Effect of Admix Ratio of Free-machining Agent 'KSX' that Contains Complex Calcium Oxide Powder on Machinability

  • Free-machining agent 'KSX' contains complex calcium oxide is developed. The effect of admix ratio of KSX on mechanical properties and machinability with two different cutting speeds is reported. KSX displays improved machinability without deterioration of mechanical properties up to 0.3mass% addition. It was observed that KSX is effective with a small addition of 0.1mass% at slower cutting speed, and increased of admix ratio is effective at faster cutting speed.

Formulation design of chloride-free cement additive by response surface methodology

  • The influences of chloride-free components of the cement additive: triethanolamine, triisopropanolamine, sodium hyposulfite and calcium gluconate on the 1d, 3d and 28d compressive strength of cement were investigated by response surface methodology. It found the early strength activators, triethanolamine and sodium hyposulfite could enhance the 1d strength of cement effectively but they did not contribute to the 3d strength enhancement, and further their interaction was able to decrease the 28d strength of cement. Calcium gluconate was not that effective for the strength enhancement on 3 and 28 days when it's simply dosed. However the interaction effect of calcium gluconate with triisopropanolamine could strongly favor the strength enhancement of cement after 3 days. Results indicated it was necessary to focus attention on the potential interactions among the chemical components. And for the concern of four chemicals studied in this paper, it was feasible to formulated a kind of chloride-free cement additive that can be effective for the early strength of cement and its the strength after 3 days.

Effect of Supplementing Sheep with Sunflower Acid Oil or its Calcium Soap on Nutrient Utilization

  • Four adult rams ($22.25{\pm}0.90kg$) were used in a $4{\times}4$ latin square design to evaluate the rations without ($T_1$) or with supplementation of sunflower acid oil at 5 ($T_2$), 10 ($T_3$) or calcium soap at 10% of dietary DM ($T_4$) on nutrient digestibility and balances of nitrogen, calcium and phosphorus. The basal ration contained 60 parts Brazilian napier grass hay and 40 parts concentrate mixture. The DM, CF, NDF and ADF digestibilities and nitrogen retention (g/d) decreased (p<0.01) by inclusion of sunflower acid oil at 5% of dietary DM. In addition, depression (p<0.01) in digestibilities of CP, nitrogen free extract (NFE), cellulose, hemicellulose, retention of calcium and phosphorus (g/d) were also observed with increasing the level of sunflower acid oil to 10% of dietary DM. The EE digestibility, total digestible nutrients (TDN) content and calcium retention (g/d) were significantly higher (p<0.01) for ration supplemented with calcium soap. It is concluded that sunflower acid oil supplementation in free form as low as 5% of dietary DM is deleterious to fibre digestion in sheep while as calcium soap, it can be fed up to 10% of dietary DM as an energy source without any adverse effect.

A Study on the Mechanism of Calcium Binding Inhibition of Cardiac Sarcoplasmic Reticulum by Oxygen Free Radicals (산소대사물에 의한 심장근 Sarcoplasmic reticulum의 칼슘운반 억제 기전에 관한연구)

  • Mechanism of calcium transport inhibition of cardiac sarcoplasmic reticulum (SR) by oxygen free radicals was examined. Effects of oxygen free radicals generated by xanthine/xanthine oxidase (X/XO) system on isolated porcine ventricle SR were studied with respect to its calcium binding, lipid peroxidation, SH-group content and alteration of membrane protein components. The results are as follows. 1) Calcium binding of isolated SR was markedly inhibited by X/XO. 2) During the incubation of sarcoplasmic reticulum with xanthine/xanthine oxidase, there were marked inclose in lipid peroxidation and reduction of SH-group content. 3) An antioxidant, p-phenylenediamine effectively prevented the lipid peroxidation but partially prevented the calcium binding inhibition of X/XO treated SR. 4) The reduction of SH-group content of SR treated with X/XO was partially prevented by p-phenylendiamine. 5) When modifying SH-group of SR by treatment with DTNB, the inhibition of calcium binding activity was partially prevented. 6) On gel-permeation chromatography of X/XO-treated sarcoplasmic reticulum, there was an increase of small molecular weight products, probably protein degradation products. 7) Semicarbazide, which prevents the cross-linking reaction of protein components, did not affect the calcium binding inhibition of X/XO-treated SR. From these results, it is suggested that the inhibition of calcium binding of SR by oxygen free radicals results from the consequence of multiple changes of SR components, which are lipid peroxidation, SH-group oxidation and degradation of protein components.

Effect of dietary Calcium Level on Cadmium and Lead Toxicity in Rats (식이내 Calcium 수준이 흰쥐의 카드뮴과 납중독에 미치는 영향)

  • This study was performed to investigate the effect of dietary calcium level on cadmium and lead toxicity in rats. Fifty-four male rats of Sprague-Dawely strain weighing 152$\pm$12g were blocked into 9 groups according to body weight, and were raised for 30 days. Nine experimental diets different with cadmium(0%, 0.04%), lead (0%, 0.071%) and calcijm(0.5%, 1.0%, 1.5%) levels were prepared. The results are summarized as follow. Weight gain, F.E.R.(food efficiency ratio), and weights of liver, kidney and femur were lower in cadmium exposed groups than those of heavy metal free groups. Weight gain F.E.R. and ash weight of lead groups were lower than those of heavy metal free groups. But, these were increased with increasing dietary calcium level. Cadmium and lead concentrations in blood, liver, kidney and femur were lower in rats fed 1.5% calcium than 0.5% calcium diet. Fecal cadmium and lead excretions were remarkably increased in 1.5% calcium groups, and cadmium and lead retention rates were decreased in 1.5% calcium groups. Metallothionein concentrations in liver, kidney and small intestine were higher in rats exposed to cadmium and lead. Calcium content in blood, femur and daily urinary and fecal calcium excretion were decreased by cadmium and lead additions, and increased in 1.5% calcium groups. Creatinine clearance were decreased with cadmium administratino and calcium addition. In conclusion, weight gain and organ weights were decreased with cadmium or lead administration. But, cadmium administration was more toxic than lead adminstration. Cadmium or lead toxicity was alleviated by increasing dietary calcium level. Especially, lead toxicity was alleviated in proportion to dietary calcium level.

  • The effects of lactose on the formation of casein phosphopeptide (CPP), the increment of soluble calcium and the enhancement of calcium absorption by dietary casein in the intestinal tract were investigated. Rats were fed a lactose-free diet, 10% lactose diet and 30% lactose diet containing powdered milk. In rats receiving a lactose-free powdered milk diet, CPP formation was confirmed by gel filtration of the intestinal content on Sephadex G -25 and the amount of soluble calcium was increased in the small intestine and calcium absorption, measured by the ligated ideal loops in situ was enhanced. However, In rats receiving a powdered milk diet containing 10% lactose or 30% lactose, the similar effects were not seen. These observations indicate that CPP-stimulated effects on soluble calcium and calcium absorption in the small intestine are not dependent upon lactose.

  • Carbon tetrachloride (CCI$_4$) induces the hepatotoxicity due to the reactive free radicals generated by cytochrome P-450 (CYP-450) enzyme, which result in destabilization of cellular membrane. Diltiazem, a calcium channel blocking agent, has been known to suppress the CYP-450 enzyme activities. To study the effect of diltiazem in $CCl_4$-treated rats, we measured the activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and glutathione S-transferase (GST), contents of bilirubin, albumin, total protein, cholesterol, triglyceride, blood urea nitrogen, creatinine malondialdehyde and calcium. Also we conducted liver histopathologic examinations. Diltiazem, when administered 1 hour prior to CCI$_4$ treaeent, significantly reduced the activities of ALT and AST, the contents of microsomal malondialdehyde and calcium in liver and microsome as compared with those of $CCl_4$-treated rats. In addition, histopathologic examination showed that diltiazem prevented the development of centrilobular necrosis induced by CCI$_4$ in liver tissue. Our results suggested that diltiazem could inhibit the formation of free radicals and the influx of calcium. Therefore diltiazem may be applied to suppress the liver damage caused by $CCl_4$.

Regulation of Blastocyst Differentiation by the Serial Exposure of Conconavalin A and $PGE_2$ (Concanavalin A와 $PGE_2$의 순차적 노출에 의한 포배의 분화 조절)

  • Differentiation of blastocyst is critical step for implantation and is under the control of regulation factors originated from embryo or reproductive tracts. The sequential communication with those factors is suspected as critical events for differentiation. It has been suggested that intracellular signaling pathways activated by calcium is essential in differentiation of blastocyst. Previously, it was known that concanavalin A (Con A) increase the levels of free calcium in blastocyst stage. However, Con A can not accelerate the hatching, although heparin-binding epidermal growth factor-like growth factor (HB-EGF), a modulator of calcium level, accelerate the hatching of blastocyst. In this study, it was investigated whether Con A or prostaglandin $E_2$ ($PGE_2$) can modulate the differentiation of blastocyst. Con A accelerated the expansion of blastocyst in both 1 hr pulse treatment group and continuous treatment group. However, Con A significantly suppressed the hatching in both groups. The inhibition was significantly strong in continuous treatment group compared with 1 hr pulse treatment group. On the other hand, $PGE_2$ induced the increase the free calcium level, but did not accelerate the expansion. In addition $10{\mu}m\;PGE_2$ inhibited hatching. However, $PGE_2$ could accelerate hatching in Con A pretreated blastocyst. $PGE_2$ also caused the increase of free calcium level in Con A pretreated blastocyst. From these results, it is suggested that changes of the free calcium level induce a different calcium-mediated signaling pathways. In addition, sequential stimulation by signal molecules may triggers the cellular mechanisms for the differentiation of blastocyst.

The Role of Na-K Pump in the Modulation of Vascular Tone in the Rabbit (혈관 긴장도 조절에 미치는 Na-K Pump에 관한 연구)

  • Force development of smooth muscle cells is directly regulated by the concentration of free calcium ions in the sarcoplasm, and the sarcoplasmic concentration of calcium ion can be modulated by electrogenic Na-K pump. The role of Na-K pump on vascular tone was studied in isolated rabbit renal artery. Helical strips of arterial muscle were prepared from left renal arteries. All experiments were performed in $HCO_3^--buffered$ Tyrode solution which was aerated with $3%CO_2-97%\;O_2$ mixed gas and kept at $35^{\circ}C$. In some experiments, rabbit was injected intraperitoneally $18{\sim}24$ hours prior to the experiments, with a large dose(5 mg/kg body wt) of reserpine, in order to eliminate the catecholamines present in intrinsic adrenergic nerve terminate. Treatment used in this experiment that inhibits Na-K pump was the exposure of strips to K-free Tyrode solution. Contractile response to K free Tyrode solution developed slowly and the time required for maximum contracture was $20{\sim}30$ minutes. This K-free contracture was rapidly relaxed by the addition of potassium to the bathing solution. No K-free contracture occurred in a Ca-free Tyrode solution. But contraction developed rapidly when calcium ion was added to the bathing solution after 30 minute exposure of the strip to Ca-free Tyrode solution. This contracture was completely inhibited by Ca-antagonist, verapamil. The K-free contracture was abolished by ${\alpha}-adrenergic$ blocker, phentolamine, as well as by the catecholamine depletion from adrenergic nerve terminals. Even in reserpinized strip, the exogenous norepinephrine-induced contraction in K-free Tyrode solution was rapidly suppressed by the addition of potassium ion. The results of this experiment suggest that K free contracture develops by norepinephrine release from adrenergic nerve terminals, while the relaxation of K-free contracture is induced by the activation of electrogenic Na-K pump.

  • This study designed to elucidate free amino acid composition and calcium, iron contents in extractive cooking. broth of boiled-dried anchovy. Composition of the free amino acid in boiled-dried anchovy, in large and medium one appeared the same tendency. Abundant free amino acid of boiled-dried anchovy were histidine, alanine, lysine, leucine, glycine and glutamic acid in order. The total free amino acid was greatly extracted from cooking broth when boiled at 30 minutes. Free amino acid, such as histidine, alanine, lysine, leucine and glutamic acid had abundant in cooking broth. The calcium and iron contents in broth were little difference between large and medium one. Calcium and iron contents were highest in cooking broth when boiled at 30 minutes. Panel test on general accetability was very good in the boiling at 30 minutes.
