Toxicity and Safety Profiles of Methanolic Extract of Pistacia integerrima J. L. Stewart ex Brandis (PI) for Wistar Rats

  • Objectives: The goals of this research were to evaluate acute (single-dose) and sub-acute (repeated-dose) toxicity profiles of methanolic extract of Pistacia integerrima J. L. Stewart ex Brandis (PI) for Wistar rats and to assess the safety profile of PI by observing physiological changes, mortality, changes in body weight, the histopathology of body organs, the hematology and the biochemistry of the animals. Methods: The toxicity profile of PI was evaluated using Wistar rats of both sexes. Animals were divided into four groups: Group 1; control group (normal saline), Group 2; PI-1 (250 mg/kg), Group 3; PI-2 (500 mg/kg), Group 4; PL-3 (1,000 mg/kg). An acute-toxicity study in which animals received a single dose of PI extract (2,000 mg/kg) and were then observed for 14 days for changes in skin, fur, eye color, mucous membrane secretions and excretions, gait, posture, and tonic or clonic movements was performed according to guideline 425 of the Organization of Economic and Corporation Development (OECD). In the repeated-dose toxicity study (OECD - 407) animals received a daily dose of PI extract for 28 days (4 weeks). The parameters observed in this study include body weight, hematology and biochemistry of the animals. Results: In the acute toxicity study, no mortalities or changes in behavior were noted in the animals. The repeated-dose toxicity study was also devoid of any toxicity in the animals during the 28 days of testing with PI extract. The extract did not alter- the body weight, hematology or biochemistry of the animals. The methanolic extract of PI was to be found safe to the no-observed-adverse-effect-level (NOAEL) for the single-dose and repeated-dose toxicity tests in rats. Conclusion: The methanolic extract of PI was devoid of toxicity; hence, it can be used for various ayurvedic preparations and treatments of diseases.

  • TS-DP2 is a recombinant human granulocyte colony stimulating factor (rhG-CSF) manufactured by TS Corporation. We conducted a four-week study of TS-DP2 (test article) in repeated intravenous doses in male and female Sprague-Dawley (SD) rats. Lenograstim was used as a reference article and was administered intravenously at a dose of $1000{\mu}g/kg/day$. Rats received TS-DP2 intravenously at doses of 250, 500, and $1000{\mu}g/kg/day$ once daily for 4 weeks, and evaluated following a 2-week recovery period. Edema in the hind limbs and loss of mean body weight and body weight gain were observed in both the highest dose group of TS-DP2 and the lenograstim group in male rats. Fibro-osseous lesions were observed in the lenograstim group in both sexes, and at all groups of TS-DP2 in males, and at doses of TS-DP2 $500{\mu}g/kg/day$ and higher in females. The lesion was considered a toxicological change. Therefore, bone is the primary toxicological target of TS-DP2. The lowest observed adverse effect level (LOAEL) in males was $250{\mu}g/kg/day$, and no observed adverse effect level (NOAEL) in females was $250{\mu}g/kg/day$ in this study. In the toxicokinetic study, the serum concentrations of G-CSF were maintained until 8 hr after administration. The systemic exposures ($AUC_{0-24h}$ and $C_0$) were not markedly different between male and female rats, between the administration periods, or between TS-DP2 and lenograstim. In conclusion, TS-DP2 shows toxicological similarity to lenograstim over 4-weeks of repeated doses in rats.

A New Training System for Improving Postural Balance Using a Tilting Bed

  • In this paper, we propose an early rehabilitation training system for the improvement of postural balance with multi-modality on a tilting bed. The integration of the visual, somatosensory and vestibular functions is significant to for maintaining the postural control of the human body. However, conventional rehabilitation systems do not provide multi-modality to trainees. We analyzed the characterization of postural control at different tilt angles of an early rehabilitation training system, which consists of a tilting bed, a visual feedback, a computer interface, a computer, and a force plate. The software that we developed for the system consists of the training programs and the analysis programs. To evaluate the characterization of postural control, we conducted the first evaluation before the beginning of the training. In the following four weeks, 12 healthy young and 5 healthy elderly subjects were trained to improve postural control using the training programs with the tilting bed. After four weeks of training, we conducted the second evaluation. The analysis programs assess (center of pressure) COP moving time, COP maintaining time, and mean absolute deviation of the trace before and after training at different tilt angles on the bed. After 4 weeks, the COP moving time was reduced, the COP maintaining time was lengthened, and the mean absolute deviation of the trace was lowered through the repeated use of vertical, horizontal, dynamic circle movement training programs. These results show that this system improves postural balance and could be applied to clinical use as an effective training system.

Productive performance of Mexican Creole chickens from hatching to 12 weeks of age fed diets with different concentrations of metabolizable energy and crude protein

  • Objective: The study aimed to evaluate the productive performance, carcass yield, size of digestive organs and nutrient utilization in Mexican Creole chickens, using four diets with different concentrations of metabolizable energy (ME, kcal/kg) and crude protein (CP, %). Methods: Two hundred thirty-six chickens, coming from eight incubation batches, were randomly distributed to four experimental diets with the following ME/CP ratios: 3,000/20, 2,850/19, 2,700/18 and 2,550/17. Each diet was evaluated with 59 birds from hatching to 12 weeks of age. The variables feed intake (FI), body weight gain (BWG), feed conversion (FC), mortality, carcass yield, size of digestive organs, retention of nutrients, retention efficiency of gross energy (GE) and CP, and excretion of N were recorded. Data were analyzed as a randomized block design with repeated measures using the GLIMMIX procedure of SAS, with covariance AR (1) and adjustment of degrees of freedom (Kendward-Roger), the adjusted means were compared with the least significant difference method at a significance level of 5%. Results: The productive performance variables BWG, mortality, carcass yield, fat and GE retention and excretion of N were not different (p>0.05) due to the diet effect. In the 3,000/20 diet, the chickens had lower values of FI, FC, crop weight, gizzard weight, retention, and retention efficiency of CP (p<0.05) than the chickens of the 2,550/17 diet. Conclusion: The Mexican Creole chickens from hatching to 12 weeks of age can be feed with a diet with 2,550 kcal ME and 17% CP, without compromising productive parameters (BWG, mortality, carcass yield) but improving retention and retention efficiency of CP.

The Effects of Scapula and Pelvis Symmetrical Reciprocal Pattern of PNF Concept on Respiratory Function and Quality of Life in the Elderly (PNF의 어깨뼈 골반 대칭 상호 패턴이 노인의 호흡기능과 삶의 질에 미치는 효과)

  • Purpose : The purpose of this study is to determine the effects of scapula and pelvis symmetrical reciprocal pattern exercise of PNF concept on respiratory function and quality of life in elderly subjects. Methods : Nineteen elderly subjects with healthy were recruited. Subjects performed scapula and pelvis symmetrical reciprocal pattern exercise of PNF concept. Exercise was appied 30 minutes three times per week for four weeks. The respiratory function and quality of life (QOL) test three times (before, two weeks, and four weeks). Respiratory function includes forced vital capacity (FVC), forced expiratory volume at one second (FEV1), Peak Expiratory Flow (PEF) and chest cage expansion test (CCET). The QOL test was measured Korean WHOQOL-BRIEF. The analysis method was analyzed through the one-way ANOVA repeated methods, and the statistical significance is α=.05. It was analyzed through the post test Bonferroni test. Results : After the 4 week scapula and pelvis symmetrical reciprocal pattern exercise of PNF concept showed statistically significant differences in the respiratory function (FVC, FEV1, PEF, and CCET), and QOL (p<.05). As a result of the post-hoc test, FVC showed a significant increase in the mid test and post test compared to the pre test (p<.05), FEV1 showed a significant increase in the post test compared to the pre test (p<.05). PEF showed a significant increase in all pre-test, mid-test, and post-test sections (p<.05), CCET showed a significant increase in all pre-test, mid-test, and post-test sections (p<.05). QOL showed a significant increase in all pre-test, mid-test, and post-test sections (p<.05). Conclusion : In this study, the scapula and pelvis symmetrical reciprocal pattern exercise of PNF concept improved respiratory function and QOL. The findings suggest that this intervention could be beneficial in improving respiratory function and QOL in the elderly.

Ovarian stimulation and liver dysfunction: Is a clinical relationship possible? A case of hepatic failure after repeated cycles of ovarian stimulation

  • Liver damage induced by ovarian stimulation has been demonstrated in some cases reported in the literature. However, there has never been a fruitful debate on this topic. The present manuscript tried to fill this gap. We reported a case of a 35-year-old nulliparous woman admitted to our obstetric emergency room for severe pre-eclampsia. She had been subjected to four cycles of controlled ovarian stimulation for intrauterine insemination. At 32 weeks of gestation, she developed severe pre-eclampsia, which led to HELLP syndrome complicated by fatal liver failure. The etiological link between ovarian stimulation and HELLP syndrome is intriguing. Further investigations are needed to understand whether repeated ovarian stimulation may represent a risk factor in pre-eclamptic patients.

The Effect of Horticultural Program on Cognitive Function, Activities of Daily Living(ADL) and Hand Grip Strength of Institutionalized Dementia Patients (원예 프로그램이 시설 내 치매환자의 인지기능, 일상생활 수행능력 및 악력에 미치는 효과)

  • Purpose: The purpose of this study was to examine the effect of use of a horticultural program on the cognitive function, activities of daily living, and hand grip strength of institutionalized dementia patients. Method: Twenty three study participants completed a five week protocol comprised of one week of baseline and four weeks of treatment. The study design was a one group repeated measurement study. For the first and fifth week of the study period, data was collected with 3 instruments: Korean Version of Hasegawa Dementia Scale (HDS-K), Katz's ADL index, hand grip strength. Data analysis was conducted by SPSS version 10.0. Result: Total HDS-K score(p=.000), Activities of daily living(p=.013) and hand grip strength(p=.001) was significantly improved after treatment compared to the baseline. Conclusion: The horticultural program was effective for improving cognitive function, ADL and hand grip strength of the dementia patients. Repeated studies utilizing a control group are needed, using various durations, plants and processes, to confirm these results and to standardize the horticultural program protocol.

  • This study was designed to evaluate a repeated dose toxicity of a new tsutsugamushi vaccine, MBRI-98305R in 4-month-old beagle dogs. Animals were intramuscularly injected with dosages of 283, 56.6, 14.15 and 0 $\mu\textrm{g}$/kg everyday for 4 weeks, respectively. There were no dose-related statistical significant changes in clinical signs, body weight changes, food or water consumption, opthalmoscopy, hematological findings and biochemical examination of all animals treated with MBRI-98305R.(omitted)

Effects of an Infection Prevention Education Program in Infant and Child for Married Immigrant Vietnamese Women (영·유아 자녀를 둔 베트남 결혼이주여성을 위한 감염예방교육 프로그램의 효과)

  • Purpose: This study was conducted to examine the impact of infection prevention program on the knowledge and performance among married Vietnamese immigrant women an infant or child. Methods: A non-equivalent control group pretest-posttest design was used. Participants were 53 Vietnamese women with an infant or a child in G city. The treatment (n=25) received an infection prevention program series that included eight sessions over a four week period with four time through telephone interviews a weekly intervals. Data were collected between October 26 and December 14, 2012, and were analyzed Chi-square, t-test, and repeated measure analysis of variance with SPSS/WIN 18.0. Results: The treatment group reported significantly higher scores in infection prevention knowledge (F=43.98, p<.001) and infection prevention performance (F=92.61, p<.001) at four and eight weeks following the treatment as compared to the control group. Conclusion: Results suggest that an infection prevention program is beneficial in increasing knowledge and performance to prevent infection.

Four-week Intravenous Toxicity Study of DA-3030, a Recombinant Human G-CSF, in Rats (재조합 인 과립구 콜로니 자극인자 DA-3030의 랫드에 대한 4주 정맥내 반복투여 독성연구)

  • This study was conducted to evaluate the repeated dose toxicity of DA-3030, a recombinant human granulocyte colony stimulating factor(rhG-CSF), in rats. DA-3030 was administered intravenously once a day for 4 weeks to 20 males and 20 females per group at doses of 0(control), 115 and 1150 $\mu\textrm{g}$/kg, and to 15 males and 15 females per group at doses of 1.15 and 11.5$\mu\textrm{g}$/kg. After the administration period, 5 males and 5 females per group in the 0,115 and 1150 $\mu\textrm{g}$/kg groups were placed on withdrawal for 2 weeks. Through-out the study, all the rats survived. The administration of DA-3030 induced, a marked increase in the number of peripheral neutrophils, elevation of serum alkaline phosphatase activity, and splenomegaly in the rats of both sexes receiving 115 or 1150 $\mu\textrm{g}$/kg. Histopathologic examination revealed extramedullary granulopoiesis in spleen and liver, and increase in the number of activated macrophages in spleen in rats of both sexes in 115 and 115 $\mu\textrm{g}$/kg groups, and increased M/E ratio in 11.5, 115 and 1150$\mu\textrm{g}$/kg groups. Most of the changes produced by DA-3030 were thought to be attributable to exaggerated pharmacological effect of the drug, and subsided or disappeared after the recovery period. Under the present condition, no effect dose of DA-3030 is estimated at 1.15 $\mu\textrm{g}$/kg/day.
