Effect of Freezing on the Physicochemical Properties of Semi-dried Red Pepper (냉동조건에 따른 반건조 홍고추의 물리.화학적 특성 변화)

  • Quality changes in semi-dried red pepper (SRP) treated with ozone water were observed upon storage (at $-18^{\circ}C$) after freezing at $-10^{\circ}C$, $-20^{\circ}C$, and $-40^{\circ}C$. Drip loss after treatment was greater than in control peppers, but no significant difference was evident between treatments (p<0.05). We observed that differences between samples decreased as storage time increased. Texture after treatment did not change significantly over a 3-month period. The redness (a-value) after treatment was greater than in the control, but no sample showed significant color alteration after the 3-month period. The capsaicinoid content decreasedas storage time increased, and was also affected by the freezing temperature. However, carotenoid content was not influenced by freezing or storage temperature. Ascorbic acid and free sugar contents showed decreases of 47% and 6.5%, respectively, after semi-drying. The results of sensory evaluation indicated no significant difference between samples in terms of color appearance.

  • In order to utilize sea mustard, Undaria pinnatifida, effectively. sea mustard jam was prepared by use of physical properties of polysaccharides, mainly alginic acid and then its chemical composition, nutritive qualifies and the stability of pigments were examined. Suitable processing condition for sea mustard jam was as fellows : as the first stage, fresh sea mustard was scalded for 20 sec at $85^{\circ}C$ and dried sea mustard was dipped for 20 min in cold water for rehydration, then both sea mustards were treated by draining and chopping. Next, after adding 4 fold of $0.5\%\;K_2HPO_4$ solution to weight of chopped sea mustards, the mixed solutions were agitated for 15 min at $95^{\circ}C$, and gelated sea mustards were filterated by pressing. Flow characteristics of those sea mustard jam were regarded mixed type having pseudoplastic type and yield stress. Judging from sensory evaluation, adding $0.375\%$ of saccharin, $2\%$ of sorbitol, $0.25\%$ of citric acid and $0.5\%$ of powder of roasted soybean to sea mustard jam were suitable for enhancing taste and flavor of product. Chemical composition of products were scarcely changed throughtout processing. Jam processed by fresh sea mustard was better than dried one in contents of chlorophyll and total carotenoid pigments. In fatty acid composition, polyenoic acids of $C_{18:3},\;C_{20:4},\;C_{20:5},\;C_{22:6}$ were held high contents as known to have lowering function of cholesterol contents. It was presumed that dominant contents in free amino acids such as lysine, alanine, glutamic acid and organic acids such as citric acid, oxalic acid, $\alpha-ketoglutaric$ acid, lactic acid and succinic acid held important role for the flavor of sea mustard jam.

Skin-Friction Drag Reduction in Wake Region by Suction Control on Horseshoe Vortex in front of Hemisphere (반구 전방에 생성된 말굽와류 흡입제어에 의한 후류영역 마찰저항 감소에 관한 연구)

  • The aim of this study was to investigate the possibility of the skin-friction reduction by vortex control. A vortical system such as a horseshoe vortex, a hairpin vortex, and a wake region was induced around a hemisphere attached on a Perspex flat plate in the circulating water channel. Hairpin vortices were developed from the wake region and horseshoe vortices were formed by an adverse pressure gradient in front of the hemisphere. The horseshoe vortices located on the flank of the hemisphere induced a high momentum flow in the wake region by the direction of their vorticity. This process increased the frequency of the hairpin vortices as well as the frictional drag on the surface of the wake region. To reduce the skin-friction drag, suction control in front of the hemisphere was applied through a hole. Flow visualization was performed to optimize the free-stream velocity, size of the hemisphere, and size of the suction hole. Once the wall suction control mitigated the strength of the horseshoe vortex, the energy supplied to the wake region was reduced, causing the frequency of the hairpin vortex generation to decrease by 36.4 %. In addition, the change in the skin-friction drag, which was measured with a dynamometer connected to a plate in the wake region, also decreased by 2.3 %.

Effects of Roasting Conditions on Components of Safflower(Carthamus tinctorius L.) Seed (홍화(紅花) 종실(種實)의 볶음 조건에 따른 성분(成分)의 변화(變化))

  • The main objective of this study was to characterize physico-chemical properties, sensory property and oxidative stability of safflower seed obtained by various roasting temperature and time. The contents of water soluble solids decreased in the higher roasting temperature and time. Sensory evaluation of safflower seed roasted in various conditions showed significant differences in taste, color, flavor and palatability. The safflower seed roasted at $190^{\circ}C$ for 20min had the best palatability. At the change of Hunter's values, L values were decreased, and a, b and ${\Delta}E$ values were increased in the higher roasting temperature and time. The content of free sugars such as sucrose and raffinose were reduced significantly in higher roasting time of $190^{\circ}C$ and $210^{\circ}C$. During the storage period after roasting treatment, peroxide values (POV) were highly increased after eight months at the all treatment except for $150^{\circ}C$. Therefore, it is inadequate over eight months after roasting treatment.

The Effect of Porous Support and Intermediate Layer on the Silica-zirconia Membranes for Gas Permeation Performance (실리카-지르코니아 분리막 성능에 대한 다공성 지지체와 중간층의 영향)

  • In this study, porous metal (O.D. = 10 mm, length = 10 mm, 316 L SUS, Mott Corp.) and ${\alpha}$-alumina tube (O.D. = 10 mm, length = 50 mm, Pall, German) support was modified with suspension sols, which were consisted of $3{\sim}4{\mu}m$ and 150 nm size of ${\alpha}$-alumina particle in the water or silica-zirconia colloidal sol. The porous support was fabricated by dip coating method for 5 seconds with suspension of alumina particles. After drying at $100^{\circ}C$ for 1 h, it was calcined at $550^{\circ}C$ for 30 min. It was repeated several times in order to decrease big pore on support. The surface roughness and largest pore size on the porous support was decreased by increasing coating times with $3{\sim}4{\mu}m$ size of ${\alpha}$-alumina particle and alumina coating with 150 nm size of ${\alpha}$-alumina particle served as further smoothening the surface and decreasing the pore size of the substrate. And the silica-zirconia membranes were successfully prepared on the modified porous metal and ${\alpha}$-alumina supports, and showed hydrogen permeance in the range of $1.8-8.4{\times}10^{-4}mol{\cdot}m^{-2}{\cdot}s^{-1}{\cdot}Pa^{-1}$ and $3.3-5.0{\times}10^{-5}mol{\cdot}m^{-2}{\cdot}s^{-1}{\cdot}Pa^{-1}$, respectively.

Upregulation of Mir-34a in AGS Gastric Cancer Cells by a PLGA-PEG-PLGA Chrysin Nano Formulation

  • Background: Nano-therapy has the potential to revolutionize cancer therapy. Chrysin, a natural flavonoid, was recently recognized as having important biological roles in chemical defenses and nitrogen fixation, with anti-inflammatory and anti-oxidant effects but the poor water solubility of flavonoids limitstheir bioavailability and biomedical applications. Objective: Chrysin loaded PLGA-PEG-PLGA was assessed for improvement of solubility, drug tolerance and adverse effects and accumulation in a gastric cancer cell line (AGS). Materials and Methods: Chrysin loaded PLGA-PEG copolymers were prepared using the double emulsion method (W/O/W). The morphology and size distributions of the prepared PLGA-PEG nanospheres were investigated by 1H NMR, FT-IR and SEM. The in vitro cytotoxicity of pure and nano-chrysin was tested by MTT assay and miR-34a was measured by real-time PCR. Results: 1H NMR, FT-IR and SEM confirmed the PLGA-PEG structure and chrysin loaded on nanoparticles. The MTT results for different concentrations of chrysin at different times for the treatment of AGS cell line showed IC50 values of 68.2, 56.2 and $42.3{\mu}M$ and 58.2, 44.2, $36.8{\mu}M$ after 24, 48, and 72 hours of treatment, respectively for chrysin itslef and chrysin-loaded nanoparticles. The results of real time PCR showed that expression of miR-34a was upregulated to a greater extent via nano chrysin rather than free chrysin. Conclusions: Our study demonstrates chrysin loaded PLGA-PEG promises a natural and efficient system for anticancer drug delivery to fight gastric cancer.

  • Anaerobic soils limit the amount of free oxygen available in the rhizosphere and therefore will impede grass root development and restrain nutrient availability for turf growth. An in-situ study was conducted on existing greens to investigate the relationship between $CO_2$ content in the rhizosphere and turf quality. Nine greens were selected in the study. On each green, five 1-m diameter circular plots were randomly selected for conducting the experiment. The greens were sampled 7 times from August, 1998 to August, 1999. Data collected from each plot included turf quality index, $CO_2$ content, and physical properties of the rooting mixtures. Turf quality declined drastically when $CO_2$ content in rhizosphere increased to $5\;to\;6{\mu}LL^{-1}$ during the late summer season. The $CO_2$ content increased as water content in the root zone increased, but was inversely related to infiltration rate. Cultivation of a golf green may reduce $CO_2$ content in the rhizosphere, but the benefit of cultivation decreased with time.

Sericin Enhances Secretion of Thyroglobulin in the Thyrocytes (갑상선세포에서 sericin에 의한 thyroglobulin의 분비증가)

  • Sericin is a type of high molecular weight water-soluble glycoprotein surrounding fibroin (silk protein) that has been used as a cell culture supplement and accelerates cell proliferation in various serum-free media. The purpose of this study was to investigate the enhancing effect of thyroglobulin (Tg) secretion by sericin in thyrocytes, FRTL-5 cells. While Tg-mRNA expression was not enhanced, a secreted form of Tg was obviously increased by sericin. In this status, expression of both endoplasmic reticulum (ER) molecular chaperones (Bip & calreticulin) and ER membrane proteins (IRE1, PERK & ATF6) was enhanced. The proximal step of IRE1, XBP1 mRNA splicing was slightly detected however, the proximal step of PERK, phosphorylation of $eIF2{\alpha}$, was changeless. In addition, sericin enhanced cell viability by the MTT assay. The above results showing the ability of sericin to promote protein production demonstrated its potential usefulness as a new biomaterial.

ESR-based Identification of Radiation-Induced Free Radicals in Gamma-Irradiated Basil and Clove Using Different Sample Pre-Treatments (감마선 조사된 바질과 정향의 전처리방법에 따른 ESR Spectra 판별 특성)

  • An improved detection of radiation-induced paramagnetic faults was developed to identify the irradiation status of basil and clove. The effectiveness of different sample pretreatments, including freeze-drying (FD), oven-drying (OD), alcoholic-extraction (AE), and water-washing and alcoholic-extraction (WAE), were examined. All non-irradiated samples showed a single central signal ($g_0$=2.006), whereas radicals representing two additional side peaks ($g_1$=2.023 and $g_2$=1.986) with a mutual distance of 6 mT were detected in the irradiated samples. AE and WAE produced the best results for irradiated clove in terms of intensities of radiation-specific ESR signals and their ratios to the central signal. However, FD provided the highest intensities of radiation-specific ESR signals for basil, whereas their ratios to the major signal were better in the cases of AE and WAE. Signal noise, particularly due to $Mn^{2+}$ signals, was observed, whereas it decreased in AE and WAE pretreatments. Based on our results, AE and WAE can improve the detection conditions for radiation-specific ESR signals in irradiated samples.

Antioxidative Effects of Campanula takesimana Nakai Extract (섬초롱(Campanula takesimana Nakai) 용매 추출물의 항산화 효과)

  • The study analyzed the total polyphenolic contents, DPPH radical scavenging activities, ABTS radical scavenging activities, reducing power levels, SOD-like activities, and tyrosinase inhibition activities of fractions from Campanula takesimana Nakai extract. Total polyphenolic content of the ethyl acetate fraction from Campanula takesimana Nakai extract was the highest. The ethyl acetate fraction of Campanula takesimana Nakai extract showed the lowest $IC_{50}$ value (0.13 mg/mL) as well as greater than 98% ABTS radical scavenging activity, which was similar to ascorbic acid. Further, reducing power was significantly higher in the ethyl acetate fraction. The SOD-like activity of edible plant was 70.56% (n-butanol fraction) and 62.27% (ethyl acetate fraction) at 1 mg/mL. Tyrosinase inhibition activity of Campanula takesimana Nakai extract was reduced to 34.77% in the n-butanol fraction and 30.85% in the ethyl acetate fraction at 5 mg/mL. These results suggest that the ethyl acetate and n-butanol fractions of Campanula takesimana Nakai extract could be applicable as food additives or cosmeceutical ingredients.
