Protein -surfactant interactions have been investigate by measuring ζ-potential of $\beta$-lactoglobulin-coated emulsion droplets and $\beta$-lactoglobulin in solution in the rpesenceof surfactant, with particular emphasis on the effect of protein heat treatment(7$0^{\circ}C$, 30min). When ionic surfactant (SDS or DATEM) is added to the protein solution, the ζ-potential of the mixture is found to increase with increasing surfactant concentration, indicating surfactant binding to the protein molecules. For heat-denatured protein,it has been observed that the ζ-potential tends to be lower than that of the native protein. The effect of surfactant on emulsions is rather complicated .With SDS, small amounts of surfactant addition induce a sharp increase in zeta potential arising from the specific interaction of surfactant with protein. With further surfacant addition, there is a gradual reductio in the ζ-potential, presumably caused by the displacement of adsorped protein (and protein-surfactant complex) from the emulsion droplet surfac by the excess of SDS molecules. At even higher surfactant concentrations, the measured zeta potential appears to increase slightly, possibly due to the formation of a surfactant measured zeta potential appears to increase slightly, possibly due to the formation of surfactant micellar structure at the oil droplet surface. This behaviour contrastswith the results of the corresponding systems containing the anionic emulsifier DATEM, in which the ζ-potential of the system is found to increase continuously with R, particularly at very low surfactant concentration. Overall, such behaviour is consisten with a combination of complexation and competitive displacement between surfactant and protein occurring at the oil-water interface. In addition, it has also been found that above the CMC, there is a time-dependent increase in the negative ζ-potential of emulsion droplets in solutions of SDS, possibly due to the solublization of oil droplets into surfactant micelles in the aqueous bulk phase.

The formation of emulsions and micelles in water/ceramide PC104/CholE $O_{20}$/C$_{16}$E $O_{20}$ and water/ceramide PC104/CholE $O_{20}$ mixtures was investigated through the phase behavior studies. The phase diagrams showed the existence of micelle and emulsion regions in both systems. The mixed surfactant system (CholE $O_{20}$/C$_{16}$E $O_{20}$) showed the wider micellar and emulsion regions than the single surfactant system (CholE $O_{20}$). From FT-IR measurements, it was found that the polyoxyethylene (POE) groups of surfactants formed the hydrogen bonds with amido carbonyl group in ceramide PC104. This result indicated that the hydrophilic part (EO) of surfactants could stabilize the lamellar structure and emulsion of ceramide PC104. The mixed surfactant system (CholE $O_{20}$/C$_{16}$E $O_{20}$) resulted in the smaller emulsion droplet size due to the effect of curvature at the interface, thus further increasing emulsion stability. With the penetration of $C_{16}$E $O_{20}$into the interfacial layer of surfactants in emulsion, the curvature of the interface might be altered for the formation of smaller emulsion droplets. The mixed surfactant system could incorporate up to 4 wt. % of ceramide PC104 into emulsion more than single surfactant system.ystem.m.
첨가제가 단백질 약물 방출 속도 및 약물 제제 제조 및 구조에 미치는 영향을 고찰하였다. 친수성 첨가제인 D-sorbitol의 경우 친유성 첨가제보다 단백질 약물 방출 속도를 감소시킬 수 있었으며 최적의 농도는 3% (w/v)로 나타났다. 또한 제제 제조시 점도를 낮게 유지할 뿐 아니라 상분리 없는 균일한 pluronic 용액상태를 유지하여 약물이 첨가될 경우에 균일한 약물제제를 만들 수 있었다. 한편 D-sorbitol은 pluronic 수용액의 CMC를 낮추고 마이셀 표면에 작용하여 구조를 강화하는 역할을 수행하는 것으로 보인다. 따라서 pluronic 제제에 D-sorbitol을 첨가하여 단백질 약물의 안정성을 향상시키고 효과적인 약물전달 시스템을 설계할 수 있었다.
Although lovastatin (LS) is widely used in the treatment of hypercholesterolemia, its bioavailability is known to be around 5%. This study was aimed to increase the solubility and dissolution-permeation rates of LS using solid dispersions (SDs) with bile salts. The solubilities of LS in water, aqueous bile salt solutions and non-aqueous vehicles were determined, and effects of bile salts on the cellulose or duodenal permeation of LS from SDs were evaluated using a horizontal permeation system. SDs were prepared at various ratios of LS to carriers, such as sodium deoxycholate (SDC), sodium glycocholate (SGC) and/or 2-hydroxypropyl-$\beta$-cyclodextrin (HPCD). The addition of bile salts (25 mM) in water increased markedly the solubility of LS by the micellar solubilization. Some non-aqueous vehicles were effective in solubilizing LS. From differential scanning calorimetric studies, it was found that the crystallinity of LS in SDs disappeared, indicating a formation of amorphous state. The SDs showed markedly enhanced dissolution compared with those of their physical mixtures (PMs) and drug alone. In the dissolution-permeation studies using a cellulose membrane, the donor and receptor solutions were maintained as a sink condition using pH 7.0 phosphate buffer containing 0.05% sodium lauryl sulfate (SLS). The flux of LS alone was nearly same as that of LS-SDC-HPCD (1:3:6) PM. However, the flux of LS-SDC-HPCD (1:3:6) SD slightly increased compared with drug alone and PM, suggesting that entrapment of LS in micelles does not significantly hinder the permeation across cellulose membrane. In the dissolution-duodenal permeation studies using a LS-HPCD-SDC (1:3:6) SD, the addition of various bile salts in donor solutions (25 mM) enhanced the permeation of LS markedly, and the fluxes were found to be $0.69{\pm}0.41$, $0.87{\pm}0.51$, $0.84{\pm}0.46$, $0.47{\pm}0.17$ and $0.68{\pm}0.32{\mu}g/cm^2/hr$ for sodium cholate (SC), SDC, SGC, sodium taurodeoxycholate (STDC) and sodium taurocholate (STC), respectively. The stepwise increase of donor SGC concentration increased the flux dose-dependently. From the relationship of donor SGC concentration and flux, the concentration of SGC initiating the permeation across the duodenal mucosa was calculated to be 11.1 mM, which is nearly same as the critical micelle concentration (CMC, 11.6 mM) of SGC. However, with no addition of bile salts and below CMC, the permeation was very limited and irratic, indicating that LS itself is very poor permeable. Higher protions of bile salt in SD such as LS-SDC or LS-SGC (1 : 49 and 1 : 69) showed highly promoted fluxes. In conclusion, SD systems with bile salts, which may form their micelles in intestinal fluids, might be a promising means for providing enhanced dissolution and intestinal permeation of practically insoluble and non-absorbable LS.
$^1H$ NMR gradient spin echo법을 이용하여 polyoxyethylene dodecyl ether[$C_{12}H_{25}(OCH_2CH_2)nOH$] 수용액에서 미셀 형성과정에 대한 것을 비교검토 하였다. 여기서 n=5($C_{12}EO_5$) 및 n=8($C_{12}EO_8$)인 비이온성 계면활성제를 사용하였으며 등방성상의 영역범위에서의 자기확산 계수는 일정온도에서 농도변화에 따른 pulsed field gradient법을 사용하여 측정하였고, 또한 여러 프로톤의 시그날에 대한 피크폭(line width)을 추적하여 액정 특성에 대한 것을 검토하였다. 알킬사슬의 메틸렌 시그날의 넓혀짐은 $C_{12}EO_5-$물 계에서는 핵사고날 액정상의 근접될 때 관찰되었지만 $C_{12}EO_8-$물 계에서는 넓혀짐이 보다 작게 관찰되었다. 낮은 온도에서 농도가 증가함에 따라 $C_{12}EO_5$에서는 막대형 미셀이 형성되지만 $C_{12}EO_8$에서는 전 농도 범위에서 작은 미셀로 이루워 진다. 계면활성제의 자기확산계수는 서서히 증가하다가 극소점에 이르러서는 농도가 증가함에 따라 급격히 감소한다. 극소점의 위치는 온도가 담점에 이르렀을 때 낮은 농도에서 나타나고 이 계에서는 두 개의 등방성 상으로 분리된다. 피크폭 연구에서 피크폭의 넓어짐은 $C_{12}EO_5$에서는 온도가 증가되었을 때 온도의 차이로 형성된다. 그 결과 담점에 이르러 계면활성제의 회합체가 커지는 것으로 생각한다. 이 회합체가 담점에 이르러 한정된 모양을 갖는 것은 아니다. $C_{12}EO_8$에서는 미셀들이 온도의 증가에 영향을 받지 않고 미셀형성이 불확실하다. 에틸렌옥사이드 일부분의 메틸렌 시그날은 일관되게 좁게 나타났다. 회합에 있어서 이들은 알킬사슬 메틸렌보다 작은 질서로 나타내었다. 회합의 크기나 모양에 있어서 여러 변화성은 상 변화에 따라 등방성과 액정상의 안정성 범위가 정하여 진다. 회합체 크기와 상 구조는 여러 온도와 농도 변화에서 분자의 효과적 모형을 고려하여 정성적인 결과에 따른다.
Sodium N-lauroyl-N-methyl taurate의 cmc에 대한 온도효과를 검토하였다. cmc값들은 처음 $20^{\circ}C{\sim}40^{\circ}C$의 온도 범위에서는 감소하다가 더욱 온도가 증가되면 다시 증가하는 경향이었다. cmc에 대한 온도의 의존성으로부터 여러가지 열역학적 파라미터들(${\Delta}H_m$, ${\Delta}G_m$, ${\Delta}S_m$)을 산출하였다. Sodium N-lauroyl-N-methyl taurate의 cmc에 대한 여러 전해질들의 첨가효과를 또한 검토하였고, 소수성결합 형성에너지 및 미셀의 해리도를 log cmc와 log conc.의 곡선으로부터 산출하였다. ${\Delta}H_m$값들은 온도가 증가함에 따라 감소하였고, $40^{\circ}C{\sim}50^{\circ}C$의 온도범위에서 부호가 양에서 음으로 변하였다. ${\Delta}G_m$값들은 전해질 농도 및 온도가 증가할수록 감소하였다.
5% corn oil과 17 mM Brij 700으로 제조한 corn oil-in-water emulsion(O/W)의 droplet의 크기는 첨가되는 phenol의 양이 많아질수록 지방구가 다소 커지는 경향을 보였으며, continuous phase의 surfactant 양은 대조구에 비해 다소 낮아졌다. O/W에 녹차로부터 분리한 phenol류를 100-200 ppm의 농도로 첨가하고 30일간 저장하며 hydroperoxide의 생성량을 측정한 결과 대조구에 비해 낮은 hydroperoxide 함량을 보여 유도기간이 길어지는 것으로 나타났으며, phenol 종류별 hydroperoxide 생성억제 효과는 BHT > procyanidin B3-3-O-gallate>(+)-gallocatechin>(+)-catechin의 순서로 나타났다. 지방 산화의 결과 발생하는 aldehyde의 함량을 측정함으로서 산화정도를 측정하기 위하여 headspace hexanal 양을 측정한 결과, 대조구는 15일까지 hexanal이 검출되지 않았으나 phenol 첨가구는 20-30일로 headspace aldehyde의 불검출기간이 길어졌으며, 농도가 증가할수록 낮은 함량을 나타내었다. Phenol 종류별 headspacealdehyde 생성 억제 효과는 BHT > procyanidin B3-3-O-gallate > (+)-gallocatechin >(+)-catechin의 순서로 나타났다.
Antimicrobial peptides (AMPs) are small but effective cationic peptides with variable length. In previous study, four hexapeptides were identified that showed antimicrobial activities against various phytopathogenic bacteria. KCM21, the most effective antimicrobial peptide, was selected for further analysis to understand its modes of action by monitoring inhibitory effects of various cations, time-dependent antimicrobial kinetics, and observing cell disruption by electron microscopy. The effects of KCM21 on Gram-negative strain, Pseudomonas syringae pv. tomato DC3000 and Gram-positive strain, Clavibacter michiganensis subsp. michiganensis were compared. Treatment with divalent cations such as $Ca^{2+}$ and $Mg^{2+}$ inhibited the bactericidal activities of KCM21 significantly against P. syringae pv. tomato DC3000. The bactericidal kinetic study showed that KCM21 killed both bacteria rapidly and the process was faster against C. michiganensis subsp. michiganensis. The electron microscopic analysis revealed that KCM21 induced the formation of micelles and blebs on the surface of P. syringae pv. tomato DC3000 cells, while it caused cell rupture against C. michiganensis subsp. michiganensis cells. The outer membrane alteration and higher sensitivity to $Ca^{2+}$ suggest that KCM21 interact with the outer membrane of P. syringae pv. tomato DC3000 cells during the process of killing, but not with C. michiganensis subsp. michiganensis cells that lack outer membrane. Considering that both strains had similar sensitivity to KCM21 in LB medium, outer membrane could not be the main target of KCM21, instead common compartments such as cytoplasmic membrane or internal macromolecules might be a possible target(s) of KCM21.
염화마그네슘 존재하의 비스(1,10-페난트롤린)구리(II) $(Cu(ph){_2}^{2+})$-도데실황산나트륨(SDS)의 전기화학적 거동들이 고찰되었다. $Mg^{2+}$의 첨가에 의한 SDS의 용액에서 $Cu(ph){_2}^{2+}$의 $E_{pa}$와 $E_{1/2}$ 값은 양의 값으로 이동했다. 1.0 mM $Cu(ph){_2}^{2+}$의 27 mM $MgCl_2$을 포함한 100mM NaCl 용액에서, ${\Delta}E_p$ 대 -log[SDS]로 도시한 그림에서 두 선의 교차점을 임계미셀농도로 결정하였다 (순환 전압전류법에 의해 3.48 mM SDS; 표면 장력법에 의해 3.34 mM SDS). $Cu(ph){_2}^{2+}$의 용액에 $Mg^{2+}$가 첨가되었을 때 유리탄소전극에서의 이중층의 거리가 감소했고 미셀형성이 지연되었다.
