Urushiol V Suppresses Cell Proliferation and Enhances Antitumor Activity of 5-FU in Human Colon Cancer Cells by Downregulating FoxM1

  • Colorectal cancer (CRC) is one of the most common malignant tumor. 5-FU is commonly used for the treatment of CRC. However, the development of drug resistance in tumor chemotherapy can seriously reduce therapeutic efficacy of 5-FU. Recent data show that FoxM1 is associated with 5-FU resistance in CRC. FoxM1 plays a critical role in the carcinogenesis and drug resistance of several malignancies. It has been reported that urushiol V isolated from the cortex of Rhus verniciflua Stokes is cytotoxic to several types of cancer cells. However, the underlying molecular mechanisms for its antitumor activity and its potential to attenuate the chemotherapeutic resistance in CRC cells remain unknown. Here, we found that urushiol V could inhibit the cell proliferation and induced S-phase arrest of SW480 colon cancer cells. It inhibited protein expression level of FoxM1 through activation of AMPK. We also investigated the combined effect of urushiol V and 5-FU. The combination treatment reduced FoxM1 expression and consequently reduced cell growth and colony formation in 5-FU resistant colon cancer cells (SW480/5-FUR). Taken together, these result suggest that urushiol V from Rhus verniciflua Stokes can suppress cell proliferation by inhibiting FoxM1 and enhance the antitumor capacity of 5-FU. Therefore, urushiol V may be a potential bioactive compound for CRC therapy.

전자파에 노출된 생쥐에서 운동량에 따른 뇌의 유전자 변화 (The Gene Expression Level Differences associated with Exercise in the Mouse Brain exposed to Radiofrequency Radiation)

  • 전자파 노출이 자발운동에 따른 뇌의 유전자 발현에 미치는 영향을 10 주간 4그룹 즉, 정상 그룹, 자발운동 그룹, 전자파 노출 그룹, 전자파 노출 및 자발운동 그룹으로 나누어 조사하였다. 선조체(striata)와 시상하부(hypothalamus)에서 RT-PCR을 수행하였으며, 타이로신수산화효소(TH), FoxO3a, AMPKα, mRNA 발현을 조사하였다. 선조체에서 TH mRNA 발현은 자발운동과 전자파 노출 조건에서 각각 감소하였고, 전자파 노출 및 자발운동 그룹에서 더 많이 감소되었다. 이 결과는 전자파 노출 및 자발운동 그룹에서의 운동량 감소가 선조체에서 도파민이 감소할 수 있음을 시사한다. 선조체에서 FoxO3a mRNA 발현은 자발운동 그룹에서 증가했지만, 전자파 노출 및 자발운동 그룹은 현저히 감소했다. 시상하부에서는 TH mRNA 유전자 발현은 전자파 노출을 받은 자발운동 그룹에서 감소가 유의했으며, FoxO3a mRNA는 발현의 현저한 증가가 있었다. 전자파가 기억력에 미치는 영향도 밝히기 위해 해마에서의 여러 단백질들의 발현을 추후 조사할 것이다.

Inhibition of melanogenesis by sodium 2-mercaptoethanesulfonate

  • Sodium 2-mercaptoethanesulfonate (mesna) is a protective agent that is widely used in medicine because of its antioxidant effects. Recently, reactive oxygen species (ROS) were shown to increase pigmentation. Thus, ROS scavengers and inhibitors of ROS production may suppress melanogenesis. Forkhead box-O3a (FoxO3a) is an antimelanogenic factor that mediates ROS-induced skin pigmentation. In this study, we aimed to investigate the whitening effect of mesna and the signaling mechanism mediating this effect. Human melanoma (MNT-1) cells were used in this study. mRNA and protein expression were measured by real-time quantitative PCR and Western blotting analysis to track changes in FoxO3a-related signals induced by mesna. An immunofluorescence assay was performed to determine the nuclear translocation of FoxO3a. When MNT-1 melanoma cells were treated with mesna, melanin production and secretion decreased. These effects were accompanied by increases in FoxO3a activation and nuclear translocation, resulting in downregulation of four master genes of melanogenesis: MITF, TYR, TRP1, and TRP2. We found that mesna, an antioxidant and radical scavenger, suppresses melanin production and may therefore be a useful agent for the clinical treatment of hyperpigmentation disorders.

GDP갭 분해기법을 이용한 변동요인 분석 (An Analysis of Factors Affecting the Variation of GDP Gap by a Decomposition Method)

  • GDP갭(gap)이란 잠재GDP와 실제GDP의 격차로서 산출갭(Output gap)이라고도 한다. 잠재GDP는 노동, 자본 등 생산요소를 완전히 활용하여 달성할 수 있는 최대GDP라고 정의할 수 있다. GDP갭은 수요-공급간 불균형을 의미한다고 할 수 있으며 이러한 특성 때문에 물가 및 고용안정을 추구하는 중앙은행들은 정책수행과정에서 GDP갭을 중요한 정보변수로 활용하고 있다. 본 연구에서는 우리나라의 GDP갭을 그간 선행연구에서 사용하였던 필터링 방법에 의해 분해함으로써 GDP갭 변동을 야기하는 주요 요인이 무엇인지 살펴보았다. Fox 등 (2003)와 Fox와 Zurlinden (2006)에서 사용되었던 Super Smoother 알고리즘을 이용하여 우리나라의 명목GDP갭을 분해해 본 결과 이론적인 설명에 부합하는 것으로 나타났다. 명목GDP갭률 변동의 상당부분은 잔차인 총요소생산성으로 설명됨을 알 수 있었는데, 이는 최근들어 급격한 기술변화 및 환경변화 등 생산성 변화가 GDP변동에 큰 영향을 주고 있음을 의미한다. 다른 나라의 경우와 마찬가지로 국내물가의 영향력도 높은 것으로 나타났지만 대외 의존도가 높은 우리 경제의 특성상 교역조건의 변동 역시 상대적으로 명목GDP 움직임에 큰 영향을 주고 있는 것으로 분석되었다.

HARD CHINE형 활주 고속선의 저항특성에 관한 고찰 (A Study on the Resistance Property of Hard Chine Type High Speed Planing Craft)

  • The resistance property of a high speed passenger craft (: "DOL-PIN HO" designed by the author in 1972) is investigated as follows; -. The Resistance property of the craft is determined by savitsky′s method and blount-Fox′s method. The theoretical results are also compared with the full scale data. The comparison reveals that the result when using blount/fox′s method are in much closer agreement with the full scale data than savitsky′s. -. The effects of ship speed on the positions of the center of pressure and of the longitudinal center of gravity (L.C.G.) are investigated. The investigation shows that the position of L.C.G. of the craft is almost constant although the ship speed is changed. -. The effect of transom flap on the Resistance property of the craft is studied using savitsky/brown′s method. From the study it is found that the resistance of the craft is decreased and hence speed gain (about 3% of the service speed) can be obtained, when using transom flap for the craft.

FOX hunting system을 이용한 배추 기능유전자 탐색 (Systematic approaches to identify functional genes using the FOX-hunting system in Chinese cabbage)

  • Full-length cDNAs are essential for the correct annotation of genomic sequences and for the functional analysis of genes and their products. To elucidate the functions of a large population of Chinese cabbage (Brassica rapa) genes and to search efficiently for agriculturally useful genes, we have been taking advantage of the full-length cDNA Over-eXpresser (FOX) gene hunting system. With oligo dT column it purify the each mRNA from the flower organs, leaf and stem tissue. And about 120,000 cDNAs from the library were transformed into $\lambda$-pFLCIII-F vector. Of which 115,000 cDNAs from the library were transformed into T-DNA binary vector, pBigs for transformation study. We used normalized full-length cDNA and introduced each cDNA into Arabidopsis by in planta transformation. Full-length Chinese cabbage cDNAs were expressed independently under the CaMV 35S promoter in Arabidopsis. Selfed seeds were harvested from transgenic Arabidopsis. We had selected 2,500 transgenic plants by hygromycin antibiotic tolerant test, and obtained a number of transgenic mutants. Each transgenic Arabidopsis was investigated in morphological changes, fertility and leaf colour. As a result, 285 possible morphological mutants were identified. Introduced cDNA was isolated by PCR amplification of the genomic DNA from the transgenic mutants. Sequencing result and BLAST analysis showed that most of the introduced cDNA were complete cDNAs and functional genes. Also, we examined the effect of Bromelain on enhancing resistance to soft rot in transgenic Chinese cabbage 'Osome'. The bromelain gene identified from FOX hunting system was transformed into Chinese cabbage using Agrobacterium methods. Transformants were screened by PCR, then RT-PCR and real time PCR were performed to analyze gene expression of cysteine protease in the T1 and T2 generations. The anti-bacterial activity of bromelain was tested in Chinese cabbages infected with soft rot bacteria. The results showed that the over-expressed bromelain gene from pineapple conferred enhanced resistance to soft rot in Chinese cabbage.

Effect of Deer Antler Extract on Muscle Differentiation and 5-Aminoimidazole-4-Carboxamide Ribonucleoside (AICAR)-Induced Muscle Atrophy in C2C12 Cells

  • The effect of deer antler extract on muscle differentiation and muscle atrophy were evaluated to minimize muscle loss following aging. Various deer antler extracts (HWE, hot water extract of deer antler; FE, HWE of fermented deer antler; ET, enzyme-assisted extract of deer antler; UE, extract prepared by ultrasonication of deer antler) were evaluated for their effect on muscle differentiation and inhibition of 5-aminoimidazole-4-carboxamide ribonucleoside (AICAR)-induced muscle atrophy in C2C12 cells. Morphological changes according to the effect of antler extracts on muscle differentiation were confirmed by Jenner-Giemsa staining. In addition, the expression levels of genes related to muscle differentiation and atrophy were confirmed through qRT-PCR. In the presence of antler extracts, the length and thickness of myotubes and myogenin differentiation 1 (MyoD1) and myogenic factor 5 (Myf5) gene expression were increased compared to those in the control group (CON). Gene expression of AMP-activated protein kinase (AMPK), MyoD1, and myogenin, along with the muscle atrophy factors muscle RING finger-1 (MuRF-1) and forkhead box O3a (FoxO3a) upon addition of deer antler extracts to muscle-atrophied C2C12 cells was determined by qRT-PCR after treatment with AICAR. The expression of MuRF-1 and FoxO3a decreased in the groups treated with antler extracts compared to that in the group treated with AICAR alone. In addition, gene expression of MyoD1 and myogenin in the muscle atrophy cell model was significantly increased compared that into the CON. Therefore, our findings indicate that antler extract can increase the expression of MyoD1, Myf5 and myogenin, inhibit muscle atrophy, and promote muscle differentiation.

A Mixture of Morus alba and Angelica keiskei Leaf Extracts Improves Muscle Atrophy by Activating the PI3K/Akt/mTOR Signaling Pathway and Inhibiting FoxO3a In Vitro and In Vivo

  • Muscle atrophy, which is defined as a decrease in muscle mass and strength, is caused by an imbalance between the anabolism and catabolism of muscle proteins. Thus, modulating the homeostasis between muscle protein synthesis and degradation represents an efficient treatment approach for this condition. In the present study, the protective effects against muscle atrophy of ethanol extracts of Morus alba L. (MA) and Angelica keiskei Koidz. (AK) leaves and their mixtures (MIX) were evaluated in vitro and in vivo. Our results showed that MIX increased 5-aminoimidazole-4-carboxamide ribonucleotide-induced C2C12 myotube thinning, and enhanced soleus and gastrocnemius muscle thickness compared to each extract alone in dexamethasone-induced muscle atrophy Sprague Dawley rats. In addition, although MA and AK substantially improved grip strength and histological changes for dexamethasone-induced muscle atrophy in vivo, the efficacy was superior in the MIX-treated group. Moreover, MIX further increased the expression levels of myogenic factors (MyoD and myogenin) and decreased the expression levels of E3 ubiquitin ligases (atrogin-1 and muscle-specific RING finger protein-1) in vitro and in vivo compared to the MA- and AK-alone treatment groups. Furthermore, MIX increased the levels of phosphorylated phosphoinositide 3-kinase (PI3K), protein kinase B (Akt), and mammalian target of rapamycin (mTOR) that were reduced by dexamethasone, and downregulated the expression of forkhead box O3 (FoxO3a) induced by dexamethasone. These results suggest that MIX has a protective effect against muscle atrophy by enhancing muscle protein anabolism through the activation of the PI3K/Akt/mTOR signaling pathway and attenuating catabolism through the inhibition of FoxO3a.

항산화 및 암전이 관련 단백질의 발현에 미치는 콩잎낙엽 에탄올 추출물의 영향 (Effect of Soybean Fallen Leaves Ethanolic Extract on Expression of Proteins Related to Antioxidant Activity and Cell Invasion)

  • 콩잎은 골다공증 및 유방암 발생을 예방한다고 널리 보고되고 있다. 이를 바탕으로 콩잎낙엽 에탄올 추출물(SBFL)을 제조하여 암 전이와 관련 있는 세포침윤에 대한 효과를 조사하기 위하여 섬유아육종세포(HT1080)에서 SBFL이 항산화와 matrix metalloproteinases (MMPs)에 미치는 영향을 분석하였다. 본 연구에서 활성산소의 소거 효과에 대한 SBFL효과는 DPPH radical, 환원력 및 지질과산화실험으로 평가되었다, 본 연구에서 SBFL은 양성 대조군으로 사용된 vitamin C 및 vitamin E와 비교 시 우수한 항산화 효과를 보여주었다. 다음으로 SBFL의 세포 독성을 측정하기 위하여 MTT assay를 수행한 결과 16 µg/ml 이상의 농도에서 세포독성을 보여주었다. SBFL은 gelatin zymography 실험에서 phorbol 12-myristate 13-acetae (PMA) 혹은 phenazine methosulfate (PMS)로 자극된 암 전이에서 중요한 MMP-9의 활성을 감소시켰다. 특히 SBFL은 단백질 발현 실험에서 SOD-1, p-FoxO-1의 발현을 증가시켰다. 더욱이 vascular endothelial growth Factor (VEGF)로 자극된 세포 침윤이 SBFL처리에 의하여 억제되는 것으로 나타났다. 이러한 연구결과를 바탕으로 SBFL은 뛰어난 항산화 효과는 산화적 스트레스를 감소시키고 MMP-9의 활성과 세포침윤을 억제시켜 암 전이의 예방을 위한 유효성분으로 이용될 수 있다는 것을 암시하고 있다.

  • In balanced design of experiment sample size is determined to detect a certain standardized size of effect with power 1-$\beta$ at the level of significance $\alpha$. Tables (Marvin, et al (1970) and Lorenzen and Anderson(1993) and charts (Odeh and Fox(1991)) are available to determine the sample size in balanced design of experiments. To simplify those tables and charts simple MATLAB program is used to find the minimal standardized detectable difference $\delta$ when $\alpha , \beta$ and sample size are given.
