ELISA-LC/MS/MS 병행에 의한 식품 중 aflatoxins 분석 (Determination of Total Aflatoxins in Foods by Parallelism of ELISA and LC/MS/MS)

  • High performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS) have been widely used to quantify aflatoxins in food, but these methods are expensive, time-consuming, unsuitable for analysis of the routine screening of large sample numbers and require derivatization and high level techniques to perform. The objective of this study is to detect aflatoxins in a large number of foods by a high efficient analytical system of combined enzyme linked immunosorbent assay (ELISA) for screening and LC/MS/MS for confirmation. The samples spiked individually with aflatoxin $B_1$ (0.5 and 1.0 ng/g) and total aflatoxins (10 ng/g) were analyzed by ELISA and LC/MS/ MS, and the recoveries for ELISA and LC/MS/MS were 71.8~119.2% and 70.8~135.3%, respectively. A total of 378 samples (grains, nuts, soybean and fermented soybean foods, pepper and fermented pepper foods) were purchased from the six major cities in Korea and analyzed by ELISA-LC/MS/MS system. Twenty two (5.8%; peanut: 11, pistachio: 2, walnut: 6, almond: 1, pepper powder: 1, pepper paste: 1) out of 378 samples were screened as aflatoxin B1 positive by ELISA, but, 4 (1.1%; peanut: 2, pistachio:1, pepper powder: 1) out of the 22 samples screened were confirmed as aflatoxins positive at levels of 1.02~52.79 ng/g by LC/MS/MS. ELISA-LC/MS/MS system provides a more rapid, accurate and cost-effective method for the detection of aflatoxins in large number of samples.

국내식품의 중금속 오염과 위해성 분셕 (Contamination and Risk Analysis of Heavy Metals in Korean Foods)

  • 인체의 중금속 노출은 식품이 대부분을 차지하고 있어 세계 어느나라나 인체건강의 유지라는 측면에서 깊은 관심을 쏟고 있다. 본 연구에서는 국내산 식품의 중금속 함량과 식이섭취량에 관련된 연구결과를 총정리하였으며 외국자료와 비교함으로써 중금속의 위해성을 평가하였다. 식품군별(해산어류, 연안패류, 담수어류, 곡류)로 비소, 카드뮴, 수은, 납의 4가지 금속원소의 함량에 대한 연차적 변화추세를 분석한 결과 1970년대보다는 1980년대, 그리고 1990년대로 올수록 중금속의 평균농도는 감소하는 것으로 나타났다. 중금속의 국내기준과 비교하였을 때 평균치로는 기준을 초과하는 경우가 없었으나 최고치로는 기준을 초과하는 경우가 드물게 발견되었다. 한국인에 의한 중금속의 식이섭취량 데이터를 요약하면 1980년대보다는 1990년대에 감소하는 경향이 있었다. 중금속 중 As, Hg의 평균섭취량은 PTWI의 6∼8%, Cd, Pb은 50∼80%수준으로 모두 허용량을 밑돌고 있으나 특정식품을 많이 먹는 극단소비량으로 계산한다면 PTWI를 초과할 수 있으므로 심도있는 평가가 요구된다. 한국인에 의한 Cd, Hg, Pb의 식이섭취량은 여러 국가에 대한 GEMS/Food 데이터와 비교시 중위권에 속하는 것으로 나타났다. 식품 중에서 수산물은 Hg, Cd 및 As의 오염이, 일반식품은 Pb의 오염이 우려되고 있으므로 이들 중금속에 대한 법적기준을 설정하여 중금속에 의한 오염 진행상황을 감시해야 될 것이다. 나아가 모든 급원(음식물, 공기, 불, 직업적 노출)으로부터 중금속의 총 노출량을 평가하고 식생활에서의 위해요소를 확인함으로써 식량자원의 안전성을 확보할 수 있게 되기를 바란다.

LC-MS/MS를 이용한 건강기능식품 원료 중 바이칼린, 엘레우테로사이드 E, 리구스틸라이드 동시분석법 개발 (Development of Analytical Method for Determination of Baicalin, Eleutheroside E, and Ligustilide by LC-MS/MS in Raw Materials of Health Functional Foods)

  • 본 연구는 건강기능식품 중 baicalin, eleutheroside E, ligustilide를 효과적으로 분석할 수 있는 방법을 확립하기 위하여 수행되었다. 이에 LC-MS/MS를 이용하여 신속하고 효율적으로 동시분석할 수 있는 시험법을 확립하였으며, 확립된 시험법에 대해 특이성, 검출한계, 정량한계, 정확도, 정밀도에 대한 검증을 통하여 유효성을 확인하고자 하였다. 표준용액을 이용하여 검량선을 작성한 결과 $r^2$ > 0.99 이상의 직선성을 확인하였고, baicalin, eleutheroside E, ligustilide에 대한 정량한계는 각각 $39.3{\mu}g/L$, $106.7{\mu}g/L$, $76.1{\mu}g/L$이었으며, 검출한계는 각각 $13.0{\mu}g/L$, $35.2{\mu}g/L$, $25.1{\mu}g/L$이었다. 또한 평균 회수율은 각 성분에 대해 108.0~109.9%, 99.8~101.3%, 91.4~97.2%로 나타났으며, 반복정밀도는 상대표준편차 5%이하, 실험실간 재현성은 9%이하로 나타나 정확성, 재현성이 우수하였으며 이는 AOAC 가이드라인에서 제시한 기준에 모두 적합한 수준이었다. 따라서 개발된 분석법은 향후 건강기능식품 중 baicalin, eleutheroside E, ligustilide를 동시분석하는데 효과적으로 활용할 수 있을 것으로 판단된다.

식품 중 발기부전치료제 및 사용금지 성분 모니터링 (Monitoring of illegal compounds and prohibited natural ingredients in foods)

  • 성기능개선, 원기증강 등에 효과가 있는 것으로 판매되고 있는 식품 등에서 발기부전치료제 및 그 유사물질 등이 불법적으로 혼입되어 검출되고 있다. 이들 부정물질이 혼입된 불량식품은 안전성과 유효성이 입증되지 않았기 때문에 국민건강을 위협할 수 있다. 그러므로 본 연구에서는 식품과 가짜 발기부전치료제에 불법 혼입된 부정물질을 모니터링하기 위하여, 해외 인터넷 사이트와 오프라인 매장에서 성기능개선, 원기증진을 표방하는 식품과 최음제, 가짜 발기부전치료제 161건을 구입하였다. 시험방법의 유효성 검증을 위하여 선택성, 직선성, 검출한계, 정량한계, 정확성, 정밀성을 검토하였다. LC-PDA와 LC-MS/MS를 이용하여 부정물질 54종을 동시분석한 결과, 48건의 시료에서 7종의 부정물질(요힘빈, 이카린, 실데나필, 타다라필, 클로로프레타다라필, 데메틸실데나필, 디메틸치오실데나필)이 검출되었다. 성기능개선이나 최음제로 판매되는 제품에는 부정물질이 혼입될 가능성이 있으므로, 이들 제품을 구입할 때는 각별한 주의가 필요하다.

근육 식품 중의 diethylstibestrol과 zeranol 분석법 (Analytical Methods for Diethylstibestrol and Zeranol in Muscle Foods)

  • 성장 촉진 호르몬제인 DES와 zeranol의 분석을 위한 조건을 검토한 결과는 다음과 같다. 질량분석기의 SIM에서 선택한 이온 중 DES는 $M^+$ 416, $D_8DES$$M^+$ 420, ${\beta}-estradiol$$M^+$ 416, zeranol은 $M^+$ 433이 가장 좋은 응신비를 나타내었고 이들 이온을 선택할 경우 효율적으로 분석이 가능하였다. 강 음이온 교환수지인 Dowex $2{\times}8$, 400 mesh 칼럼 1 cc에 통과시킨 경우 방해물질이 효율적으로 제거되었고 회수율이 가장 높았다. $D_8DES$을 내부표준물질로 사용하여 회수율을 측정한 결과 DES는 $85{\sim}110%$이었고, zeranol은 $75{\sim}110%$이었으며 ${\beta}-estradiol$을 내부표준물질로 사용한 경우 DES의 회수율은 $82{\sim}105%$, zeranol은 $65{\sim}120%$이었다. 또한 $D_8DES$ 대신에 ${\beta}-estradiol$을 내부표준물질로 사용하여 DES와 zeranol을 분석하는 것이 가능하였다. DES와 zeranol의 LOD를 측정한 결과 각각 0.05 ng/g, 0.2 ng/g었으며 LOQ를 측정할 결과 DES는 0.5 ng/g, zeranol은 1.0 ng/g이었다. 국내산과 수입산 쇠고기, 국내산 돼지고기, 닭고기, 오리고기, 양식광어와 양식 송어를 분석한 결과 DES와 zeranol은 잔류되지 않았다.

일간 신문의 기사와 광고에 나타난 식품영양정보의 현황 조사 (2002년) (Analysis of Food and Nutrition Information for Articles and Advertisements in the Daily Newspapers (Year 2002))

  • The object of this study is to investigate and analyze the quantity and quality of informations on health, food and nutrition reported by newspapers. Six different major domestic daily newspapers(Hankook Ilbo, Donga IIbo, Chosun IIbo, Kyunghyang Shinmun, Hankyoreh Shinmun, JoongAng Ilbo), were monitored from 1st of May to 31st of Oct. 2002.. The results of monitoring in the newspapers were summarized as follows; 1. The total percentage of the articles on health and food nutrition was 2.7% and the percentage of the advertisements on health, food and nutrition was 17.5% of total area of the newspaper. The ratio of the number of food and nutrition topic on the total number of health and food nutrition topic was 35.8% for the articles, was 62.9% for the advertisements. Among advertisements on food and nutrition, the percentage of advertisements on healthy foods was the highest (3,481 or 55.0%). 2. Contents of 340 articles(26.1%) were reported as suitable informations. Contents of 259 articles(19.9%) were reported as inaccurate informations on health, food and nutrition. In the analysis of advertisements, the number of advertisements without sufficient reliable sources was 2,488 cases(23.0%), and with exaggerated contents was 2,268 cases(21.0%). The articles and advertisements should be backed by scientific research or reliable sources and also the opinions of people with expertise in order to report accurate informations to the general public. In order to achieve these results, there should be continuing monitoring activity for the newspapers.

Individualized Exercise and Diet Recommendations: An Expert System for Monitoring Physical Activity and Lifestyle Interventions in Obesity

  • This paper proposes an exercise recommendation system for treating obesity that provides systematic recommendations for exercise and diet. Five body indices are considered as indicators for recommend exercise and diet. The system also informs users of prohibited foods using health data including blood pressure, blood sugar, and total cholesterol. To maximize the utility of the system, it displays recommendations for both indoor and outdoor activities. The system is equipped with multimode sensors, including a three-axis accelerometer, a laser, a pressure sensor, and a wrist-mounted sensor. To demonstrate the effectiveness of the system, field tests are carried out with three participants over 20 days, which show that the proposed system is effective in treating obesity.

Comparison of Near-Infrared Spectroscopy with Raman Spectroscopy from the Point of Nondestructive Analysis of Biological Materials

  • Recently, near-infrared (NIR) spectroscopy and Raman spectroscopy have received keen interest as powerful techniques for nondestructive analysis of biological materials. The purpose of this review paper is to compare the advantages of NIR and Raman spectroscopy in the nondestructive analysis. Both methods are quite unique and often complementary. For example. NIR spectroscopy is very useful in monitoring in situ the content of components inside biological materials while Raman spectroscopy is very suitable for identifying micro-components on the surface of biological materials. In this article specific characters of the two spectroscopic methods are discussed first and then several examples of applications of NIR and Raman spectroscopy to the biological nondestructive analysis are introduced.

Development of Convenient Software for Online Shelf-life Decisions for Korean Prepared Side Dishes Based on Microbial Spoilage

  • User-friendly software was developed to determine the shelf-life of perishable Korean seasoned side dishes in real time based on growth models of spoilage and pathogenic microorganisms. In the program algorithm, the primary spoilage and fastest-growing pathogenic organisms are selected according to the product characteristics, and their growth is simulated based on the previously monitored or recorded temperature history. To predict the growth of spoilage organisms with confidence limits, kinetic models for aerobic bacteria or molds/yeasts from published works are used. Growth models of pathogenic bacteria were obtained from the literature or derived with regression of their growth rate data estimated from established software packages. These models are also used to check whether the risk of pathogenic bacterial growth exceeds that of food spoilage organisms. Many example simulations showed that the shelf-lives of the examined foods are predominantly limited by the growth of spoilage organism rather than by pathogenic bacterial growth.

  • Farmers need timely information on the nutritional status of their animals and the nutritive value of pastures and supplementary feeds if they are to apply successfully this existing nutritional information. Near infrared reflectance(NIR) spectroscopy has been used over the last forty years to analyse accurately protein, fiber, and other organic components in animal foods. NIR spectroscopy is a rapid, non-destructive, and non-polluting technology. When properly calibrated, NIR spectroscopy is used successfully with both concentrate and forage feeds. NIR methods predict in vitro digestibility accurately and precisely, and can predict in vivo digestibility at least as well as conventional "wet chemistry" methods such as in vivo digestion or the pepsin-cellulase method, and much more rapidly. NIR technology has been applied to the routine monitoring (through analysis of feces samples) of the nutritional status of cattle and other grazing animals. This report reviews the use of near infrared reflectance(NIR) spectroscopy to monitor the nutritive value of animal feeds and the nutritional status of grazing animals.
