Isomeric Folate-Conjugated Polymeric Micelles Bind to Folate Receptors and Display Anticancer Effects

  • The present study aimed to prepare and evaluate polymeric micelles conjugated with folic acid through ${\alpha}$- or ${\gamma}$-carboxyl groups for antitumor efficacy. The isomeric block copolymers, ${\alpha}$- and ${\gamma}$-folate-polyethyleneglycol-distearoyl phosphatidylethanolamine (${\alpha}$- and ${\gamma}$-Fol-PEG-DSPE), were produced by solid phase peptide synthesis. Three types of doxorubicin (DOX)-loaded polymeric micelles (MPEG-DSPE-DOX and ${\alpha}$- / ${\gamma}$-Fol-PEG-DSPEDOX micelles) were prepared via the film formation method. Compared with MPEG-DSPE-DOX micelles, the ${\alpha}$- / ${\gamma}$-Fol-PEG-DSPE-DOX micelles presented a higher cellular uptake behavior in the live cell study. Cell viability percentages were 81.8%, 57.3%, 56.6% at 2 hours for MPEG-DSPE-DOX, ${\alpha}$- and ${\gamma}$-Fol-PEG-DSPE-DOX micelles, respectively (p<0.05). Using the KB xenograft tumor model, both ${\alpha}$- and ${\gamma}$-folate-conjugated micelles were found to have better antitumor effects with lower toxicity in comparison with MPEG-DSPE-DOX micelles. No difference in in vivo antitumor efficacy was found between ${\alpha}$- and ${\gamma}$-Fol-PEG-DSPE-DOX micelles. The folate-conjugated micelles might be a potentially useful strategy for tumor targeting of therapeutic agents, whether grafting with folic acid through ${\alpha}$- or ${\gamma}$-carboxyl groups.

처리 방법에 따른 참나물의 이화학적 특성 및 항산화 활성 (Antioxidant Activity and Physicochemical Characteristics of Pimpinella brachycarpa Nakai with Treatments Methods)

  • 본 연구에서는 참나물의 처리 조건에 따른 이화학적 특성 및 항산화 활성의 변화를 살펴보기 위하여 씻기, 데치기 및 찌기에 따른 특성 변화를 살펴보았다. 원료 참나물의 조단백질 함량은 24.43%, 조지방은 9.02%, 회분은 17.74% 그리고 식이섬유는 33.50%이었다. pH는 5.99~6.49 범위로 데친 참나물이 높았고, 찐 참나물이 5.99로 낮았다. 색의 L 값은 열처리에 따라 감소하였으며, 녹색을 나타내는 -a값은 데친 참나물이 크게 나타났다. ${\beta}$-Carotene은 원료에 비해 씻은 참나물은 18%, 데친 참나물은 54% 그리고 찐 참나물은 10% 증가하였다. 엽산은 씻은 참나물, 원료, 찐 참나물 및 데친 참나물이 각각 848.87, 772.16, 271.54 및 260.74 mg/100 g으로 씻은 참나물이 높게 나타났다. 주요 무기성분은 K, Ca, Na 및 Mg이었으며, K이 93.13~244.38 mg/100 g으로 가장 높았다. 총 polyphenol 및 flavonoid 함량, DPPH radical 소거능 모두 데친 참나물, 찐 참나물, 씻은 참나물, 원료 참나물 순으로 높았으며, 비가열 처리보다 가열 처리에서 항산화 성분 및 생리활성이 높게 나타났다.

국내에서 판매되는 임산부용 비타민·무기질 보충제에 함유된 영양소의 종류와 함량 및 적절성 평가 (Nutrient Composition and Content of Vitamin and Mineral Supplements and Their Appropriateness for Pregnant and Lactating Women in Korea)

  • Objectives: Use of dietary supplements containing vitamins and minerals is growing in Korean adults, especially in pregnant and lactating women. Vitamin and mineral supplements are available in different composition and in a wide range of contents. The purposes of the study were to examine nutrient composition and content of vitamin and mineral supplements for pregnant and lactating women and assess their appropriateness as dietary supplements. Methods: Information on the name, manufacturer, nutrient composition, and usage of vitamin and mineral supplements for pregnant and lactating women were obtained from the homepage of the Food Safety Information Portal managed by the Ministry of Food and Drug Safety, and Korean Index of Medical Specialties. A total of 264 products were identified. Results: Among 264 products, 26.1% were single nutrient products, and 73.9% were multinutrient products. The most commonly included nutrient was iron (70.1%), folic acid (66.3%), vitamin $B_{12}$ (45.8%), vitamin C (38.6%), and vitamin $B_6$ (38.6%). Although more than 50% of products contained nutrients less than 150% of Recommended Nutrient Intakes or Adequate Intakes for daily use, some products contained inappropriately high amounts of nutrients. When a maximum daily dose of supplements was taken as described on the label, iron in 73 products (39.5%), folic acid in 14 products (8.0%) were likely to be consumed in amounts greater than Tolerable Upper Intake Levels. Most products were assessed as inappropriate for pregnant women due to the possibility of excessive intake of vitamins or minerals when compared with Dietary Reference Intakes. Conclusions: Pregnant and lactating women need to carefully select dietary supplements containing adequate amounts of vitamins and minerals. Nutritionists should provide guidelines regarding selection of appropriate vitamin and mineral supplements for pregnant and lactating women.

햇순나물의 소비확대를 위한 조리기술 개발 및 영양성 평가 (Development of recipe for promoting consumption of Haetsun sprouts and their nutrient evaluation)

  • 본 연구는 햇순나물의 이용성 증진 및 소비 촉진을 위하여 햇순나물을 이용하여 4종의 햇순요리를 개발하고 이들의 관능적 특성 및 영양성을 평가하였다. 햇순요리에 대한 관능평가를 실시한 결과, 4종의 햇순요리의 전반적인 기호도가 3.0 이상으로 만족 수준이었으며, 그 중 '엄나무순닭고기냉채'의 기호도는 5점 만점에 4.48점으로 가장 높은 기호를 보였다. 엄나무 순을 이용한 '엄나무순닭고기냉채'는 단백질, 비타민 A, 비타민 E, 비타민 $B_6$, 엽산의 함량이 권장량보다 높았으며, 비타민 C 및 아연의 함량은 권장량보다 낮았으나 권장량의 50% 이상은 충족하였다. 두릅의 새순을 이용한 '두릅곶감무침'은 곶감과 함께 무쳐 두릅의 쓴맛과 떫은맛을 약화시켰으며, 비타민 E, 비타민 C, 비타민 $B_6$, 엽산 및 철의 함량이 권장량보다 약 1.2~1.7배 정도 높았다. 오가피 순을 이용한 '오가피순꽁치조림'은 무와 오가피순을 첨가하여 조리함으로써 꽁치의 비린내를 줄여 기호도를 높였다. '오가피순꽁치조림'은 단백질 함량이 권장량의 1.5배, 비타민 A, E, C 등 각종 비타민의 함량이 권장량의 1.4~3.6배 함유되어 있었다. 참죽순을 이용한 '참죽만두'는 고기를 넣지 않고 두부와 각종 채소를 넣어 만들었으며, 비타민 및 무기질이 풍부하고 다양한 영양소가 함유되어 있었다. '참죽만두'는 햇순나물 요리 중 가장 열량이 높았으며, 비타민 A, 비타민 E, 비타민 C, 티아민, 인과 철의 함량이 권장량보다 높았다.

합성배지(合成培地)를 이용(利用)한 고온성(高溫性) 느타리버섯의 자실체(子實體) 형성(形成)에 관한 연구(硏究) (Fruit-body Formation of Pleurotus florida on the Synthetic Medium)

  • 합성배지(合成培地)에서 고온성(高溫性) 느타리 버섯균(菌)의 균사생육(菌絲生育)과 자실체형성(子實體形成)에 관한 영양적(營養的) 특성(特性)과 생리화학적(生理化學的) 성질을 검토한 결과를 요약하면 다음과 같다. 1. 균사생육(菌絲生育)과 자실체형성(子實體形成)이 우수한 당류(糖類)는 mannitol과 sucrose이었고 arabinose, lactose, inulin은 균사생육(菌絲生育)이 빈약할 뿐만 아니라 자실체(子實體)가 형성(形成)되지 않았으며 mannitol의 최적 온도(溫度)는 2%였다. 2. 질소원(窒素源)중에서 peptone이 균사생육(菌絲生育)과 자실체형성(子實體形成)이 빠르고 자실체(子實體)의 수량(收量)이 많았으며 아질산태(亞窒酸態) 질소(窒素)는 저해적이었다. peptone의 최적 농도(濃度)는 0.2%이었다. 3. 균사생육(菌絲生育)과 자실체형성(子實體形成)이 우수한 Vitamin은 thiamine과 folic acid이었고 $KH_2PO_4$$MgSO_4{\cdot}7H_2O$의 최적 농도(濃度)는 각각 0.2%와 0.02%이었으며 그 밖의 무기염류(無機鹽類)는 효과가 없었다. 4. 광조도(光照度)는 $100{\sim}500\;lux$, 균사생육(菌絲生育) 온도(溫度)는 $25^{\circ}C$, 자실체형성(子實體形成) 온도(溫度)는 $20^{\circ}C$, pH는 6.0에서 균사생육(菌絲生育)과 자실체형성(子實體形成)이 우수하였다.

원자력 발전소 주변지역 거주민의 가구소득별 영양섭취 (Nutrient Intakes Differences of the People Living Near the Nuclear Plant by the Household Income Level)

  • This study was conducted to measure and evaluate the food and nutrient intakes of the people living near the nuclear plant and to investigate the relationship between the household income level and the food and nutrient intake patterns. A total of 552 cases (263 males and 289 females) were surveyed during the period from April 1 to December 21 of 2005. Dietary intake was measured by means of the 24-hour recall method. The data were analyzed using SPSS Windows (ver. 14.0). The household income level of the subjects was classified into two groups : Low income group (LIG; $\leq$2,000,000 won) and high income group (HIG; > 2,000,000). The subjects at large had less energy and nutrient intakes than did the population in town and village who participated in the 2005 National Health and Nutrition Survey. The intake of calcium, zinc, vitamin A, riboflavin, vitamin $B_6$, vitamin C, and folic acid was less than the Estimated Average Requirement in case of $50{\sim}95%$ of the subjects. The LIG consumed less beans, vegetables, fruits, meats, and beverages than did the HIG in male, while the LIG consumed less eggs and beverages than did the HIG in female. The LIG consumed less nutrients than did the HIG in male, except for carbohydrate, while the LIG consumed less nutrients including zinc, vitamin A, riboflavin, vitamin B6, vitamin C, folic acid than did the HIG in female. In addition, the LIG had higher percentage energy consumption from carbohydrate. These results suggest that higher food and nutrient intake is associated with higher income.

COVID-19 팬데믹 전후 한국 성인 남녀의 건강 및 식생활행태 비교연구: 국민건강영양조사 제8기(2019-2021년도) 자료 활용 (Comparative study on the health and dietary habits of Korean male and female adults before and after the coronavirus disease 2019 pandemic: utilizing data from the 8th Korea National Health and Nutrition Examination Survey (2019-2021))

  • Objectives: This study aims to compare changes in physical factors, health behaviors, eating habits, and nutritional intake among Korean male and female adults over a period of three years (2019-2021) before and after the outbreak of coronavirus disease 2019 (COVID-19). Methods: This study utilized raw data from the 8th Korea National Health and Nutrition Examination Survey (2019-2021). The participants in this study included 6,235 individuals in 2019, 5,865 individuals in 2020, and 5,635 individuals in 2021. Individuals whose daily energy intake was less than 500 kcal or exceeded 5,000 kcal were excluded from the study. Results: In comparison to 2019, overweight/obesity rates, weight, waist circumference, weekend sleep hours, and resistance exercise days/week increased in both male and female during the COVID-19 pandemic. Regarding eating habits, the proportions of people skipping breakfast, not eating out, consuming health supplements, and recognizing nutritional labels increased in 2020 and 2021, whereas the rate of skipping dinner decreased. Total energy intake has continued to decrease for the two years since 2019. A comparison of nutrient intake per 1,000 kcal before and after the outbreak of COVID-19 revealed that intake of nutrients, including protein, phosphorus, iron, vitamin A, riboflavin, and niacin increased, while folic acid intake decreased. In male, calcium, phosphorus, riboflavin, and niacin intakes increased, whereas iron, vitamin C, and folic acid intakes decreased. In female, phosphorus, iron, vitamin A, and riboflavin intake increased significantly, while protein and niacin intake decreased significantly. Conclusions: After COVID-19, the obesity rate, breakfast skipping rate, health supplement intake, and nutritional label use increased, while the frequency of eating out, dinner skipping rate, and total energy intake decreased. These environmental changes and social factors highlight the need for nutritional education and management to ensure proper nutritional intake and reduce obesity rates in the post-COVID-19 era.

표고버섯을 이용한 조미료의 주요성분에 관한 연구 (A Study on the Composition of Seasoning Using Lentinus edodes.)

  • 표고버섯(IfHDHHS efodes)조미료 중의 조성물질의 성분을 규명하고자 무기질, 구성아미노산, 유리아미노산, 비타민 등을 분석 검토한 결과는 다음과 같았다. 무기질은 Na가 5.04g%로 가장 많이 함유되어 있고, K, Ca, p, Mg, fe, Zn, Mn, Cu의 순으로 함유되어 있었다. 구성 아미노산을 분석한 결과 총 19종의 아미노산을 함유하고 있었고 인체에 필수한 9종의 아미노산을 모두 함유하고 있었으며, glutamic acid가 4.62%로 가장 많았었고 그 다음으로 glysine, leucine, alanine, aspartic acid, lysine, valine, proline 등의 순으로 함유되어 있으며 그 중 필수아미노산의 총함량은 41.78%였다. 또한 유리 아미노산은 20종으로 분석되었으며 glycine, glutamic acid, asparamine, proline, taurine과 aspartic acid, arginine, leucine, threonine과 serine phenylalanine 등의 순으로 함유되어 있으며 총 함유량은 71.9 mg%이었으며 인체 필수아미노산의 총 함유량은 12.80%이었다. 비타민의 경우 괴혈병의 치료제로 많이 알려져 있는 vitamin C가 224.78 mg%로 가장 많고 판토텐산은 159.09 mg%, vitamin B6 15.08 mg%, 나이아신 3.19 mg%, vitamin E 1.51 mg%, 엽산 1.14 mg%, vitamin B$_{l}$ 1.10 mg%, vitamin B$_{2}$ 0.44 mg%, vitamin D$_{3}$ 0.28 mg%, vitamin A 0.15 mg%, vitamin B$_{12}$ 0.04 mg%, vitamin $K_{l}$ 이 0.02 mg%으로 비타민의 총 함유량은 506.82mg%이었다.다.

In vitro infection of Cryptosporidium parvum to four different cell lines

  • To determine a suitable condition for in vitro infection model of cryptosporidium parvum, four different cell lines, AGS, MDCK, HCT-8 and Caco-2, were used as host cell lines which were cultured at various concentrations of added supplements. These supplement include fetal bovine serum (FBS), sodium choleate, ascorbic acid, folic acid, calcium pantothenate, para-aminobenzoic acid and pyruvate and their effects on the cell lines which were infected with C. parvum were evaluated. The results of this study showed that the AGS cell line was most susceptible to C. parvum whereas the Caco-2 cells appeared to be least susceptible to C. parvum. In regards to the serum condition, 10% FBS was suitable for the growth of AGS and HCT-8 cells, and 1% FBS was good for the growth of the MDCK cells when they were inoculated with C. parvum. Vitamines had a positive effect on the AGS cells, and pyruvate also showed positive effects on all of the cell lines except for Caco-2. Modified medium for each cell line was prepared by adding appropriate amounts of each supplement which resulted in the highest parasite infection number. Modified media increased the number of parasites infected on AGS cells to 2.3-fold higher when compared to the control media. In this study, we found that the AGS cell line was a suitable host model for evaluating C. parvum in vitro study and the media contents for the optimal infection conditions were suggested.

The strategies for the supplementation of vitamins and trace minerals in pig production: surveying major producers in China

  • Objective: Adequate vitamin and trace mineral intake for pigs are important to achieve satisfactory growth performance. There are no data available on the vitamin and trace mineral intake across pig producers in China. The purpose of this study was to investigate and describe the amount of vitamin and trace minerals used in Chinese pig diets. Methods: A 1-year survey of supplemented vitamin and trace minerals in pig diets was organized in China. A total of 69 producers were invited for the survey, which represents approximately 90% of the pig herd in China. Data were compiled by bodyweight stages to determine descriptive statistics. Nutrients were evaluated for vitamin A, vitamin D, vitamin E, vitamin K, thiamine, riboflavin, vitamin B6, vitamin B12, pantothenic acid, niacin, folic acid, biotin, choline, copper, iron, manganese, zinc, selenium, and iodine. Data were statistically analyzed by functions in Excel. Results: The results indicated variation for supplemented vitamin (vitamin A, vitamin D, vitamin E, vitamin K, vitamin B12, pantothenic acid, niacin, and choline) and trace minerals (copper, manganese, zinc, and iodine) in pig diets, but most vitamins and trace minerals were included at concentrations far above the total dietary requirement estimates reported by the National Research Council and the China's Feeding Standard of Swine. Conclusion: The levels of vitamin and trace mineral used in China's pig industry vary widely. Adding a high concentration for vitamin and trace mineral appears to be common practice in pig diets. This investigation provides a reference for supplementation rates of the vitamins and trace minerals in the China's pig industry.
