene, policosanols, and isomers of phytosterol were analyzed using GC, and isomers of tocols (tocopherol and tocotrienol) and ${\gamma}$-oryzanol were quantified using HPLC from both brown rice oil (BRO) and germinated brown rice oil (GBRO). The contents of phytosterol isomers, campesterol, stigmasterol, and ${\beta}$-sitosterol were increased by 8.3%, 31.6%, 3.3% in GBRO, respectively. Furthermore, the squalene content showed the highest increase of up to 2.4 fold in GBRO compared to BRO. In addition, linoleic and linolenic acid composition increased whereas oleic and palmitic acid decreased in the GRBO. However, the contents of tocols (tocopherol and tocotrienol) in GBRO were lower than those in BRO, and there was no significant difference in policosanol and ${\gamma}$-oryzanol between GBRO and BRO. These results suggest that GBRO has the potential as a healthy and functional source due to its lipid profile on improved lipid metabolism.

Characteristics of Squid Viscera Oil (오징어 내장의 지방질조성)

  • The oil content and composition of squid visera were determined to obtain data for utilization of this by-product. There was no significant difference in the glycolipid (GL) and phospholipid (PL) content in Illex argentinus and Todarodes pacificus, but neutral lipid (NL) was different (p<0.05). The viscera oil of I. argentinus contained $30.50\%$ total lipid which consisted of $96.24\%$ NL, $2.63\%$ GL, $2.37\%$ PL, and contained $644mg\%$ cholesterol. The viscera oil of T. pacificus contained $30.20\%$ total lipid which consisted of $94.82\%$ NL, $2.85\%$ GL, $2.34\%$ PL, and contained $1,224\;mg\%$ cholesterol. The NL, GL and PL of viscera oil in I. argentinus mainly consist of triglyceride $(44.01\%)$, esterified steryl glycosides $(58.95\%)$ and phosphatidyl cholines $(32.36\%)$, respertively. Those of viscera oil in T. pacificus mainly consist of triglyceride $(39.63\%)$, monogalactosyl diglycerides $(51.67\%)$ and phosphatidyl cholines $(31.98\%)$, respectively. The major fatty acids of the viscera oil of I. argentinus and T. pacificus were C16 : 0, $C16\;:\;0,\;C18\;:\;1\omega9,\;C20\;:\;4\omega6,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$. In Illex argentinus, the fatty acids of NL mainly were $C16\;:\;0,\;C18\;:\;1\omega9,\;C20\;:\;4\omega6,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$. PL were $C16\;:\;1\omega7,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$ and GL were $C18\;:\;1\omega9,\;C20\;:\;5\omega3,\;22\;:\;6\omega3$. The major fatty acids of NL in T. pacificus were $C16\;:\;0,\;C18\;:\;1\omega9,\;C20\;:\;4\omega6,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$, PL were $C16\;:\;1\omega7,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$, and GL were $C18\;:\;1\omega9,\;C20\;:\;5\omega3,\;C22\;:\;6\omega3$.

Changes of Seed Germination Rate and Lipid Components in Different Brown Rices during Ageing (저장에 따른 생태형별 현미의 발아율과 지질 성분의 변화)

  • The objective of this study was to determine quality changes in seed germination rates and lipid components in Japonica and Tongil (indica/japonica) type brown rices with different tocol composition during storage. Moisture content, seed germination rate, crude lipid content, fat acidity, tocol content, free fatty acid composition of brown rice and pH of milled rice were measured to evaluate their quality after storage. Seed germination rates of stored Japonica and Tongil type brown rices were decreased by 22.4% and 59.7%, respectively, after 8 weeks of storage. The fat acidity of Japonica rice was significantly (p<0.05) higher than that of Tongil type brown rices after storage. The major tocol homologue of Tongil type was ${\gamma}-tocotrienol$, whereas, major tocol homologues of Japonica brown rices were ${\alpha}-tocopherol$ and ${\alpha}-tocotrienol$ throughout the entire storage period.

Changes of Chemical, Bacteriological, and Allergenicity of Raw Milk by Gamma Irradiation (감마선 조사 처리에 의한 우유의 화학적${\cdot}$세균학적 및 항원성 변화)

  • Effects of heat and gamma irradiation on chemical, microbiological, and immunological changes of raw milk were compared. Free fatty acid content of milk showed increasing tendency according to the increase of heating temperature and irradiation dose, and showed similarity in UHT (ultra high temperature) and 5 kGy irradiation. Total bacterial counts and coliforms were not detected after treatment of LTLT (low temperature long time), HTST (high temperature short time), UHT, and irradiation from 1 to 10 kGy in the milk with initial microbial load at $10^3$ CFU/mL initially, but after 7 day storage, were not detected in UHT milk and that irradiated at 3 kGy or above. Heat treatment decreased (p<0.05) arginine, asparate, iso-leucine, lysine, and methionine content compared to raw milk while irradiation decreased (p<0.05) asparate, histidine, iso-luecine, leucine, and lysine content, which means irradiation could change primary structure of milk proteins. It was concluded that f kGy gamma irradiation treatment of raw milk could give a similar effect to UHT treatment in chemical and microbiological viewpoint, and may reduce allergenicity of raw milk.
