Asymmetric Mannich-type Reactions of Fluorinated Ketoesters with Binaphthyl-Modified Thiourea Catalysts

  • The catalytic enantioselective Mannich-type reaction promoted by chiral binaphthyl-modified bifunctional organocatalysts is described. The treatment of ${\alpha}$-fluoro-${\beta}$-ketoesters with N-Boc imines under mild reaction conditions afforded the corresponding ${\beta}$-aminated ${\alpha}$-fluoro-${\beta}$-ketoesters with excellent enantioselectivities (up to 98% ee).

  • Novel 9-[2-fluoro-4-hydroxy-3-hydroxymethyl-2-butenyl]adenine and its related compounds were designed and synthesized as open-chain analogues of neplanocin A. Alkylation of adenine or pyrimidine bases with the mesylate 4 was chosen as a simple approach to the synthesis of 2-fluoro-2-butenylated nucleosides. Mesylate 4 was prepared from dihydroxyacetone dimer via four steps in 58% overall yield. The synthesized compounds were evaluated their antiviral activity against HSV, HIV and Polio viruses.

Syntheses of Novel Liquid Crystalline Compounds with Partially Fluorinated Side Chains

  • A new series of three ring type liquid crystalline compounds containing partially fluorinated alkenyl or alkyl side chains together with fluorine substituted cyclohexylbiphenyls were designed and synthesized in this study. The structures of synthe sized compounds were established by 1 H, 13 C and 19 F NMR spectroscopy. The phase transition temperatures of fluorinated liquid crystalline compounds were determined by cross-polarizing mi-croscopy equipped withhot stage. All compounds were found to have nematic liquid crystalline phase with rel-atively low phase transition temperature and wide liquid crystalline temperature range. The dependence of phase transition temperatures on the chainlength falls into three categories; (a) decreasing transition tempera-tures for 4-fluoro-4'-[4-fluoro-4-(1-fluoroalkyl)cyclohexyl]biphenyl (15) series, (b) higher transition tempera-tures for odd numbered chains for 4-fluoro-4'-[4-fluoro-4-(1-fluoroalk-1-enyl)cyclohexyl]biphenyl (14) series, (c) higher transition temperatures for even numbered chains for 4-[4-(1,2-difluoroalk-1-enyl)-4-fluorocyclo-hexyl]-4'-fluorobiphenyl (16) series.

Effects of pyrimidine salvage inhibitors on uracil incorporation of Toxoplasma gondii (Toxoplasma gondii의 활성화된 uracil 도입 과정에 미치는 pyrimidine 대사 억제제의 영향)

  • Metabolic inhibitors which act in the process of pyrimidine salvage influenced on the uracil incorporation into nucleic acids of Toxoplasma. Inhibitors of dihydrofolate reductase, pyrimethamine and methotrexate, and inhibitors of thymidylate synthase, fluoro-uridine, fluoro·dUMP and fluoro-uracil, diminished isotopic uracil uptake in dose-dependent manners. Azauridine which suppresses do novo pyrimidine biosynthesis did not affect the salvage even in a relatively high dose. These results suggested that the activation of uracil salvage should be closely related with the function of TMP biosynthetic enzymes. The pattern of thymidine uptake had no differences between control HL-60 cells and Toxoplasma infected cells, which did not reject the specific proliferation of Texoplasma. It can be exploited to characterize the elects of various compounds related with the proliferation of Toxoplasma, especially its DNA synthesis. Herbicidal Activity and Molecular Similarity of 1-(4-chloro-2-fluoro-5-propargyloxyphenyl)-3-thiourea Derivatives (1-(4-chloro-2-fluoro-5-propargyloxyphenyl)-3-thiourea 유도체의 제초활성과 분자 유사성)

  • In the search for third generation herbicidal cyclic imide derivatives, the average values of herbicidal activity ($pI_{50}$) in vivo (pre-emergence) of 40 new peroxidizing herbicides, 1-(4-chloro-2-fluoro-5-propargyloxyphenyl)thiourea derivatives (1-40) against rice plant (Orysa sativa) and barnyard grass (Echinochlor crus-galli) were studied. The molecular similarity between protoporphyrinogen IX (protogen) as the substrate of protox enzyme and Urea derivatives (1-40) was discussed quantitatively. The diallyl (20) and 3-nitro substituent (33) showed the selective herbicidal activity against barnyard grass. Allyl substituent (8) and their molecular similarity in dice (S=0.81) showed the highest levels of herbicidal activity ($pI_{50}$=4.71). Also, similarity indices (S) and superimposed volume (C) of protogen and aryl-substituents (21-40) showed good correlation.

The Application of ParalluxTM System for Multi-Detection of (Fluoro)quinolone Class Antibiotics Residues in Raw Bovine Milk

  • This study aimed to apply the Parallux system to detect (fluoro)quinone antibiotics residues in raw bovine milk. The immunogen enabled the generation of a specific antiserum with a titer of 1/40,000. The $Parallax^{TM}$ kit using the antibody displayed $IC_{50}$ value of 10 to 150 ppb for (fluoro)quinolone antibiotics. $Parallax^{TM}$ kit was also sensitive for the detection of incurred (fluoro)quinolone at Korean Maximum Residual Levels in raw bovine milk as the result of dose response test. Cross reactivities of the antibody with the common (fluoro)quinolones were determined to be norfloxacin, 100%; enrofloxacin, 100%; ciprofloxacin, 100%; danofloxacin, 100%; nalidixic acid, 40%. Lower detection limit (LOD) values of the $Parallax^{TM}$ kit in raw bovine milk were determined to be norfloxacin, 4 ppb; enrofloxacin, 5 ppb; danofloxacin, 5 ppb; ciprofloxacin, 5 ppb and nalidixic acid, 10 ppb. The $Parallax^{TM}$ kit was run 8 times with five different concentrations of norfloxacin to determine the coefficient of variation (CV, %) of intra-assay, which was between 2.7% and 11.8%. To confirm the precision among kit batches for the inter-assay, five different batch kits were tested with 2 different concentration of norfloxacin. The CVs of the inter assay were 4.2% at 50 ppb, and 7.2% at 10 ppb norfloxacin, respectively.

  • A series of N-[4-cyano-2-fluoro-5-(2-pyrimidinyloxy, 2-benzyloxy or 2-pyridinyloxy)phenyl]-3,4,5,6-tetrahydrophthalimides was synthesized, and the herbicidal activities of those deivatives were evaluated through pre- and post-emergence application under upland conditions in a greenhouse. The results showed that most compounds resulted in stronger herbicidal activity on broadleaf weeds than on grass weeds and higher through post-emergence than pre-emergence application. The N-[(4-cyano-2-fluoro-5-(2-pyrimidinyloxy) phenyl]-3,4,5,6-tetrahydrophthalimide showed the best weed control efficacy and marginal corn safety at a rate of 60 g/ha through pre-emergence application.
