Investigation of Residual Organochlorine Pesticides in Apple and Pear Orchard Soil and Fruit (사과 및 배 과수원의 토양 및 과실 중 잔류성유기염소계 농약류 잔류량 조사)

  • BACKGROUND: Residual organochlorine pesticides (OCPs) are designated as persistent organic pollutants (POPs) by Stockholm Convention because they bioaccumulate through the food web, and pose a risk of causing adverse effect to human health and the environment. Apple and pear is economic crop in agriculture, and its cultivation area and yield has been increased. Therefore, we tried to investigate the OCPs residue in apple and pear orchard soils and fruits. METHODS AND RESULTS: Extraction and clean-up method were developed using the modified QuEChERS method for residual organochlorine pesticides (ROCPs) in apple and pear orchard soil and fruits. Recovery and limit of quantitation (LOQ) of ROCPs in soil and fruits were 75.4-101.4 and 76.9-93.4%, 0.03-0.21 and $0.6-1.2{\mu}g/kg$, respectively. Detected ROCPs in apple and pear orchard soil was 2,4-DDT, 4,4-DDD, 4,4-DDE, 4,4-DDT, and endosulfan sulfate, the residues were 2.2, 1.9-48.0, 1.3-84.1, 90.6-863.1, and $11.3-239.0{\mu}g/kg$, respectively. But five pesticides in all fruit samples were not detected. CONCLUSION: These results showed that ROCPs residues in apple and pear orchard soil had no effect on safety of agricultural products.

Study on fatty acids composition by latent fingerprint deposition (유류된 잠재지문의 지방산조성에 관한 연구)

  • In order to investigate the information for effective detection and developing of latent fingerprints, we identified fatty acids composition of latent fingerprints on non-porous evidence surface and the chemical changes of latent fingerprint residue after print deposition during 7 months. Fingerprints from eight Korean male donors (aged 29-50 years) and one female donor (aged 36 years) were collected. All fingerprints were found to contain lauric acid (C12:0), myristic acid (C14:0), palmitic acid (C16:0), stearic acid (C18:0), elaidic acid (C18:1n9t), oleic acid (C18:1n9c), linoleic acid (C18:2n6c), arachidic acid (C20:0), linolenic acid (C18:3n3), erucic acid (C22:1n9) and docosadienoic acid (C22:2) and primarily palmitic acid (35.45-48.37%), oleic acid (14.84-28.49%), stearic acid (9.71-24.96%) and linoleic acid (7.68-18.8%) occupied 75% of total fatty acids. When the fingerprints were deposited at dark room for 7 months, total fatty acids components decreased about 12-25%. It can be explained that significant degradation of long-chain fatty acids such as elaidic acid (C18:1n9t), arachidic acid (C20:0), linolenic acid (C18:3n3), erucic acid (C22:1n9), and docosadienoic acid (C22:2) resulted in the generation of myristic acid (C14:0), myristoleic acid (C14:1) and pentadecanoic acid (C15:0).

Antibacterial and Therapeutic Effects of Houttuynia cordata Ethanol extract for Murine Salmonellosis (어성초 ethanol 추출물의 마우스 살모넬라 감염증에 대한 항균 및 치료효과 규명)

  • Salmonellosis is a major bacterial zoonosis that causes a variety of disease syndromes, self-limited enteritis to fatal infection in animals and food-borne infection and typhoid fever in humans. Recently, the emergence of multidrug resistant strains of Salmonella spp. causes more serious problems in environment and public health. The present study was investigated the antibacterial effect of Houttuynia cordata ethanol extract(HCEE) for murine salmonellosis. In the cytotoxic effect of HCEE on RAW 264.7 cells, there was no detectable effect with any concentrations between 25 and 100 ${\mu}g/ml$ after 8 h incubation. The bacteriocidal effect of HCEE was not showed on a Salmonella enterica serovar Typhimurium(S. typhimurium). HCEE makes morphological change of the RAW 264.7 cells, and there was significant decreased bacterial uptake and intracellular replication within Salmonella infected cells. And further nitric oxide(NO) production of Salmonella infected RAW 264.7 cells with HCEE was decreased comparing to RAW 264.7 cells without HCEE until 8 h post infection. Oral administration of HCEE showed a therapeutic effect for S. typhimurium infected BALB/c mice. The mortality of HCEE treated mouse was 80% until 12 days, while that of HCEE untreated mouse was 100 % until 8 days after lethal dose of S. typhimurium infection. These data suggested that HCEE has a potency treatment for intracellular replicative pathogen including salmonellosis, brucellosis, tuberculosis, listeriosis etc., and the application of HCEE makes new strategies for safety medicine development without antibiotic resistance bacterial appearance and residue problem in food and solves the public health problem from antibiotic mis- and over use.

Residual Characteristics and Processing Factors of Difenoconazole in Fresh Ginseng and Processed Ginseng Products (인삼 및 가공품 중 difenoconazole의 잔류특성 및 가공계수)

  • This study was carried out to elucidate the residual characteristics and calculate processing factors of difenoconazole in ginseng and its processed products, such as dried ginseng, red ginseng and their water and alcohol extracts. The pesticide was sprayed onto the ginseng according to its pre-harvest intervals in 2009 (four-year-old ginseng) and 2010 (five-year-old ginseng). Harvested ginseng was processed to dried ginseng, red ginseng and their extracts according to the commercially well-qualified conventional methods provided by the Korea Ginseng Corporation. Limit of detection (LOD) and limit of quantitation (LOQ) of difenoconazole in fresh ginseng were 0.001 and 0.003 mg/kg, respectively. In case of processed ginseng products, their levels were 0.002 and 0.007 mg/kg, respectively. Concentration of difenoconazole in both fresh ginseng and its processed products increased with the experimental period. Processing factors, calculated as a ratio of difenoconazole concentration in processed products to fresh ginseng were found to be 1.71 to 2.17 and 1.62 to 2.03 in case of dried and red ginseng, respectively, while those for their extracts ranged from 1.76 to 2.98. In case of five-year-old dried ginseng and red ginseng as well as their extracts, the ranges of processing factor of difenoconazole were found to be 2.9 to 3.1, 1.9 to 2.2 and 2.4 to 4.7, respectively.

Processing Factor of Matrine in Chilli Pepper (홍고추 중 matrine의 가공계수)

  • This study was carried out to investigate the residual characteristics and calculate processing factor of the environment friendly material matrine in fresh chilli pepper by drying. Spray solution of matrine was prepared by dilution of the commercial product (2% active ingredient) with water at 1 : 1000 (v/v) ratio and sprayed onto chilli pepper plants at seven day intervals. Samples were collected at 0, 1, 3, 5 and 7 days after last application and then dried using a hot air dry oven at $60^{\circ}C$ for 36 hours until the water content was reduced to 14%. Recoveries and storage period stabilities of matrine in the samples ranged from 106.6 to 119.1% and 106.6 to 113.1%, respectively. The residual concentrations of matrine in fresh chilli pepper and dried chilli peppers treated only once were found to be from less than 0.01 to 0.11 and from 0.03 to 0.25 mg/kg, respectively. In case of plants sprayed twice with matrine, the residual concentrations ranged from 0.02 to 0.12 and from 0.04 to 0.4 mg/kg, respectively. Processing factor of matrine in the fresh chilli pepper by drying was found to be from 1.5 to 3.3, indicating that the residual concentration of matrine in dried chilli pepper increased about two or three times by drying.

Changes in Chemical and Microbiological Properties of Spring Waters in Tongyeoung Area (통영시내 약수의 화학적 및 세균학적 품질변화)

  • This study was carried out to changes in chemical and microbiological properties of spring waters in Tongyeoung area. In this paper, ninety spring water samples were collected from 9 station for 11 month to evaluated chemical and bacteriological water quality. Range and mean values of constituents of the samples are as followed; water temperature 5.2~25.8$^{\circ}C$, 16.3$^{\circ}C$, pH 6.0~7.2, 6.7, total residue 33.6~210 mg/1, 90.6 mg/1, turbidity 0.35~5.48, 1.45NTU, KMnO4 consumed 0.51~4.21 mg/1, 1.39 mg/1, chloride ion 6.23~42.5, 16.7 mg/l, phosphate-phosphorus ND-0.04, 0.02 mg/1, nitrite-nitrogen ND~0.02, 0.01 mg/1, nitrate-nitrogen ND~3.56, 1.42 mg/1, ammonia-nitrogen ND~0.20, 0.14 mg/1, dissolved total nitrogen ND~3.78, 1.57 mg/1, iron 0.04~0.28, 0.13ppm, zinc 0.03~0.66, 0.20ppm, mangan ND~0.01, allumium 0.14~0.58, 0.39ppm, copper ND~0.01, 0.01, lead ND~0.01, 0.01ppm, Arsenic ND~0.01, 0.01ppm, mercury ND~0.02, chrome not detected, cadmium not detetced respectively. The viable cell counts of the spring waters ranged 5.0~760/m1(means 130/m1). Range and mean value of total coliform and focal coliform MPN's of the spring waters were 0~2,400MPN/100 ml, 73MPN/100 ml and 0~540MPN/100 ml, 21MPN/100 ml. Spring water quality was usually poor with viable cell counts exceeding 130 CFU/liter and the coliform counts in spring waters of 73 MPN/liter. Composition of coliform by IMViC reaction was 33.3% E. coli, 15.6% Citrobacter freundii, 35.6% Klebsiella aerogenes and others.

Quality Characteristics of Bijijang in Different Fermentation Conditions (발효 조건을 달리한 비지장의 품질특성)

  • Changes in quality characteristics of Bijijang (fermented soybean curd residus) prepared at $35^{\circ}C\;and\;40^{\circ}C$ for 0, 12, 24, 36, and 48 hr were investigated. Acidity of Bijijang increased, whereas pH and Hunter's color values decreased during fermentation. Immediately after Bijijang preparation, ${\alpha}-and\;{\beta}-amylase$ activities were very low, ${\beta}-Amylase$ activity during fermentation increased rapidly, with those fermented at $40^{\circ}C$ higher than at $35^{\circ}C$. Neutral pretense activity was significantly higher than acidic pretense activity, and increased gradually after 12 hr. Change in total nitrogen content in Bijijang was insignificant, whereas contents of amino-type and water-soluble nitrogens increased significantly during fermentation. Major free amino acids of Bijijang were Arg, Pro, Glu, Thr, Ser, and Lys at initial fermenting stage, and, as fermentation progressed, contents of Cys, Met Glu, Ile, Leu, and Phe increased. Reducing sugar contents of Bijijang fermented at $40^{\circ}C$ were higher than those fermented at $35^{\circ}C$. Sucrose content decreased and glucose content increased. Glucoside (genistin and daidzin) contents decreased and aglycone (genistein and daidzein) contents increased during preparation of Biji and fermentation of Bijijang. Contents of free sugars and isoflavones were higher in Bijijang fermented at $40^{\circ}C$ than at $35^{\circ}C$. Based on these results, fermentation at $40^{\circ}C$ for 48 hr was determined to be optimum fermentation condition for Bijijang.

Ecological Risk Assessment of Residual Petroleum Hydrocarbons using a Foodweb Bioaccumulation Model (먹이연쇄 생물축적 모형을 이용한 잔류유류오염물질의 생태위해성평가)

  • Residual petroleum hydrocarbons after an oil spill may accumulate in the marine benthic ecosystem due to their high hydrophobicity. A lot of monitoring data are required for the estimation of ecosystem exposure to residual petrochemicals in an ecological risk assessment in the affected region. To save time and cost, the environmental exposure to them in the affected ecosystem can also be assessed using a simple food-web bioaccumulation model. In this study, we evaluated residual concentrations of four selected polycyclic aromatic hydrocarbons (phenanthrene, anthracene, pyrene, and benzo[a]pyrene) in a hypothetic benthic ecosystem composed of six species under two exposure scenarios. Body-residue concentration ranged 5~250 mg/kg body depending on trophic positions in an extreme scenario in which the aqueous concentrations of PAHs were assumed to be one-tenth of their aqueous solubility. In addition, bioconcentration factors (BCFs) and bioaccumulation factors (BAFs) were evaluated for model species. The logarithm of bioconcentration factor (log BCF) linearly increased with increasing the logarithm of 1-octanol-water partition coefficient (log $K_{OW}$) until log $K_{OW}$ of 7.0, followed by a gradual decrease with further increase in log $K_{OW}$ without metabolic degradation. Biomagnification became significant when log $K_{OW}$ of a pollutant exceeded 5.0 in the model ecosystem, indicating that investigation of food-web structure should be critical to predict biomagnifications in the affected ecosystem because log $K_{OW}$ values of many petrochemicals are higher than 5.0. Although further research is required for better site-specific evaluation of exposure, the model simulation can be used to estimate the level of the ecosystem exposure to residual oil contaminants at the screening level.

Evaluation of Greenhouse Gas Emissions in Cropland Sector on Local Government Levels based on 2006 IPCC Guideline (2006 IPCC 가이드라인을 적용한 지자체별 경종부문 온실가스 배출량 평가)

  • This study was conducted to estimate the greenhouse gas emissions on local government levels from 1990 to 2010 using 2006 IPCC guideline methodology. To calculate greenhouse gas emissions based on the 16 local governments, emission factor and scaling factor were used with default value and activity data came from the food, agricultural, forestry and fisheries statistical yearbook of MIFAFF (Ministry for Food, Agriculture, Forestry, and Fisheries). The total emissions in crop sector gradually decreased from 1990 to 2010 due to a decline in agricultural land and nitrogen fertilizer usage. The annual average emission of greenhouse gas was the highest in Jeonnam (JN) with 1,698 Gg $CO_2$-eq and following Chungnam (CN), Gyungbuk (GB), Jeonbuk (JB) and Gyunggi (GG). The sum of top-six locals emission had occupied 83.4% of the total emission in cropland sector. The annual average emissions in 1990 by applying 2006 IPCC guideline were approximately 43% less than the national greenhouse gas inventory by 1996 IPCC guideline. Jeonnam (JN) province occupied also the highest results of greenhouse gas emission estimated by gas types (methane, nitrous oxide and carbon dioxide) and emission sources such as rice cultivation, agricultural soil, field burning of crop residue and urea fertilizer.

Processing of Oleoresin Onion (양파 Oleoresin의 가공)

  • The purpose of this study was to investigate the extraction yield and quality stability as to the oleoresin process with large amount of onion at one time. The first mixed-product is raw onion juice which was reduced the compression and concentrated by Brix 70% mixed together wit the residue which was extracted and concentrated by ethanol, the second product manufactured by the same method above after the autoclaving with onion, and the other product is made by grinding by 50mesh to freeze-dried onion. Each of yields were 7.3, 9.1 and 0.8% and each of total sugar content was 616.4, 712.3 and 150.3mg/g. Therefore the product extracted by ethanol from freeze-dried onion was very low in yield and total sugar content. By the index of the overall odor intensity, contents of total pyruvate were 1,733.7, 520.6, and 2,716.5$\mu\textrm{g}$/g for each product. As a result, oleoresin onion processing that desired to use raw onion was remarkable for odor recovery. For the homogenous mixture with concentrate of onion juice and ethanol extract were emulsified by the addition of 2% of PGDR(polyglycerol condensed ricinoleate) and agitation(10,000rpm, 30 minutes). At this time, interfacial tension was 1.9 dyne/cm and the formation of emulsion was for 96.2% when left over 24hours in 6$0^{\circ}C$. When it was to be centrifuged(2,000$\times$G, 80 minutes) after emulsification, the volume of emulsion level without seperation was 92.6%, and very high in emulsification stability. The induced heating-oxidize with soy bean oil and sesame oil added to 1% of onion oleoresin, induction-time extension effect appeared with antioxidant activity that was applicable for 80.8~82.2% as to the effect of addition of 0.02% BHA.
