흰쥐 해마 절편에서 저산소증에 의한 [$^3H$-5-Hydroxytrytamine의 유리 변동에 미치는 superoxide dismutase/catalase의 영향 (Effect of Superoxide Dismutase on the Release of [$^3H$]-5-Hydroxytrytamine after Hypoxia from Rat Hippocampal Slices)

  • Many factors are known to be responsible for cerebral ischemic injury, such as excitatory neurotransmitters, increased intraneuronal calcium, or disturbance of cellular energy metabolism. Recently, oxygen free radicals, formed during ischemia/reperfusion, have been proposed as one of the main causes of ischemia/reperfusion injury. Therefore, to investigate the role of oxygen free radical during ischemia/reperfusion, in the present study the effect of endogenous oxygen free radical scavenger, superoxide dismutase / catalase(SOD / catalase) on the release of [$^3$H]-5-hydroxytryptamine([$^3$H]-5-HT) during hypoxia/reoxygenation in rat hippocampal slices was measured. The hippocampus was obtained from the rat brain and sliced 400 gm thickness with manual chopper. After 30 min's preincubation in the normal buffer, the slices were incubated for 20 min in a buffer containing [$^3$H]-5-HT(0.1 $\mu$M, 74 $\mu$Ci) for uptake, and washed. To measure the release of [$^3$H]-5-HT into the buffer, the incubation medium was drained off and refilled every ten minutes through a sequence of 14 tubes. Induction of hypoxia for 20 min (gassing it with 95% N$_2$/5% CO$_2$) was done in the 6th and 7th tube, and oxygen free radical scavenger, SOD / catalase was added 10 minutes prior to induction of hypoxia. The radioactivity in each buffer and the tissue were counted using liquid scintillation counter and the results were expressed as a percentage of the total activity. When slices were exposed to hypoxia for 20 min, [$^3$H]-5-HT release was markedly decreased and a rebound release of [$^3$H]-5-HT was observed on the post-hypoxic reoxygenation period. SOD / catalase did not changed the release of [$^3$H]-5-HT in control group, but inhibited the decrease of [$^3$H]-5-HT release in hypoxic period and rebound increase of [$^3$H]-5-HT in reoxygenation period. This result suggest that superoxide anion may play a role in the hypoxic-, and reoxygenation-induced change of [$^3$H]-5-HT release in rat hippocampal slices.

  • Hwang Chil (Dendropanax morbifera Lev.) belongs to Aralia elata group. This plant is only found in Korea and is considered to be along with ginseng. The physiological activities of it have been reported as having scavenging effect on oxygen free radicals and anti-carcinogenic effect. As most of the anti-oxidants were effective on protecting hepatotoxicity from alcohol, hard tissues restoring and skin whitening, the effectiveness of Hwang Chil extracts was screened. (omitted)

  • The surgically ovariectomized rat induces aging by reactive oxyyen species(ROS) generation. Free oxygen radicals have been proposed as important causative agents of aging. The purpose of this study was to investigate the effect of Chondroitin Sulfate(CS) to prevent ovariectomy(OVX) induced oxidative stress and atherosclerosis.   • Reactive oxygen species(ROS) or free radicals are produced in the pathogenesis of human diseases including atherosclerosis, diabetes, cancer, and aging. Antioxidants are associated with the prevention of ROS-induced tissue and cellular damage in the various diseases. This study investigated the antioxidative activities of the methanol extract of Alisma plantagoaquatica var. orientale and its main component under conditions of radical generation using allophycocyanin and ferric-thiocyanate assay. Alisol B 23-acetate as a main component was isolated from the methanol extract of Alisma plantago-aquatica var. orientale. In results, the extract of Alisma plantago-aquatica var. orientale showed inhibitory activity on AAPH [2,2'-azobis(2-amidinopropane)dihydrochloride]-induced protein oxidation. Also, the extract of Alisma plantago-aquatica var. orientale and alisol B 23-acetate inhibited lipid peroxidation. These results indicate that Alisma plantago-aquatica var. orientale and alisol B 23-acetate show promise as therapeutic agents for various damages involving free radical reactions.

  • Antioxidants either scavenge superoxide and free radicals or stimulate the detoxification mechanisms within cells, resulting in increased detoxification of free radicals formation. Hyperin, isolated from the stem of Uncaria rhynchophylla, prevented oxygen radical formation and inhibited lipid oxidation. The effective concentrations were 31.3 $\mu$M for a radical scavenging assay and 2.2 $\mu$M for a microsome assay. cDNA microarray analysis to determine which genes were modulated by hyperin found that 50 genes were upregulated and 37 genes were downregulated in SNU-668 human gastric cancer cells. Among these genes, thirteen genes that were significantly affected by hyperin were verified by RT-PCR for their effect of genetic reprogramming.

  • Recently, low energy ion beam irradiation has been adopted for surface modification. Low energy ion beam irradiation has many advantages in polymer engineering such as weak damage, good adhesion, noticeably-enhanced wettability(less than 15 degree), good reproducibility and so on. In this experiment, chemical reactions between free radicals and environment gas species have been investigated using angle-resolved XPS and TRIM code. In the case of low ion beam energy (around 1 keV), energy loss in polymer is mainly originated from atomic collisions instead of electronic interference. Atomic collisions could generated displaced atoms and free radicals. Cold cathode-ion source equipped with 5cm convex grid was used in an O2 environment. Base and working pressure were 5$\times$10-6 and 2.3$\times$10-4 Torr. Flow rates of argon and oxygen were fixed at 1.2 and 8 sccm. target materials are polyethylene polyvinyidenefluoride and polytetrafluoroethylene.

Antioxidants as alleviating agents of in-vitro embryo production oxidative stress

  • Despite numerous advances in in-vitro embryo production (IVP), many documented factors have been shown to influence the development of mammalian preimplantation embryos and the success of IVP. In this sense, elevated levels of reactive oxygen species (ROS) correlate with poor outcomes in assisted reproductive technologies (ART) due to oxidative stress (OS), which results from an imbalance between ROS production and neutralization. Indeed, excessive production of ROS compromises the structural and functional integrity of gametes and embryos both in vivo and in vitro. In particular, OS damages proteins, lipids, and DNA and accelerates cell apoptosis. Several in-vivo and in-vitro studies report an improvement in qualityrelevant parameters after the use of various antioxidants. In this review, we focus on OS and the source of free radicals and their effects on oocytes, sperm, and the embryo during IVP. In addition, antioxidants and their important role in IVP, supplementation during oocyte in vitro maturation (IVM), in vitro culture (IVC), and semen extenders were discussed. Nevertheless, various methods for determining the level of ROS in germ cells have been briefly described. Still, it is crucial to develop standardized antioxidant supplement systems to improve overall IVP success. Further studies should explore the safety, efficacy, mechanism of action, and combination of different antioxidants to improve IVP outcomes.
