Preparation and Characterization of Resveratrol Nanoemulsions Stabilized by Self-assembly and Complex Coacervation Consisting of Sodium Alginate, Chitosan, and β-Cyclodextrin

  • Resveratrol was incorporated into various combinations of single- and double-layer nanoemulsions, prepared by self-assembly emulsification and complex coacervation with chitosan, alginate, and ${\beta}$-cyclodextrin, respectively. Resveratrol nanoemulsions were composed of medium-chain trigacylglycerols (MCTs), $Tween^{(R)}$ 80, water, chitosan, alginate, and ${\beta}$-cyclodextrin. The corresponding mixtures were formulated for the purpose of being used as a nutraceutical delivery system. Resveratrol nanoemulsions were obtained with particle sizes of 10-800 nm, with the size variation dependent on the emulsification parameters including the ratio of aqueous phase and surfactant ratio. Resveratrol nanoemulsions were characterized by evaluating particle size, zeta-potential value, stability, and release rate. There were no significant changes in particle size and zeta-potential value of resveratrol nanoemulsions during storage for 28 days at $25^{\circ}C$. The stability of resveratrol in the double-layer nanoemulsions complexed with chitosan or ${\beta}$-cyclodextrin was higher, compared with the single-layer nanoemulsions.

Production of DNA polymerase from Thermus aquaticus in recombinant Escherichia coli

  • Among dozens of DNA polymerases cloned from thermophilic bacteria, Taq DNA polymerase from Thermus aquaticus has been most frequently used in polymerase chain reaction (PCR) that is being applied to gene cloning, DNA sequencing, gene expression analysis, and detection of infectious and genetic diseases. Since native Taq DNA polymerase is expressed at low level in T. aquaticus, recombinant Escherichia coli system was used to produce Taq DNA polymerase in a large amount. Taq DNA polymerase was expressed as a soluble form under the control of tac promoter in E. coli, and purified by heat treatment and ion exchange chromatographies. The purified Taq DNA polymerase was nearly homogeneous and exhibited a similar DNA amplification activity with a commercial Taq DNA polymerase.

  • Objectives Fish vitellogenin (VTG) is produced in the female liver during oogenesis through the estradiol cycle and produced in the male liver by endocrine disrupting chemicals (EDCs) such as alkylphenols. In this study, we propose that the VTG concentration in the pale chub could be detected using monoclonal antibodies and polyclonal antibodies against vitellin (Vn) in a VTG enzyme-linked immunosorbent assay (ELISA) system. Methods Monoclonal antibodies and polyclonal antibodies were produced using the Vn extracted from the matured ovum of the ovary. The VTG was extracted from the plasma of the male pale chub. The Vn and VTG were confirmed by measuring the molecular weight of their proteins using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and the specificity of the antibodies was checked through western blotting methods. The assay system was validated with respect to optimal assay concentrations, specificity, recovery, and intra- and inter-assay variations. Results The Vn consisted of two protein bands with apparent molecular weights of 64 and 37 kDa. The SDS-PAGE indicated protein weights of 146 and 77 kDa in the VTG. The assay range was 15.6 ng/mL to 2,000 ng/mL, and the value of the intra- and inter-assay variations were within 10.0% and 14.7%, respectively. The recovery rate was $99.5{\pm}5.5%$. Conclusions A sandwich ELISA was developed that could be used to qualify the VTG of pale chub in screening for EDCs. Pale chub is an ideal species for observing estrogen activity in the environment because of its extensive habitat and extensive food chain. The ELISA developed here would be more favorable than those for other species for determining the effect of long-term food chain accumulation of EDCs in aquatic environments.

공급사슬관리 (SCM) 운영 변화의 저항에 영향을 미치는 선행요인 평가 : 식품제조 중소기업 K사 사례를 중심으로 (An Examination of Multi-Dimensional Constructs of Resistance to Supply Chain Management (SCM) Change for a Small and Medium Sized Food Production Company)

  • The present study identified several antecedents that create perceived resistance to supply chain management (SCM) change. This work particularly emphasizes SCM change, which is notable given its central role in reacting market orientation and varied environmental and managerial conditions. A careful case study on a small and medium sized food production company in South Korea leads to the formulation of our framework including one second-order construct of 'resistance' as well as eight first-order constructs of 'implementing the payment terms', 'balancing of business process', 'fear of responsibility', 'business sustainability transparency', 'past experience of failure', 'competence of work personnel', 'cooperation with third parties' and 'sharing personnel information with partner'. The hypothesized relation of first- and second-order construct was validated using survey sample data collected from 350 respondents who completed their questionnaire instrument. Results from confirmatory factor analysis revealed that nomological validity was established at statistical significance level by identifying six first order constructs of 'implementing the payment terms', 'fear of responsibility', 'business sustainability transparency', 'past experience of failure', a conbined construct of 'competence of work personnel/cooperation with third parties' and 'sharing personnel information with partner'. The findings from our work are expected to provide important insights to the strategy for SCM risk management for small and medium sized company.

A Decentralized and Non-reversible Traceability System for Storing Commodity Data

  • In the field of traceability systems, researchers focus on applications in the agricultural food traceability and scanning commodities. The purposes of this paper, however, is to propose an efficient and reliable traceability system that can be applied to all kinds of commodities. Currently, most traceability systems store data in a central server, which is unreliable when the system is under attack or if the administrator tampers with the data for personal interests. Therefore, it is necessary to design a system that can eliminate these threats. In this paper, we propose a decentralized and non-reversible traceability system for storing commodity data. This system depends on blockchain technology, which organizes data in the form of chains without a central server. This chain-style storage mechanism can prevent malicious modifications. In addition, some strategies are adopted to reduce the storage pressure and response time when the system has stored all kinds of commodity data.

Pheflinus baumii 자실체 추출물의 Sarcoma-180 에 대한 항암 및 면역효과 (Antitumor and immunomodulatory effects of a hot water extract of Phellinus baumii using Sarcoma-180 in the mouse)

  • 국내에서 재배되고 있는 약용버섯인 상황(P. baumii) 자실체의 열수 추출물로 부터 건강 기능성 식품 및 의약품개발의 기초자료를 제공하고자 항암실험 및 면역효과에 대한 실험을 수행하였다. 본 연구에서 사용한 PMO-P4의 경우 rDNA의 ITS부위의 염기서열 분석을 통한 계통분석결과 P. baumii로 판명 되었으며, PMO-P4 자실체로부터 열수 추출한 성분(HWE-P4)을 160mg/kg/day의 농도로 하여 Sarcoma-180 암세포를 유발한 마우스에 경구투여한 결과 A군의 경우, 종양억제효과는 35.3%로 대조군보다 유의성 있게 감소하였으며(p<0.05), 생명연장 효과는 156%증가한 것으로 나타났다. 시료의 경구투여 방법에 있어서도 시료를 먼저 2주간 투여한 후 암세포를 접종한 A군의 경우가 시료 투여와 암세포 접종을 동시에 실시한 B군의 경우 보다 종양억제효과 및 생명연장효과가 높았음을 알 수 있었다. A군의 경우 대조군에 비하여 CD4/CD8의 비율이 71.4%증가 하였으며, CD25(IL-2receptor chain)분자의 발현을 5배정도 증가시키는 것으로 관찰되었다. 이러한 결과로 보아 P. baumii로 밝혀진 PMO-P4 자실체로부터 열수 추출한 성분의 항암효과는 암세포를 직접 공격하여 항암효과를 나타내기 보다는 T cell등의 면역세포를 활성화시킴으로써 암세포를 억제 혹은 사멸시키는 것으로 생각된다.

푸드뱅크 기탁식품의 안전성 확보를 위한 HACCP 제도 적용 (Implementation of HACCP System for Safety of Donated Food in Foodbank Organization)

  • The purpose of this study was to analyze the problems arising from the actual conditions of the Foodbank, and to implement the HACCP system as a solution in terms of increasing the safety of donated food within the Foodbank. In order to apply HACCP system, the entire Foodbank working process such as preparation, collection, transportation, division, and distribution was considered and analyzed to decide the application point for CCPs. Donated foods mainly consisted of processed foods, raw materials, lunch boxes, and cooked foods from mass catering establishments, which dominated over the others in terms of quantity. Cooked foods were divided into three groups based on menu-types and processing methods. Temperature, pH, and aw were measured on cooked foods, and Total Plate Count, Coliforms, E. coli, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, and E. coli O157:H7 were conducted in order to apply a HACCP plan. From these experiments, temperature, pH, and $a_w$ of donated food were likely contributed to microbial growth. Donated foods before HACCP implementation showed high numbers in terms of total plate count and Coliforms, both well over the acceptable standard levels. By setting the CCPs on maintenance of donated food below $10^{\circ}C$ and using a $75^{\circ}C$ reheating method, microbiological hazard levels were able to be controlled and lowered. From these results, it is concluded that in order to guarantee food safety, foods donated to the Foodbank must not only maintain a reasonable level of initial microbiological growth, but also must be handled properly through time and temperature controls within the Foodbank system. Furthermore, in terms of implementing the HACCP plan within the Foodbank management structure, basic food safety and sanitation measures, such as reheating facilities and various cold chain systems such as refrigerated vehicle for food transportation are importantly needed. The training and education of Foodbank personnel and management in areas such as awareness of hygiene and safe food handling and practice are also required and necessary.

엉겅퀴로부터 분리 정제한 Silymarin 및 Silybin의 지질 과산화에 대한 항산화 효과 (Antioxidative Effects of Silymarin and Silybin Purified from Silybum marianum on Lipid Peroxidation)

  • 본 연구는 지질의 과산화에 대한 항산화 효과를 조사하기 위하여 엉겅퀴(Silybum marianum)로부터 silymarin 및 silybin을 정제하여 실험하였다. Silymarin 및 silybin은 xanthine oxidase system에서 superoxide anion의 생성을 억제하였다. 쥐의 간 mitochondria에서는 silymarin 및 silybin은 reduced nicotinamide adenine dinucleotide phosphate(NADPH)에 의해 효과적 또는 ascorbic acid 또는 Fenton's reagent에 의하여 비효소적으로 유도되는 지질의 과산화를 억제하였다. 또 mitochondria의 지질과산화도 silymarin 및 silybin에 의하여 억제되었고 NADPH 의존 cychrome P-450 reductase에 의한 Fe2+의 산화도 silymarin 및 silybin에 의하여 억제되었다. Silymarin 및 silybin은 microsome의 효소 시스템 및 linoleic acid hydroperoxide induced peroxidation system에서 지질의 과산화의 연쇄반응에서 유리기의 억제효과가 있었다.

  • 포스트 코로나 시대의 수요 변화와 급변하는 물류 환경 속에서 콜드체인 물류 섹터의 중요성이 부각되고 연관 산업도 빠르게 성장하고 있다. 콜드체인의 범위는 식품에만 한정되지 않고 의약품, 반도체, 화훼 등 다양한 분야로 확장되고 있으며, 최근에는 백신 보관과 수송에 대한 수요가 급증하고 있다. 수요 증가에 기인한 국내 저온시설 및 관련 시설 신설과 개보수 급증 현상은 장기적 관점에서는 콜드체인 연관 산업이 향후 포화상태에 이르러 성장 둔화로 이어질 수 있다. 이에 대비하여 AI, 블록체인, IT 기술과 연계한 콜드체인 인프라 구축에 대한 노하우 축적, 한국의 선진국 진입과 방역 우수국가 등의 이점을 활용하여 잠재적 틈새시장인 유엔 조달시장 진출을 고려해 볼만 하다. 유엔 조달시장에서 콜드체인 시장은 저개발 국가를 중심으로 수요가 증가하고 있고, 지속 성장세를 전망한다. 본 논문에서는 국내 콜드체인 연관기업의 역량을 분석하고 국내외 콜드체인 물류 시장 동향, 해외시장 진출 현황을 조사하고 전문가 심층 설문조사를 실시하여 국내 콜드체인 산업의 유엔 조달시장 진출 전략을 제시하였다.

농산물 유통과정에서 복숭아의 품질유지를 위한 마스터 포장 시스템 (A Master Packaging System for Preserving Qualities of Peaches in the Fresh Produce Supply Chain)

  • 신선 농산물 공급체인에서 최종적으로 소비자에게 신선한 복숭아를 공급할 수 있는 통합적 마스터 포장 시스템을 구성하고 그 효과를 실험적으로 검증하고자 하였다. 마스터 포장 시스템은 $35{\mu}m$ 두께의 LDPE 필름 bag에 6개의 300 g 낱개 복숭아의 $30{\mu}m$ OPP 개별포장을 포함하는 것으로 구성하였다. 개별포장에 직경 $59{\mu}m$의 미세공을 1, 3, 7개로 두는 변수조건을 두어서 $5^{\circ}C$의 저온단계 저장과 $20^{\circ}C$의 유통판매단계에서 포장기체조성과 품질변화를 측정하여 비교하였다. $5^{\circ}C$에서 일정기간 저장한 마스터 포장은 해체하여 $20^{\circ}C$ 조건으로 옮겨서 3일간 보관하는 조건으로 실험하였다. 대조구로서 필름에 싸지 않고 골판지 상자에 복숭아를 6개씩 담아서 같은 온도조건에서 보관하였다. 7개 미세공을 가진 포장이 $5^{\circ}C$의 저온단계 저장과 $20^{\circ}C$의 유통판매단계에서 6~10%의 $O_2$ 농도와 11~19% $CO_2$ 농도를 유지시켜 가장 적정 MA 조건에 가까운 기체조성을 유지할 수 있는 것으로 판단되었다. 이보다 작은 미세공을 가진 포장은 일련의 연속적인 저장에서 혐기적인 조건을 유발하였다. 대조구와 비교하여 마스터 포장의 모든 처리구는 낮은 중량손실과 양호한 경도 유지를 보여주었다.
