Effect of Ginseng Components on Content of Cholesterol and Activity of Acyl CoA.Cholesterol Acyltransferase in Hep G2 Cells Cultured in Cholesterol Rich Medium (고콜레스테를 조건으로 배양한 Hep G2세포의 콜레스테를 함량변동과 Acyl CoA : Cholesterol Acyltransferase의 활성에 미치는 인삼성분의 영향)

  • A human hepatoma cell line, hep G2, was used to investigate the mechanism of serum cholesterol reduction by ginseng total saponin, ginsenoside-$Rb_1$, - $Rb_2$, and non-saponin fraction (ether extraction). Hep G2 cells were incubated in 10 $\mu\textrm{g}$/ml of cholesterol containing serum free-RPMl1640 medium with various concentration of ginseng components. The amounts of cholesterol in Hep G2 cells were decreased to maximum 51% in total saponin or two ginsenoside-treated groups while there was 137% increase in cholesterol level of control group as compared with that of normal group. Nonsaponin groups did not show the same effect. In order to elucidate the observed changes in the amount of cholesterol, the activity of amyl CoA : cholesterol acyltransferase (ACAT) in groups showing remarkable reduction in cholesterol amount, i.e., total saponin 10-6%, ginsenoside-$Rb_1$ $10^{-4}$%, ginsenoside-$Rb_2$, $10^{-4}$%, and non-saponin fraction $10^{-4}$%, was assayed using [1-$^{-14}C$%]oleic acid as enzyme substrate. The activity of ACAT was increased in all groups tested as compared with that of control group except for non-saponin group cultured in water soluble cholesterol containing medium. The serum cholesterol lowering effects of ginseng components can partially be attributed to the increased hepatocellular ACAT activity.

The Effects of Vitamin E and C on Serum Cholesterol and Antioxidative enzyme in ovariectomized rat (난소 절제 흰쥐의 혈청 콜레스테롤 및 항산화효소에 미치는 비타민 E와 비타민 C의 효과)

  • In order to observe the bioactivity of ovariectomized rats, ovariectomized group (Ovx), nonovariectomized group (Sham), ovariectomized Vitamin C-treat group (Ovx+Vit C), ovariectomized Vitamin E-treat group (Ovx+Vit E) and ovariectomized Vitamin C+Vitamin E-treat group(Ovx+Vit C+E) were made. Lipidperoxides of liver and kidney, serum total cholesterol and HDL-cholesterol were investigate as follows. Lipidperoxides of liver and kidney in Ovx group were 1.78 times and 1.61 times increased compared to Sham group respectively. But, they were significantly decreased in Ovx+Vit C group, Ovx+Vit E group, Ovx+Vit C+E group compared to Ovx group. Serum total cholesterol in Ovx group was increased 2.57 times compared to Sham group. Injections of each substance such as ascorbate, tocopherol, mixture (C+E) make data of Cholesterol become low. When especially Vit C is injected, the data of cholesterol lowed by about 94%. Serum HDL-cholesterol in Ovx group decreased 36.7% compared to Sham group. And as the result of the measurement of SOD, Catalase, and GPx which are antioxidant enzyme, SOD and Catalase activities in Ovx group much higher than in Sham group. Based on the results, it is supposed that more produced free radicals increased antioxidant enzyme. And it is also thought that vitamin can inhibit aging by reducing antioxidant enzyme.

Effect of Dandelion Leaf Extracts on Lipid Metabolism in Rats Fed High Cholesterol Diet (민들레잎추출물의 흰쥐 체내 지질대사 개선 효과)

  • The effect of dandelion leaf extracts on lipid metabolism was investigated in high cholesterol-fed rats. Four groups of rats were given a high cholesterol diet containing 10g cholesterol/kg and 2.5 g sodium cholate/kg for 6 weeks. The control group received without dandelion leaf extracts and the other three groups received with one of dandelion leaf extracts (water, ethyl acetate, ether), respectively. Weight gain and feed intake were significantly higher in water group than control group. Serum triglyceride, total cholesterol, cholesteryl ester, chylomicron and VLDL-cholesterol concentratins were lower, whereas HDL-cholesterol and phospholipid concentrations were significantly higher in dandelion leaf extract supplemented-groups than control group. Dandelion extract increased HDL-C/total-C ratio and lowered atherogenic index. Hepatic triglyceride, total cholesterol, free cholesterol and cholesteryl ester concentrations lowered in dandelion leaf extract groups compared to control group. Especially, hepatic triglyceride and free cholesterol levels were lower in ethyl acetate extract group than in water and ether extract groups. Dandelion leaf extract supplements did not affect hepatic phospholipid cencentration. Hepatic HMG-CoA reductase activity was significantly lower in water extract group than the others. ACAT activity was significantly decreased by dandelion extract supplements.

Korean Curcuma longa L. induces lipolysis and regulates leptin in adipocyte cells and rats

  • BACKGROUND/OBJECTIVES: Turmeric (Curcuma longa L.) has been reported to have many biological functions including anti-obesity. Leptin, peptide hormone produced by adipocytes and its concentration is increased in proportion to the amount of the adipocytes. In the present study, we examined the effects of Korean turmeric on the regulation of adiposity and leptin levels in 3T3-L1 adipocytes and rats fed a high-fat and high-cholesterol diet. MATERIALS/METHODS: Leptin secretion, free fatty acid and glycerol contents in 3T3-L1 adipocytes were measured after incubation of cells with turmeric for 24 hours. Rats were divided into four experimental groups: a normal diet group (N), a high-fat and high-cholesterol diet group (HF), a high-fat and high-cholesterol diet group supplemented with 2.5% turmeric extracts (TPA group) and a high-fat and high-cholesterol diet group supplemented with 5% turmeric extracts (TPB group). Serum samples were used for the measurement of leptin concentration. RESULTS: Contents of free fatty acid and glycerol showed concentration dependent increase in response to turmeric extracts. Effects of turmeric extracts on reduction of lipid accumulation in 3T3-L1 cells were examined by Oil Red O staining. Treatment with turmeric extracts resulted in increased expression levels of adipose triglyceride lipase and hormone-sensitive lipase mRNA. The concentration of leptin from 3T3-L1 adipocytes was significantly decreased by turmeric. Proportional abdominal and epididymal fats weights of the turmeric 5% supplemented group, TPB has significantly decreased compared to the HF group. The serum levels of leptin in the TPA and TPB groups were significantly lower than those of the HF group. CONCLUSIONS: Based on these results, we suggested that Korean turmeric may contribute to the decreasing of body fat and regulating leptin secretion.

A 8-Week, Randomized, Double-Blind, Placebo-Controlled Human Trial to Evaluate the Efficacy and Safety of Punica granatum L.·Actinidia chinensis Planch. Mixed Extract on Body Fat (석류·참다래 혼합추출물의 체지방 감소에 대한 유효성 및 안전성을 평가하기 위한 8주, 무작위 배정, 이중눈가림, 위약-대조 인체적용시험)

  • Objectives: This study was conducted to evaluate efficacy and safety of Punica granatum L. Actinidia chinensis Planch. mixed extract on body fat. Methods: Thirty women were recruited ($25kg/m^2{\leq}body$ mass index [BMI]${\leq}29.9kg/m^2$) ($19yr{\leq}age{\leq}38yr$) and randomized to receive Punica granatum L. Actinidia chinensis Planch. mixed extract or placebo for 8 weeks. During the test, they visited four times including screening. Body fat mass, body fat rate, anthropometric dimensions at waist and hip, waist-hip ratio, total cholesterol, triglyceride, low density lipoprotein-cholesterol (LDL-C), and high density lipoprotein-cholesterol (HDL-C) were measured at screening and 8 weeks after screening. Adverse reaction were also checked each visit. Results: There were no significant differences in all index. But when divide experimental group by BMI index, age, and compliance, less than BMI $27kg/m^2$ group decreased body weight, body fat mass and more than BMI $27kg/m^2$ group also decreased body weight, body fat mass and waist circumference. Less than age 30 years group showed decline of waist circumference, fat free rate and T-cholesterol and more than age 30 years group showed decline of fat free mass. Group with compliance more than 95% was decreased BMI, fat free mass, and hip circumference. Conclusions: There was no significant efficacy as compared with placebo group, but as divide experimental group according to BMI index, age, compliance, mixed extract might be effective to reduce obesity index.

Properties of Cholesterol-reduced Butter and Effect of Gamma Linolenic Acid Added Butter on Blood Cholesterol

  • The present study was carried out to develop cholesterol-reduced and gamma linolenic acid (GLA)-added butter and to examine the changes in chemical and sensory properties, and cholesterol lowering effect of GLA addition. The cholesterol removal rate reached 93.2% by $\beta$-cyclodextrin in butter before GLA addition. The thiobarbituric acid value of cholesterol-reduced and GLA-added butter increased slowly up to 4 week and plauteaued thereafter. TBA value was significantly increased with 2% GLA addition, compared with no GLA addition. The production of short-chain free fatty acids (FFA) increased with storage in all treatments. From 4 weeks storage, the amount of short-chain FFA in 2% GLA-added group was significantly higher than those in other groups. Among sensory characteristics, color, greasiness and overall acceptability were mostly affected by GLA addition, however, the rancidity value of 2% GLA addition was significantly different from those of control and GLA-unadded and cholesterol-reduced butter at 0, 6 and 8 week storage. Among groups, no difference was found in texture in all storage periods. The smallest increase of total blood cholesterol in rats was found in the group fed 2% GLA-added and cholesterol-reduced butter for 8 week, compared with that in controls. The present results showed the possibility of cholesterol-reduced and GLA-added butter development without much difference in chemical, rheological and sensory properties, and indicated a slow increase effect on blood total cholesterol in rats.

Effects of the Feeding Hijikia fusiforme(Harvey) Okamura on Lipid Composition of Serum in Dietary Hyperlipidemic Rats (톳(Hijikia fusiforme(Harvey) Okamura)이 식이성 고지혈증 흰쥐의 혈청 지질성분에 미치는 영향)

  • This study was designed to observe the effects of the feeding of Hijikia fusiforme(Harvey) Okamura juice extract on the improvement of lipids in the serum of cholesterol-supplemented diet induced dietary hyperlipidemic rats fed experimental diets for 4 weeks. Concentration of total cholesterol and LDL-choloesterol in serum was significantly lower in the Hijikia fusiforme juice extract group than in the cholestero diet group. Concentration of HDL-cholesterol in serum was higher in the Hijikia fusiforme juice extract group than the cholesterol supplemented diet group. Those in the Hijikia fusiforme juice extract group were higher than those in the cholesterol diet group. In the ration of HDL-cholesterol concentration to total cholesterol concentration, Hijikia fusiforme juice extract administration group was higher percentage than the other groups. Atherosclerotic index was lower in the Hijikia fusiforme juice extract group than in the cholesterol diet group. Concentration of free cholesterol and cholesteryl ester in serum was rather lower in the Hijikia fusiforme juice extract group than the other groups. Triglyceride concentration in serum was significantly lower in the Hijikia fusiforme juice extract group than in the cholesterol diet group. Concentration of phospholipid in serum was more decreased in the Hijikia fusiforme juice extract group than in the cholesterol diet group. The activities of aspartate aminotransferase and alanine aminotransferase in serum were rather lower in the Hijikia fusiforme juice extract group than the cholesterol supplemented diet group. From the avove research, the Hijikia fusiforme juice extracts were effective on the improvement of the lipid compositions in serum of high fat diet induced dietary hyperlipidemic rats.

Effects of Callophyllis japonica Extracts on the Glucide and Lipid Metabolism in Hypercholesterolemic Rats (고콜레스테롤혈증 흰쥐의 당질 및 지질 대사에 볏 붉은잎(Callophyllis japonica)추출액이 미치는 영향)

  • This study was designed to observe the effects of the feeding Callophyllis japonica extracts on the improvement of the blood glucose, lipid in the serum of dietary hypercholesterolemic rats(S.D. strain, ?) fed the experimental diets for 4 weeks. Concentrations of total cholesterol and LDL-cholesterol in serum were significantly lower in the Callophylis japonica extracts group than in the choloesterol supplemented diet group. Concentrations of HDL-cholesterol in serum were higher in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. Concentrations of free cholesterol, triglyceride, phospholipid and blood glucose in serum were significantly lower in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. The activities of aspatate aminotransferae(AST), alanine aminotransferase(ALT), lactate dehydrogenase(LDH) and alkaline phosphatase(ALP) in serum were lower in the CAllophyllis japonica extracts supplemented group than in the cholesterol diet group. From the above research, the physiological activity substances in Callophylis japonica extracts were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hypercholesterolemic rats.

Effect of Soybean Powder on Lipid Metabolism and Enzyme Activities in Induced Hyperlipidemic Rats (대두분 급여가 고지혈증 흰쥐의 지질대사 및 효소 활성도에 미치는 영향)

  • To investigate the effects of yellow soybean powder on serum lipid level and enzyme activities, male Sprague-Dawley(SD) rats were given hyperlipidemic diets for 6 weeks. Rats(n=8) were divided into three groups consisting the normal group(N), hyperlipidemic diets group(C) and hyperlipidemic diets and yellow soybean powder group(YSP). YSP group lowered triglyceride, total lipid, phospholpid, ester-cholesterol and free-cholesterl than control group. YSP group lowered total-cholesterol, VLDL-cholesterol and LDL-cholesterol but increased HDL-cholesterol of the serum compared to the control group. Athergenic Index(AI) and LHR(LDL-cholesterol/HDL-cholesterol ratio) of YSP group was decreased by control group (p<0.05). On the other hand, glutamic oxaloacetate transaminase(GOT), glutamic pyruvate transaminase (GTP), lactate dehydrogenase(LDHase) and alkaline phosphatase(ALP) enzymes activities were decreased by hyperlipidemic diets group (C). These results suggest that yellow soybean powder fed a lipid lowering effect in serum of rats.

Properties and Cholesterol Lowering Effect of Cholesterol-reduced Milk Supplemented with Evening Primrose Oil

  • This study was carried out to investigate the changes of chemical and sensory properties and cholesterol lowering effect of milk treated with $\beta$-cyclodextrin to reduce cholesterol and supplemented with evening primrose oil (EPO). The average cholesterol removal rate was 93.5%. The thiobarbituric acid (TBA) absorbance increased proportionally to storage period and amount of EPO addition. TBA absorbance was significantly different in the EPO-added groups from that in unadded groups in all periods. The production of short-chain free fatty acids (FFA) increased with longer period of storage. From 6 days of storage, the amounts of short-chain FFA in 6 and 10% EPO-added groups were significantly different from other groups. The scores for all sensory characteristics indicated that sensory quality decreased with both storage time and increasing amount of EPO. Oxidative off-flavor and off-taste were more intense with higher amounts of EPO addition and longer storage period. Also, the higher the amount of EPO addition, the lower overall scores throughout the 15 day storage. Feeding 10% EPO-supplemented cholesterol-reduced milk increased high density lipoprotein (HDL) in male Sprague-Dawley rats by 76%, which is significantly different from the control (27%).
