Identification of Differentially Expressed Genes in Bovine Follicular Cystic Ovaries

  • Follicular cystic ovary (FCO) is one of the most frequently diagnosed ovarian diseases and is a major cause of reproductive failure in mammalian species. However, the mechanism by which FCO is induced remains unclear. Genetic alterations which affect the functioning of many kinds of cells and/or tissues could be present in cystic ovaries. In this study, we performed a comparison analysis of gene expression in order to identify new molecules useful in discrimination of bovine FCO with follicular cystic follicles (FCFs). Normal follicles and FCFs were classified based on their sizes (5 to 10 mm and $\geq25mm$). These follicles had granulosa cell layer and theca interna and the hormone $17{\beta}$-estradiol ($E_2$)/ progesterone ($P_4$) ratio in follicles was greater than one. Perifollicular regions including follicles were used for the preparation of RNA or protein. Differentially expressed genes (DEG) that showed greater than a 2-fold change in expression were screened by the annealing control primer (ACP)-based PCR method using $GeneFishing^{TM}$ DEG kits in bovine normal follicles and FCFs. We identified two DEGs in the FCFs: ribosomal protein L15 (RPL15) and microtubule-associated protein 1B (MAP1B) based on BLAST searches of the NCBI GenBank. Consistent with the ACP analysis, semi-quantitative PCR data and Western blot analyses revealed an up-regulation of RPL15 and a down-regulation of MAP1B in FCFs. These results suggest that RPL15 and MAP1B may be involved in the regulation of pathological processes in bovine FCOs and may help to establish a bovine gene data-base for the discrimination of FCOs from normal ovaries.

  • The etiology of mesiodens is unknown but the most widely accepted theory is the hyperactivity of the dental lamina. Complications of mesiodens are delayed or prevented eruption of maxillary central incisors, displacement or rotation of permanent teeth, crowding of affected region, abnormal diastema or permanent space closure, dilaceration or abnormal root development of permanent teeth, primordial or follicular cyst formation, root resorption of adjacent teeth, eruption into nasal cavity. If mesiodens rotate the maxillary central incisors, space deficiency is not common and relapse is very common. So overcorrection is needed. To prevent the rotational relapse, early treatment, overcorrection, long retention period, properly formed proximal surface, use of coupled force, and surgical techniques have been suggested. The authors present two cases, whose chief complain were severely rotated maxillary incisors by mesiodens, treated by orthodontic and surgical technique and showed good results.

Development of Differential Diagnosis and Treatment Method of Reproductive Disorders Using Ultrasonography in Cows V, Differential Diagnosis of Ovarian Cysts in Slaughtered Cows (초음파검사에 의한 소의 번식장애 감별진단 및 치료법 개발 V, 도축우에서 난소낭종의 감별진단)

  • To establish the differential diagnosis and treatment method in bovine ovarian cysts, specially ovarian cysts with corpus luteum, serum progesterone (P$_4$) concentration and ultrasonography for measuring the cystic wall thickness and diameter of cyst and corpus luteum were investigated from slaughtered cows with ovarian cysts. Ovarian cysts were classified 8 types by the number of cyst, cystic wall thickness and present of corpus luteum. Ovarian cysts with corpus luteum were 11 (13.6%) of 81 cows and ovarian cysts without corpus luteum were 70 (86.4%) cows. The incidence rates of 8 various types of ovarian cysts were as follows; 2Ba 33.3%, 2Aa 25.9% and 2Bb 14.8%, respectively. The incidence rates of ovarian cysts without corpus luteum were follicular cyst 59.2% and luteal cyst 27.2%. The cystic wall thickness were 2Ab 3.7 mm and 2Bb 3.5 mm, and the serum P4 concentrations were above 2.0 ng/ml in 1Aa, 1Ab, 1Ba, 2Ab and 2Bb, respectively. In ovarian cysts with corpus luteum, the correlation coefficients between corpus luteum area and serum P$_4$ concentration were 0.45. In ovarian cysts without corpus luteum, there was significantly positive correlations between cystic wall thickness and serum P$_4$ concentration ($r^2$= 0.54, p<0.01). These results indicate that PGF$_2$$\alpha$ analogues can be choice for treatment of ovarian cysts with corpus luteum and above 3 mm the cystic wall thickness because serum P$_4$ concentrations were above 2.0 ng/ml in ovarian cysts with corpus luteum and thickened cystic wall. In conclusion, it is suggest that ultrasonography is useful diagnostic tool for diagnosis and selection of treatment remedy in cystic ovaries of bovine.

$H^1$ Magnetic Resonance Spectroscopy of Cystic Ovarian Lesions (낭성 난소병변의 자기공명분광술)

  • On $H^1$ MRS (magnetic resonance spectroscopy), malignant tumors show higher concentration of metabolite than benign lesions. Lactate double peak was detected in malignant tumor and endometriosis, and more prominent high concentration was demonstrated in endometriosis. Tuboovarian abscesses and salpingitis do not show prominent peak. Dermoid cysts show high levels of lipid peak. Paratubal cyst and follicular cyst can be showed the lipid peak, however, the concentration of lipid is lower than that of dermoid cyst. $H^1$ MRS of ovarian cystic lesions can give valuable information about the presence of metabolites of ovarian cystic lesions.

Upregulation of Glutathion S-Transferase mu 1 in Bovine Cystic Follicles

  • Follicular cystic follicles (FCFs) show delayed regression with persistent follicle growth. However, the mechanism by which follicles are persistently grown remains unclear. Glutathione S-transferases (GSTs) are drug-metabolizing and detoxification enzymes that are involved in the intracellular transport and metabolism of steroid hormones. In this study, a proteomic analysis was performed to identify whether GST expression is changed in bovine FCFs and to predict the interactions between GST and other proteins. Normal follicles and FCFs were classified based on their sizes (5 to 10 mm and 25 mm). In bovine follicles, GST mu 1 (GSTM1) was detected as a differentially expressed protein (DEP) and significantly up-regulated in FCFs compared to normal follicles (p<0.05). Consistent with the proteomic results, semi-quantitative PCR data and western blot analysis revealed an up-regulation of GSTM1 in FCFs. Expression levels of aromatase and dehydrogenase proteins were changed in FCFs. These results show that the up-regulation of GSTM1 that is observed in bovine FCFs is likely to be responsible for the persistent follicle growth in FCFs as the activity of aromatase and the dehydrogenases.

  • Purpose: Nevus comedonicus is a rare disease involving abnormal development of the pilosebaceous unit, clinically appearing confluent clusters of open comedones. It is characterized by follicular epidermic invaginations filled with keratin, with atrophic pilose or sebaceous structures which are open to the lower pole of the invagination. We report a child with inflammatory pustules and cysts correlated to the nevus comedonicus. Methods: A 15-month-old girl was referred for treatment of a nevus comedonicus that has been developed since birth. There were periodic episodes of erythema and swelling of the plaque. The patient had treated previously with retinoid cream for 2 weeks without obvious benefit at dermatology clinic. We excised the nevus comedonicus with Y-shape leaving some lesions. Results: When we incised skin, we could see multiple large whitish cysts beneath the nevus comedonicus. Histopathologic findings showed deep invagination of epidermis filled with keratin plugs, keratinous cyst in the dermis, rudimentary hair follicle. Conclusion: We treated rare nevus comedonicus with multiple large cysts as surgical excision.

A repeatedly recurrent desmoplastic ameloblastoma after removal and allobone graft: Radiographic features compared with histological changes

  • A 40-year-old man suffered from a repeatedly recurrent desmoplastic ameloblastoma in the right maxillary anterior and premolar regions. During the first visit, the patient was provisionally histopathologically diagnosed with a developmental cyst, and it was confirmed to be unicystic ameloblastoma and resected. Four years later, the lesion recurred, and was diagnosed as a desmoplastic type of ameloblastoma and removed again. Then, 5 years after the second surgery, the lesion recurred again, and was diagnosed as a type containing a follicular pattern, recurrent ameloblastoma. A panoramic radiograph showed a multilocular and mixed radiolucent/radiopaque expansile lesion at the first visit, a unilocular cystic lesion confined to the premolar area at the second visit, and a small soap bubble appearance in the molar area in the final visit. Cone-beam computed tomographic images of the final recurrence of the tumor revealed multiple small cyst-like structures in the right maxillary anterior and posterior regions.

Clinical evaluation of thymic tumors: a report of 10 cases (흉선에 발생한 종양 10례에 대한 임상적 고찰)

  • Thymic tumor is the most common neoplasm originating in the anterior mediastinum. Histologically, thymic tumors and tumorlike conditions have been classified into thymic cyst, thymolipoma, true thymic hyperplasia, thymoma and carcinoid tumor of the thymus gland. We have experienced several tumors and a few tumorlike conditions of the thymus gland in 10 patients: thymoma 6, thymic hyperplasia 2, carcinoid tumor 1 and thymic cyst 1. The age distributions were ranging from 30 to 60 years except for one 3 year old child [malignant thymoma, lymphocytic type], and male to female ratio was 1:1.5. In 6 cases, thymic lesions were removed through lateral thoracic incision [right 2, left 4] because the mediastinal masses were growing far into the pleural space, meanwhile, medially located lesions [4 cases] through median sternotomy incision. Among these 10 patients, 3 were malignant thymomas, in which 2 were died of recurrence after tumor resection. Myasthenia gravis was all noted in 3 females [thymoma 2, thymic follicular hyperplasia 1], in which only two revealed remission in symptoms following thymectomy. The authors would like to recommend early radical thymectomy through median sternotomy incision whenever patients demonstrate suspicious thymic tumor lesions on the chest roentgenogram or generalized myasthenic symptoms.

  • Fine structures of the testis and vas deferens in the fall-web worm, Hyphantria cunea Drury, are studied with electron microscope. Adult worms have single testis close to the midlines of the abdomen. Testis is composed of 4 follicles which are incompletly separated from each other and bounded together by a peritoneal sheath. The peritoneal sheath consisted of outer cuticular layer and two kinds of inner layers, in which glycogen particles are dispersed commonly. These two layers are divided by the morphology of cytoplasmic granules. Follicular epithelium forming the wall of the follicles have melanin pigment granules, and trachea or tracheoles are extended through this epithelium. In the cysts of adult testis, matured spermatozoa are grouped together in bundles and after releasing the sperm bundles to the vas deferens, lamellar shaped lysosomes appeared in the cytoplasms of the cyst cells. The number of spermatozoa per cyst is exactly 256 ($2^8$), this number is characteristics of the Lepidoperan species. Vas deferens is a tube with a fairly thick bounding epithelium, a basement membrane and a layer of circular muscle outside it. At the apical portion of the epithelial cells, microvilli are well developed. And in the cytoplasms of these cells, numerous excretory granules are observed.
