DNA subfragments, sopA, sopB, and sopC supporting stable maintenance of an oriC plasmid, were derived from mini-F plasmid DNA (EcoRI restriction fragment, f5) after digestion with restriction endonucleases, and cloned in vector plasmid pBR322. The recombinant plasmid obtained were introduced into E. coli KY7231 and E. coli CSR603, and proteins specified by the mini-F fragments were analysed by SDS-polyacrylamide gel electrophoresis. Two proteins encoded by the F fragments were detected, having molecular weights of 41,000 and 37.000. The sopA protein (41K) encoded by a plasmid pXX288 was observed in the cytoplasm, whereas the sopB protein (37K) encoded by a plasmid pXX157 was in the membrane fraction. There was no novel protein band detected in the cell with a plasmid pXX300, which contained sopC fragment. Gene products of a plasmid pXX167, which is comprised of sopA, sopB, and sopC, were not detectable. Fluorography after one and two dimensional gel electrophoresis of the lysates showed that these two proteins were overproduced in the cells which were allowed to incorporate radioactive amino acid after plasmid amplification by chloramphenicol treatment. The isoelectric points of the sopA and sepB proteins were 6.6 and 7.0, respectively.
The purpose of this study is to develop instructional methods for the formalized algorithm through informal knowledge in teaching division of fractions. The following results have been drawn from this study: First, before students learn formal knowledge about division of fractions, they knowledge or strategies to solve problems such as direct modeling strategies, languages to reason mathematically, and using operational expressions. Second, students could solve problems using informal knowledge which is based on partitioning. But they could not solve problems as the numbers involved in problems became complex. In the beginning, they could not reinvent invert-and-multiply rule only by concrete models. However, with the researcher's guidance, they can understand the meaning of a reciprocal number by using concrete models. Moreover, they had an ability to apply the pattern of solving problems when dividend is 1 into division problems of fractions when dividend is fraction. Third, instructional activities were developed by using the results of the teaching experiment performed in the second research step. They consist of student's worksheets and teachers' guides. In conclusion, formalizing students' informal knowledge can make students understand formal knowledge meaningfully and it has a potential that promote mathematical thinking. The teaching-learning activities developed in this study can be an example to help teachers formalize students' informal knowledge.
It has been widely reported that carbide-free bainitic steels or super-bainite TRIP (SB-TRIP) steels for the automotive industry are a new family of steels offering a unique combination of high strength and ductility. Hence, it is important to exactly evaluate the volume fraction of RA and to identify the 3-D morphology of constituent phases, because it plays a crucial role in mechanical properties. Recently, as electron back-scattered diffraction (EBSD) equipped with focused ion beam (FIB) has been developed, 3-D EBSD technique for materials science are used to these steels. Moreover, newly developed atom probe tomography (APT) technique can provide the exact distribution and chemical concentration of alloying elements in a sub-nm scale. The APT analysis results indicate exactly the distribution and composition of alloying elements in the austenite and bainite phases of SB-TRIP steels with the atomic-scale resolution. And thus, no partitioning of aluminum and manganese atoms was showed between the austenite containing $7.73{\pm}0.39$ at% C and the bainitic ferrite associated with $0.22{\pm}0.06$ at% C in SB-TRIP steel.

The agricultural soil, meets soil environmental standards whereas agricultural product from the same soil does not meet permissible level of contaminants, is identified in the vicinity of the abandoned mine in Korea. This study estimated the stabilization efficiency of Cd and Pb using limestone through the flood pot test for this kind of agricultural paddy soil. We had the concentration of the monitored contaminants in soil solution for 4 months and analyzed fractionations in soil and concentrations in rice plant. In soil solution of plow layer, the reductive Mn had been detected constantly unlike Fe. The concentrations of Mn in limestone amended soil was relatively lower than that in control soil. This reveals that the reductive heavy metals which become soluble under flooded condition can be stabilized by alkali amendment. This also means that Cd and Pb associated with Mn oxides can be precipitated through soil stabilization. Pb concentrations in soil solution of amended conditions were lower than that of control whereas Cd was not detected among all conditions including control. In contaminants fractionation of soil analysis, the decreasing exchangeable fraction and the increasing carbonates fraction were identified in amended soil when compared to control soil at the end of test. These results represent the reduction of contaminants mobility induced by alkali amendment. The Cd and Pb contents of rice grain from amended soil also lower than that of control. These result seems to be influenced by reduction of contaminants mobility represented in the results of soil solution and soil fractionation. Therefore contaminants mobility (phytoavailability) rather than total concentration in soil can be important factor for contaminants transition from soil to agricultural products. Because reduction of heavy metal transition to plant depends on reduction of bioavailability such as soluble fraction in soil.
From the total methanolic extract of Gardenia jasminoides (Rubiaceae), various antioxidative characteristics were identified in terms of nitrite scavenging ability, 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radical cation inhibition, superoxide dismutase (SOD)-like activity, and elongation effect of lipid peroxidation using Rancimat. After successive partitioning with n-hexane, chloroform, n-butanol, and water, potent nitrite scavenging abilities were shown in the n-butanol fraction and water fraction, and $IC_{50}$ values were 183 ppm and 194 ppm, respectively. As for ABTS radical cation inhibition, the chloroform fraction was most potent and its $IC_{50}$ was 159 ppm. SOD-like activity was slightly low in all of the fractions. The elongation effect of lipid peroxidation also increased dose-dependently and the antioxidative index (AI) of the total methanolic extract was 2.93 in 1000 ppm, which was more effective than 1.66 of butylated hydroxy anisol in the same concentration. The compounds I and II were isolated through silica gel column chromatography of the active fractions, and identified as geniposide and crocin, respectively, by $^1H-NMR$ spectral data. The $IC_{50}$ values for the nitrite scavenging abilities of geniposide and crocin were 940 ppm and 77 ppm, respectively. In ABTS radical cation inhibition, the $IC_{50}$ values of geniposide and crocin were 684 ppm and 549 ppm, respectively. And the $EC_{50}$ value for SOD-like activity of crocin was 259 ppm, which was much smaller than 453 ppm by the positive control, chlorogenic acid. The $EC_{50}$ value of geniposide could not be identified.
This research was performed to evaluate the extraction characteristics of heavy metals for soil washing of mine tailings-contaminated soil according to particle size distribution and the chemical distributional existence of the metals. As the soil particle size was decreased, the extracted concentrations of heavy metals was increased except Fe and Mn. Most of all heavy metals were extracted within 6 h by soil washing with 0.05 M EDTA. Extraction efficiency of metals was decreased for Pb, Cu, and Zn with decreasing of particle size. Significant difference was not observed in extraction efficiency for Cd according to particle size distribution. Extraction efficiency for Cd was the highest as 86~91%, while the lowest as 5~14% for Fe. Most metals of the soil without soil washing was distributed as reducible, oxidizable, and residual fractions. Pb, Zn, and Cd existed as reducible (Fe/Mn oxide) and residual fractions and Cu existed as oxidizable and residual fractions after soil washing treatment with 0.05 M EDTA. As the soil particle size was decreased, residual fraction was increased for Pb and Cu. About 90% of reducible fraction in Pb, Zn, and Cd was removed by soil washing with 0.05 M EDTA. As the results, it was founded that soil particle size was the important parameter to effect on distributional fraction and extraction efficiency of metals in mine tailings-contaminated soil.

The extraction characteristics of heavy metals(HM) from a contaminated soil at existing lead smelters were investigated with ethylene diamine tetraacetic acid(EDTA), citrate and HCl as washing solutions. EDTA was more effective for Pb than for other heavy metals. As the mol ratio of EDTA/HM increased, the removal efficiency of heavy metals became higher. When the mol ratio of EDTA/HM approached to 6.5, it removed Pb most effectively. Citrate was effective especially in extracting Zn. The removal efficiency of HCl was comparatively high in almost all heavy metals, and at 0.3N concentration it was the highest. After soil washing process by the use of EDTA, the great part of exchangeable fractions and most of heavy metals of weakly adsorbed like carbonate fraction were extracted. For washing with citrate and HCl, four heavy metals showed the similar exchange of chemical partitioning and the exchangeable fractions of Pb which has weakly adsorbed to soil were more increased than before the process. As removal efficiency of citrate washing process depends upon the distribution of non-detrital fractions, so it can be contended that only the amount of non-detrital fractions could be removed from all the heavy metal content. EDTA and HCl could remove most of non-residual fractions in all heavy metals except Zn. As a result of EDTA washing, toxicity characteristic leaching procedure(TCLP) concentration of the processed soil met the USEPA Pb limit of 5.0 mg/L.

The environmental behaviors of polycyclic aromatic hydrocarbons (PAHs) are mainly governed by their solubility and partitioning properties on soil media in a subsurface system. In surfactant-enhanced remediation (SER) systems, surfactant plays a critical role in remediation. In this study, sorptive behaviors and partitioning of naphthalene in soils in the presence of surfactants were investigated. Silica and kaolin with low organic carbon contents and a natural soil with relatively higher organic carbon content were used as model sorbents. A nonionic surfactant, Triton X-100, was used to enhance dissolution of naphthalene. Sorption kinetics of naphthalene onto silica, kaolin and natural soil were investigated and analyzed using several kinetic models. The two compartment first-order kinetic model (TCFOKM) was fitted better than the other models. From the results of TCFOKM, the fast sorption coefficient of naphthalene ($k_1$) was in the order of silica > kaolin > natural soil, whereas the slow sorbing fraction ($k_2$) was in the reverse order. Sorption isotherms of naphthalene were linear with organic carbon content ($f_{oc}$) in soils, while those of Triton X-100 were nonlinear and correlated with CEC and BET surface area. Sorption of Triton X-100 was higher than that of naphthalene in all soils. The effectiveness of a SER system depends on the distribution coefficient ($K_D$) of naphthalene between mobile and immobile phases. In surfactant-sorbed soils, naphthalene was adsorbed onto the soil surface and also partitioned onto the sorbed surfactant. The partition coefficient ($K_D$) of naphthalene increased with surfactant concentration. However, the $K_D$ decreased as the surfactant concentration increased above CMC in all soils. This indicates that naphthalene was partitioned competitively onto both sorbed surfactants (immobile phase) and micelles (mobile phase). For the mineral soils such as silica and kaolin, naphthalene removal by mobile phase would be better than that by immobile phase because the distribution of naphthalene onto the micelles ($K_{mic}$) increased with the nonionic surfactant concentration (Triton X-100). For the natural soil with relatively higher organic carbon content, however, the naphthalene removal by immobile phase would be better than that by mobile phase, because a high amount of Triton X-100 could be sorbed onto the natural soil and the sorbed surfactant also could sorb the relatively higher amount of naphthalene.

Objective: The objective of this study was to describe a methodological procedure to quantify the heat production (HP) partitioning in basal metabolism or fasting heat production (FHP), heat production due to physical activity (HPA), and the thermic effect of feeding (TEF) in roosters. Methods: Eighteen 54-wk-old Hy Line Brown roosters (2.916±0.15 kg) were allocated in an open-circuit chamber of respirometry for O2 consumption (VO2), CO2 production (VCO2), and physical activity (PA) measurements, under environmental comfort conditions, following the protocol: adaptation (3 d), ad libitum feeding (1 d), and fasting conditions (1 d). The Brouwer equation was used to calculate the HP from VO2 and VCO2. The plateau-FHP (parameter L) was estimated through the broken line model: HP = U×(R-t)×I+L; I = 1 if t<R or I = 0 if t>R; Where the broken-point (R) was assigned as the time (t) that defined the difference between a short and long fasting period, I is conditional, and U is the decreasing rate after the feed was withdrawn. The HP components description was characterized by three events: ad libitum feeding and short and long fasting periods. Linear regression was adjusted between physical activity (PA) and HP to determine the HPA and to estimate the standardized FHP (st-FHP) as the intercept of PA = 0. Results: The time when plateau-FHP was reached at 11.7 h after withdrawal feed, with a mean value of 386 kJ/kg0.75/d, differing in 32 kJ from st-FHP (354 kJ/kg0.75/d). The slope of HP per unit of PA was 4.52 kJ/mV. The total HP in roosters partitioned into the st-FHP, termal effect of feeding (TEF), and HPA was 56.6%, 25.7%, and 17.7%, respectively. Conclusion: The FHP represents the largest fraction of energy expenditure in roosters, followed by the TEF. Furthermore, the PA increased the variation of HP measurements.

A validation study was conducted to evaluate the efficacy and mechanism of the cryo-precipitation, monoclonal anti-FVIIIc antibody (mAb) chromatography, Q-Sepharose chromatography, and lyophilization steps involved in the manufacture of high purity factor VIII (GreenMono) from human plasma, in the removal and/or inactivation of hepatitis A virus (HAV). Samples from the relevant stages of the production process were spiked with HAV and subjected to scale-down processes mimicking the manufacture of the high purity factor VIII concentrate. Samples were collected at each step and immediately titrated using a 50% tissue culture infectious dose (TCID$\_$50/) and then the virus reduction factors were evaluated. HAV was effectively partitioned from factor VⅢ during cryo-precipitation with the log reduction factor of 3.2. The mAb chromatography was the most effective step far removal of HAV with the log reduction factor of $\geq$4.3. HAV infectivity was not detected in the fraction of factor VⅢ, while most of HAV infectivity was recovered in the fractions of flow through and wash during mAb chromatography. Q-Sepharose chromatography showed the lowest efficacy for partitioning HAV with the log reduction factor of 0.7. Lyophilization was an effective step in inactivating HAV with the log reduction factor of 2.3. The cumulative lag reduction factor, $\geq$10.5, achieved for tile entire manufacturing process was several magnitudes greater than the potential HAV load of current plasma pools.
